활성화된 단핵구 및 대식세포의 항원제시기능에 대한 Kaempferitrin의 조절 효과 (Modulatory Effect of Kaempferitrin, a 3,7-Diglycosylflavone, on the LPS-Mediated Up-regulation of Surface Co-stimulatory Molecules and CD29-Mediated Cell-cell Adhesion in Monocytic- and Macrophage-like Cells)

  • Kaempferitrin, isolated from Kenaf (Hibiscus cannabinus), was examined to evaluate its modulatory effects on antigen-presenting cell functions of macrophages/monocytes such as phagocytosis of foreign materials, up-regulation of costimulatory molecules (CD40, CD80 and CD86), adhesion molecule activation, and antigen processing and presentation. Kaempferitrin strongly blocked up-regulation of CD40, CD80 and CD86, but not pattern recognition receptor (PRR) (e.g., TLR2). It also suppressed functional activation of CD29 (${\beta}1$-integrins), as assessed by cell-cell adhesion assay, required for T cell-antigen-presenting cell (APC) interaction. Furthermore, this compound did not block a simple activation of CD29, as assessed by cell-fibronectin adhesion assay. However, the compound did not diminish phagocytic uptake, an initial step for antigen processing, and ROS generation in RAW264.7 cells. In particular, to understand molecular mechanism of kaempferitrin-mediated inhibition, the regulatory role of LPS-induced signaling events was examined using immunoblotting analysis. Interestingly, this compound dose dependently suppressed the phosphorylation of $I{\kappa}B{\alpha}$, Src, Akt and Syk, demonstrating that it can negatively modulate the activation of these signaling enzymes. Therefore, our data suggested that kaempferitrin may be involved in regulating APC function-relevant immune responses of macrophages and monocytes by regulating intracellular signaling.

  • Eight compounds were isolated from the n-BuOH soluble fraction of the leaves of Weigela subsesillis. On the basis of spectral data, they wεre identified as $kaempferol-O-3-{\alpha}-L-(3-O-acetyl)\;rhamnopyranosyl-7-O-{\alpha}-L-rhamnopyranoside$ (1), sutchuenoside A (2), kaempferitrin (3), astragalin (4), kaempferol 7-O-rhamnoside (5), scopolin (6), farxin (7), kaempferol 3-O-{\alpha}-L-rhamnosyl-7-O-{\beta}-D-glucoside (8), respectively.

  • 본 연구에서는 kaempferol 및 그 배당체의 항산화 및 항염증 효능을 평가하기 위해 free radical 소거활성, ROS inhibition assay, TARC 생성 억제 관련 실험을 수행하였다. 또한 kaempferol과 그들의 rhamnosides (${\alpha}$-rhamnoisorobin, afzelin, kaempferitrin)의 구조에 따른 생리적 활성의 상관관계를 조사하였다. Kaempferol과 ${\alpha}$-rhamnoisorobin은 free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거활성($FSC_{50}$)에서 각각 62.5, 50.0 ${\mu}M$이 측정되었으며 afzelin과 kaempferitrin은 농도와 상관없이 free radical 소거활성을 나타내지 않았다. ROS inhibition assay에서는 시료 농도를 10, 50, 100 ${\mu}M$을 처리한 결과 kaempferol은 대조군(100 %)에 비하여 ROS 발현 농도를 각각 97.5, 57.8, 47.8 %로 감소하였으며 ${\alpha}$-rhamnoisorobin은 93.1, 59.1, 41.4 %의 감소를 나타내었다. TARC (thymus and activation regualted chemokine) 생성 억제능 실험을 통해 시료의 항염증 효능을 평가한 결과 kaempferol은 10, 50, 100 ${\mu}M$ 농도에서 대조군(100 %)에 대비하여 각각 48.8, 5.5, 4.4 %로 농도 의존적으로 TARC 생성을 감소시켰으며, ${\alpha}$-rhamnoisorobin은 88.1, 19.0, 1.0 %로 TARC 생성의 감소 효능을 나타내었다. 결론적으로 kaempferol과 그들의 rhamnoside 중 kaempferol과 ${\alpha}$-rhamnoisorobin은 항산화, 항염증에 대한 우수한 효능을 나타내는 것으로 사료되며 더 나아가서는 항노화 및 항염증에 효과가 있는 기능성 화장품에 응용 가능성이 있음을 시사한다.

  • In this study, kaempferol and its rhamnosides (${\alpha}$-rhamnoisorobin, afzelin, kaempferitrin) were isolated electively by bioconversion technology from Hibiscus cannabinus L. leaves to evaluate the anti-wrinkles effects and anti-microbial effects. In order to evaluate anti-wrinkles activity, reduction of expression matrix metalloproteinase-1 (MMP-1) protein and proliferation/ pro-collagen production were investigated. Kaempferol and ${\alpha}$-rhamnoisorobin showed inhibition activity of MMP-1 generated to compared to positive control. In HaCaT cell proliferation assay, kaempferol and ${\alpha}$-rhamnoisorobin significantly promoted cell proliferation in a concentration-dependent manner. In addition, procollagen synthesis assays (by HDF-N cell) showd that TGF-${\beta}$ induced procollagen production and also, all four kinds of experimental significantly promoted procollagen synthesis in a concentration-dependent manner. Kaempferol and ${\alpha}$-rhamnoisorobin exhibited strong antimicrobial activities on five of microbes, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans and Aspergillus niger.

현초에서 분리된 페놀성 화합물들의 흰쥐 수정체 유래 알도즈 환원효소 및 갈락티톨 생성 억제 효과 (Inhibitory Effect of the Phenolic Compounds from Geranium thunbergii on Rat Lens Aldose Reductase and Galactitol Formation)

  • We evaluated the inhibitory effects of extracts and components of Geranium thunbergii on aldose reductase (AR) and galactitol formation in rat lenses with high levels of galactose as a part of our ongoing search of natural sources for therapeutic and preventive agents for diabetic complications. The inhibitory effects of water, methanol and ethanol extracts of G. thunbergii on rat lens AR (RLAR) were determined. Comparing inhibitory effects of various solvent extracts, ethanol extract showed RLAR inhibitory activity ($IC_{50}$ values, 5.24 and $6.39{\mu}g/m{\ell}$, respectively). The ethanol extract was fractionated to chloroform, ethyl acetate and water. Of these, the ethyl acetate fraction from ethanol extract of G. thunbergii exhibited RLAR inhibitory activity ($IC_{50}$ value, $2.64{\mu}g/m{\ell}$). In order to identify the bioactive components of ethyl acetate soluble fraction of ethanol extract from G. thunbergii, eight compounds, namely gallic acid (1), protocatechuic acid (2), p-hydroxybenzoic acid (3), brevifolin carboxylic acid (4), geraniin (5), ellagic acid (6), kaempferol-3-O-arabinofuranosyl-7-O-rhamnopyranoside (7), kaempferitrin (8) were isolated. The isolates were subjected to in vitro bioassays to evaluate their inhibitory activity on RLAR and galactitol formation in rat lenses. The ellagic tannins (5, 6) and flavonoid (7) exhibited strong inhibitory effects on RLAR. Also, these three compounds (5, 6 and 7) suppressed galactitol accumulation in rat lens under high galactose conditions, demonstrating the potential to prevent galactitol accumulation exo vivo. These results suggest that the extracts and components of G. thunbergii are a promising agent in the prevention or treatment of diabetic complications.

페놀성 화합물을 이용한 현지초의 UPLC 다성분 동시분석 개발 (Simultaneous Analysis of Phenolic Compounds in Geranium thunbergii Using UPLC)

  • The aim of this study was to develop a UPLC method for simultaneous analysis of 8 phenolic compounds including gallic aicd (1), protocatechuic acid (2), methyl gallate (3), ellagic acid (4), kaempferol-3-arabinofranosyl-7-rhamnoside (5), kaempferitrin (6), afzelin (7) and kaempferol-7-rhamnoside (8) isolated from Geranium thunbergii which has been traditionally used as anti-diarrheal agent. The UPLC method was optimized and validated using Halo C18 column ($4.6{\times}100mm$, $2.7{\mu}m$) consisting of MeOH and 0.1% formic acid at 260 nm in 25 minutes. In quantitative analysis of 8 compounds in MeOH extract of G. thunbergii, contents of 4-6 were 12.39, 20.52 and 21.45 mg/g, respectively. These compounds were measured as major phenolic compounds in G. thunbergii and can be useful as marker compounds for its quality control. These results suggest that the UPLC method can be contributed as basic data for quality evaluation of herbal preparations.
