OPTIMIZATION OF PARAMETERS IN MATHEMATICAL MODELS OF BIOLOGICAL SYSTEMS

  • Under pathological stress stimuli, dynamics of a biological system can be changed by alteration of several components such as functional proteins, ultimately leading to disease state. These dynamics in disease state can be modeled using differential equations in which kinetic or system parameters can be obtained from experimental data. One of the most effective ways to restore a particular disease state of biology system (i.e., cell, organ and organism) into the normal state makes optimization of the altered components usually represented by system parameters in the differential equations. There has been no such approach as far as we know. Here we show this approach with a cardiac hypertrophy model in which we obtain the existence of the optimal parameters and construct an optimal system which can be used to find the optimal parameters.

  • Three-dimensional flow analysis and numerical optimization methods are presented for the design of an axial-flow fan. Steady, Incompressible, three-dimensional Reynolds-averaged Wavier-Stokes equations are used as governing equations, and standard k-$\epsilon$ turbulence model is chosen as a turbulence model. Governing equations are discretized using finite volume method. Steepest descent method, conjugate gradient method and BFGS method are compared to determine the searching directions. Golden section method and quadratic fit-sectioning method are tested for one dimensional search. Objective function is defined as a ratio of generation rate of the turbulent kinetic energy to pressure head. Sweep angle distributions are used as design variables.

  • Three phase squirrel cage induction motor is generally adopted as a traction motor to drive high speed trains because of its robustness for surrounding environment and easy maintenance. In the design of traction molar, reduction of weight is very important in order to reduce kinetic energy to accelerate the vehicle. Therefore, in this paper, design variables of a preliminary designed traction motor to minimize its weight is determined using the optimization technique. Before the optimization process, rotor slot number is determined to reduce vibration and noise by the analysis of magnetic force. As a result, a design example to reduce weight by 12% than that of the preliminary designed motor is presented.

  • 본 연구에서는 상용 니켈-알루미나 촉매를 이용한 메탄가스-수증기 개질반응에서의 고유반응속도 상수를 결정하였다. 반응메커니즘을 반영하기 위해 Langmuir-Hinshelwood chemisorption 이론에 기반한 반응속도식을 사용하였고 반응온도($630{\sim}750^{\circ}C$) 및 반응물의 분압(S/C ratio = 2.7~3.5)을 실험변수로 설정하였다. 실험을 통해 얻어진 데이터를 기반으로 효율적인 최적화 알고리즘을 이용하여 최적 고유반응속도상수들을 결정하였다. 최종적으로 제안된 이 수학적 반응 모델은 촉매반응기의 설계 및 운전조건 최적화에 활용 가능하다.

초고속 충돌을 받는 모자형 보강 패널의 충격 저감 특성에 관한 연구 (A Study of Impact Reduction Characteristics of Hat-Shaped Stiffened Panel Under Hypervelocity Impact)

  • 본 논문은 사각 보강 패널에 사용되는 모자형 보강재의 크기 최적화에 대한 결과를 제시하였다. 보강 패널은 1500 ~ 2500 m/s 의 속도를 가지는 발사체와 충돌한다. 모자형 보강재의 크기를 결정하기 위해서 크기 최적화가 수행되었다. 크기 최적화를 수행하기 위해서는 3 개의 함수들로 구성되어 있다. 이 함수들은 목적함수, 제한함수 그리고 설계 함수로 이루어져 있다. 목적 함수는 보강 패널의 1 차 고유 진동수의 최대화가 되도록 하는 것이다. 제함 함수는 보강재의 부피가 사각 패널 부피의 10 % 이내가 되도록 하는 것이며, 설계 변수로는 모자형 보강재의 치수가 된다. 최적화된 모자형 보강재를 이용하한 보강 패널을 사용하여 초고속 충돌에 대한 시뮬레이션을 수행하였으며, 최적화된 보강재에 대해서 속도와 운동 에너지 변화에 대한 결과를 얻었다. 보강 패널의 충돌 저감을 평가하기 위해서 발사체의 운동 에너지와 속도를 무차원화 계산을 수행하여 비교 분석 하였다.

Robustness of optimized FPID controller against uncertainty and disturbance by fractional nonlinear model for research nuclear reactor

  • In this study, a fractional order proportional integral derivative (FOPID) controller is designed to create the reference power trajectory and to conquer the uncertainties and external disturbances. A fractional nonlinear model was utilized to describe the nuclear reactor dynamic behaviour considering thermal-hydraulic effects. The controller parameters were tuned using optimization method in Matlab/Simulink. The FOPID controller was simulated using Matlab/Simulink and the controller performance was evaluated for Hard variation of the reference power and compared with that of integer order a proportional integral derivative (IOPID) controller by two models of fractional neutron point kinetic (FNPK) and classical neutron point kinetic (CNPK). Also, the FOPID controller robustness was appraised against the external disturbance and uncertainties. Simulation results showed that the FOPID controller has the faster response of the control attempt signal and the smaller tracking error with respect to the IOPID in tracking the reference power trajectory. In addition, the results demonstrated the ability of FOPID controller in disturbance rejection and exhibited the good robustness of controller against uncertainty.

Optimization of Reflectron for Kinetic and Mechanistic Studies with Multiplexed Multiple Tandem (MSn) Time-of-flight Mass Spectrometry

  • Photoexcitation of a precursor ion inside a cell floated at high voltage installed in a tandem time-of-flight (TOF) mass spectrometer provides triple tandem mass spectrometric information and allows kinetic and mechanistic studies. In this work, the factors affecting, or downgrading, the performance of the technique were identified. Ion-optical and computational analyses showed that an optimum instrument could be designed by utilizing a reflectron with linear-plus-quadratic potential inside. Theoretical predictions were confirmed by tests with instruments built with different ion-optical layout. With optimized instruments, masses of intermediate ions in the consecutive dissociation of a precursor ion could be determined with the maximum error of $\pm5$ Da. We also observed excellent agreement in dynamical parameters (critical energy and entropy) for the dissociation of a model peptide ion determined by instruments with different ion-optical layout operated under optimum conditions. This suggests that these parameters can be determined reliably by the kinetic method developed previously when properly designed and operated tandem TOF instruments are used.

Pilot 규모의 모의 관망에서의 염소 농도 예측 (Prediction of Chlorine Concentration in a Pilot-Scaled Plant Distribution System)

  • The chlorine's residual concentration prevents the regrowth of microorganism in water transport along the pipeline system. Precise prediction of chlorine concentration is important in determining disinfectant injection for the water distribution system. In this study, a pilot scale water distribution system was designed and fabricated to measure the temporal variation of chlorine concentration for three flow conditions (V = 0.88, 1.33, 1.95 m/s). Various kinetic models were applied to identify the relationship between hydraulic condition and chlorine decay. Genetic Algorithm (GA) was integrated into five kinetic models and time series of chlorine were used to calibrate parameters. Model fitness was compared by Root Mean Square Error (RMSE) between measurement and prediction. Limited first order model and Parallel first order showed good fitness for prediction of chlorine concentration.

하수처리장의 고도처리 upgrading 설계와 공정 최적화를 위한 다변량 통계분석 (Design of a Wastewater Treatment Plant Upgrading to Advanced Nutrient Removal Treatment Using Modeling Methodology and Multivariate Statistical Analysis for Process Optimization)

  • 하수처리 시스템에서의 생물학적 영양염류 기준이 강화됨에 따라, 표준활성슬러지공법으로 운전 중인 하수처리장의 고도처리 공법으로의 개보수 필요성이 증가하고 있다. 그러나 실제 하수처리 시스템에서의 다양한 유입조건 및 운전조건의 복잡한 반응 구성으로 인해 실험을 통하여 개보수된 고도처리공법의 최적조건을 찾는 것은 쉽지 않은 일이며, 이는 많은 시간과 비용을 소모하여 비효율적이다. 따라서 본 연구에서는 활성슬러지공정모델(ASMs)을 기반으로 한 하수처리장의 모델링 및 시뮬레이션 기법을 통하여 하수처리장의 고도처리공법으로의 upgrading 설계를 수행하며, 이를 통계적이며 체계적으로 접근하기 위해 반응표면분석법(Response surface method)을 통한 고도처리공법의 설계 최적화를 수행하였다. 또한 실규모 하수처리장에서의 운전 최적화를 위해서는 하수처리의 동력학적 매개변수에 대한 정확한 분석이 수행되어야 한다. 본 연구에서는 다변량 통계분석 기법인 부분최소승자법(PLS)을 통하여 하수처리 시스템의 동력학적 매개변수 간의 상관관계를 파악하며, 고도처리공법 하수처리장의 운전 결과에 가장 큰 영향을 미치는 매개변수를 도출하였다. 본 연구를 통해 하수처리장의 고도처리공법 upgrading 설계 및 운전 최적화를 위한 방법론을 제시하였으며, 이를 통하여 설계시간 및 경비 절감 등 고도처리공법으로의 고효율적인 개보수가 가능할 것으로 예상된다.

곡관부 하류에 핀휜이 부착된 회전 냉각유로의 최적설계 (Optimization of a Rotating Two-Pass Rectangular Cooling Channel with Staggered Arrays of Pin-Fins)

  • This study investigates a design optimization of a rotating two-pass rectangular cooling channel with staggered arrays of pin-fins. The radial basis neural network method is used as an optimization technique with Reynolds-averaged Navier-Stokes analysis of fluid flow and heat transfer with shear stress transport turbulent model. The ratio of the diameter to height of the pin-fins and the ratio of the streamwise spacing between the pin-fins to height of the pin-fin are selected as design variables. The optimization problem has been defined as a minimization of the objective function, which is defined as a linear combination of heat transfer related term and friction loss related term with a weighting factor. Results are presented for streamlines, velocity vector fields, and contours of Nusselt numbers, friction coefficients, and turbulent kinetic energy. These results show how fluid flow in a two-pass square cooling channel evolves a converted secondary flows due to Coriolis force, staggered arrays of pin-fins, and a $180^{\circ}$ turn region. These results describe how the fluid flow affects surface heat transfer. The Coriolis force induces heat transfer discrepancy between leading and trailing surfaces, having higher Nusselt number on the leading surface in the second pass while having lower Nusselt number on the trailing surface. Dean vortices generated in $180^{\circ}$ turn region augment heat transfer in the turning region and in the upstream region of the second pass. As the result of optimization, in comparison with the reference geometry, thermal performance of the optimum geometry shows the improvement by 30.5%. Through the optimization, the diameter of pin-fin increased by 14.9% and the streamwise distance between pin-fins increased by 32.1%. And, the value of objective function decreased by 18.1%.
