절수시 Mongolian Gerbil 콩팥에서 TonEBP와 Urea transporter의 발현 변화 (Renal Expression of TonEBP and Urea Transporter in the Water-deprived Mongolian Gerbil(Meriones unguiculatus))

  • Tonicity responsive enhancer binding protein(TonEBP)는 콩팥에서 osmolyte의 세포내 축적을 촉매해 주는 전사조절인자로 높은 삼투농도에서 세포를 보호하는데 중요한 역할을 수행한다. 고장성환경은 TonEBP의 양적 증가와 핵 내 분포의 증가를 통해 TonEBP의 활성을 자극한다. 또한 TonEBP는 콩팥 수질내 요소축적에 중요한 역할을 하는 UT-A의 전사를 조절하는 것으로도 알려져 있다. 따라서 본 연구에서는 콩팥수질내 TonEBP와 UT-A의 기능과 상관관계를 밝히는 연구의 일환으로 다른 동물보다 급수가 제한된 환경에서 더 오래 살아남을 수 있는 수분대사능력을 가지고 있는 Mongolian gerbil을 이용하여, 절수로 인한 고장성환경의 유발에서 TonEBP와 UT-A에 대한 발현 변화를 관찰하고자 하였다. 절수에 따른 TonEBP와 UT-A의 발현 양상을 연구하기 위해, 먼저 Mongoian gerbil 각 5마리씩 3그룹으로 나누어 절수 실험을 실시하였고, 면역조직화학법을 실시하여 다음과 같은 결과를 얻었다. 정상대조군에서 TonEBP의 면역반응성은 속수질 세포들의 핵 내에 주로 분포하였으며, 절수 7일군에서 면역조직화학검사 결과, 속수질집합관에서의 염색성은 대조군에 비해 증가하였고, 특히 바깥수질 부위에 속수질에서 요세관의 가는 부분에서의 증가가 두드러졌다. 절수 14일군에서 염색성이 대조군보다 오히려 감소하였으며, 콩팥의 조직학적 손상이 관찰되었다. UT-A의 경우 바깥수질 속줄무늬층의 짧은-헨레고리가는내림부분과 정상군에서는 미약한 양성반응을 나타낸 속수질 초기부분의 긴-헨레고리가는내림부분에서 강한 발현 양상을 나타내었고, 속수질의 초기에서 중간부위의 속수질집합관도 강한 발현 양상을 확인할 수 있었다. 그러나 속수질 말단부위의 속수질집합관은 콩팥유두 끝으로 갈수록 발현량이 감소하는 것을 확인할 수 있었다. 이상의 결과는 Mongolian gerbil을 이용한 절수모델에서 증가된 콩팥수질의 Tonicity에 의해 TonEBP의 발현이 증가하고 이에 따라 UT-A의 발현도 동반하여 증가하는 것을 확인하였고, 또한 이렇게 증가된 TonEBP는 UT의 전사를 조절하여 UT를 증가시켜 오줌농축기전을 향상시키는 것으로 생각된다. 이는 속수질 세포의 스트레스에 대한 세포방어기전으로 생각된다. 그러나 절수가 계속되면 이런 적응반응에 한계를 지나쳐 오히려 TonEBP와 UT-A의 발현이 감소함을 확인 할 수 있었다.

콩팥에서 Erythropoietin 투여로 인한 HSP70의 발현 변화 (Expression of HSP70 Immunoreactivity in EPO Treated Rat Kidney)

  • Heat shock protein 70 (HSP70)은 다양한 질병상태와 치명적인 열 손상에서 세포 및 조직을 보호하는데 중요한 역할을 하며, 또한 외부의 stress로부터 세포내 단백질의 파괴와 변화를 감소시키는 단백질로 알려져 있다. 본 연구에서는 오랜 기간 동안 조혈기관의 치료제로 콩팥에서도 세포보호효과가 있는 것으로 알려진 Erythropoietin(EPO)을 투여하여 콩팥내의 HSP70의 발현변화를 세포수준에서 관찰하고자 하였다. Sprague-Dawley계 흰쥐를 사용하여 전자현미경적 면역조직화학법으로 rHuEPO투여군과 대조군에서 HSP70의 발현변화를 관찰하였다. 대조군에서 HSP70은 콩팥의 바깥수질과 속수질에서 관찰되었으며, 특히 속수질에서 강하게 발현되었고 그 부위는 속수질집합관세포와 헨레고리의 내림가는 부분이었다. EPO 투여군에서는 속수질과 바깥수질의 내림가는 부분에서는 발현변화가 관찰되지 않았으나, 바깥수질의 집합관 세포에서 발현이 급격히 증가함을 관찰되었다. 특히 대조군에서의 핵주변부위뿐 아니라 세포내 핵상부분을 비롯한 세포막주변부위에도 강한 면역 염색성을 나타내었다. 이러한 결과는 콩팥의 바깥수질에서 stress성 단백질인 HSP70의 조절기전이 EPO에 의해 매개됨을 보여주면, 세포 stress및 질병상태에서도 이러한 기전이 작용할 것으로 생각된다.

포유동물의 신유두 및 신우상피에 대한 비교형태학적 연구 (Comparative Renal Papillary and Pelvic Epithelial Morphology of the Mammalian Kidney)

  • Anatomical features of the renal papilla and pelvis and ultrastructures of the epithelium covering these areas in four species of mammals were studied by means of light, scanning and transmission electron microscopy. In terms of the morphology of mammalian kidney types distinguished by Sperber(1944), Pfeiffer(1968) and Schmidt-Nielsen(1977), the kidneys of animal species used in this experiment were; 1) the mouse kidney with the fornix between a long conical papilla and the funnel-shaped pelvis, 2) the guinea pig kidney with the peripelvic column and pelvic pouch between a short conical papilla and the funnel-shaped pelvis, 3) the dog kidney with the peripelvic column and pelvic pouch between the crest-shaped papilla and the funnel-shaped pelvis, and 4) the cattle kidney which is divided into multiple renculi with minor and major calyces and pelvis. The renal papilla was lined with the simple or pseudostratified columnar epithelium which covered the inner zone of the renal medulla. The epithelial cells with numerous short microvilli on the surface contained a few organelles. In the mouse, the fornix was lined with one to two cell-layered cuboidal epithelium which covered the outer zone of the renal medulla and a part of the cortex. The epithelial cells of the fornix with numerous short microvilli or microridges on the surface had well-developed organelles. In the guinea pig, the peripelvic column was lined with the simple cuboidal or low columnar epithelium which covered the outer zone of the renal medulla. The epithelial cells with numerous short microvilli on the surface contained well-developed organelles. The pelvic pouch was lined with the pseudostratified columnar epithelium which was composed of four kinds of cells; the secretory cell with small electron-dense granules (310 nm), the secretory cell with large granules (720 nm) showing various electron densities, the mitochondria-rich cell with a single cilium, and the basal cell. Pelves of the mouse and guinea pig, peripelvic column, pelvic pouch and pelvis of the dog, and minor and major calyces and pelvis of the cattle were lined with the transitional epithelium. The fusiform vesicles in the superficial cells of the epithelium were highly developed in the dog, relatively well developed in the mouse and guinea pig, and poorly developed in the cattle. From the above findings, it is suggested that the transport of solutes and water of the urine in the pelvic cavity can take place through the epithelia covering the renal papilla and fornix of the mouse, papilla and peripelvic column of the guinea pig, and papillae of the dog and the cattle. And specialized cell types in the epithelium of the guinea pig pelvic pouch, two kinds of secretory cells and mitochondria-rich cell with a single cilium, could have peculiar functions in the renal pelvis, respectively.

숙지황 전탕액이 백서의 신장내 cGMP 생성과 ANP 수용체에 미치는 영향 (Effects of Shudihuang Water Extracts on the cGMP Production and Receptors for Atrial Natriuretic Peptide in the Kidney in Rats)

  • The purpose of this study is to investigate the effect of Shudihuang water extracts on the cGMP production of medulla and cortical membranes, receptors for atrial natriuretic peptide in the kidney by in vitro autoradiography in rats for 8, 16 weeks. The cGMP production of medullary and cortical membranes of the kidney decreased after the administration of Shudihuang water extracts. The density of receptors for atrial natriuretic peptide in the kidney decreased after the administration of Shudihuang water extracts only for 16 weeks. These results suggest that the long term administration of Shudihuang water extracts has decreased plasma renin activity and plasma levels of aldosterone modulated cGMP production of medullary and cortical membranes, density of receptors for atrial natriuretic peptide in the kidney.

개에서 신우신염의 방사선학적 및 초음파학적 진단 2례 (Radiogyaphic and Ultrasonographic Diagnosis of Pyelonephritis in 2 dogs)

  • A female 8-year-old Pug weighing 7.3 kg and a female 10-year-old Maltese dog weighing 3.5 kg showing anorexia and vomiting for a few weeks were referred to Veterinary Medical leaching Hospital, Seoul National University. Radiographic findings were an enlarged right kidney in a pug dog and a radiopaque material on the right ureteral region lateral to the third lumbar vertebrae with indefinite right kidney contour in a Maltese dog, repectively. Excretory urography performed in a Pug dog revealed a poor opacified enlarged right kidney with absent of pelvic recesses and pelvic dilation with proximal ureteral dilation on contralateral kidney. Ultrasonographic findings were enlarged kidney with dilated pelvis and echogenic sediment within the medulla in both dogs and especially an engorged proximal ureter and a thin rim of functional renal tissue remains in a Maltese dog. Those diagnostic findings indicated high possibility of pyelonephritis and these were confirmed by pathologic examination. Radiography and ultrasonography, although not giving final diagnosis for pyelonephritis, are useful for assessment and diagnosis of pyelonephritis.

신장 조직의 브라디키닌 결합부위 발현 (Expression of Bradykinin Binding Sites within the Mammalian Kidney Tissues)

  • Based upon the previous experiments showing that kidney and lung tissues of rat had relatively abundant bradykinin binding sites, we tried to characterize and determine the densities of the bradykinin binding sites in the rabbit kidney tissue and proximal tubular cells under different growing conditions. Among the kidney tissue renal medulla segments showed the highest bradykinin binding sites. To determine which growth factors are to add in the serum free culture medium to express selectively the bradykinin binding sites in the rabbit kidney proximal tubular cells, we tried so called hormone-deletion approach and in here insulin, hydrocortisone, transferrin, triiodothyronine and prostaglandin $E_1$ are examined. By performing receptor binding assay and determination of protein concentrations, we may conclude that the most required hormones in the expression for bradykinin binding sites are insulin and transferrin, and fetal bovine serum is shown to be less effective in this regard.

비글견에 있어서 간, 신장 및 비장의 초음파 히스토그램 비교 (Comparison of Ultrasound Histogram in Liver, Kidney and Spleen in Beagle Dogs)

  • 초음파 히스토그램의 내부 장기에 대한 임상적용을 알아보기 위하여 다주파수 탐촉자의 초음파기기를 이용하여 실험하였다. 9마리의 정상 비글견을 우측전복부의 간과 우측신장, 좌측전복부의 간, 비장과 신장를 검사하였다. 같은 깊이의 B-모드 영상으로 간, 신장의 피질, 비장, 그리고 각 신장의 피질과 수질의 수치를 비교하였다. 우측신장피질의 메아리발생치는 간에 비하여 유의적으로 $15{\%}$ 낮았다. 비장의 메아리발생치는 좌측신장피질보다 $23{\%}$ 높았으며, 간은 좌측 신장피질보다 $30{\%}$ 높았다. 각 신장의 피질과 수질의 비교에서는 피질이 우측과 좌측에서 각각 $47{\%}$$65{\%}$ 높게 나타났다(P<0.05). 우측신장피질과 간실질간의 메아리발생치 비율은 $0.86{\pm}0.11$, 비장과 좌측신장간에는 $1.37{\pm}0.47$, 간실질과 좌측신장피질간에는 $1.30{\pm}0.19$이었으며, 이들 모두다 유의적인 차이가 있었다(p<0.05). 초음파 히스토그램은 간, 비장, 신장 같은 내장장기의 구조물의 초음파적 평가에서 정량적으로 평가하기에 단순하고 유용하고 실질적이다.

급성 신장 빈혈이 신장의 유기성 대사에 미치는 영향 (Effects of Acute Renal Ischemia on Aerobic Metabolism of Rabbit Kidney Homogenates)

  • This experiment was carried out to investigate systematically how the aerobic metabolic capacity of renal tissue reduced by the effects of a period of induced ischemia. Aerobic metabolic studies were performed on homogenates of cortex and medulla of rabbits. Ischemia was induced by occluding the renal vein or renal artery of the left kidney for an hour. The right kidney used as a paired control. Aerobic metabolism was asesssed by measuring the oxygen consumption using the Warburg's manometric apparatus. The results are summarized as follows: 1. One hour of occlusive ischemia does not increase in the kidney weight in the renal arterial occlusion but increase in the renal venous occlusion. 2. Occlusion of either the renal vein or renal artery for an hour did not reduce to any significant degree the level of endogenous substrate in cortical homogenates as measured the rates of $0_2$ consumption. 3. A significant reduction in the rate of $C_2$ consumption was noted in the medullary homogenates of renal venous occluded kidneys while renal arterial occlusion had less of an effect. 4. The capaciy of homogenates for aerobic metabolism is not reduced by acute ischemia, because of the higher rate of oxygen consumption induced by exogenous glucose in renal vein occlusion. 5. The oxygen consumption of medullary homogenate more decreased to acute ischemia than cortical homogenates. The results of this investigation suggest that one hour circulatory stasis does not reduce major potential capacity of renal cortical tissue at the subcellular level to produce energy. In contrast, the aerobic metabolism of medullary tissue is reduced by renal ischemia. Further, both cortex and medulla appear to be more sensitive to ischemia induced by renal venous occlusion than by renal arterial occlusion.

성인 다낭종신 환자 1례에 대한 임상적 고찰 (Clinical Study on 1 Case of patient with Adult Polycystic Kidney Disease)

  • One case of patient with adult polycystic kidney disease was reported in this clinical study. After the study, the results were as follows: The adult polycystic kidney disease(APCK) is inheritant disease in which cysts are distributed throught the cortex and medulla of both kidneys. It belongs to Hematuria in oriental medicine field. For this patient, it was effective to appliment acupuncture Hap-kok, Kok-chi, Oe-gwan, Chok-samni, Chok-imup, Sam-umgyo, Su-bun, Tae-chung and Hae-gye showed a great effect on decreasing the pain and increasing the volume of urine. For this patient, it was effective to dispense Gamiwuiryungtang(加味胃苓湯), Ohryungsan(五苓散) and Chungsimyeonjaeum(淸心蓮子飮), Dosubokryungtang(導水茯苓湯).

개의 ethylene glycol 중독 (Ethylene glycol toxicosis in a dog)

  • A five-month old female Great Dane with a history of depression and convulsion was submitted to the Pathology and Diagnosis Reference Division of National Veterinary Research and Quarantine service. The dog had ingested concentrated antifreeze and died 12 days after the sudden onset of clinical signs. Main gross lesion was limited to swollen kidneys with diffusely fine granularity on the cut surface. Principle microscopic lesions were focused on the kidneys including dilated Bowman's space, thickened glomerular basement membrane, many light yellowish crystals, and proteinaceous casts in some tubules. Some neutrophils and lymphocytes were focally infiltrated in interstitium of the cortex and medulla. Crystals were angular or quite irregular in shape and arranged in sheave or rosette form. Under polarized light, the crystals were birefringent, which is typical of calcium oxalate crystals. Crystals were more concentrated in the cortex than in the medulla. A few crystals were also observed in the small blood vessels of the brain. Based upon the history of ingestion of antifreeze and the identification of calcium oxalate crystals in the tissue sections of the kidney and brain, this case was diagnosed as ethylene glycol toxicosis in a dog.
