Differentiation of Colletotrichum spp. Causing Anthracnose on Capsicum annum L. by Electrophoretic Method (전기영동법을 이용한 고추탄저병균의 분류)

  • The present researches were carried out to differentiate the species of Colletotrichum by elecrophoretic method. C. gloeosporioides, C. dematium and Gloeosporium fructigenum could be differentiated by the isozyme patterns of esterase, leucine aminopeptidase, acid phosphatase and glutamic oxaloacetic transaminase. Especially. G-strain and R-strain of C. gloeosporioides were differentiated by the enzyme patterns. The G­strain damaged all stage fruits (green and red fruits) of Capsicum annum. The R-strain could not infect the unripe (green) fruits, but it could damage only ripe (red) fruit of Capsicum annum.

The Characterization and Activity Changes of Phosphatases in Amoeba sp. to the Light Stimuli and Its Response Pattern (광자극에 대한 Amoeba 의 반응양상과 phosphatase 의 특성 및 활성도 변화)

  • Amoeba sp. was cultured under the light and the dark conditions, and the activity of phosphatases was investigated. There was a linear correlation between the early reaction time and the activity of phosphatases when phosphatases were incubated at 30℃. Then the activity of acid phosphatase was about 2 times higher than that of alkaline phosphatase. The activity of phosphatase was optimal at pH 5.0 in acidic part and at pH 8.0 in alkaline part, respectively. The optimal temperature of phosphatases was near the 40℃. The isozyme patterns of cytoplasmic acid phosphatase were compared with those of membraneous one. Both the isozyme patterns were shown to bo polymorphic on the polyacyamide gel, but different band patterns were observed in the isozymes of the cytoplasmic and the membraneous acid phosphatases. The number of Amoeba sp. under the light stimulus for 48 hours decreased negative exponentially from the illumination. The activity of acid and alkaline phosphatases under the illumination of light incresed 1.7 and 1.5 times higher, respectively, than the activity of those under the dark condition. This result apperars to be related to the mechanism of the autophosphorylation.

Isozyme Variability in Two Species of Freshwater Viviparid Snails in Korea : Cipangopaludina chinensis malleata and C. Japonica (한국산 논우렁이과 ( Family Viviparidae ) 2종에서의 동위효소 변이)

  • A horizontal starch gel electrophoresis for enzyme proteins extracted from 2 species of Korean viviparid snails; Cipangopaludina chinensis malleata and C. japonica was carried out in order to elucidate their genetic relationships. A total of 10 enzymes were employed in three different kinds of buffer systems. Two loci from each enzyme of alcohol dehydrogenase, esterase, glucose phosphate isomerase, isocitrate dehydrogenase, iditol dehydrogenase, malate dehydrogenase and peptidase(VL); and only one locus dach from two enzymes, glycerlo-3-phosphate dehydrogenase and phosphoglucomutase were detected; but, four loci from peptidase(LGG) were observed. Most of loci in two viviparid species showed homozygous monomorphic banding patterns and some of them were specific as genetic markers between two different species. However, EST-1, MDH-1, PEP(VL)-1loci showed polymorphic banding patterns. Foru populations of C. chinensis malleata were more closely clustered in a dendrogram within the range of genetic identity values of 0.928-1.00, and these clusters were lineated with C. japonica at the value of 0.355. In summarizing the above results, two viviparid snail species dmployed in this study mostly showed monomorphic enzyme protein banding patterns, and genetic differences specific between two species.

Changes in Isozyme Patterns of Peroxidase and Esterase during Regeneration of Pepper (Capsicum annuum L.) (고추 (Capsicum annuum L.)의 재분화에 따른 Peroxidase와 Esterase 동위효소의 양상 변화)

  • The plantlet was regenerated on MS medium containing BAP (2 mg/I) and 1M (1 mg/I) from leaf discs of pepper after 3 weeks of culture. And then, we investigated the activity of peroxidase and esterase and the pattern of their isozymes from leaf, stem and root in order to observe physiological and biochemical changes on the developemental stage, respectively. The peroxidase was expressed with tissue specificity because peroxidase activity according to the developemental stage of the tissue was not only highest in the leaf of the pepper at 10 days after it germinated but also 2 new bands of its isozyme were found in pI 7.2 and pI 5.2. However, a new pI 3.4 band was found in the leaf and root of the pepper after 14 days of germination, and in the stem was found out pI 5.2 band. As regeneration of leaf dises was progressed, its peroxiase activity was increased about 80% more than that of control after 14 days of culture and new pI 3.2 and 6.5 bands of it isozyme were found. The results suggested that peroxidase would be connected with regeneration of pepper. Also, esterase activity was increased about 50% more than that of control after 14 days of culture, the pattern of esterase isozyme was shown to be 3 cathodic bands and 1 anodic band after 7 days of culture.ulture.

  • Studies were carried out to compare from the viewpoint of peroxidase isoenzyme patterns by ages, locations and organs. In addition, the relationships among six cultivars peroxidase isoenzyme patterns were compared and the relationships between cultivars and their hybrids were examined. The results are summarized as follows. 1. The zymogram was not affected by the ages of 'Fuji' cultivar. 2. There was no locational difference in zymogram among Suwon, Taejon and Yesan in 'Fuji' and 'Tsugaru' cultivars. 3. Significant difference in isozyme patterns was found between stems and leaves in 'Fuji' and 'Tsugaru' cultivars. 4. There were some differences in zymogram among 'Ralls Genet', 'Fuji', 'Jonathan', 'Starking', 'Golden Delicous' and 'Tsugaru' cultivars. 5. Classification was made by the peroxidase isozyme development between cultivars and their hybrids. 1) The type showed both parent's isozyme pattern. 2) The type showed a loss of a part of parent's isozyme bands. 3) The type developed new isozymes which were not detected in parent plants.

The Protein and Isozyme Patterns During in vitro Plant Regeneration of Yooja (Citrus junos Sieb.) and Trifoliate Orange (Poncirus trifoliata Rafin.)

  • In this study, plant regeneration through in vitro culture from plantlet stems of Yooja (C. junos Sieb.) and trifoliate orange (P. trifoliata Rafin.) was attempted to make mass-production system of virus-free plants having the same genotype with mother plant. In order to investigate physiological change depending on the developmental stage of plant regeneration, the changes of total protein, peroxidase and esterase activity and their isozyme patterns as well were examined in 1/2 MS medium. The results are as follows : 1. The MS medium for the optimal callus induction and shoot formation was utilized. The medium was supplemented either with 2,4-D and Kinetin or with BA and NAA. The optimal concentrations were the combination of 1.0mg/ 2,4-D +0.3mg/ Kinetin and 1.0mg BA +0.3mg NAA in callus induction and shoot formation, respectively. 2. For the plant regeneration from somatic embryos, 1/2 MS medium was used with supplements of growth regulators (free, 1.0mg/ IBA +1.0mg/ BA ,0.5mg/ IBA +0.5mg/ BA). Shooting and rooting were the best in the treatment of 0.5mg/ IBA and 0.5mg/ BA combination. 3. The total protein content has a tendency of increase with the developmental stage of embryo, but it was decreased at the plantlet. Also it was the highest at 8 and 6 weeks stage in C. junos Sieb. and P. trioliata Rafin, respectively. In the SDS-PAGE pattern of protein, C. junos Sieb. showed bands of 29.0 and 40kDa at 10 weeks. The 45,66 and 97.4 kDa bands at 10 weeks of culture were shown in P. trifoliata Rafin. 4. The highest esterase activity was shown at the 6 and 8 weeks of culture in C.junos Sieb. and P. trifoliata Rafin.., respectively. 5. Esterase isozyme patterns were shown difference according to the developmental stage. In C. junos Sieb. a new band was observed at pl 7.7 following 4 weeks culture. On the other hand, new bands in P. trifoliata Rafin. were observed at pl 7.5~6.5 following 4 and 6 weeks culture, respectively.

  • Isozyme patterns of leucine aminopeptidase, esterase and protein from 16 isolates of Ganoderma lucidum were observed by electrophoresis for characterization of the isolates. Even in the same isolate, the patterns of the isozymes from mycelium and cap were different. The esterase patterns from the mycelium could differentiate the isolates. Of 16 isolates, four isolates showed identical patterns. It was assumed that these had the same genetic background.

Alterations of Glutathione Peroxidase Patterns by Stressor Treatment in Rice Seedling Roots (스트레스 물질에 의한 벼 glutathione peroxidase 활성패턴 변화)

  • The effect of various stressors such as reductant ascorbic acid, signalling molecules (salicylic acid and methyl jasmonic acid), heavy metals $(NiCl_2,\;and\;MnSO_4)$ and NaCl on the glutathione peroxidase (GPX) activities and isoenzyme expression patterns were investigated in rice seedling roots. Total GPX activity increased according to the increase of ascorbic acid concentration. Prominent enhancement of GPX1 isozyme due to ascorbic acid contributed to the increase of total GPX activity. GPX showed different reactivity toward salicylic acid and methyl jasmonic acid. GPX activity increased at 0.1 mM salicylic acid, and then decreased thereafter. However, GPX increased gradually in a methyl jasmonic acid concentration-dependent manner, and 3 fold increase of GPX activity was found at 1 mM methyl jasmonic acid. Moreover, GPX1 isozyme increased according to the increase of salicylic acid, while GPX1 isozyme decreased according to the increase of methyl jasmonic acid. When metal ions were treated, GPX activity increased considerably according to the increase of $NiCl_2$ concentration, however, GPX activity increased about 2 fold at 0.5 mM $CuSO_4$ and then decreased. Enhancement of GPX1 isozyme contributed to the increase of total GPX activities in $NiCl_2-treated$ and $MnSO_4-treated$ rice seedlings. Total GPX activity increased 1.7 fold in response to 300 mM NaCl. Especially GPX2 isozyme showed gradual increase according to the increase of NaCl concentration.
