각·결막염 유발균에 대한 Iysozyme의 항균활성 (The Antimicrobial Activity of Lysozyme against the Microorganisms Causing Conjunctivitis and/or Keratitis)

  • 안구의 각막과 결막은 외계에 노출되어 있어서 손상을 받기 쉽고, 손상부위에 병균이 감염되면 염증이 유발되어 심한 시력장애를 초래하기도 하는데, 누액 속에 포함되어 있는 lysozyme은 이러한 병균에 대해 항균작용을 가지므로 평소에는 안구가 노출되어 있어도 각 결막염이 발생하지 않는다. 본 연구는 lysozyme의 항균효과를 조사하고자 각 결막염의 원인균 중 5가지를 선별하여 배양하였고, 그 배양액에 lysozyme을 농도별로 첨가하면서 생육저해 정도를 측정하였다. 그 결과, 배지 중의 lysozyme 농도가 증가할수록 공시균의 생육은 저해되었다. 1mM 농도의 lysozyme에 의해 Pseudomonas는 43%, Neisseria는 41%, KlebsielIa는 35%, Staphylococus는 22% 생육이 저해되었고, 그람음성균은 그람양성균인 Staphylococus보다 lysozyme에 대한 감수성이 약 1.5~2.0배 큰 것으로 나타났다. 그러나 lysozyme은 진균인 Fusarium의 생육에는 매우 미약한 저해양상을 나타내었다.

버섯 추출물로부터 Acanthamoeba castellanii의 세포외 Serine 단백질분해효소 저해제 탐색 (Screening of Inhibitors of Extracellular Serine Protease of Acanthamoeba castellanii from Mushroom Extracts)

  • 콘텍트렌즈의 착용이 증가하면서 알칸사아메바에 의한 각막염이 급증하지만 아직도 이병을 치료하기가 매우 어려운 실정이다. 이 아메바에서 나오는 serine 단백질분해효소는 각막염의 유발에 중요한 역할을 하고 있다고 알려졌다. 본 연구에서는 A. castellanii의 세포외 serine 단백질분해효소의 활성을 저해하는 능력이 우수한 추출물을 230종 버섯의 메탄올 추출물로부터 선별하였다. 버섯을 메탄올과 물($65^{\circ}C$)로 추출한 후 여과.농축하여 이 추출물을 0.3 mg/ml의 농도로 효소와 반응시킨 후 기질 N-succinylala-ala-pro-phe p-anilide을 넣고 효소의 활성을 측정하여 우수한 저해능을 가진 추출물을 선별하였다. 이렇게 선별된 추출물을 가지고 성질을 연구하였는데, 한 추출물로 활성이 완전하게 억제되지 않았으나 두 추출물을 조합하여 사용하면 활성을 완전하게 억제할 수 있었다. 이 추출물은 다른 serine 단백질분해효소인 인간과 소의 트롬빈과 일반적인 단백질분해효소인 Protease K에는 영향을 미치지 않았다.

Testing of the Safety and the Effectiveness of Using Samjeong Pharmacopuncture Solution as Eye drops

  • Objective: This experimental study was designed to investigate the safety and the effectiveness of Samjeong pharmacopuncture solution (SPS) manufactured by using a the lowtemperature extract on process. Methods: To identify the safety and the effectiveness of using SPS as eye drops, we performed applied eye irritation tests on rabbits and antibacterial tests for Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, and Candida albicans. The eye irritation test was performed according to the toxicity testing regulation of the Korea Food & Drug Administration (2009. 8. 24, KFDA 2009-116). After SPS had been applied on the left eye of the rabbits, eye irritation in the cornea, iris and conjunctiva was observed on the 1st, 2nd, 3rd, 4th & 7th day. After SPS had been dropped on bacterial species that cause keratitis, the minimum inhibition concentration and the size of the inhibition zone were measured. The anti-bacterial potency was also measured by taking the size of inhibition zone. Results: After SPS had been administered on the left eye of the rabbits, none of nine rabbits were found to show abnormal signs or weight changes. After SPS had been administered on the left eye of the rabbits, no eye irritation in the cornea, iris and conjunctiva was observed on the 1st, 2nd, 3rd, 4th & 7th day. No specific response was detected in MIC for bacterial species Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, and Candida albicans after SPS had been applied. Conclusions: This study suggests that SPS is a non-toxic and non-irritant medicine that does not cause any of eye irritation in rabbits, but it has no antibacterial effects on bacterial species that are well known to cause keratitis. These results suggest that more research is required on extracts from herbal medicines for treating keratitis.

Specific Detection of Acanthamoeba species using Polyclonal Peptide Antibody Targeting the Periplasmic Binding Protein of A. castellanii

  • Acanthamoeba keratitis (AK) is a rare ocular disease, but it is a painful and sight-threatening infectious disease. Early diagnosis and adequate treatment are necessary to prevent serious complications. While AK is frequently diagnosis via several PCR assays or Acanthamoeba-specific antibodies, a more specific and effective diagnostic method is required. This study described the production of a polyclonal peptide antibody against the periplasmic binding protein (PBP) of A. castellanii and investigated its diagnostic potential. Western blot analysis showed that the PBP antibody specifically reacted with the cell lysates of A. castellanii. However, the PBP antibody did not interact with human corneal epithelial (HCE) cells and the other 3 major causative agents of keratitis. Immunocytochemistry (ICC) results revealed the specific detection of A. castellanii trophozoites and cysts by PBP antibodies when A. castellanii were co-cultured with HCE cells. PBP antibody specificity was further confirmed by co-culture of A. castellanii trophozoites with F. solani, S. aureus, and P. aeruginosa via ICC. The PBP antibody specifically reacted with the trophozoites and cysts of A. polyphaga, A. hatchetti, A. culbertsoni, A. royreba, and A. healyi, thus demonstrated its genus-specific nature. These results showed that the PBP polyclonal peptide antibody of A. castellanii could specifically detect several species of Acanthamoeba, contributing to the development of an effective antibody-based AK diagnostics.

다종의 안질환 사용 처방이 각막염 유발균인 Staphylococcus epidermidis에 미치는 영향 (The Experimental Study on Anti-bacterial Potency of Variou Herb-medicine used for eye disease on Staphylococcus epidermidis Keratitis)

  • Objective : This experimental study was performed to investigate the effect of Herbe-medicine used for eye disease(Sean-tang, Jinpi-san, Tangpo-san, Copitdis Rhizoma) on Staphylococcus epidermidis keratitis. Methods : After administering various herbal eye drops on Staphylococcus epidermidis, I measured MIC and the size of inhibition zone. MIC was measured by dropping from 20 to $50{\mu}{\ell}$ according to density. Anti-bacterial potency was measured by the size of inhibition zone with change of volume under 2 days culture condition. Also continuous anti-bacterial potency of herbal eye drops was measured by the size of inhibition zone according to 2 days and 6 days culture each under the $50{\mu}{\ell}$ condition. Results : 1. MIC on Staphylococcus epidermidis in Sean-tang, Jinpi-san was 1%, $50{\mu}{\ell}$ 2. MIC on Staphylococcus epidermidis in Tangpo-san, Coptidis rhizoma was 10%, $50{\mu}{\ell}$ 3. MIC on Staphylococcus epidermidis in Cravit was 0.1%, $20{\mu}{\ell}$. 4. Under the 2 days culture condition, the size of inhibition zone on Staphylococcus epidermidis by volume for Sean-tang was 10.0mm in $50{\mu}{\ell}$, Jinpi-san was 16.0mm in $50{\mu}{\ell}$, Tangpo-san was 17.5mm in $50{\mu}{\ell}$, Coptidis rhizoma was 31.0mm in $50{\mu}{\ell}$ and Cravit was 34mm in $50{\mu}{\ell}$, showed the highest anti-bacterial potency. 5. Under the $50{\mu}{\ell}$ condition, the size of inhibition zone on Staphylococcus epidermidis by 2 and 6 culture days for Sean-tang was 47.5mm in 6days, Jinpi-san was 36.0mm in 6days, Tangpo-san was 45.0mm in 6 days and Cravit was 48.0mm in 6 days, which showed the highest anti-bacterial potency. 6. Under the $50{\mu}{\ell}$ culture condition, the size of inhibition zone on Staphylococcus epidermidis by 2 and 6 culture days for Coptidis rhizoma was 31.0mm in 2 days and 6 days, which showed the same anti-bacteria1 potency. Conclusions : The present author think that Herbe-medicine used for eye disease can be used to cure Staphylococcus epidermidis keratitis and if further study is performed, the use of the Herbe-medicine used for eye disease will be valuable and beneficial in the clinical medicines.

황련해독탕 추출액의 각막염 원인균에 대한 항균효과 (Antimicrobial Activity of Hwangryunhaedok-tang Extract against Keratitis)

  • Objectives: The goal of this project was to measure the antibacterial effectiveness of Hwangryunhaedok-tang (barberry root decoction for detoxification) and its constituents. All of the active ingredients including the final product were prepared using high-pressure sterilization for use as medicinal eye drops. The varying forms of Hwangryunhaedok-tang (barberry root decoction for detoxification) were used to treat Staphylococcus epidermidis and Enterococcus faecalis which are found in keratitis. Methods: The antibacterial effect was measured by observing the presence or absence of antimicrobial activity when treated with varying concentrations of Hwangryunhaedok-tang extract. The tests were performed using a dosage of $70{\mu}l$ dosages of 100%, 50%, 10% and 1% the extracted solution by the minimum growth inhibitory concentration measurement. Antimicrobial activity was measured by examining the correlation between dosage strength and bacterial activity from $70{\mu}l$ to $10{\mu}l$ at the same concentration. Results: 1. Hwangryunhaedok-tang (barberry root decoction for detoxification), Phellodendri Cortex, and gardenia didn't show any antimicrobial effects against S. aureus, S. epidermidis, or E. faecalis. 2. Barberry root showed antimicrobial effects against S. aureus and S. epidermidis depending on the levels of concentration but didn't show any antimicrobial effects against E. faecalis. 3. Skullcap showed antimicrobial effects against S. aureus and S. epidermidis when a dosage of 100% extract $70{\mu}l$ was used. However, did not show any antimicrobial effects at all against E. faecalis. Conclusions: Hwangryunhaedok-tang (barberry root decoction for detoxification) and its constituents such as barberry root, phellodendri cortex, skullcap, and gardenia, can be used as an alternative to antibiotic medicinal eye drops to treat keratitis. However, further research on effective uses of and efficient extraction methods are needed.

고양이 증식성 호산구성 각막염에서 국소적인 사이클로스포린과 코르티코스테로이드 점안 1례 (A Case of Feline Proliferative Eosinophilic Keratitis Treated by Topical Cyclosporine and Corticosteroids)

  • 6 년령 중성화 암컷 페르시안 고양이가 3개월동안 우안의 재발성 각막궤양, 각막혼탁, 각막 혈관화를 주증상으로 내원하였다. 병변은 그 기간동안 국소적인 항생제와 비스테로이드성 소염제 점안에 치료반응이 없었다. 우안에서 유루증, 안검경련과 같은 안구통증 증상이 확인되었다. 미세 틈새등 현미경 검사에서 우안의 각막 전체에 분홍색에서 흰색을 띄는 부종성의 불규칙한 덩어리와 미약한 결막염을 확인하였다. 임상증상을 통해 고양이 증식성 호산구성 각막염으로 잠정 진단하였다. 각막의 세포학적 검사결과에서 다수의 호산구와 비만세포가 관찰되어 잠정적으로 내렸던 진단을 확진하였다. 본 환자는 국소적인 항생-코르티코스테로이드 합제, 사이클로스포린 연고, 트리플루리딘 점안과 전신적인 L-lysine 의 경구투여로 치료하였다. 치료시작 18일 후 내원 시 병변은 눈에 띄게 호전되었다. 국소적인 코르티 코스테로이드와 사이클로스포린의 병용치료는 고양이 증식성 호산구성 각막염에 유용한 치료법으로 생각된다.

다종의 점안약이 각막염 유발균인 Staphylococcus aureus에 미치는 항균효과에 대한 실험적 연구 (The Experimental Study on Anti-bacterial Potency of Various Herbal Eye drops on Staphylococcus aureus induced keratitis)

Methods : After administering various herbal eye drops on Staphylococcus aureus, I measured MIC and the size of inhibition zone. Under the 2 days culture condition, the size of inhibition zone on Staphylococcus aureus by volume for Sean-tang was 12.0mm in $50{\mu}{\ell}$, Jinpi-san was 19.0mm in $50{\mu}{\ell}$, Tangpo-san was 15.0mm in $50{\mu}{\ell}$, Coptidis rhizoma was 32.7 mm in $50{\mu}{\ell}$ and Cravit was 31mm in $50{\mu}{\ell}$, Coptidis rhizoma showed the highest anti-bacterial potency. 5. Under the $50{\mu}{\ell}$ condition, the size of inhibition zone on Staphylococcus aureus by 2 and 6 culture days for Sean-tang was 12.0mm, Jinpi-san was 19.0mm, Tangpo-san was 15.0mm, Coptidis rhizoma was 32.7mm and Cravit was 31.0 mm, which showed sameness anti-bacterial potency in 2 days and 6 days. Conclusions : The present author think that the herbal eye drops can be used to cure Staphylococcus aureus keratitis and if further study is performed, the use of the herbal eye drops will be valuable and beneficial in the clinical medicines.

In vitro cytotoxicity of Acanthamoeba spp. isolated from contact lens containers in Korea by crystal violet staining and LDH release assay

  • In order to observe the cytotoxicity of Acanthamoeba spp., which were isolated from contact lens containers as ethiological agents for the probable amoebic keratitis in Korea, the crystal violet staining method and LDH release assay were carried out. In the crystal violet staining method, among eight contact lens container isolates, isolate 3 (Acanthauloeba KA/LS5) showed 83.6% and 81.8% of cytotoxicity, and isolate 7 (Acanthamoeba KA/LS37) showed 28.2% and 25.1% of cytotoxicity, in 1 mg/ml and 0.5 mg/ml Iysate treatments, respectively. Acanthamoeba cutbertsoni and A. healyi showed 84.0% and 82.8% of cytotoxicity. Similar results were observed in A. costellunii and A. hafchefti which showed 83.6% and 75.5% or cytotoxicity. Acanthamoeba roureba and A. polyphaga showed 9.0% and 1.7% of cytotoxicity. In the LDH release assay, isolate 3 (20.4%) showed higher cytotoxicity than other isolates in 1 mg/ml Iysate treatment. The results provide that at least isolate 3 has the cytotoxic effect against CHO cells and seems to be the pathogenic strain.

Efficacy of Korean Multipurpose Contact Lens Disinfecting Solutions against Acanthamoeba castellanii

  • Acanthamoeba keratitis has been increasing in recent years. Main risk factors are contact lens wear and their cleaning solutions. Most contact lens wearers use multipurpose disinfecting solutions (MPDS) for cleansing and disinfecting microorganisms because of its convenience. We determined amoebicidal effects of MPDS made in Korea and their cytotoxicity on human corneal epithelium cells. Fifteen commercial MPDS (A to O) were tested for their amoebicidal effects on Acanthamoeba castellanii trophozoites and cysts by using a most probable number (MPN) technique. Among them, 7 kinds of MPDS showed little or no amoebicidal effects for 24 hr exposure. Solutions A, B, G, H, L, and O showed positive amoebicidal effects, and solutions M and N killed almost all trophozoites and cysts after 24 hr exposure. However, 50%-N solution showed 56% cytotoxicity on human corneal epithelial cells within 4 hr exposure, and 50%-O solution also showed 62% cytotoxicity on human cells within 4 hr exposure. Solution A did not show any cytotoxicity on human cells. These results revealed that most MPDS made in Korea were ineffective to kill Acanthamoeba. The solutions having amoebicidal activity also showed high levels of cytotoxicity on human corneal epithelial cells. New formulations for improved MPDS that are amoebicidal but safe for host cells are needed to prevent Acanthamoeba keratitis.
