Synthesis and Degradation Behaviors of PEO/PL/PEO tri-block Copolymers

  • Poly (ethylene oxide)/polylatide/poly(ethylene oxide) (PEO/PL/PEO) tri-block copolymers, which each block is connected by ester bond, were synthesized by coupling reaction of PL with PEO in the presence of pyridine. PL/PEO/PL tri-block copolymer was synthesized by ring opening polymerization of L-lactide initiated by PEO in the presence of stannous octoate. Degradation behavior of the copolymers was investigated in a pH 7.4 phosphate buffer saline (PBS) at 37$\pm$1 $^{\circ}C$. Gel permeation chromatography (GPC) and $^1$H-nuclear magnetic resonance (NMR) were used to monitor the change of mass loss, molecular weight and composition of copolymers. In hydrolytic degradation, the PEO/PL/PEO tri-block copolymer with high PEO contents affected the increase of its mass loss, and resulted in the decrease of its molecular weight as well as PEO composition. However, when PL/PEO/PL and PEO/PL/PEO tri-block copolymers had similar PEO contents, PEO/PL/PEO decreased faster in molecular weight and PEO composition than PL/PEO/PL.

  • oxyethylene/oxypropylene 공중합체의 존재하에 L-lactide를 중합시킴에 의해 poly(L-lactic acid) (PLLA) (A)와 polyether (B)로 이루어진 A-B-A block copoly(ester-ether)를 합성하였으며, 이들 블록 공중합체는 세그멘트를 도입함에 의해 PLLA에 유연성이 부여되었고, 표면에서의 미세상분리 구조로 인한 항혈전성의 개질을 확인하기 위하여 AFM사진을 관찰한 결과, PLLA와 비교하여 블록 공중합체는 필름표면의 요철성이 현저하게 저하하여 매끄러운 것을 확인하였으며 따라서, 표면의 요철이 항혈전성의 증가와 깊은 관계가 있음을 확인하였다.

Comparative study on long-term stability in mandibular sagittal split ramus osteotomy: hydroxyapatite/poly-ʟ-lactide mesh versus titanium miniplate

  • Background: Resorbable devices have recently been adopted in the field of orthognathic surgery with controversies about their postoperative skeletal stability. Hence, we determined the long-term skeletal stability of unsintered hydroxyapatite/poly-ʟ-lactic acid (HA/PLLA) mesh for osteofixation of mandibular sagittal split ramus osteotomy (SSRO), and compared it with that of titanium miniplate. Methods: Patients were divided into resorbable mesh and titanium miniplate fixation groups. A comparative study of the change in the mandibular position was performed with preoperative, 1-day, 6-month, and 2-year postoperative lateral cephalograms. Results: At postoperative 6 months-compared with postoperative 1 day, point B (supra-mentale) was significantly displaced anteriorly in the titanium-fixation group. Moreover, at postoperative 2 years-compared with postoperative 6 months, point B was significantly displaced inferiorly in the titanium-fixation. However, the HA/PLLA mesh-fixation group did not show any significant change with respect to point B postoperatively. Conclusions: The HA/PLLA mesh-fixation group demonstrated superior long-term skeletal stability with respect to the position of mandible, when compared with the titanium-fixation group.

In vitro Cartilage Regeneration using Primary Chondrocytes Cultured within Porous Poly(lactide-co-glycolide) Scaffolds

  • Cartilage injuries are frequent nowadays. The previous surgical treatment of cartilage defect was limited. Another approach in the treatment of cartilage injuries is the use of reconstitute cartilage consisting of chondrocytes cultured in suitable biodegradable scaffolds. Current studies have demonstrated the compatibility of chondrocytes with different biomaterials and the chondrogenesis in various types of porous scaffolds. The cell ingrowth into the porous scaffolds is modulated by initial cell loading efficiency. Therefore, well-interconnected pore structure and even pore distribution of the scaffolds are essential for efficient cell seeding. According to our previous work, well-interconnected macroporous scaffolds can be prepared by gas-foaming/salt-leaching method using ammonium bicarbonate salt as porogen additives. In this work, primary chondrocytes were cultured in PLGA 65/35 scaffolds fabricated by using our method. Cells seeded in the scaffolds showed well distribution by agitated seeding method. Histochemical staining of proteoglycans present in the scaffolds was used to visualize the chondrocyte ingrowth in the scaffolds. At 3 weeks, the population of chondrocytes was increased for the most part of the scaffolds, and extra cellular matrix (ECM) secretion was increased as culture periods progressed.

Delayed degradation according to the location of fixation with using an absorbable plate

  • Background: The ideal absorbable plating system should provide sufficient rigidity and then be absorbed within a timely manner. The Resorb-X has been recently developed as a plating system with a mixture ratio of 50:50 poly(D, L-lactide). Methods: We present seven of 121 patients who experienced delayed degradation with this absorbable plate. One hundred twenty-one patients with facial bone fracture underwent surgical treatment from March 2011 to March 2015, and rigid fixation was achieved with the Resorb-X. Results: Of 121 patients, seven (5.8%) developed complications at the surgical sites. Six of 102 cases underwent fixation of the infraorbital rim and one of 73 underwent fixation of the frontozygomatic buttress; the other sites of fixation did not develop delayed degradation. Foreign body granuloma developed at the earliest by postoperative 20 months and at the latest by postoperative 28 months (average, 23.5 months). Conclusion: We observed that the use of absorbable plates in incision sites or areas with thin skin can increase the possibility of delayed degradation. When performing surgery in these areas, the normal skin above the fixed location should be covered sufficiently.

Enhanced Antigen Delivery Systems Using Biodegradable PLGA Microspheres for Single Step Immunization

  • To demonstrate their possibilities as an enhanced vaccine delivery system, protein-loaded Poly lactide glycolide copolymer (PLGA) microspheres were prepared with different physical characteristics. Ethyl acetate (EA) solvent extraction process was employed to prepare microspheres and the effects of process parameters on drug release properties were evaluated. The biodeuadability of microspheres was also evaluated by the pH change and GPC (Gel permeation chromatography). Primary IgG antibody responses in BALB/c mice were compared with protein saline solutions as negative controls and adsorbed alum suspensions as positive controls after single subcutaneous injection for in vivo studies. The microspheres showed a erosion with a highly porous structure and did not keep their spherical shape at 45 days and this result could be confirmed by GPC. In vitro release of proteinous drug showed initial burst effect in all batches of microspheres, followed by gradual release over the next 4 weeks. PLGA microspheres were degraded until 45 days and the secondary structure of OVA was not affected by the preparation method. Enzyme-linked immunosorbent assays demonstrated that the single subcutaneous administrations of OVA-loaded PLGA microspheres induced enhanced serum IgG antibody response in comparison to negative and positive controls. These results demonstrated that microspheres providing the controlled release of antigens might be useful in advanced vaccine formulations for the parenteral carrier system.

  • Restenosis after percutaneous coronary intervention (PCI) continues to be a serious problem in clinical cardiology. To solve this problem, drug eluting stents (DES) with antiproliferative agents have been developed. Variable local drug delivery systems in the context of stenting require the development of stent manufacture, drug pharmacology and coating technology. We have worked on a system that integrates electrophoretic deposition (EPD) technology with the polymeric nanoparticles in DES for local drug delivery and a controlled release system. The surface morphology and drug loading amount of DES by EPD have been investigated under different operational conditions, such as operation time, voltage and the composition of media. We prepared poly-D,L-lactide-co-glycolic acid (PLGA) nanoparticles embedded with curcumin, which was done by a modified spontaneous emulsification method and used polyacrylic acid (PAA) as a surfactant because its carboxylic group contribute negative charge to the surface of CPNPs (?53.5 ± 5.8 mV). In the process of ‘trial and error' endeavors, we found that it is easy to control the drug loading amount deposited onto the stent while keeping uniform surface morphology. Accordingly, stent coating by EPD has a wide application to the modification of DES using various kinds of nanoparticles and drugs.

생분해성 iodophor microsphere 함유 유두침지 소독제의 건유기 젖소에서의 유방내 신규감염 예방 효과 (Effects of teat dipping disinfectant containing biodegradable iodophor microspheres on preventing dry period intramammary infection in dairy cows)

  • 1% iodophor loaded microspheres of PLGA (Poly[DL-Lactide-co-Glycolide]) were prepared by solvent evaporation method and were applied to the cows on dry period for evaluating it's preventive effects on intramammary infections. The morphology of the microspheres were evaluated using scanning electron microscopy and their releasing patterns were investigated. On investigating idophor releasing patterns of the microsphere, burst releasing pattern was detected until 2 days after in vitro incubation and sustained releasing was observed until 4 weeks. In field trial of teat dipping solution containing idophor loaded microspheres in dry cows showed significant preventive effects of intramammary infection caused by S. aureus, S. agalactiae, coagulase negative Staphylococci and coliform bacteria (p<0.05).

Photocatalytic performance of graphene/Ag/TiO2 hybrid nanocomposites

  • To improve photocatalytic efficiency, graphene/Ag/TiO2 nanotube catalyst was synthesized, and its surface characteristics and photocatalytic activity investigated. For deposition of Ag nanoparticles on the TiO2 nanotubes, a polymer compound containing CH3COOAg/poly(L-lactide) was utilized, and the silver particles were precipitated by reducing the silver ions during the annealing process. Graphene deposition on the Ag/TiO2 nanotubes was achieved using an electrophoretic deposition process. Based on the dye degradation results, it was determined that the photocatalytic efficiency was significantly affected by deposition of silver particles and graphene on the TiO2 catalyst. Highly efficient destruction of the dye was obtained with the new graphene/Ag/TiO2 nanotube photocatalyst. This may be attributed to a synergistic effect of the graphene and Ag nanoparticles on the TiO2 nanotubes.

  • Triblock copolymers from poly(ethylene glycol) (PEG) and D,L-lactide or $\varepsilon$-carprolactone were synthesized to prepare semi-interpenetrating polymer network (semi-IPN) with chitosan by U.V. irradiation method. Then, solute permeation through these semi-IPNs hydrogels were investigated. The structures of semi-IPNs were confirmed by FT-IR spectroscopy and wide angle X-ray diffractometer (WAXD). Equilibrium water content (EWC) of these hydrogels was in the range of 67-75%. The crystallinity, thermal properties and mechanical properties of semi-IPNs hydrogels were studied. All the hydrogels revealed a remarkable decrease in crystallinity as compared with PEG macromer itself. The tensile strengths of semi-IPNs hydrogels in dry state were rather high, but those of hydrogels in wet state decreased drastically. The permeabilities of solutes of hydrogels followed the swelling behaviors and were regulated by solute size.
