Intraspecific Polymorphism and Classification of Paeonia Iactiflora Based on the Giemasa C-banding Patterns

  • On the basis of karyotypic analysis performed by conventional staining and Giemas C-banding technique, cytological relationship was inferred for 21 lines of Paeonia lactiflora Pal. cultivated in Korea. It was very difficult to infer their organized karyotypic classification system using the composition of somatic chromosomes involving sat-chromosomes, relative length of chromosomes, arm ratio and karyotypic formulae by conventional staining. From the distribution and number of Giemsa C-bands on the chromosomes b and c, 21 lines can be subclassified into 5 groups. It seems that the karyotypic polymorphism is observed in 21 lines of cultivated P. lactiflora because peony mainly propagates by outbreeding.

Variations in Karyotypic Characteristics of Different Breed Groups of Water Buffaloes (Bubalus bubalis)

  • Karyotype analysis was carried out on blood samples of 30 water buffaloes belonging to different breed groups (i.e. Philippine Carabao (PC), Indian Murrah (IM), Bulgarian Murrah (BM), "$F_1$ 50% IM-50% PC", "$F_1$ 50% BM-50% PC" and "75% IM-25% PC"), using the modified Leucocyte Culture Technique. The modal chromosome numbers of the PC, "$F_1$ 50% IM-50% PC", "$F_1$ 50% BM-50% PC", IM, BM and "75% IM-25% PC" were 2n=48, 49, 49, 50, 50 and 50, respectively. The water buffalo chromosomes are mostly acrocentric (79.67%) and the remainder submetacentric (20.33%). Results of the ordinary least square analysis showed significant breed effects (p<0.01) on other karyotypic characteristics (i.e. relative length, arm ratio and centromeric index). Significant correlation between karyotypic characteristics and some animal performance traits were also found. The significant correlation values imply that karyotypic characteristics can be used as important criteria to select potentially productive young water buffaloes. In the future, more production and reproduction traits from non-institutional herds should be included in the analysis to reveal meaningful correlations with various karyotypic characteristics.

  • The present study demonstrates karyotype based on H-patterns of A.fistulosum and A. ascalonicum using Giemsa technique. The results obtained in this study are summarized as follows: I). Karyotypic analysis of A. fistulosum is 6VII+$JII^t+JII$ and that of A. ascalonicum collected from a local farm in the suberbs of Taegu city clearly heterozygous as $13V+J_1^t+J_2+i. ii$). The heterochromatin of both species is generally located distally in both arms of chromosomes and each chromosome type possesses some variations on H-patterns. iii). The percentage of heterochromatin to total chromosome length in cell is about 14.6% in A. fistulosum, 12.8% in A. ascalonicum. The number of bands is revealed about 38 in A. fistulosum and 33 in A. ascalonicum. Also in the amounts of chromocenters per nucleus, the former is somewhat more than the latter.

  • To clarify the cytogenetic features of the chicken grunt, Parapristipoma trilineatum, small yellow croaker, Larimichthys polyactis, and brown croaker, Miichthys miiuy, in Jeju, Korea, karyotype analysis was performed. The karyotype of chicken grunt and brown croaker consists of 24 pairs of telocentrics [2n=48, fundamental number (FN)=48], and small yellow croaker consists of 3 pairs of submetacentrics and 21 pairs of telocentrics (2n=48, FN=54). No evidence of polymorphism, including aneuploidy or sex-related heteromorphic chromosomes, was found in any specimens examined in this study.

  • Karyotypic analysis was performed for Labridae fishes, Pseudolabrus japonicus, Halicho eres tenuispinis, H. poecilopterus and Pteragogus flagellifera from coastal area of Cheju Island in Korea. The chromosome numbers(Karyotypes) were 42(4M+24SM+14ST, A), 48(2M+2SM+44ST, A), 48(2M+46ST, A) and 42(4M+24SM+14ST, A) in P. japonicus, H. tenuispinis, H. poecilopterus and P. flagellifera, respectively. Heteromorphic sex chromosome was not found in both sexes of each Labridae fishes. However, large satellites were located on the largest subtelocentrics in P. japonicus and P. flagellifera.

  • The growth charateristics and karyotypes of Aster spathulifolius collected from 5 sites including coastal and island region on the Korean peninsula, were analysed. Several morphological characteristics of the plants such as leaf length, leaf width, top internode, medium internode, spike branching, flower diameter, number of petal, leaf color, leaf form, stem and leaf hair, viscosity, and serration of the plants were distinctly different depending on the native region from which they were collected. Karyotypic analysis showed that the chromosome number was all diploid (2n=18), with one pair of submetacentric satellite chromosomes. The chromosome composition included 7 pairs of metacentric chromosomes and 2 pairs of submetacentric chromosomes in all plants. However, chromosome order and the ranges of the chromosome lengths were a little different from plant to plant according to their native growing regions. The plants from Geoje-Do especially showed large differences in the chromosome lengths between the longest and the shortest compared to the plants from other places. This results provide important data to support the classification of the species into several sub-species.

  • Karyotypic analysis and FISH (fluorescence in situ hybridization) with 45S and 5S rRNA genes were carried out in Persicaria tinctoria H Gross. The somatic metaphase chromosomes were ranged from 2.25 ${\mu}m$ to 1.50 ${\mu}m$ in length. Chromosome number was 2n = 4x = 40 with the basic number of x = 10. The chromosome complement of the species consisted of 16 pairs of metacentrics (chromososomes 1,2,3,4,6,7,8,9, 10, 11, 12, 13, 15, 18, 19 and 20) and 4 pairs of submetacentrics (chromosome 5, 14, 16 and 17). The karyotype formula was K(2n) = 4x = 32 m + 8 sm. In FISH analysis, three pairs of 45S rRNA gene loci on the terminal region of submetacentrics (chromosomes 5, 16 and 17) and two pairs of 5S rRNA gene loci on the centromeric region of metacentrics (chromosomes 9 and 11) were detected, respectively.

  • The calli obtained from the bulbs of A. victorialis var. platyphyllum on MS based medium containing 2 mg/L 2, 4-D and 1mg/L BA. Plants from calli were regenerated on MS basal media supplemented with combinations of NAA and four kinds of cytokinin, BA, zeatin, kinetin and 2iP, and with only each cytokinin. The good response of shoot regeneration was observed in the media with combinations of NAA and BA or zeatin, and with only BA or zeatin. Shoot regenerating response in the medium with combinations of NAA and BA or zeatin, and with only BA or zeatin. Shoot regerating response in the medium with combinations of NAA and BA or zeatin was about two times higher than that in the mediuim with only BA or zeatin. From the karyotypic analysis of the regenerated plants, abnormal diploids, aneuploids and mixoploid with structural aberrations were investigated. The somaclonal variants (AV 16-04, AV 13-03) were shown the considerable differences from normal diploid regenerant (AV 18-03) in the external morphology.

Nager syndrome associated with 45,X monosomy

  • Nager syndrome is a rare malformation complex characterized by facial, limb, and skeletal morphogenesis. The mode of inheritance has not been definitely established. Major karyotypic abnormalities were seldom associated with this syndrome. We report on an infant with Nager acrofacial dysostosis that was associated with 45,X monosomy. This baby was born to a 36-year-old multigravid woman after 37 weeks of gestation and with maternal hydramnios. The baby girl died of airway obstruction due to retruded tongue 3 hours after birth. Phenotypically, this this patient had mandibulofacial dysostosis, radioulnar synostosis, hypoplasia and aplasia of thumbs, peripheral edema and apparently normal genital organs. We confirmed that major chromosomal anomaly including 45,X monosomy could be associated with Nager syndrome, although its pathogenetic significance remains unanswered.

  • A detailed karyotype analysis was carried out in five Korean native Iridaceae species; Iris pseudoacorus, I. pallasii. var. chinensis, I. tectrum, I. dichotoma and Belamcanda chinensis. Chromosome compositions of the five species showed that they have different karyotypic formulae; I. pseudoacorus 2n=34=10m+16smn+8st including two pairs of satellite chromosomes, I. pallasii var. chinensis 2n=40=26m+12sm+2st including two pairs of satellite chromosomes, I. tectrum 2n=30=14m+16sm including five pairs of satellite chromosomes, I. dichotoma 2n=32=22m+10sm including two pairs of satellite chromosomes, and B. chinensis 2n=32=20m+10sm+2st including one pair of satellite chromosomes. These results will supplement the previous cytogenetic reports in Iridaceae species and enhance our understanding on the genetic structure, which will be useful in clarifying the unique characteristics of each species for practical breeding programs for horticultural and pharmaceutical purposes.
