  • Recently, many people have demanded reliable nutritional data even for minor-components. On the other hand, an analytical method for the analyses of vitamin $B_5$ and $B_6$ is lacking. Therefore, this study attempted to validate with accuracy and precision the analysis of vitamin $B_5$ and $B_6$ using a high-performance liquid chromatography (HPLC) method. The vitamin $B_5$ and $B_6$ contents were analyzed using an Agilent 1260 series HPLC system. YMC-Pack ODS-AM ($250{\times}4.6mm$ I.D.) and YMC-Pack Pro RS $C_{18}$ ($250{\times}4.6mm$ I.D.) columns were used for the analyses of vitamin $B_5$ and $B_6$, respectively. In the case of vitamin $B_5$, the flow rate was set to 1.0 mL/min by isocratic elution using the 50 mM $KH_2PO_4$ solution (pH 3.5)/acetonitrile (ACN) (95:5, v/v) with monitoring at 200 nm using HPLC/DAD, whereas the flow rate for vitamin $B_6$ was set to 1.0 mL/min of flow rate by isocratic elution using a 20 mM $CH_3CO_2Na$ solution (pH 3.6)/ACN (97:3, v/v) with monitoring by excitation at 290 nm and emission at 396 nm using HPLC/FLD. The column temperature was set to $30^{\circ}C$. The injection volume was $20{\mu}L$ for each experiment. The specificity of the accuracy and precision for vitamin $B_5$ and $B_6$ were also validated by HPLC. The results showed high linearity in the calibration curve for vitamin $B_5$ ($R^2=0.9998^{{\ast}{\ast}}$), the limit of detection (LOD) and limit of quantitation (LOQ) were 0.4 mg/L and 1.3 mg/L, respectively, In contrast, for the calibration curve of vitamin $B_6$, which showed high linearity ($R^2=0.9999^{{\ast}{\ast}}$), the LOD and LOQ were 0.006 mg/L and 0.02 mg/L, respectively.

  • Microcapsules(Mc) of cephalexin (CEPH), using Eudragit RS, RL, L, S and polyethylene glycol 1540, were evaluated biopharmaceutically. The area under the curve of CEPH-Eudragit RS/RL Mc administered orally once was larger than that of cephalexin powder twice every 6 hrs. Controlledrelease effectiveness and the absorption rate effectiveness, two important parameters of Vallner's method, of CEPH-Eudragit RS/RL Mc indicate that these Mc can be good sustained-release preparations. And a simple pharmacokinetic model is introduced which allows the gastric emptying and intestinal-transit rates of a drug itself and a solid-state drug contained in Mc. Decreasing $K_r$, without change in $K_a$, showed that the rate-limiting step of absorption moved from absorption step to releasestep.

  • The prediction of impending collapse of deep tunnel is one of the most difficult problems. Collapse mechanism of deep tunnel in layered soils is derived using a new curved failure mechanism within the framework of upper bound theorem, and effects of seepage forces are considered. Nonlinear failure criterion is adopted in the present analysis, and the possible collapse shape of deep tunnel in the layered soils is discussed in this paper. In the layered soils, the internal energy dissipations along velocity discontinuity are calculated, and the external work rates are produced by weight, seepage forces and supporting pressure. With upper bound theorem of limit analysis, two different curve functions are proposed for the two different soil stratums. The specific shape of collapse surface is discussed, using the proposed curve functions. Effects of nonlinear coefficient, initial cohesion, pore water pressure and unit weight on potential collapse are analyzed. According to the numerical results, with the nonlinear coefficient increase, the shape of collapse block will increase. With initial cohesion of the upper soil increase, the shape of failure block will be flat, and with the lower soil improving, the size of collapsing will be large. Furthermore, the shape of collapsing will decrease with the unit weight decrease.

  • This paper deals with the precision grinding for aspherical surface of optical parts. A parallel grinding method using the spherical wheel was suggested as a new grinding method. In this method, the wheel axis is positioned at a $\pi$/4 from the Z-axis in the direction of the X-axis. An advantage of this grinding method is that the wheel used in grinding achieves its maximum area, reducing wheel wear and improving the accuracy of the ground mirror surface. In addition, a truing by the CG (curve generating) method was proposed. After truing, the shape of spherical wheel transcribed on the carbon is measured by the Form-Talysurf-120L. The error of the form in the spherical wheel which is the value ${\Delta}x$ and $R{^2}{_y}$ inferred from the measured profile data is compensated by the re-truing. Finally, in the aspherical grinding experiment, the WC of the molding die was examined by the parallel grinding method using the resin bonded diamond wheel with a grain size of #3000. A form accuracy of 0.16${\mu}m$ P-V and a surface roughness of 0.0067${\mu}m$ Ra have been resulted.

  • The objective of this study is to investigate fracture toughness and fatigue crack propagation behavior in the Reduced Activation Ferritic Steel (RAFs) JLF-I. The fracture toughness tests were performed with various size(plane size and thickness) and various side groove of specimens. The fatigue crack propagation behavior of the JLF-I steel was investigated by the constant-amplitude loading test for the stress ratios R=O.I, 0.3 and 0.5 respectively. The effects of stress ratios and specimen size on the fatigue crack growth behaviors for JLF-I steel were discussed within the Paris law. The test results showed the standard CT specimen with the side groove of 40 % represented a valid fracture toughness. The fracture resistance curve increased with increasing plane size and decreased with increasing thickness. However, the fracture resistance curve of half size specimen was similar to that of the standard specimen. The fatigue crack propagation rate of a half size specimen was similar to that of a full size specimen at the stress ratios of 0.1, 0.3 and 0.5 respectively. The fatigue crack propagation behavior of this material were evaluated by using a half size specimen.

  • A rapid, sensitive and selective liquid chromatography-tandem mass spectrometric (LC-MS/ MS) method for the determination of lithospermic acid B (LSB) in rat serum was developed. LSB and internal standard, 7-hydroxy-3-phenyl-chromen-4-one (HPC) were extracted from rat serum with methyl-tert-butyl ether at acidic pH and analyzed on a Luna $C_8$ column with the mobile phase of acetonitrile-ammonium formate (10 mM, pH 6.5) (50:50, v/v). The analytes were detected using a negative electrospray ionization tandem mass spectrometry in the multiple- reaction-monitoring mode. The standard curve was linear $(r^2 = 0.997)$ over the concentration range of 10.0-500 ng/mL. The coefficient of variation and relative error for intra- and interassay at three QC levels were 1.1~6.2% and -10.3~-2.7%, respectively. The recovery of LSB from serum sample ranged from 73.2 to 79.5%, with that of HPC (internal standard) being 75.1 %. The lower limit of quantification for LSB was 10 ng/mL using 50 ${\mu}L$ of serum sample.
