Oviposition and Feeding Preference in Monochamus Saltuarius (Coleoptera: Cerambycidae) (북방수염하늘소의 산란부위 및 식이 선호성)

  • The average number of oviposion scars in lower (facing the Land) and upper part (facing the Sun) of P. koraiensis logs were $16.3{\pm}13.9$ and $3.3{\pm}3.6$, respectively. Oviposition preference of Monochamus saltuarius adults was lower part of Pinus koraiensis. Estimated average number of larvae and egg from oviposition scars ($64.7{\pm}23.8$) in P. koraiensis logs was $18.8{\pm}10.0$ in the laboratory condition. However, oviposition scars were $63.6{\pm}31.1$ and average number of larvae and egg was $2.0{\pm}2.6$ in field condition. When we fed with inner bark (phloem) and xylem of P. koraiensis, body weight of M. saltuarius larvae was normal. However, when we only fed xylem of P. koraiensis, body weight and size were reduced but they were recovered after feeding inner bark (phloem). Esterase isozymes were assessed from larvae fat body of M. saltuarius and Est1 were specifically activated when we fed with inner bark (phloem) and xylem.

Studies on the Effect of Copper Ion on the Lactate Dehydrogenase Isozyme Patterns in Organs of Catfish, Parasilurus asotus (메기 (Parasilurus asotus)의 장기내 젖산수소이탈효소 아이소자임 형에 미치는 동이온의 영향에 관한 연구)

  • The effect of copper ion on the lactate dehydrogenase isozyme patterns in the heart, liver, kidney, skeletal muscle, and ovary of catfish, Parasilurus asotus, was studied by cellulose acetate gel electrophoresis. 1. The LDH-1 and LDH-2 of heart type appeared in the heart muscle of control fish. When the fish were exposed to copper ion, however, the LDH-1, LDH-2, LDH-3, and LDH-4 appeared. The amount of LDH-1 was decreased and those of LDH-2, LDH-3, and LDH-4 were increased. 2. There was one band of LDH-4 in the liver of normal fish. But the amount of LDH-4 was decreased and additional new LDH-5 appeared by exposure to copper ion. 3. There were LDH-1, LDH-2, and LDH-4 in the kidney tissue of both control and experimental groups. The LDH-1 was increased, whereas LDH-2 and LDH-4 were decreased after exposure to copper ion. 4. There was broad band of LDH-5 in the skeletal muscle of the control fish. However, the LDH-4 and LDH-5 with M sub-band appeared by the exposure to copper ion. 5. There was LDH-3 band only in the ovary of control, wheras all five LDH isozymes appeared in the ovary of the fish exposed to copper iion. 6. During the period of exposure to copper ion, the LDH isozyme of heart type which associated with aerobic metabolism was decreased, but the LDH isozyme of muscle type of anaerobic metabolism was increased in most of heart, liver, and skeletal muscle. It seems that these organs are related to some of important functions for anaerobic metabolism during the copper poisoning period.

Kinetics of Thermal Inactivation of Peroxidases and Polyphenol Oxidase in Pineapple (Ananas comosus)

  • The heat tolerance and the inactivation kinetics of peroxidase (POD) and polyphenol oxidase (PPO) in pineapples (Ananas comosus) were studied in the temperature range $45-95^{\circ}C$. The kinetic parameters, such as deactivation rate constant (k), activation energy ($E_a$), and decimal reduction rate (D) of the thermal inactivation process, were determined. POD in pineapples showed biphasic inactivation behavior at temperatures range $45-75^{\circ}C$ but was monophasic at $85-95^{\circ}C$. This indicate that POD has 2 isozymes, namely heat labile and heat resistant, with $E_a$ of 68.79 and 93.23 kJ/mol, respectively. On the other hand, the heat denaturation of pineapple PPO could be described as simple monophasic first-order behavior with $E_a$ of 80.15 kJ/mol. Thus, the results of this study is useful in blanching technology where it shows a shortened time with higher temperature can be applied. The determination of the heat tolerance and inactivation POD and PPO, at different temperature range as done in the present work, was very important to improve the blanching process. This also will help to optimize the pineapple canning process which is one of the most important food industries in many tropical regions.

Characterization of Sporulation-Specific Glucoamylase of Saccharomyces diastaticus (Saccharomyces diastaticus의 포자형성 특이 글루코아밀라제의 특성)

  • The yeast strains of Saccharomyces diastaticus produce one of three isozymes of an extracellular glucoamylase I, II or III, a type of exo-enzyme which can hydrolyse starch to generate glucose molecules from non-reducing ends. These enzymes are encoded by the STA1, STA2 and STA3 genes. Another gene, sporulation-specific glucoamylase (SGA), also exists in the genus Saccharomyces which is very homologous to the STA genes. The SGA has been known to be produced in the cytosol during sporulation. However, we hypothesized that the SGA is capable of being secreted to the extracellular region because of about 20 hydrophobic amino acid residues at the N-terminus which can function as a signal peptide. We expressed the cloned SGA gene in S. diastaticus YIY345. In order to compare the biochemical properties of the extracellular glucoamylase and the SGA, the SGA was purified from the culture supernatant through ammonium sulfate precipitation, DEAE-Sephadex A-50, CM-Sephadex C-50 and Sephadex G-200 chromatography. The molecular weight of the intact SGA was estimated to be about 130 kDa by gel filtration chromatography with high performance liquid chromatography (HPLC) column. Sodium dedecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis showed it was composed of two heterogeneous subunits, 63 kDa and 68 kDa. The deglycosylation of the SGA generated a new 59 kDa band on the SDS-PAGE analysis, indicating that two subunits are glycosylated but the extent of glycosylation is different between them. The optimum pH and temperature of the SGA were 5.5 and $45^{\circ}C$, respectively, whereas those for the extracellular glucoamylase were 5.0 and $50^{\circ}C$. The SGA were more sensitive to heat and SDS than the extracellular glucoamylase.

Biochemical Changes in Brassica Seedlings Due to Cold Acclimation Treatment (Brassica속 작물 유묘에서 장기 저온 순화처리에 따른 생화학적 변화)

  • This study was aimed for determining the biochemical mechanism of cold tolerance in crops and for searching the biochemical genetic marker related with cold tolerance by the analysis of isozyme pattern. We investigated various biochemical changes induced by the long-term cold acclimation in cold sensitive rape (B. napus) and in cold tolerant 'Sandongchae'(B. campestris) seedlings. The cold shock after long-term cold acclimation to B. napus and B. campestris greatly increased the activities of peroxidase 157% and 50% in root fraction and, 201% and 205% in hypocotyl, respectively. Simultaneously, the activity of superoxide dismutase was largely increased in hypocotyl fraction, too. Protein contents of hypocotyl fractions in B. napus and B. campestris were also increased by 11.4% and 57.8%, respectively. The band of pl 6.4 among peroxidase isozymes newly biosynthesized during long-term cold acclimation was emerged in the hypocotyl fraction of cold tolerant B. campestris as well as in the root of both species. From above and previous results, we presented a model of interconversions of molecular oxygen species due to the cold injury and biochemically inferred the mechanism of cold tolerance in crops.

  • In order to obtain the basic data for clarifing the mechanism of cold tolerance in crops, we analyzed various biochemical changes according to the Uniconazole treatment in Brassica seedling. Peroxidase activity in the root fraction of Brassica seedling was about 3 to 4 times higher than that in hypocotyl fraction, while catalase activity in those fractions showed opposite trend to the peroxidase activity. The content of hydrogen peroxide in root fraction was higher than that of hypocotyl fraction as being a reciprocal proportion with catalase activity. Especially in all fractions, peroxidase· activity of 'Sandongchae' (B. campestris) seedling, known as cold tolerant, was two-fold higher than that of cold sensitive rape(B. napus). The elongation rate of hypocotyl after germination was faster in B. napus than in B. campestris. The application of Uniconazole at 0.3 to 1.0 ppm to B. napus suppressed 43 to 46% of hypocotyl elongation and increased 65 to 73% of peroxidase activity in hypocotyl fraction. The shortening rate of hypocotyl length due to Uniconazole treatment was positively correlated with the increasing rate of peroxidase activity in hypocotyl fraction. Superoxide dismutase was not induced upon Uniconazole treatment and has only 3 isozymes in any fractions. Its activity was observed in the order of cotyledon>root>hypocotyl fraction.

  • Callus was induced from the petioles of scented-geranium (Pelargonium graveolense) in MS medium containing various concentrations of plant growth regulators. The highest frequency of more than 70% of callus was induced in 2 mg/l NAA and 0.5 mg/l BAP or 2 mg/l 2,4-D and 0.5 mg/l BAP combined treatment, while not in 2,4-D, NAA or BAP alone. When the callus was transferred to the MS medium containing 0.05 mg/l NAA and 0.5 mg/l BAP, were highest intensity of shoot formation, 14 shoots/callus, was induced after 5 weeks. The highest rooting was observed on hormone-free rooting media from the regenerated shoots after 3 weeks, indicating that the regeneration from geranium callus might be possible by changing the hormone ratios. Peroxidase (POD) specific activities in callus induced from 2 mg/l NAA and 0.5 mg/l BAP were higher than those of 2 mg/l 2,4-D and 0.5 mg/l BAP callus during the entire culture periods. POD isozyme C3 was the main cathodic POD isozyme expressed in NAA and BAP callus, while C1 was the main in 2,4-D and BAP callus. However, anodic POD isozymes, A1, A2 and A3 were expressed with similar activities in both hormone combinations.

  • Schisandra nigra Max. has been cultivated far a medical use as well as food. It is an endemic species which has a unique habitat at the altitude of 600-1,400 m in Cheju island. In this study, three natural populations of S. nigra were investigated by using of starch-gel electrophoresis to determine the extent and distribution of genetic diversity. Except 2 monomorphic locus (Mdh-2 and Pgi-1), 4 of the 6 isozyme locus (Idh, Mdh-2, Mnr, and Pgi-2), verified from 4 isozymes, revealed polymorphism in the three populations of S. nigra. The mean number of allele per locus was 1.7 and the percentages of polymorphism loci were 38.9% at 95% level and 50% at 99% level respectively. The observed and estimated heterozygosities were 0.141 and 0.147 respectively. Although plants which were in the face of crisis and distributed in the restricted area, have been known to the very low degree of genetic variation, S. nigra showed higher genetic variation than others. Genetic variation was mostly allocated within population and individuals than that among populations. The result of Wright's F analysis estimates of $F_{IT}$ and $F_{IT}$ showed that S. nigra population revealed Hardy-Weinberg steady state.

Characterization of Anionic Peroxidase Induced by Low Host-Specific Elicitor in Suspension Cultures of Rose (Rosa sp.) (장미(Rosa sp.) 현탁배양세포에서 숙주 특이성이 낮은 Elicitor에 의해 유도되는 Anionic Peroxidase의 특성)

  • Whereas cationic extracellular peroxidases (PODs) were observed in the suspension cultures of rose (Rosa sp. L. cv Pual's scarlet) grown under normal conditions, new anionic isozymes were induced within 24 hr by the treatment of low host-specific elicitor (10 mg glucan/L media) prepared from yeast cell wall. Prominent anionic (pI 6.1) and cationic POD (pI 8.4) were purified and characterized to understand the physiological role of the enzymes. Both enzymes were purified (ca.200 fold) by the ammonium sulfate precipitation, ion exchange chromate-graphy and gel filtration chromatography. The Km values of the purified anionic POD for ferulic acid and $\textrm{H}_2\textrm{O}_2$ were 4.64 mM and 0.72 mM, whereas those of the cationic POD were 1.38 mM and 0.48 mM, respetively. The activity of the anionic POD as NADH oxidase was twice higher than that of cationic POD. The NADH oxidation in the anionic POD fraction was inhibited by 60% on the addition of 0.1 mM coniferyl alcohol, while that in the cationic fraction was inhibited by 15%.

Inheritance of Isozymes IDH, ME and PGI in Pinus densiflora and Pinus thunbergii in Kyungpook Province (경북지방(慶北地方) 소나무 및 곰솔에 대한 동위효소(同位酵素) IDH, ME 및 PGI의 유전(遺傳))

  • This study was conducted to estimate the inheritance of allozyme variants in IDH, ME and PGI, using megagametophyte tissue of seeds of Pinus densiflora and Pinus thunbergii in Kyungpook province. Two alleles at a single locus were detected for MH in Pinus densiflora, but Pinus thunbergii showed no varition, showing only one band. Four alleles at one locus were determined for ME in Pinus densiflora and Pinus thunbergii. Two loci were found for PGI and no variation seemed to occurred in A-locus as the presence of one single band. Five alleles were presented in B-locus in Pinus densiflora, but two alleles $B_1$ and $B_2$ in Pinus thunbergii. In PGI, an allozyme composed of multiple bands is observed from haploid megagamtophyte tissues. The isozyme variants of IDH, ME and PGI were appeared to segregate into 1 : 1 ratio for all zones.
