Kinesin-13, a Motor Protein, is Regulated by Polo-like Kinase in Giardia lamblia

  • Kinesin-13 (Kin-13), a depolymerizer of microtubule (MT), has been known to affect the length of Giardia. Giardia Kin-13 (GlKin-13) was localized to axoneme, flagellar tips, and centrosomes, where phosphorylated forms of Giardia polo-like kinase (GlPLK) were distributed. We observed the interaction between GlKin-13 and GlPLK via co-immunoprecipitation using transgenic Giardia cells expressing Myc-tagged GlKin-13, hemagglutinin-tagged GlPLK, and in vitro-synthesized GlKin-13 and GlPLK proteins. In vitro-synthesized GlPLK was demonstrated to auto-phosphorylate and phosphorylate GlKin-13 upon incubation with [γ-32P]ATP. Morpholino-mediated depletion of both GlKin-13 and GlPLK caused an extension of flagella and a decreased volume of median bodies in Giardia trophozoites. Our results suggest that GlPLK plays a pertinent role in formation of flagella and median bodies by modulating MT depolymerizing activity of GlKin-13.

농촌지역의 가족/친족의례 실태 분석 (An Analysis of the Family/Kin Rites in Rural Area)

  • The purpose of this study was to investigate the general tendency of family/kin ritual transition and to find out related variables. For this purpose, 593 subjects of rural family were interviewed with questionnaires. The major results of the study were summarized as follows: 1. It was discovered that the characteristics of the ritual transition among the rural families is same as the urban families, i.e. westernization, commercialization, and socialization. But the speed of rural area was relatively slow. 2. In contemporary family/kin rites, traditional structure coexisted with external westernized aspects by the group who were elderly, lowly educated, lowly earned and Buddhist. 3. The family/kin rites were more significantly different depending on the age than gender, and the religion was the significant variable to the family/kin rites. So the family/kin ritual management education programs were needed.

도시지역의 가조/친족의례 실태 분석 (An Analysis of the Family/Kinship Rites in Urban Area)

  • The purpose of this study is to investigate actual conditions of family/kin ritual transition and to find out relationships to socio-economic variables. For this purpose, 716 subjects of urban family were interviewed with questionnaires. The results of this study may be summarized as follows: 1. It was discovered the pervasive themes of family/kin ritual transition: westernization, commercialization, socialization, and cultural anomie. 2. In contemporary family/kin rites, traditional structure coexists with external westernized aspects under the influnce of industrialization and commercialization. The rites were continued to provide a place where participants reproduce the ideology of patriarchal family group, especially the strong parent-child ties and narrow kin relationships. 3. Respondents who support traditional ritual style are old, lower class, and Buddhist. 4. Respondents who support westernization and socialization of family/kin rites are young, higher class, and Christian. But family/kin rites are not affected by sex relatively.

가족의 사회관계망 유형화 연구 - 도시 핵가족 주부를 중심으로 - (A Study on the Types of Social Networks of Housewives in Urban Nuclear Families)

  • The purpose of this study was to identify the types of social networks of urban housewives according to different network composition patterns and to analyze the structural and functional characteristics of identified types. The data used in this study were collected from 589 full-time housewives residing in Taejeon city. The major findings are as follows: 1) The social networks of housewives in urban nuclear families were classified into eight types: the kin network, the non-kin network, the kin-centered network, the friend-centered network, the neighbor-centered network, the associate-centered network, the parallel network, and the decentralized network. 2) The structual characteristics (size, density, homogeneity, duration, proximity, frequency, closeness, direction) varied according to the type. The kin network type and the non-kin network type showed extreme degrees in network characteristics. The parallel network type and the decentralized network type showed an average level of network characteristics. The kin-, friend-, neighbor-, and the associate-centered types showed network characteristics of an intermediate level between the single-category types and the decentralized type. 3) The average levels of function of social network types were different in only two(service support, interference) of the six function areas(emotional support, service support, material support, information support, social companionship support, interference). The average level of service support by the non-kin network type was higher than other types. The average level of interference by the kin-centered network type was higher than other types, and that of the neighbor-centered network type was lower than other types. On the other hand, the total amount of function performance of social network types was different in all function areas. The total amount of social support given by the decentralized network type was greater than the other types. The total amount of interference given by the non-kin network type was smaller than the other types.

한국아동의 친척명 분류, 서열, 군집 수행의 비교 (Comparison of Performance in Classification, Seriation, and Grouping of Kin Terms in Korean Children)

  • This study investigated developmental change with reference to continuity theory in the acquisition of concepts of kin relation, task difficulty with reference to cognitive complexity, and interrelationships in the performance of cognitive tasks of kinship concepts with reference to cognitive parallelism. The subjects consisted of 6-, 8-, 10, and 12-year-old randomly selected children attending kindergartens or elementary schools in Seoul. The schools were located in various residental areas regarded as either middle or lower class. The 81 boys and 80 girls participated in 3 experiments on classification, seriation, and grouping. The instrument for the classification, seriation, and grouping tasks was composed of 10 10cm black on white line drawings of the head and upper torso area of persons in kin relationship. The data was analyzed with MANOVA. A significant age effect was found in the 3 quasi- experiments. There were significant effects on task difficulty. The biosocial power distribution indirectly influenced children's acquisition of kin relational concepts; that is, children performed better in male-kin than in female-kin tasks. There was a high correlation in performance between the 3 cognitive tasks. These findings support the continuity theory (except for seriation), a model which arranges kin-names in order of cognitive load, the centric status of men in society, and the theory of cognitive developmental parallelism.

Longitudinal Patterns of Financial Kin Support and Welfare to Single Mother Families

  • This study examines longitudinal patterns of financial kin support and welfare given to single mother families using life table techniques. Drawing on the 1985-1993 Panel Study of Income Dynamics(PSID), I followed 560 single mothers from the time they began a single motherhood spell. About 30 percent of these single mothers received financial support from relatives, and 40 to 50 percent received AFDC and Food Stamps, respectively. Results indicate that kin support is more likely to be given in a transitional period and tends to decrease over time. In comparison, receiving welfare appears positively time-dependent.

American And Korean Consumers Perceived Importance of Group Identity on Gift Giving Purchase Behavior

  • This study examines the importance of group identity (kin, friends, co-workers) when gift recipient changes among American and Korean consumers. Female college students and academicians completed a self-administered questionnaire. Four hundred fifty-eight respondents evaluated the perceived importance of group identity when buying an apparel gift for kin, friends, and co-workers. The results suggest that the importance of group identity may influence the type of gift a recipient receives. The results show that when buying a gift for kin, friends, and co-workers that both young and older American consumers place greater importance on self rather than the opinion of other group. The older and younger Korean respondents rated the opinion of each group (kin, friends, and co-workers) to be more important than self. Recommendations for future research on the affect of culture on consumer purchases are suggested.

In Vitro Propagation Through Nodal Explants in Helicteres isora L., a Medicinally Important Plant

  • Helicteres isora is medicinally important plant effective against asthma, diabetes, hypolipidemia, HIV, besides a good source of diosgenin. Seed dormancy and low rate of natural fruit production make this plant a perfect candidate for developing an in vitro method useful for its clonal propagation and further biotechnological developments. This is the first report on in vitro production of this plant. Nodal explants obtained from aseptically germinated seedlings were cultured on MS medium (Murashige and Skoog 1962) fortified with indole-3-acetic acid (IAA) ($0.57-22.83\;{\mu}M$), indole-3-butyric acid (IBA) ($0.41-16.58\;{\mu}M$), 6-benzylaminopurine (BA) ($0.44-17.75\;{\mu}M$) and kinetin (Kin) ($0.46-13.94\;{\mu}M$) either singly or in combinations of IAA + BA, IAA + Kin and BA + Kin. Combinations of cytokinins (BA and Kin) were most suitable for multiple shoot induction and $13.94\;{\mu}M\;Kin\;+\;13.31\;{\mu}M\;BA$ was optimum (79% frequency) associated with high number of microshoots (7.1 shoots per explant) after 20 days of culture. Maximum shoot elongation and proliferation (10 shoots per explant with 4.8 cm average height) was achieved on MS media containing $2.32\;{\mu}M\;Kin\;+\;2.22\;{\mu}M\;BA\;+\;2.85\;{\mu}M\;IAA$. High rooting frequency (70%) was achieved on MS medium (1/2 basal strength) fortified with $4.14\;{\mu}M$ IBA, while activated charcoal showed inhibitory effects on rooting. Hardening was done with 76% survival rate and these plants were growing without any visual defects and morphologically mimicking the naturally growing plants.

한국의 가족 및 친족간의 접촉빈도와 사회적 지원의 양상: 국제간 비교의 맥락에서 (Contact frequency and Social Supports among Korean Kin: From the comparative perspective)

  • 본 연구의 목적은 한국의 가족 및 친족관계의 특성을 국제 비교적 맥락에서 분석하는 것이다. ISSP (International Social Survey Program)의 모듈인 '사회관계와 지원체계'를 조사한 2004년 한국일반종합조사의 자료를 바탕으로 하여, 한국의 친족 간 접촉빈도와 사회적 지원에서 친족이 차지하는 위치를 비교적 시각에서 분석하였다. 연구결과는 한국의 대면적 친족접촉빈도가 조사대상 국 중 최하위 권에 있으며, 한국의 가족 및 친족이 정서적 지원보다는 도구적 지원의 대상으로 작동하는 성격이 보다 강함을 보여주고 있다. 한국의 낮은 친족접촉빈도는 한국의 경제발전 수준이나, 강한 가족주의의 전통으로는 쉽게 설명되기 어렵다는 점에 주목하여, 유교적 전통 중 위계적 집합주의가 섬소한 대면적 접촉빈도를 설명할 수 있음을 주장하였다. 사회적 지원과 관련하여서는 한국의 가족주의가 도구적 가족주의의 성격을 가지고 있음을 확인하는 한편, 친족집단의 낮은 정서적 지원정도와 위계적 집합주의와의 관련이 논의되었다. 또, 이러한 낮은 정서적 지원의 양상이 반드시 한국의 가족관계가 비정서적인 것을 의미하는 것은 아니라는 것이 지적되었다.

Conservation of Thymus pallidus Cosson ex Batt. by shoot tip and axillary bud in vitro culture

  • Here, we describe an efficient and rapid protocol for the micropropagation of Thymus pallidus Cosson ex Batt., a very rare medicinal and aromatic plant in Morocco. After seed germination, we tested the effect of different macronutrients, cytokinins alone or in combination with gibberellic acid (GA3) or auxins, on T. pallidus plantlet growth. We found that Margara macronutrients (N30K) had the best effect on the in vitro development of the plantlets. The addition of 0.93 μM/L 1,3-diphenylurea (DPU), 0.46 μM/L adenine (Ad), and 0.46 and 0.93 μM/L kinetin (Kin) resulted in the best shoot multiplication and elongation. In addition, the combination of 0.46 μM/L Kin, DPU, or Ad with gibberellic acid, in particular, 0.46 μM/L Ad + 0.58 μM/L GA3 and 0.46 μM/L Kin + 1.15 μM/L GA3, led to better bud and shoot multiplication. Moreover, the integration of the combinations of 0.46 μM/L Kin and auxins, namely 0.46 μM/L Kin + 2.85 μM/L indole-3-acetic acid (IAA), 0.46 μM/L Kin + 2.85 or 5.71 μM/L indole-3-butyric acid (IBA), and 0.46 μM/L Kin + 0.3 or 0.57 μM/L 1-naphthaleneacetic acid (NAA), in the culture medium led to better root development and optimized aerial growth. Finally, the in vitro plants from the medium containing N30K + 0.46 μM/L Kin + 2.85 μM/L IAA were successfully acclimatized; these plants served as a source for repeating in vitro culture.
