Laccase activity is influenced by copper (Cu) as an inducer. In this study, laccase was immobilized on Cu and Cu-magnetic (Cu/Fe2O4) nanoparticles (NPs) to improve enzyme stability and potential applications. The Cu/Fe2O4 NPs functionally activated by 3-aminopropyltriethoxysilane and glutaraldehyde exhibited an immobilization yield and relative activity (RA) of 93.1 and 140%, respectively. Under optimized conditions, Cu/Fe2O4 NPs showed high loading of laccase up to 285 mg/g of support and maximum RA of 140% at a pH 5.0 after 24 h of incubation (4℃). Immobilized laccase, as Cu/Fe2O4-laccase, had a higher optimum pH (4.0) and temperature (45℃) than those of a free enzyme. The pH and temperature profiles were significantly improved through immobilization. Cu/Fe2O4-laccase exhibited 25-fold higher thermal stability at 65℃ and retained residual activity of 91.8% after 10 cycles of reuse. The degradation of bisphenols was 3.9-fold higher with Cu/Fe2O4-laccase than that with the free enzyme. To the best of our knowledge, Rhus vernicifera laccase immobilization on Cu or Cu/Fe2O4 NPs has not yet been reported. This investigation revealed that laccase immobilization on Cu/Fe2O4 NPs is desirable for efficient enzyme loading and high relative activity, with remarkable bisphenol A degradation potential.

The nucleotide sequence witl~out introlls was 47Y0 identical to that of lee1 gene of Pametes wllosa; lhe identity in amino acid sequences of the two was 7994 The deduced amino acid seqoence was also sunilar to those of Coriolus versicolo~ kc3 (79%); Co~,iolz~s hirsutus phenolouiduse (78%), Trainetes vel.srcoloi. lccl (77%), Trametes ~!i/Iosa Ice2 (77%) and Trametes vemicolor kc4 (66%).
The culture conditions needed to maximize the production of laccase from Pholiota highlandensis mycelia were investigated. Among the tested media for laccase production, Coriolus versicolor medium (CVM; 2% dextrose, 0.4% peptone, 0.6% yeast extract, 0.046% KH2PO4, 0.1% K2HPO4, 0.05% MgSO4·7H2O) showed the highest activity for the enzyme. Then, to optimize culture conditions for laccase activity, the influences of various carbon, nitrogen, phosphorus, and inorganic salt sources in CVM were investigated. The optimum culture medium was 2% fructose, 0.4% peptone with 0.6% yeast extract, 0.05% NaH2PO4, and 0.05% MgSO4·7H2O as carbon, nitrogen, phosphorus, and inorganic salt sources, respectively. Several aromatic compounds in the medium enhanced laccase activity to varying degrees. Guaiacol induced maximum laccase production, yielding 114.1 U/ml laccase activity after cultivation for 11 days at 25℃. The optimum pH and temperature for laccase production were 8.0 and 35℃, respectively. Native polyacrylamide-gel electrophoresis (PAGE) followed by laccase-activity staining with 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) as the substrate was performed to identify the presence of laccase under the optimum conditions studied. Zymogram analysis of the supernatant culture showed an enzymatic band with a molecular mass of about 90 kDa.
The purpose of this study was to identify and characterize the laccase genes of Flammulina velutipes var. lupinicola. Five laccase genes (g1934, g1937, g2415, g2539, g5858) were selected based on the copper binding site and signal peptide analysis results using the laccase gene selected from the F. velutipes var. lupinicola genome. The size of the laccase genes of F. velutipes var. lupinicola were 1,488 bp~1,662 bp. As a result of cDNA sequence analysis, 14 to 17 introns were identified in the laccase genes. The cleavage site predicted as the signal peptide of the laccase gene was found to be located between 20 bp and 34 bp from the N-terminus. In addition, separation and purification were performed to characterize the F. velutipes var. lupinicola laccases, and the optimal activity of the separated and purified proteins were analyzed by pH, temperature and time. Five bands with laccase activity were found from zymogram analysis. The optimal pH of the reaction was 5.5, the optimal temperature was found to be 40℃. Therefore, characterization of the laccase genes identified in this study should help in better understanding the biomass decomposition of F. velutipes var. lupinicola.
The production media, partial purification and the enzymatic characteristics of laccase from Pleurotus ostreatus were investigated. Among various media tried the double strength onion media showed much higher production of laccase compared to glucose and/or CMC added media. The laccase from Pleurotus ostreatus has the optimum pH of 5.94 for the activity and appears to be stable at relatively broad pH spectrum (4.7-8.7). The experiments on the temperature stability shows that above 90% activity could be preserved after holding at $30^{\circ}C$ for 40 min., 60% activity was remained after 40 min. at $50^{\circ}C$. The Km value of the laccase from Pleurotus ostreatus was estimated to be 3.209mM and the molecular weight of laccase was estimated to be 55,000 by SDS-Polyacrylamide gel electrophoresis.
For Trametes sp. CJ-105, a kind of white-rot fungi which was collected from the mountain of Korea and was proven to be effective in decolorizing a wide range of structurally different synthetic dyes, the optimum conditions for mycelial growth and laccase(E.C. 1.10.3.2) production were investigated. Among various carbon sources, glucose showed the highest potential for the mycelial growth and laccase production, the optimum concentration being 2% glucose. For the nitrogen source, asparagine was good for the mycelial growth, while ammonium tartrate for laccase production(optimum concentration: 0.04%). The addition of thiamine and biotin increased both th emycelial growth and laccase production. When 2,5-xylidine was added as an inducer after the first day of culture, the production of alccase was seven-times higher than that in the absence of the inducer. The optimum pH and temperature conditions for laccase production by Trametes sp. CJ-105 were pH 5.0 and $25^{\circ}C$, respectively. In the 5L fermentation, the production of laccase reached a maximum of 340U/ml at the time when the ammonium ion was being rapidly depleted.
The gene encoding Thermus thermophilus HJ6 laccase (Tt-laccase) was cloned, sequenced, and comprised of 1,389 nucleotides encoding a protein (462 amino acids) with a predicted molecular mass of 51,049 Da. The deduced amino acid sequence of Tt-laccase showed 99.7% and 44.3% identities to the Thermus thermophilus HB27 laccase and Synechococcus sp. RS9917 laccase, respectively. Tt-laccase gene was expressed as a fusion protein with six histidine residues in E. coli Rosetta-gami (DE3) cells, and the recombinant protein was purified to homogeneity. UV-Vis spectrum analysis revealed that the enzyme has copper atoms, a type I Cu(II) and a type III binuclear Cu(II). The optimum pH for the oxidation of guaiacol was 5.0 and the optimum temperature was $90^{\circ}C$ The half-life of heat inactivation was about 120 min at $90^{\circ}C$ The enzyme reaction was inhibited by sodium azide, L-cystein, EDTA, dithiothreitol, tropolone, and kojic acid. The enzyme oxidized various known laccase substrates, its lowest $K_m$ value being for 4-hydroxyindole, highest $k_{cat}$ value for syringaldazine, and highest $k_{cat}/K_m$ for guaiacol.

The enzyme production from F. fraxinea was reached to the highest level after the cultivation for 10 days at $25^{\circ}C$ and initial pH 8. The enzyme activity of the culture supernatant was most active at $50^{\circ}C$ and pH 5.

Among various carbon and nitrogen sources, 2% glucose and 0.4% peptone showed the highest production of the enzyme, respectively. The enzyme production was reached to highest level after the cultivation for 8 days at $25^{\circ}C$. Zymogram analysis of the culture supernatant showed a laccase band with molecular mass of 52 kDa. The optimum pH and temperature for the enzyme activity were $80^{\circ}C$ and pH 3.0.
Laccase was applied to improve the drying speed of urushi lacquer to confirm a potential use of laccase as an enzyme additive. As a result of applying laccase of lacquer and white-rot fungi, drying times for both Korean and Chinese specimens were reduced. All of the specimens to which laccase was applied were dried(set to touch) within 60 minutes from the start of the drying, and the drying rate was improved as the content ratio of laccase increased. In addition, although there were differences in hardening drying time between Korean and Chinese lacquers, it was confirmed that hardening drying took place even at room with temperature of 20 ± 2 ℃ and humidity of 55-60%. As a result of lacquer layer analysis of the dried specimens, the drying speed was improved when the content ratio of laccase to urushiol was 5:1, and there were no differences in chromaticity and adhesion, confirmed that the layers were stable. It was observed that the gloss of both Korean and Chinese lacquers were reduced when laccase was applied. However, according to the analysis of FT-IR and Py-GC/MS, the changes in components were not as sufficient to affect the physical properties. Thus, its applicability as an additive was confirmed.
