작약에 의해 유도되는 HSP72 및 HO-1 유전자의 간독성 보호 효능 (Paeonia lactiflora Pall Pprevents $H_2O_2$-induced Hepatotoxicity by Increasing HSP72 and HO-1)

  • In Korea, China, and Japan, Paeonia lactiflora Pall (PL) has been used in the treatment of rheumatoid arthritis, hepatitis, and fever for more than 1200 years. It has been reported that PL has protective effects against $H_2O_2$-induced oxidative stress and LPS-induced liver inflammation. However cellular and molecular mechanism of PL protection against oxidative stress has not fully been elucidated. Here, we describe that the water-soluble extract of PL decreased $H_2O_2$-induced hepatotoxicity. This hepatoprotective effect of PL is reason to decrease the level of intracellular reactive oxygen species (ROS) and increase expression of heme oxygenase 1 (HO-1) and heat shock protein 72 (HSP72) which proteins are involved in protecting the cells from stress like as oxidative stress. We also elucidated that hepatoprotective effect of PL was abolished by knock down of HO-1 and HSP72 by siRNA. These results suggest that the increasing of HO-1 and HSP72 protein by PL treatment might be participated in hepatoprotective effect against oxidative stress such as $H_2O_2$.

Effect of Modulation of hnRNP L Levels on the Decay of bcl-2 mRNA in MCF-7 Cells

  • It has been shown that CA repeats in the 3'-untranslated region (UTR) of bcl-2 mRNA contribute the constitutive decay of bcl-2 mRNA and that hnRNP L (heterogenous nuclear ribonucleoprotein L) interacts with CA repeats in the 3'-UTR of bcl-2 mRNA, both in vitro and in vivo. The aim of this study was to determine whether the alteration of hnRNP L affects the stability of bcl-2 mRNA in vivo. Human breast carcinoma MCF-7 cells were transfected with hnRNP L-specific shRNA or hnRNP L-expressing vector to decrease or increase hnRNP L levels, respectively, followed by an actinomycin D chase. An RT-PCR analysis showed that the rate of degradation of endogenous bcl-2 mRNA was not affected by the decrease or increase in the hnRNP L levels. Furthermore, during apoptosis or autophagy, in which bcl-2 expression has been reported to decrease, no difference in the degradation of bcl-2 mRNA was observed between control and hnRNP L-knock down MCF-7 Cells. On the other hand, the levels of AUF-1 and nucleolin, transacting factors for ARE in the 3'UTR of bcl-2 mRNA, were not significantly affected by the decrease in hnRNP L, suggesting that a disturbance in the quantitative balance between these transacting factors is not likely to interfere with the effect of hnRNP L. Collectively, the findings indicate that the decay of bcl-2 mRNA does not appear to be directly controlled by hnRNP L in vivo.

Expression of Cu/Zn SOD Protein Is Suppressed in hsp 70.1 Knockout Mice

  • Heat shock proteins (HSPs) are known to protect cells from oxidative stress and other types of injuries. We previously reported the neuroprotective effect of HSP70 following cerebral ischemia and reperfusion using hsp 70.1 knockout (KO) mice. However, the precise role of HSP70 in neuroprotection has not been established yet. The purpose of this study was to investigate the relationship between HSP70 and antioxidant enzymes using hsp 70.1 KO mice. The activities of both SOD-1 and SOD-2 were significantly decreased in hsp 70.1 KO mice than in the wild type (WT) littermates. SOD-1 protein level in the hsp 70.1 KO mice was lower than that of WT. We speculate that HSP70 might be involved in regulation of expression of SOD-1 at the level of transcription or by post-transcriptional modification.

급속압축팽창기를 이용한 DME의 HCCI 연소 (HCCI Combustion of DME in a Rapid Compression and Expansion Machine)

  • Compression ignition of homogeneous charges in IC engines indicates possibilities of achieving the high efficiency of DI diesel engines with low level of NOx and particulate emissions. The objectives of this study are to further understand the characteristics of the HCCI(Homogeneous charge compression ignition) combustion and to find ways of extending the rich HCCI operation limit in an engine-like environment. DME fuel is supplied either in the form of premixture with air or directly injected in the combustion chamber of a rapid compression and expansion machine under the conditions of various equivalence ratio and injection timing. The cylinder pressure is measured and the rate of heat release is computed from the measured pressure for the analysis of the combustion characteristics. The experimental data show that the RCEM can operate without knock on mixtures of higher equivalence ratio, when DME is directly injected in the combustion chamber than introduced as a fraction of a perfect or nearly perfect premixture. Very early fuel injection timings usually employed in HCCI operation are seen to have only insignificant effects in control of ignition timing.

A Study on Furniture Design for Disassembly

  • Modernity which is superficial phenomenon set off the mass scale for mass consumption and provide uniformly artificial environment. But natural destruction, environment pollution, resources exhaustion and so on has been caused by this and now ecology is threatened by destruction and damage beyond the limitation and human beings survival is even threatened. Accordingly furniture development for environment preservation considered environment problem is the urgent real situation. Recent paradigm is the concept of Eco-design which is the green design possible to live together in symbiosis, and new types of alternative furniture are needed in Korea as well. 'Furniture for disassembly' is presented as new method for alternative furniture. Furniture for disassembly can be presented by mainly two directions. The first main characteristic is what is assembled by the use of woodworking joints technique as an assembly structure system without any hardware. The second is what is presented as the structure possible to be assembled by simple manual tools with hardware without any glue. The advantages of furniture for disassembly are environment preservation, space application, transportation efficiency and shapeliness. In manufacture method which is different from present furniture, the application of traditional truss technique which uses various types of custom-made and connection technique in case of assemble structure system without hardware is the typical differences. This assembly method expects not only interest induction about assembly and disassembly of diagram per sub materials but also the development of emotion, the improvement of collaboration, space perception ability and shape sense, the improvement of solid body structure insight and so on, when it use in the furniture for children with the application to many kinds of structure with BANGDOOSANJ (Wedged), JUMUGJANGBU (Dovetail) or NABIEUNJANG (Dovetail Keys) and so on.

Analysis of Semen Parameters in α1,3-Galactosyltransferase-/- Boars

  • It is very difficult to get the information about semen quality analysis in transgenic pigs because of limited numbers and research facilities. Therefore, in the present study, we analyzed the semen quality of transgenic boars generated for xenotransplantation research. Briefly, the semen samples were collected from 5 homozygous ${\alpha}1,3$-Galactosyltransferase knock-out ($GalT^{-/-}$) transgenic boars and immediately transported to the laboratory. These semen samples were decupled with DPBS and conducted to analyze semen parameters by a computer-assisted semen analysis (CASA) system. The boar semen were examined all 12 parameters such as total motility (TM), curvilinear velocity (VCL), straight line velocity (VSL), average path velocity (VAP), and hyperactivated (HYP), etc. In results, among the 5 $GalT^{-/-}$ boars, three boars (#134, 144, and 170) showed normal range of semen parameters, but #199 and 171 boars showed abnormal ranges of semen parameters according to standard ranges of semen parameters. Unfortunately, #171 boar showed azoospermia symptom with rare sperm counts in the original semen. Conclusively, assessment of semen parameters by CASA system is useful to pre-screening of reproductively healthy boar prior to natural mating and artificial insemination for multiplication and breeding.

Multi-zone 모델링을 통한 온도성층화와 농도성층화가 존재하는 DME HCCI 엔진의 운전영역에 관한 수치해석연구 (Comparison of DME HCCI Operating Ranges for the Thermal Stratification and Fuel Stratification based on a Multi-zone Modeling)

  • This work investigates the potential of in-cylinder thermal stratification and fuel stratification for extending the operating ranges in HCCI engines, and the coupling between thermal stratification and fuel stratification. Computational results areemployed. The computations were conducted using both a custom multi-zone version and the standard single-zone version of the Senkin application of the CHEMKINII kinetics rate code, and kinetic mechanism for di-methyl ether (DME). This study shows that the potential of thermal stratification and fuels stratification for extending the high-load operating limit by a staged combustion event with reduced pressure-rise rates is very large. It was also found that those stratification offers good potential to extend low-load limit by a same mechanism in high-load. However, a combination of thermal stratification and fuel stratification is not more effective than above stratification techniques for extending the operating ranges showing similar results of fuel stratification. Sufficient condition for combustion (enough temperature for) turns misfire in low-load limit to operate engines, which also leads to knock in high-load limit abruptly due to the too high temperature with high. DME shows a potential for maximizing effect of stratification to lower pressure-rise rate due to the characteristics of low-temperature heat release.

Interleukin-10 UP-regulates TRAIL Gene Expression in the Mammary Epithelial Cell at the Involution Stage

  • Interleukin-10 (IL-10) is known as a regulator of inflammation and pathogenesis in mammalian organs, but its precise role is little known in the mammary gland. Our initial experiment showed that IL-10 expression levels in mice decreased at the lactation stage otherwise increased at the involution stage. To reveal the effects of IL-10 on the involution of mammary gland, expression profiles of the apoptosis-related genes were examined in transgenic mice expressing human IL-10 as well as in knock-out mice (IL-10-/-). Mild inflammatory legions by lymphocytes were observed in the mammary glands of transgenic lines at the lactation stage. The expression of TRAIL (Tumor necrosis factor-Related Apoptosis-Inducing Ligand) among the apoptosis-related genes was highly elevated in the transgenic mice while others were not significantly changed. Furthermore, TRAIL was down regulated by four fold in the IL-10-/- mice at the involution stage. The expression of DR4 was elevated at the involution stage of normal mice. DR4 was detected in the milk of transgenic mice but absent in that of normal mice. Our results proposed that the elevated IL-10 at the involution stage recruit lymphocytes and induce TRAIL and DR4 genes, therefore, lead to enter involution stage of mammary glands.

Bis Is Involved in Glial Differentiation of PI9 Cells Induced by Retinoic Acid

  • Previous observations suggest that Bis, a Bcl-2-binding protein, may playa role the neuronal and glial differentiation in vivo. To examine this further, we investigated Bis expression during the in vitro differentiation of P19 embryonic carcinoma cells induced by retinoic acid (RA). Western blotting and RT-PCR assays showed that Bis expression was temporarily decreased during the free floating stage and then began to increase on day 6 after the induction of differentiation. Double immunostaining indicated that Bis-expressing cells do not express several markers of differentiation, including NeuN, MAP-2 and Tuj-1. However, some of the Bis-expressing cells also were stained with GFAP-antibodies, indicating that Bis is involved glial differentiation. Using an shRNA strategy, we developed bis-knock down P19 cells and compared them with control P19 cells for the expression of NeuroD, Mash-1 and GFAP during RA-induced differentiation. Among these, only GFAP induction was significantly attenuated in Pl9-dnbis cells and the population showing GFAP immunoreactivity was also decreased. It is noteworthy that distribution of mature neurons and migrating neurons was disorganized, and the close association of migrating neuroblasts with astrocytes was not observed in P19-dnbis cells. These results suggest that Bis is involved in the migration-inducing activity of glial cells.

TonEBP suppresses adipocyte differentiation via modulation of early signaling in 3T3-L1 cells

  • TonEBP belongs to the Rel family of transcription factors and plays important roles in inflammation as well as kidney homeostasis. Recent studies suggest that TonEBP expression is also involved in differentiation of several cell types such as myocytes, chondrocytes, and osteocytes. In this study, we investigated the roles of TonEBP during adipocyte differentiation in 3T3-L1 cells. TonEBP mRNA and protein expression was dramatically reduced during adipocyte differentiation. Sustained expression of TonEBP using an adenovirus suppressed the formation of lipid droplets as well as the expression of FABP4, a marker of differentiated adipocytes. TonEBP also inhibited the expression of $PPAR{\gamma}$, a known master regulator of adipocytes. RNAi-mediated knock down of TonEBP promoted adipocyte differentiation. However, overexpression of TonEBP did not affect adipogenesis after the initiation of differentiation. Furthermore, TonEBP expression suppressed mitotic clonal expansion and insulin signaling, which are required early for adipocyte differentiation of 3T3-L1 cells. These results suggest that TonEBP may be an important regulatory factor in the early phase of adipocyte differentiation.
