Draft genome sequence of lytic bacteriophage KP1 infecting bacterial pathogen Klebsiella pneumoniae (병원균 Klebsiella pneumoniae를 감염시키는 용균 박테리오파지 KP1의 유전체 염기서열 초안)

  • Klebsiella pneumoniae is a Gram-negative, rod-shape bacterium causing disease in human and animal lungs. K. pneumoniae has been often found to gain antimicrobial resistance, thus it has been difficult to treat K. pneumoniae infection with antibiotics. For such infection, bacteriophage can provide an alternative approach for pathogenic bacterial infection with antimicrobial resistance, because of its sensitivity and specificity to the host bacteria. Bacteriophage KP1 was isolated in sewage and showed specific infectivity to K. pneumoniae. Here, we report the draft genome sequence of Klebsiella pneumoniae phage KP1. The draft genome of KP1 is 167,989 bp long, and the G + C content is 39.6%. The genome has 295 predicted ORFs and 14 tRNA genes. In addition, it encodes various enzymes which involve in lysis of the host cell such as lysozyme and holin.

The Inhibitory Effect of Fermented Kalopanax pictus by Bioconversion on Endotoxemia and the Competitive Inhibitor Activity on LPS (생물전환을 통한 음나무발효물의 LPS에 대한 경쟁적 억제제 효과 및 내독소혈증 억제 효과)

  • The objective of this study was to evaluate the effect of fermented Kalopanax pictus (KP-F) on macrophage activation and its effect as a competitive inhibitor of LPS and inhibitory effect on endotoxemia. The results showed that KP-F could activate macrophage in a dose-dependent manner, and KP-F was confirmed to act as a ligand for TLR4. Also, it was found that KP-F did not exhibit the same biotoxicity as LPS in intraperitoneal injection, and that it could suppress the neutrophil migration induced by LPS administration. In normal mice, the body weight, tissue weight, and amount of nitrite and pro-inflammatory cytokines in serum showed no significant changes with KP-F diet for 2 weeks, confirming that administration of KP-F in normal mice did not lead to over activation of immune response and biotoxicity. In the mouse model of endotoxemia induced by LPS and D-galactosamine(D-GalN) in sub-lethal dose, the diet of KP-F effectively inhibited the amount of nitrite and cytokines in the blood, and thus was found to be able to relieve the hepatic and kidney injury. In addition, in the endotoxemia mouse model induced by LPS and D-GalN of lethal dose, the survival rate was increased by KP-F diet in a dose-dependent manner.

The Knowledge Process and Performance of Knowledge Management Systems (지식 프로세스와 지식관리시스템의 성과)

  • This study examines the impact of knowledge processes (KP) on the performance of knowledge management systems (KMS). It posits that task needs and available functionality of technology existing in an organization could influence the usability of KP and the KMS performance. A firm-level structural model was developed based on data collected from corporate KM users. Survey-based research was carried out to test this model. Following questionnaire development, validation, and pretest with a pilot study, data were collected from 886 knowledge management (KM) users including directors, managers, and workers in a South Korea-based company, Korea Asset Management Corporation (KAMCO), to measure the task needs and available functionality of technology to improve the KMS performance. Results show that the matching between the two factors-technology and task-had a significant influence on the usability of KP and the KMS performance, and a better usability of KP has positive impact on the KMS performance. Implications on KM practices and KMS designs are also discussed.

Relationship between Korean Peninsula Landfalling Tropical Cyclones and Interannual Climate Variabilities

  • The relationship between two interannual climate variabilities and the frequency of tropical cyclone (TC) that landed over the Korean Peninsula (KP) has investigated for the period of 1951-2004. In the analysis of the relationship between KP-landfall TC frequency and the ENSO phase, most TCs of C-14 (TCs that do not pass through mainland China before landing the KP) and C-23 (TCs that pass through mainland China before landing the KP) tended to more land in the warm phase than normal and cold phases. However, TC intensity at landfall was stronger in the cold and normal phases. In the analysis of the relationship between KP-landfall TC frequency and Arctic Oscillation (AO) phase, the TCs of C-14 tended to more land in the positive (POS) phase of AO and the negative (NEG) phase of AO for C-23. It was found that AO index was negatively correlated with the Ni$\tilde{n}$o-3.4 index. And then the TCs of C-14 landed more frequently over the KP in the AO POS - Ni$\tilde{n}$o-3.4 NEG phases and in the AO NEG - Ni$\tilde{n}$o-3.4 POS phases for the TCs of C-23.

Bleaching of Kraft Pulp with Xylanase and Laccase-Mediator System

  • Xylanase (X) derived from Aurreobasidium pullulans and laccase-mediator system (LM) using Trichophyton sp. LKY-7 laccase (TrL) and N-hydroxy-2-pyridone analogue (NHP) as a mediator were applied in hardwood kraft pulp (HwKP) bleaching. The individual and the synergistic effects of X and LM stage were investigated in the enzymatic bleaching of HwKP. Also, the effects of subsequent alkaline extraction (E) and alkaline/hydrogen peroxide treatment (P) were examined. In X or LM treatment alone, an appreciable bleaching effect of HwKP was not observed, whereas subsequent E or P stage enhanced the increase of brightness and the decrease of kappa number. Especially, P stage significantly enhanced the bleaching effect of pulp. Bleaching of HwKP with XLM sequentially gave significantly higher pulp brightness and lower kappa number than that obtained after the treatment of HwKP with X+LM simultaneously. When HwKP was sequentially treated with XLM followed by P stage, the brightness increased by about 11% ISO and the kappa number decreased by about 3.6 in comparison with the initial pulp. Xylanase and laccase were strongly inactivated by NHP both in the absence and the presence of pulp.

  • A bacterial strain KP-364 producing extracellular keratinolytic protease was isolated from the soil of the poultry fac-tory. It was identified as Pseudomonas sp. based on its morphological and physiological characteristics, The optimal culture conditions for the production of keratinolytic protease by Pseudomonas sp. KP-364 were investigated. The composition of optimal medium for the keratinolytic protease was 2.0% glucose, 0.5% soybean meal. 0.5% $NaNO_3$ and 0.2% KCI Optimal initial pH for production of Keratinolytic protease production were 6.5 and $37^{\circ}C$ respec- tively. The keratinolytic protease production reached a maximum of 1,270 U/ml/hr after 48 hours cultivation under the optimal culture conditions.

Detection of Aromatic Pollutants by Bacterial Biosensors Bearing Gene Fusions Constructed with the dnaK Promoter of Pseudomonas sp. DJ-12

  • Gene fusions were constructed by the transcriptional fusion of the dnaK promoter of pseudomonas sp. DJ-12 or E. coli to the lux or luc marker gene. The dnaKp-DJ::luxCDABE bioluminescent fusion in the biosensor using the Pseudomonas sp. DJ-12 dnaK promoter exhibited about 5-fold more extensive response to ethanol than that of dnaKp-EC::luxCDABE. The bioluminescent response of the dnaK-DJ::luc fusion to ethanol was much weaker than those of the other fusions. The biosensor harboring the dnaKp-DJ::luCDABE fusion was examined for its bioluminescence production based on exposure to aromatic compounds, such as biphenyl, 4-chlorobiphenyl (4CB), 4-hydroxybenzoate (4HBA), and catechol. In particular, the bioluminescence produced by the dnaKp-DJ::luxCDABE fusion was most sensitive to 1 mM biphenyl and 4CB when exposed for 80 min, and the responses were also very strong to other aromatics. Therefore, the biosensor bearing the dnaKp-DJ::luxCDABE fusion would appear to be the most useful for the detection of aromatics and other pollutants.

A New Proposition on the Definition of the Tropical Cyclone Influence on the Korean Peninsula (한반도 영향 태풍의 정의에 대한 새로운 제안)

  • A new proposition on the definition of the tropical cyclone (TC) which influences the Korean Peninsula (KP) is presented. The definition is based upon the TC track and intensity, 34 wind swath considering the TC size, and the line of 200 nautical mile (NM) from the KP shore which is the boundary of the official warning of Korea Meteorological Administration. Four types are proposed. First type is TC that hits the KP inland. Second is TC that falls within the 200-NM boundary. Third type is TC that passes outside the 200-NM boundary but large enough to significantly influence the KP. Last, the cases for a TC which are downgraded to the midlatitude cyclone and hit the KP are included. 30-year reanalysis reveals that 21 tropical cyclones should be included in the TC list that influenced the KP during the period from 1977 to 2006, which corresponds to 3.93 TCs per year. Among them, number of type I, II, III and IV TCs turn are to be 36, 47, 10, and 16, respectively. The net increase found in the current reanalysis is 2, 5, 7, and 7 for each type.

Quality Characteristics of Kochujang Prepared with Commercial Protease (시판 Protease를 이용한 고추장의 품질 특성)

  • To study the characteristics and processing of Kochujang which is rapidly fermented by commercial enzymes, three kinds of Kochujang(KP-FA, KP-FN, and KP-BN) using commercial proteases and one Kochujang(KM) using Meju were prepared and their qualities investigated. There were only small differences in pH and acidity between each Kochujang. The moisture contents were high tendency in the three kinds of Kochujangs using the commercial proteases at 20 days of fermentation. Reducing sugars had a tendency to decrease during the fermentation in the Kochujangs using the proteases. During the first half of fermentation, the Kochujangs made with proteases showed higher amino nitrogen contents than the Kochujang(KM) made using Meju. Acidic protease activity was high in KP-FA at 20 days of fermentation and neutral protease activity was high in KP-FN and KP-BN at the beginning of fermentation. The Kochujangs made using the proteases, through 20 days of fermentation, obtained high preference in the sensory evaluation for color, texture, and overall acceptability. However, the hot taste was stronger in these Kochujangs during the fermentation.

A Study on the Machinability of High Hardness Steel in Ball End Milling (볼 엔드밀 가공에서 고경도 강재의 절삭특성에 관한 연구)

  • The STD11 and KP4 are important steels and applied to the manufacturing of the die and mold. The purpose of this study is to investigate the machinability of tool steels of STD11(HRC60) and KP4(HRC32) when machining them by using ball end milling tools coated with TiAlN. Cutting forces by using a Kistler piezo-cell type tool dynamometer, surface roughness and tool wear by using tool microscope are used in the tests. The results from the cutting tests of KP4 specimens show that 85m/min. of cutting speed and 0.32mm/rev. of feed per revolution are optimum conditions for the higher productivity and 0.26mm/rev. with the same cutting speed are optimum conditions for better surface finishing. The results from machining STD11 workpiece at 30m/min. of cutting speed and 0.17m/rev. of feed per revolution show recommended for the higher productivity. The KP4 shows relatively smaller cutting forces than STD11 and STD11 shows the better surface finish than KP4.
