Recent Progress in Development of Vaccines against Avian Coccidiosis (조류 콕시듐증의 백신개발에 대한 최근의 진보)

  • Protozoa of the genus Eimeria are the etiologic agents of avian coccidiosis, the most economically important Parasitic disease for the poultry industry. Coccidia multiply in intestinal epithelial cells of a wide range of hosts, including livestock in addition to poultry. Chemotherapy is extensively used to control coccidiosis. However, development of drug resistance by Eimeria parasites, the intensive cost and labor involved in the identification of new anticoccidial compounds and public awareness of drug residues in foods warrant alternative methods to prevent coccidiocic in the fast growing poultry industry. For these reasons, there is a great interest in developing vaccines against avian coccidiosis. Live Eimeria vaccines confer protective immunity, however a significant disadvantage of using these types of vaccines is their pathogenicity. Live parasites with attenuated pathogenicity also usually produce immunity but may revert back to a pathogenic form and may be contaminated with other pathogenic organisms. Killed Eimeria vaccines are safer but, unlike live attenuated vaccines, are not able to generate cytotoxic T lymphocyte responses. Recombinant vaccines are biochemically purified proteins produced by genetic engineering that consist of particular epitopes or metabolites of Eimeria. Unlike live attenuated organisms, recombinant vaccines do not possess as much risk and generally are able to induce both humoral and cell mediated immunity. DNA vaccines consist of genes encoding immunogenic proteins of pathogens that are directly administered into the host in a manner that the gene is expressed and the resulting protein generates a protective immune response. Although all of these different types of vaccines have been applied to coccidiosis, this disease continues to cause substantial morbidity and mortality in the poultry industry. Future development of an effective vaccine against coccidiosis will depend on further investigation of protective immunity to Eimeria infection and identification of important immundgenic parasite molecules.

Effect of Immersion and Oral Vaccination using Formalin-killed Edwardsiella tarda against Eel Anguilla japonica (뱀장어(Anguilla japonica)에 대한 Edwardsiella tarda 불활화 백신의 침지 및 경구 투여 효과)

  • Edwardsiellosis has become a serious diseases problem in cultured eels for many years. This study was performed to investigate possibility of vaccination against edwardsiellosis caused by Edwardsiella tarda. We conducted a immersion and/or oral vaccination using formalin-killed E. tarda in eel Anguilla japonica. Three groups of fish ($26.8{\pm}1.2g$, $7.1{\pm}0.7g$ and $2.2{\pm}0.4g$) were used in this study. The protection (relative percentage survival, RPS) and serum antibody response (agglutination titer) were evaluated in the vaccinated fish. No correlation between agglutination titer and survival rate was observed in vaccinated fish. However, there was a satisfactory protective (RPS>50%) in vaccinated fish. Immersion (10 mg/mL, 1 hr) and immersion (10 mg/mL, 1 hr) plus oral (10 mg/g, 10 days) of $26.8{\pm}1.2g$, immersion (10 mg/mL, 1 hr) plus oral (10 mg/g, 10 days) of $7.1{\pm}0.7g$ showed RPS of 62.6%, 52.2% and 56.8%, respectively.

Effect of Formalin Inactivation on Viral Hemorrhagic Septicemia Virus (VHSV) (Viral Hemorrhagic Septicemia Virus (VHSV)에 대한 포르말린 불활화 의 영향)

  • Killed vaccines, developed by inactivation with formalin, have been investigated for many fish viruses. In this study, the inactivation of viral hemorrhagic septicemia virus (VHSV) by formalin was investigated based on the infectivity titer. When viral cell culture supernatants were used, the infectivity titer decreased 1,000-fold at 1 d after treatment with 0.1% (v/v) formalin, but was below the detection limit at 7 and 14 d. Moreover, neither the N nor G gene were detectable by RT-PCR immediately after formalin treatment. In western blot analysis, N protein was not detected by rabbit antiserum against VHSV KR-9225 from 2 d after formalin treatment. On the other hand, when we used a virus that was purified and concentrated ~100 times, the infectivity titer was maintained at 106.05 TCID50/mL, even at 14 d after formalin treatment, and no change in the viral structural proteins was observed. This study provides important data on the production and use of formalin-inactivated vaccines.

Immune Response of Olive flounder, Paralichthys olivaceus against Miamiensis avidus (Ciliophora: Scuticociliatida) (스쿠티카충 Miamiensis avidus에 대한 넙치의 면역반응)

  • Miamiensis avidus is a scuticociliate causing mortality in olive flounder Paralichthys olivaceus. To evaluate immune response of olive flounder against M. avidus, 2.6×106cells/fish of Formalin killed cell (FKC) was intraperitoneally (i.p.) injected, and 2.4 × 106cells/㎖ of sonicated FKC was immersion immunized to 14.9 cm (26.8g) fish. Fish were immunized 2 times with 2 weeks intervals. Antiserum from immunized fish caused agglutination and immobilization of the ciliate. In ELISA test, immunized group exhibited higher titers than control group. In addition, i.p. immunized fish showed higher phagocytic ratio, phahgocytic index and chemotaxic activity. To evaluate in vivo efficacy of FKC on fish, 1.43 × 105cells/fish was i.p.. injected, and 2.2 × 105 cells/㎖ of sonicated cell was immersed into 8.6cm (6.3g) fish. After 2 times immunization with 2 weeks intervals, fish were infected with 2.0 × 104 and 2.0 × 103 cells/ml of live ciliates by immersion. After 3 weeks, cumulative mortality was lower in the i.p. immunized group and mortality was delayed in immersion immunized group. In conclusion, specific immune response of oliver flounder against M. avidus was elevated after immunization and these immune response may prevent and/or delay for the M. avidus infection to olive flounder.

  • A cluster of severe pneumonia of unknown etiology in Wuhan City, Hubei province in China emerged in December 2019. A novel coronavirus named severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) was isolated from lower respiratory tract sample as the causative agent. The current outbreak of infections with SARS-CoV-2 is termed coronavirus disease 2019 (COVID-19) by the World Health Organization (WHO). COVID-19 rapidly spread into at least 114 countries and killed more than 4,000 people by March 11, 2020. WHO officially declared COVID-19 a pandemic on March 11, 2020. There have been 2 novel coronavirus outbreaks in the past 2 decades. The outbreak of severe acute respiratory syndrome (SARS) in 2002-2003 caused by SARS-CoV had a case fatality rate of around 10% (8,098 confirmed cases and 774 deaths), while Middle East respiratory syndrome (MERS) caused by MERS-CoV killed 858 people out of a total 2,499 confirmed cases between 2012 and 2019. The purpose of this review is to summarize known-to-date information about SARS-CoV-2, transmission of SARS-CoV-2, and clinical features of COVID-19.

Effect of Dietary Vitamin E Supplementation on Serum α-Tocopherol and Immune Status of Crossbred Calves

  • An experiment was conducted with twenty crossbred male calves (7-15 days old) divided into 4 different experimental groups on the basis of body weights to study the effect of vitamin E supplementation on the serum ${\alpha}$-tocopherol concentration and immune response of the calves. All the calves were fed on milk and calf starter up to 13 weeks and afterwards, they were fed on concentrate mixture and oat hay up to 32 weeks of age. In addition, the calves in groups I, II, III and IV were supplemented with 0, 125, 250 and 500 IU feed grade DL-${\alpha}$-tocopheryl acetate, respectively. Blood samples were collected at 0 day and subsequently at 1, 2, 3, 4, 6 and 8 months of age to monitor the serum ${\alpha}$-tocopherol concentration in crossbred calves. After 24 weeks of experimental feeding, 4 animals from each group were intramuscularly inoculated with single dose (3 ml) of Haemorrhagic septiceaemia (Pasteurella multocida P52 strain) oil adjuvant vaccine. The cumulative group mean serum ${\alpha}$-tocopherol concentration (${\mu}g/100ml$) was 88.12, 210.11, 235.21 and 294.02 in-groups I, II, III and IV, respectively and differed significantly (p<0.001) among the four groups. Lymphocyte stimulation indices (LSI) did not differ among the groups significantly. The pooled mean ELISA antibody titer against Pasteurella multocida (P52 strain) was 788.02, 926.85, 1,214.00 and 1,109.51 for group I, II, III and IV, respectively, which indicated higher antibody titer in groups supplemented with vitamin E as compared to the control group. It may be concluded that vitamin E supplementation increased the ${\alpha}$-tocopherol concentration in serum and dietary supplementation of vitamin E at higher level has a humoral immune enhancing effect against killed bacterial antigen.

  • 2004 summer, most of Korean were shocked from serial killer Ryo young chul's criminal behavior. He had kept anger to society and selected the victim randomly. He killed many females and senior persons. Korean National Police's investigation method to homicide depend on traditional homicide investigative methods. But serial killer's criminal behaviour is evolving frequently. Therefore traditional investigative method is no effective to serial homicide anymore. If traditional investigative method is old version virus vaccine, serial killer's criminal behaviour is newly appeared computer virus. Fortunately all of criminal has modus operandi independently. Any person comes into contact with an object or another persons, a cross-transfer of physical evidence happens. In the future it is impossible to escape from confrontation with serial killer. It can be prospect that more severe copycat serial killer will appear in Korean society. Based on the Ryo young chul's criminal behavior. I'll try to analyse the serial killer's modus operandi through various aspects.

Evaluation of the cost-effectiveness of ASF detection with or without the use of on-field tests in different scenarios, in Sardinia

  • African swine fever (ASF) is a highly contagious disease of domestic pigs and wild boars (WBs). Without a vaccine, early antibody and antigen detection and rapid diagnosis are crucial for the effective prevention of the disease and the employment of control measures. In Sardinia, where 3 different suid populations coexisted closely for a long time, the disease persists since 1978. The recent ASF eradication plan involves more stringent measures to combat free-ranging pigs and any kind of illegality in the pig industry. However, critical issues such as the low level of hunter cooperation with veterinary services and the time required for ASF detection in the WBs killed during the hunting season still remain. Considering the need to deliver true ASF negative carcasses as early as possible, this study focuses on the evaluation and validation of a duplex pen-side test that simultaneously detects antibodies and antigens specific to ASF virus, to improve molecular diagnosis under field conditions. The main goal was to establish the specificity of the two pen-side tests performed simultaneously and to determine their ability to detect the true ASF negative carcasses among the hunted WBs. Blood and organ samples of the WBs hunted during the 2018/2019 hunting seasons were obtained. A total of 160 animals were tested using the pen-side kit test; samples were collected for virological and serological analyses. A specificity of 98% was observed considering the official laboratory tests as gold standards. The new diagnostic techniques could facilitate faster and cost-effective control of the disease.

해조류 첨가사료가 돌돔의 성장과 면역기능에 미치는 영향

  • 양식 생물의 질병 제어는 예방과 치료를 통해 이루어진다. 그러나 항생제의 오ㆍ남용으로 인하여 약제 내성균의 증가와 치료 효과가 감소하고, 나아가 식품안전성 문제로 양식생물의 소비를 위축시켜왔다. 따라서 질병이 발생한 후에 시행하는 화학약품 치료법은 이미 한계에 달하였다. 이에 따라 사전에 질병을 차단하는 예방법인 vaccine이나 질병에 대한 저항능력을 키우는 비특이적 면역 증강물질에 대한 관심이 꾸준히 높아지고 있다. 해조류에는 다량의 lectin이 포함되어 있는데, Lectin은 당에 결합하는 단백질의 총체적인 이름으로, defence molecule로 작용하여 어류가 섭취할 경우 외래 항원으로 인식하여 지속적으로 면역 기능을 활성화시키는 것으로 알려져 있다. 따라서 본 연구는 돌돔사료에 kelf meal을 첨가하여 공급한 후 이들의 성장과 비특이적 면역기구에 미치는 효과를 조사하였다. 실험사료는 상업용 kelf meal을 0%, 2%, 5% 첨가하고, 2반복으로 500L FRP 탱크에서 8주간 사육하였다. 실험어류는 40g 내외의 돌돔을 대상으로 하였고, 사료공급 4주 및 8주에 각 실험구의 돌돔을 대상으로 비특이적 면역기구에 미치는 효과를 조사하였다. 체액성 면역 조사로는 혈청 및 점액 lysozyme 활성과 Escherichia coli에 대한 혈청 내 보체의 살균 능력을 조사하였고, 세포성 면역 조사는 전신 식세포의 NBT 환원 실험과 말초혈액의 식세포 식균능을 측정하였다. 식균능 시험은 Escherichia coli FKC (formalin killed cell)에 대한 식균율 및 식균지수로 나타내었다. 나아가 돌돔 말초혈액의 혈구조성에 미치는 영향을 조사하기 위하여 적혈구 5,000 세포당 임파구, 전구, 호중구의 수를 계수하였다. 8주간의 사육결과 사료계수는 각각 1.20, 1.25, 1.42로 대조구와 2% 첨가구는 큰 차이가 없었으나, 5% 첨가구는 나빠지는 경향을 보였다. 사료섭취량은 kelf meal의 첨가량이 늘어나면서 감소하는 경향이었다. 비특이적인 면역 기구에 대한 자극 효과는 체액성 면역의 지표로 측정한 lysozyme의 활성도는 혈청과 점액 모두에서 대조구보다 실험구에서 활성이 높았고, 혈청 lysozyme 활성은 5% 첨가구에서 가장 높았다. 혈청 내 보체의 살균능은 실험구간 차이가 없었다. 세포성 면역의 지표로 측정한 전신 식세포의 $O_2$ 생성능(NBT 환원능)은 5% 첨가구에서 가장 높았다. 말초 혈액의 식균능은 식균율과 식균지수 모두 실험구에서 높았고, 특히 5% 첨가구에서 가장 높았다. 말초 혈액 혈구조성의 변화는 임파구와 전구의 경우 실험구에서 높았으나 호중구에서는 차이가 없었다. 이상의 결과, 양식 돌돔에 kelf meal을 2% 첨가하는 것이 사료계수에 큰 영향을 미치지 않고, 돌돔의 체액성 및 세포성 면역을 적절히 자극하는 것으로 나타나, 돌돔의 기능성 사료 첨가물로서 이용 효과가 있을 것으로 기대된다.

Effect of sodium butyrate on performance, immune status, microarchitecture of small intestinal mucosa and lymphoid organs in broiler chickens

  • Objective: This study aimed to examine the effect of sodium butyrate (SB) on growth performance, immune status, organs weights, and microarchitecture of lymphoid organs and small intestine. Methods: A total of 120, 1-d-old broiler chicks were distributed into the following four treatment groups: corn-soy based basal diet (BD) without supplement (control), or the same BD supplemented with 0.1 g/kg zinc bacitracin (ZnB), 0.5 g/kg SB (SB-0.5), or 1.0 g/kg SB (SB-1), respectively. Six birds/group were killed on d-21 and d-35, and samples were collected. Results: Cell-mediated immune response at 48 h post-Phytohemagglutinin-P injection, and antibody titer against Newcastle disease vaccine and sheep red blood cells on d-35 was noted higher (p<0.05) in SB-1 compared to ZnB and control. Lower (p<0.05) feed conversion ratio (FCR) was attained by the supplemented groups. Thymus and spleen weighed more (p<0.05) in SB-1, and bursa registered more (p<0.05) weight in both SB groups compared to control. On d-21, areas of thymus medulla and spleen germinal centers were noted higher (p<0.05) in SB-1 group. The villus height and villus surface area increased (p<0.05) in duodenum and jejunum in both SB groups on d-21, and in SB-1 on d-35, respectively compared to ZnB and control. On d-21, number of goblet cells containing mucins of acidic nature increased (p<0.05) in all the segments of small intestines in SB-1 group compared to control, and on d-35 in ileum compared to other groups. Conclusion: In conclusion, SB improved growth performance and immunity as well as modulated morphology of lymphoid organs and gut mucosa in broiler chickens.
