Cronobacter sakazakii is an opportunistic pathogenic bacterium found in powdered infant formula and is fatal to neonates. Antibiotic resistance has emerged owing to overuse of antibiotics. Therefore, demand for high-yield bacteriophages as an alternative to antibiotics has increased. Accordingly, we developed a modified mass-production method for bacteriophages by introducing a two-stage self-cycling (TSSC) process, which yielded high-concentration bacteriophage solutions by replenishing the nutritional medium at the beginning of each process, without additional challenge. pH of the culture medium was monitored in real-time during C. sakazakii growth and bacteriophage CS01 propagation, and the changes in various parameters were assessed. The pH of the culture medium dropped to 5.8 when the host bacteria reached the early log phase (OD540 = 0.3). After challenge, it decreased to 4.65 and then recovered to 4.94; therefore, we set the optimum pH to challenge the phage at 5.8 and that to harvest the phage at 4.94. We then compared phage production during the TSSC process in jar-type bioreactors and the batch culture process in shaker flasks. In the same volume of LB medium, the concentration of the phage titer solution obtained with the TSSC process was 24 times higher than that obtained with the batch culture process. Moreover, we stably obtained high concentrations of bacteriophage solutions for three cycles with the TSSC process. Overall, this modified TSSC process could simplify large-scale production of bacteriophage CS01 and reduce the unit cost of phage titer solution. These results could contribute to curing infants infected with antibiotic-resistant C. sakazakii.
산업적 유산균 배양의 가격대비 효율성을 높이기 위하여 질소원, 탄소원 및 무기염물을 기본량으로 기본배지를 제조하고, 효율적인 고농도 유산균체를 수득하기 위하여 pH 조절제로서 따로 첨가되어지는 합성 화학물질을 대신하여 미역을 실험에 사용하였다. 미역의 최적의 첨가 방법을 찾고자 추출, 착즙, 분말의 형태로 배지를 제조하여 유산균증식능 평가한 결과, 착즙의 형태에서 유산균 증식능이 가장 큰 것으로 확인되었다. 이러한 실험 결과를 통해, 폐기되어 환경오염의 원인이 되는 가식부 외 미역을 유산균 배양배지에 착즙형태로 첨가함으로서 종래 pH 조절을 위해 완충액으로 첨가되는 합성 화학물질을 대체하여 활용될 수 있음을 확인하였다. 또한 미역첨가 기본배지 선정에 따른 배지 단가를 절감하면서 높은 균체 수율을 얻는 등 충분한 경제적 장점을 기대할 수 있다. 이러한 연구결과는 미역을 이용함으로써 배양 시 해조류의 식이섬유 등이 정장작용에 유산균의 시너지 효과뿐만 아니라 폐기되는 천연 식물자원을 활용함으로써 새로운 부가가치 창출과 환경오염을 예방하는 효과를 기대할 수 있을 것이다.
2020년 기준 한국의 곡물자급률은 20.2%에 불과하지만 곡물수출국에서 발생하는 가뭄이 국내에 미치는 영향은 아직 면밀히 분석되지 않았다. 본 연구에서는 증발산 기반 가뭄지수인 Evaporative Stress Index (ESI)를 이용해 세계 주요 곡물생산지역의 농업가뭄의 발생빈도, 장기추세, 자연진동과의 상관성을 분석하였다. 또한 국제 곡물거래가격과 작물생산지역의 가뭄면적을 비교하여 해외에서 발생한 가뭄이 한국 경제에 미치는 영향을 정성적으로 평가하였다. ERA5 기후재분석자료로 산정된 ESI는 전지구적으로 토양수분과 강한 상관성을 보였으며 특히 작물재배 지역에서의 둘의 상관성이 매우 강하게 나타났다. 작물재배지역에서의 높은 상관성은 강한 지면-대기결합을 의미하며, 이 때문에 작은 토양수분 부족이 상대적으로 큰 수확량 손실로 연결될 가능성이 크다. 1991-2022 기간 작물재배지역에서 ESI는 뚜렷한 감소추세를 보였으며 지구온난화와 함께 가뭄면적이 증가할 가능성이 있다. 2012년과 2022년에 급격히 상승한 국제곡물가격은 수출국에서 발생한 대규모 가뭄과 밀접한 관계가 있는 것으로 분석되었으며 한국의 생산자물가지수를 상승시킨 주요 원인 중 하나로 판단된다. 본 연구는 해외지역에서 일어나는 가뭄의 영향을 줄이기 위해 감시와 위험관리 전략이 필요함을 시사한다.
이상기상으로 인해 노지 작물이 스트레스 상황에 노출되는 빈도가 증가하고 있다. 우리나라에서도 대표적인 벼 재배지역에서 대규모의 병해가 발생하는 사례가 나타났으며, 특정 시기에 대규모 필지에서 발생하는 피해를 현장방문으로 조사하는 것은 한계가 있다. 위성 기반의 원격탐사 기법은 시군 영역을 대상으로 작물을 모니터링하기에 유용하나 작물의 생육이상에 따른 민감도 평가가 선행되어야 한다. 본 연구에서는 벼 병해 발생 지역에서 서로 다른 공간해상도를 가지는 위성 기반의 정규화식생지수(normalized difference vegetation index, NDVI)를 드론 영상을 이용하여 평가하였다. 10 m와 30 m의 공간해상도를 가지는 Sentinel-2, Landsat-8 위성 영상을 평가하였으며, 드론 영상은 약 8-10 cm의 공간해상도를 가졌다. 위성 영상에 맞춰 리샘플링(resampling)된 드론 NDVI는 Sentinel-2 NDVI 와 0.867-0.940의 상관관계를 가졌으며, Landsat-8 NDVI와는 0.813-0.934의 상관관계를 가졌다. 센서의 차이, 관측 시점의 차이 등으로 인한 편향(bias) 영향을 최소화하였을 때, Sentinel-2 NDVI는 Landsat-8 NDVI에 비해 드론 NDVI와 0.2-2.8% 더 적은 정규화된 평균 제곱근 오차를 가졌다. 또한, Sentinel-2 NDVI는 드론 NDVI와 병해 피해 정도와 관계없이 일정한 오차를 가졌으나 Landsat-8 NDVI는 병해 피해 정도에 따라 드론 NDVI와 오차 특성이 다르게 나타났다. 농경지 경계에서 오차가 크다는 것을 고려했을 때 공간해상도가 높은 영상을 활용하는 것이 작물 모니터링에 효과적이라고 판단된다.
The composition of dissolved gases and nutrients in a liquid medium were determined for establishment of the optimum conditions for in vitro culture of Helicobacter pylori. A microaerobic condition facored by the organism was prepared by adjusting the partial pressure of the gas, agitation speed, and viscosity of the medium. The gaseous concentrations were controlled by utilizing CampyPak Plus that reduced oxygen while augmenting carbon dioxide. Agitation of the broth facilitated the oxygen transfer to the cells, yet inhibited the growth at high rates. An increase of viscosity in the medium repressed the culture although this variable was relatively insignificant. The chemical constituents of the liquid broth were examined to establish an economic model for H. pylori cultivation. The microbe required a neutral pH for optimum growth, and yet was also able to proliferate in an acidic condition, presumably by releasing the acidity-modulating enzyme, urease. Cyclodextrin and casamino acid were investigated as growth enhancers in place of serum, while yeast extract unexpectedly inhibited the cells. A low concentration of glucose, the unique carbon source for the organism, increased the cell density, yet high concentrations resulted in an adverse effect. Under optimally dissolved gas conditions, the cell concentration in brucella broth supplemented with serum substitutes and glucose reached $1.6{\times}10^8$ viable cells/ml which was approximately 50% higher than that obtained in the liquid medium added with only cyclodextrin or serum.
Interest in commercial cultivation and product development of Cordyceps species has shown a recent increase. Due to its biochemical and pharmacological effects, Cordyceps militaris, commonly known as orange caterpillar fungus, is being investigated with great interest. Cultivation of C. militaris has been practiced on a large scale in order to fulfill a demand for scientific investigation and product development. Isolates of C. militaris can be easily established from both spores and tissue. For isolation of spores, ascospores released from mature stromata are trapped in sterile medium. Multi-ascospore isolates, as well as combinations of single ascospore strains, are used for production of fruiting bodies. Progeny ascospore strains can be isolated from artificial fruiting bodies, thus, the cycle of fruiting body production can be continued for a long period of time. In this study, we examined fruiting body production from multi-ascospore isolates and their progeny strains for three generations. $F_1$ progeny strains generally produced a larger number of fruiting bodies, compared with their mother multi-ascospore isolates; however, $F_2$ and $F_3$ progeny strains produced fewer fruiting bodies. Optimum preservation conditions could help to increase the vitality of the progeny strains. In order to retain the fruiting ability of the strains, further testing of various methods of preservation and different methods for isolation should be performed.
Under the light intensity of 25-72$\mu$E/ m$^{2}$/s Botryococcus braunii UTEX 572 grew faster than Botryococcus sp. GE 24 isolated from a freshwater lake. The specific growth rate ($\mu$) of B. braunii UTEX 572 was highest at 0.260 (1/day) on a dry weight basis in Chu 13 medium from 1 to 9 days of incubation and then continuously decreased. Carbohydrate concentration and cellular nitrogen and phosphorus contents of B. braunii UTEX 572 gradually decreased with light intensity over a range of 25-72$\mu$E/m$^{2}$2/s, whereas the concentrations of protein and cellular N:P ratio increased with light intensity. Chlorophyll-a concentration showed a decreasing tendency with light intensity. The dry weight of B. braunii UTEX 572 increased in the highest rate of 83 mg/l/day at pH 8.0. When the N: P ratio of Chu 13 medium was adjusted to 50: 1 by addition of nitrogen source, dry weight increasing rate was 115 mg/l/day between 20 and 28 days of incubation which was the highest value during the cultivation. Cell growth in an open culture of B. braunii UTEX 572 was highest with Chu 13 medium, whereas that with Chu 13 medium adjusted to pH 7.0, containing 250 mg/l penicillin, or containing 1% glucose was reduced on a large scale. However, this result shows the possibility of the mass cultivation of B. braunii UTEX 572 in an open system competing with other microorganisms.
최근 폐열에너지와 스마트 그리드 연계를 통한 에너지 효율 향상 기술 수요가 증가하고 있는 추세이다. 또한 컨테이너 온실을 이용한 식물공장 개념의 스마트팜과 폐열 에너지의 실시간 활용을 위한 융합기술에 대한 연구를 통하여 고부가가치 작물을 재배에 대한 투자가 증가하고 있다. 이런 관점에서 에너지 효율 측면에서 LED 기반의 식물공장에 대한 경제성을 높여 농가에 실질적인 도움이 되는 실용화 기술에 대한 연구를 수행 하였다. 이는 향후 자동화될 대규모의 스마트 팜과 연계할 수 있는 기회를 제공할 수 있다. 본 연구에서는 폐열 에너지를 이용한 컨테이너 온실에서 냉방기술을 이용하여 작물재배의 경제성에 대한 연구이다. 컨테이너 온실, 폐열을 이용한 흡수식 냉동기를 IOT 기술을 이용하여 실시간 모니터링/제어하기 위하여 라즈베리 파이 기반의 게이트웨이를 모바일 기반에서 운영함으로써 냉방 에너지의 효용을 최대화 할 수 있는 새로운 형태의 농업 기술에 대한 연구를 제안하고자 한다.

Global warming has accelerated in recent decades due to the continuous consumption of petroleum-based fuels. Cyanobacteria-derived biofuels are a promising carbon-neutral alternative to fossil fuels that may help achieve a cleaner environment. Here, we propose an effective strategy based on the large-scale cultivation of a newly isolated cyanobacterial strain to produce phycobiliprotein and biodiesel, thus demonstrating the potential commercial applicability of the isolated microalgal strain. A native cyanobacterium was isolated from Goryeong, Korea, and identified as Pseudanabaena mucicola GO0704 through 16s RNA analysis. The potential exploitation of P. mucicola GO0704 was explored by analyzing several parameters for mixotrophic culture, and optimal growth was achieved through the addition of sodium acetate (1 g/l) to the BG-11 medium. Next, the cultures were scaled up to a stirred-tank bioreactor in mixotrophic conditions to maximize the productivity of biomass and metabolites. The biomass, phycobiliprotein, and fatty acids concentrations in sodium acetate-treated cells were enhanced, and the highest biodiesel productivity (8.1 mg/l/d) was achieved at 96 h. Finally, the properties of the fuel derived from P. mucicola GO0704 were estimated with converted biodiesels according to the composition of fatty acids. Most of the characteristics of the final product, except for the cloud point, were compliant with international biodiesel standards [ASTM 6761 (US) and EN 14214 (Europe)].
