Amphiregulin (AREG) Genotypes, Allele Frequencies and the First Parity Litter Size in the Pig

  • Amphiregulin (AREG), a glycoprotein that is a member of the epidermal growth factor (EGF) family, is expressed by the porcine conceptus and endometrium. AREG genotypes were determined based on an SNP in the intron 3 of the gene. Contradictory effects of AREG genotypes on reproductive traits in different pig breeds were reported previously. G allele had undesirable effect on reproductive trait in Meishan breed, while it had favorable effects in Polish Landrace and Large White. We determined AREG genotypes of 179 pigs including the Duroc, Landrace, Yorkshire, Korean native pig (KNP), and Meishan breeds. Two new SNPs were identified near the previously reported SNP in the intron 3 of AREG. Frequencies of AREG alleles among the Duroc, Landrace, Yorkshire, and KNP sows were significantly different (p<0.001), indicating association between AREG genotypes and pig breeds. The first parity litter size was significantly affected by the breeds (p=0.014), but not by AREG genotypes (p=0.148). However, there were breed and AREG genotype associated trends in the first parity litter size. The first parity litter size appeared to be higher in Duroc and KNP sows with G allele, while it appeared to be lower in Landrace sows with G allele. Significant variability of AREG alleles among pig breeds, for the first time in Duroc and KNP sows, was identified. AREG genotypes may influence reproductive traits differentially for each breed and thus, AREG genotypes may need to be considered when sows are bred to increase litter size.

Effects of Crossbreeding and Gender on the Carcass Traits and Meat Quality of Korean Native Black Pig and Duroc Crossbred

  • This study was conducted to observe the effects of crossbreeding and gender on the carcass traits and meat quality of Korean Native Black Pig (KNP) and $KNP{\times}Duroc$ crossbred ($KNP{\times}D$). A total of 50 pigs comprising seven KNP barrows, eight KNP gilts, twenty $KNP{\times}D$ barrows and fifteen $KNP{\times}D$ gilts were used in this study. Animals were reared in the same housing condition with same feed diet for six months prior to slaughter. After an overnight chilling, the carcasses were graded, and samples of Musculus longissimus dorsi were obtained for meat quality analysis. The slaughter and carcass weights and dressing percentage of $KNP{\times}D$ were higher (p<0.001) than those of KNP. The slaughter and carcass weights and backfat thickness of barrows were higher (p<0.01) than those of gilts. There were no significant difference in carcass conformation and quality grade between KNP and $KNP{\times}D$ as well as barrow and gilt. Fat content of $KNP{\times}D$ was higher (p<0.001) than that of KNP. Fat content of barrow was higher (p<0.001) than that of gilt. There was interaction between crossbreeding and gender on the fat content. KNP gilt showed higher fat content than KNP barrow whereas $KNP{\times}D$ barrow showed higher fat content than $KNP{\times}D$ gilt. Lightness, redness, yellowness, chroma and hue angle values and color preference of meat of $KNP{\times}D$ were lower (p<0.001) than those of KNP. Redness, yellowness and chroma values of meat of barrow were lower (p<0.05) than those of gilt. It is concluded that crossbreeding KNP with Duroc increases carcass productivity and meat fat but decreases meat color values and preference. Crossbreeding of KNP with Duroc produces a better fat deposition in meat of barrows than in gilts.

Dietary Chromium-methionine Chelate Supplementation and Animal Performance

  • Chromium has emerged as an essential trace mineral in nutrition. However, it readily causes toxicity because of slightly excessive dose and/or form of chromium supplement. Therefore, developing a noble form of chromium supplement which is capable of not only an increased availability but also a reduced toxicity has been a critical issue in chromium nutrition. Chromium-methionine chelate has been, so far, one of the latest developments in its kind. Although not much information is available for the chromium-methionine chelate, especially in view of animal performances upon dietary supplementation, several studies indicated chromium methionine chelate could be effective to improve meat quality by increasing muscle mass but decreasing body fat. Highly-graded beef was produced by dietary chromium methionine supplementation during fattening stage of Korean native steers. Body muscle was increased in replace of decreasing body fat in both pig and rat that were dietary supplemented with chromium methionine chelate. However, a pig farm study did not show any significant improvement of body gain upon supplementation of chromium methionine. Immune responses of pig and rat were not always dependent upon chromium form but were varied by species. These results suggest there could be a different mode of responses due to species as well as onset time of dietary supplementation of chromium methionine. It is still early to conclude the bio-efficacy of chromium methionine chelate presumably due to its recent appearance into the field. But the chelate is certainly worth more application to animal since it certainly reduced the application level of dietary chromium.

출하체중에 따른 재래돼지와 랜드레이스의 도체 및 돈육의 이화학적 특성 비교 (Comparison of Meat Quality and Physicochemical Characteristics of Pork between Korean Native Black Pigs (KNBP) and Landrace by Market Weight)

  • Results of meat quality, physicochemical characteristics, fatty acid and amino acid composition of the Korean Native Black Pig(70 and 90 kg of slaughter weight; KNBP) versus Landrace(110kg of slaughter) are as below. Compared with Landrace, the KNBP exhibited a greater lean meat yield, with no difference in backfat thickness between Landrace and KNBP. There were no difference in chemical composition between Landrace and KNBP, but pH, and cooking loss in the KNBP were superior value than those of the Landrace. These results, KNBP were affected in sensory property and cooking yield. Amino acid content of KNBP and Landrace were significant differences between Landrace and KNBP. Except for the methionine, essential amino acid contents of KNBP had significantly higher than those of Landrace. In the change of fatty acid composition, the Landrace had significantly higher percentages of palmitic(l6: 0), stearic(18: 0) and linolenic(18: 3) acids than that of KNBP, but linoleic acid(l8: 2) was decreased.

우리흑돈 종모돈이 삼원교잡종 돼지의 성장, 체형 및 부분육 생산수율에 미치는 영향 (Effects of Woori Black Pig Sire on Growth Performance, Body Shape, and Retail Cut Yield of Crossbred Pigs)

  • 우리흑돈(Woori black pig, W)은 재래흑돼지와 두록(Duroc, D)을 이용하여 재래흑돼지의 낮은 경제적 특성을 개선시키기 위해 국립축산과학원에서 개발한 신품종이다. 따라서 본 연구는 교배 종료종모돈으로써 우리흑돈이 삼원교잡종의 성장, 체형 및 부분육 생산수율에 미치는 영향을 평가하기 위해 수행되었다. 총 32두의 삼원교잡종을 교배 종료 종모돈에 기반하여 2처리구로 완전임의 배치하였다. 교배 종료종모돈으로 두록과 우리흑돈에 기반한 2개의 다른 삼원교잡종은 LYD[(Landrace×Yorkshire)×Duroc]와 LYW[(Landrace×Yorkshire)×Woori black pig]이였다. 본 연구는 시험 개시 후 53일 동안 수행되었다. 우리흑돈과 두록 종모돈은 삼원교잡종의 53일 체중, 총증체량, 일당증체량 및 90 kg 체중 도달일령에 영향을 미치지 않았다. 체장, 체고 및 흉심에 차이가 발견되지 않았으나, 등지방두께에서 LYD(17.29 mm)와 LYW(18.96 mm) 간의 유의적인 차이가 있었다(p<0.05). LYW 삼원교잡종의 등심수율(13.11%)이 LYD(13.85%)에 비해 유의적으로 낮게 나타났다(p<0.05). LYW의 목심 수율(8.99%)은 LYD(8.21%)에 비해 유의적으로 높게 나타났다(p<0.05). 본 연구결과, 우리흑돈은 삼원교잡종의 사양성적, 체형 및 생산수율에 부정적인 영향을 미치지 않으며, 교배 종료종모돈으로 사용할 수 있을 것으로 사료된다.

  • This study was carried out to compare farrowing rate and litter traits for European and American lines with boar sperm frozen in straws. Farrowing rate, litter size and mean pig weght at birth and 21 days were investigated. A total of 36 gilts Landrace, Large white and Duroc were investigated at the Chungnam Provincial Animal Breeding Station. We obtained higher farrowing rate and litter traits for European line boars compared to American line boars.

한우와 돼지에서 간의 약물 대사효소의 활성 비교 (Comparative liver drug metabolizing enzymes activities between Korean native cattle and swine)

  • Drug-metabolizing activities of Korean native cattle and swine were investigated from viewpoints of the cytochrome P-450's level, their dependent mixed function oxidase activities, the reactive oxygen species formation and cytosolic enzyme acitivities from each liver homogenates. Level of cytochrome P-450 in the liver microsome of Korean native cattle was $0.28{\pm}0.05nmole/mg$ and that in pigs $0.35{\pm}0.03nmole/mg$. Level of cytochrome $b_5$ of Korean native cattle was $0.24{\pm}0.06nmole/mg$, and that of pigs $0.2{\pm}0.05nmole/mg$, showing no difference between two species. NADPH P-450 reductase were higher in Korean native cattle ($58.3{\pm}5.3nmole/mg/min$) than in pigs ($29.9{\pm}3.8nmole/mg/min$)(p<0.01). The activities of cytochrome P-450 dependent monooxygenases such as ethoxyresorufin O-deethylase (cattle, $96.5{\pm}12.5nmole/mg/min$ ; pigs, $13.6{\pm}2.1nmole/mg/min$), N-benzphetamine N-demethylase (cattle, $5.23{\pm}0.82nmole/mg/min$ ; pigs, $0.76{\pm}0.3nmole/mg/min$) and aniline hydroxylase (cattle, $0.95{\pm}0.1nmole/mg/min$ ; pigs, $0.33{\pm}0.08nmole/mg/min$) were much higher in Korean native cattle than in swine(p<0.01). However, the activity of testosterone $7{\alpha}$-hydroxylase was higher in swine ($90.4{\pm}1.2nmole/mg/min$) than cattle (cattle, $32.8{\pm}1.2nmole/mg/min$). Interestingly, testosterone $16{\alpha}$-hydroxylase, a marker enzyme for P-450 IIA was not detected in both animal species. These results suggest that Korean native cattle and pigs have high contents of P-450 IA1 and P-450 IIIA. Total sulfhydryl compound (cattle, $10.3{\pm}1.1nmole/mg$ ; Pigs, $14.5{\pm}1.8nmole/mg$) and glutathione related enzymes except glutathione reductase (cattle, $38.1{\pm}7.9nmole/mg/min$; swine, $22{\pm}3.6nmole/mg/min$) showed higher levels in swine than in Korean native cattle. Superoxide dismutase (cattle, $7.64{\pm}0.84nmole/mg/min$ ; pigs, $4.47{\pm}0.94nmole/mg/min$) and catalase (cattle, $30.4{\pm}3.7nmole/mg/min$ ; pigs, $17.2{\pm}1.8nmole/mg/min$) were remarkably higher in Korean native cattle than in swine (p<0.05).

제주도 한라산에 서식하는 도입종 야생멧돼지에 대한 분자유전학적 분석 (A Molecular Genetic Analysis of the Introduced Wild Boar Species (Sus scrofa coreanus) on Mount Halla, Jeju Island, Korea)

  • 제주도에서는 절멸된 것으로 간주되었던 멧돼지들이 최근 한라산 인근지역에서 발견되었다. 본 연구는 분자유전학적 실험기법을 바탕으로 한라산 멧돼지들이 가축돼지들과 이종교배된 것들인지를 조사하였다. 또한 동일 종내에서의 유전적 유연관계와 분자 성판별을 시험하였다. 가축돼지 품종들(Landrace, Large White, Berkshire, Hampshire, Duroc)과의 교배여부는 핵 DNA와 미토콘드리아 DNA에서 4 종류의 분자 표지인자(MC1R, KIT, 조절영역, ND2)를 적용하여 시험하였다. 야생멧돼지 집단의 모든 개체들이 동일한 mtDNA 조절영역 서열을 나타내었고, 그 서열들은 중국 동북부 재래돼지들과 동일하였으나 기존에 보고된 한반도 멧돼지의 서열들과는 다른 것으로 확인되었다. 이상의 연구결과는 한라산 멧돼지집단이 중국 재래돼지 품종들과 근연이면서, 기존에 연구되지 않았던 유전적 계통에서 유래한 것으로 사료된다. 분자 성판별 결과 수컷에 비해 암컷이 2 배 이상으로 확인되어, 한라산 야생멧돼지 집단이 팽창하고 있으며, 조절하지 않으면 집단 규모는 극적으로 증가할 것이다.

Porcine Fatty Acid Synthase Gene Polymorphisms Are Associated with Meat Quality and Fatty Acid Composition

  • We assessed the effects of single-nucleotide polymorphisms (SNPs) within the porcine fatty acid synthase (FASN) gene regarding meat quality and fatty acid composition in two pig populations: Korean native pigs (KNP) were crossed with Yorkshire (YS) $F_2$, and KNP were crossed with Landrace (LR) $F_2$. Direct DNA sequencing using eight KNP and eight YS pigs revealed two SNPs: c.265C>T (silent) in exon 4 and c.6545A>C (Asn${\rightarrow}$His) in exon 39. The frequency of the two SNPs was analyzed using the polymerase chain reaction-restriction fragment length polymorphism method in seven pig breeds and their association with meat quality traits and fatty acid composition was studied. In the $KNP{\times}YSF_2$ population, both SNPs were significantly associated with the level of monounsaturated fatty acids, including palmitoleic (C16:1) and oleic acid (C18:1) (p<0.005). c.6545A>C was associated with intramuscular fat content in both populations. Our results indicate that variations in c.265C>T and c.6545A>C of the pig FASN can be used to select animals with better fatty acid composition and meat quality. Moreover, KNP was a useful breed for identifying genetic factors affecting meat quality and fatty acid composition and for producing high quality pork.

Comparison of prediction accuracy for genomic estimated breeding value using the reference pig population of single-breed and admixed-breed

  • This study was performed to increase the accuracy of genomic estimated breeding value (GEBV) predictions for domestic pigs using single-breed and admixed reference populations (single-breed of Berkshire pigs [BS] with cross breed of Korean native pigs and Landrace pigs [CB]). The principal component analysis (PCA), linkage disequilibrium (LD), and genome-wide association study (GWAS) were performed to analyze the population structure prior to genomic prediction. Reference and test population data sets were randomly sampled 10 times each and precision accuracy was analyzed according to the size of the reference population (100, 200, 300, or 400 animals). For the BS population, prediction accuracy was higher for all economically important traits with larger reference population size. Prediction accuracy was ranged from -0.05 to 0.003, for all traits except carcass weight (CWT), when CB was used as the reference population and BS as the test. The accuracy of CB for backfat thickness (BF) and shear force (SF) using admixed population as reference increased with reference population size, while the results for CWT and muscle pH at 24 hours after slaughter (pH) were equivocal with respect to the relationship between accuracy and reference population size, although overall accuracy was similar to that using the BS as the reference.
