Isozyme Analysis and Relationships Among Three Species in Malaysian Trichoderma Isolates

  • Isozyme and protein electrophoresis data from mycelial extracts of 27 isolates of Trichoderma harzianum, 10 isolates of T. aureoviride, and 10 isolates of T. longibrachiatum from Southern Peninsular Malaysia were investigated. The eight enzyme and a single protein pattern systems were analyzed. Three isozyme and total protein patterns were shown to be useful for the detection of three Trichoderma species. The isozyme and protein data were analyzed using the Nei and Li Dice similarity coefficient for pairwise comparison between individual isolates, species isolate group, and for generating a distance matrix. The UPGMA cluster analysis showed a higher degree of relationship between T. harzianum and T. aureoviride than to T. longibrachiatum. These results suggested that the T. harzianum isolates had high levels of genetic variation compared with the other isolates of Trichoderma species.

Changes of Glucose-6-Phosphate Dehydrogenase on Alloxan-induced Diabetic Rat (Alloxan으로 유발된 당뇨쥐의 간조직 Glucose-6-Phosphate Dehydrogenase 효소의 변화)

  • The hypoglycemic and metabolic effects of Commelina communis L. extract were investigated in alloxan-induced diabetic rats. The increased blood glucose level in the diabetic rats was sinificantly lowered with the treatments of the plant protein extract. Administration of the plant extract ellicited the significant increase of glucose-6-phosphate dehydrogenase (G6PD) activity in liver of alloxan-induced rats. Three isozyme patterns(band I, II & III : in order decreasing mobility) of G6PD were found when normal rat liver extract were subjected to electrophoresis on native polyacrylamide gel. On the other hand, G6PD band patterns of alloxan-induced rat liver extract were found band II isozyme missing. By treatment of plant extract in alloxan-induced rats has been showed pattern the recovery of missing band patterns. This indicates that changes of the G6PD isozyme might be related to the cellular process of diabetes.

Taxonomic Position and Affinities of Isopyrum mandshuricum within Korean Isopyroideae (Ranunculaceae) Based on Molecular Data

  • To examine the taxonomic position and affinities of Isopyrum mandshuricum (Ranunculaceae) and related taxa, genetic analysis were carried out on the basis of isozyme patterns and ITS sequences. Molecular data, both isozyme patterns and ITS sequences suggest that I. mandshuricum is closely related to Enemion raddeanum than to Semiaquilegia adoxoides. The estimation of genetic identities by isozyme analysis reveals that I. manshuricum is genetically distant from E. raddeanum. The phylogenetic tree based on molecular data is rather congruent with the phenogram based on quantitative morphological characteristics, but not consistent with one based on qualitative morphological characteristics. Incongruencies between molecular and qualitative morphological data provide clues to re-evaluate several morphological features.

  • Isozyme comparisons of mycelial extracts from Pleurotus ostreatus were undertaken using isoelectric focusing. Enzyme isozyme patterns were Used to describe the extent of geographical diversity and degree of intraspecific variation in these extracts. A total of 77 bands were resolved from six different enzymes. Cluster analyses were performed using the zymograms for esterase (EST), glucose phosphate isomerase (GPI), leucine aminopeptidase (LAP), malate dehydrogenase(MDH), peroxidase (POX), and phosphoglucomutase (pGM). EST gave multiple banding patterns, while less variability was observed for GPI, MDH, and PGM. Cluster analyses demonstrated that strains of P. ostreatus from geographically different origins are genetically divergent, supporting the idea that there is little or no gene flow between these geographically distant population groups.

  • In this study, we calculated the genetic relationships of Pestalotiopsis species collected from various places in southern part of Korea through isozyme analyses. As a result, EST showed the largest number of band, and the number of bands were ranged from 5 to 7 on the average. All the isozymes used in this study showed distinctive band patterns for each isolates. Similarities among the compared isolates ranged from 48 to 93%. Isolates SP7, SP19 and SP23 showed more than 90% similarities, and most isolates showed similarities ranging from 65 to 82%.

  • A series of experimental groups has been studied in the state of hypolycemia caused by a single intraperitoneal injection of insulin after 24 hrs of fasting albino rats and then the variation of LDH activities and LDH isozyme patterns in the liver, muscle and serum had been reported. The total LDH activity has been elevated in the liver and the muscle above the average level for control group, but increased continusly during 20 min and decreased in the 20-45 min intervals and increased again 45 min in the serum. The change of LDH isozyme patterns had been shown that in the liver LDH$_{5}$ was increased, LDH$_{4}$ was decreased and in the muscle LDH$_{1}$ was diminished by 30 min ws restored again after 45 min and LDH$_{2}$ decreased about 94 percentage at 30 min, decreased by 45 min and increased greater again after 45 min and in the muscle LDH$_{3}$, LDH$_{4}$, and LDH$_{5}$ were increased to the greatest by 20 min, decreased in 20-45 min intervals and increased again after 45 min.

  • A cellulose acetate electrophoretic survey of Korean Anura has revealed the presence of diverse lactate and malate dehydrogenase isozymes. The LDH and MDH isozymes in the tissues of the brain, heart, stomach, skeletal muscle and liver of the six species of Anura examined show the species specific patterns which differ from those of mammals and birds. Two isozymic forms of LDH and MDH exist in both Rana nigromaculata and Rana nigromaculata coreana, respectively, with almost the same pattern. LDH of Bombina orientalis has five isozymic forms, and that of Rana temporaria ornativentris contains four isozymes. Bufo sp. has 3 to 5, and Rana rugosa has 3 to 4 isozymic bands according to the tissues. MDH's of all animals have two isozymic forms with different spacing on the zymograms.

Effect of Metals on Anti-Oxidase Activity and Isozyme patterns in Brassica juncea

  • To study the effects of metal ions on the activity of anti-oxidase enzymes, the activity of superoxide dismutase (SOD) and peroxidase (POD) and isozyme patterns of Brassica juncea have been studied after treating with CD, Cu, Zn, and Al. The activity of SOD after treating with metal ions was higher than that of untreated control. SOD activity in leaves increased by treatment of 50 ppm of Zn and 500 ppm of Al. POD in stems gave highest activity after treating with 500 ppm of Cu. When the activity was compared by plant parts, lowest POD activity was observed in leaves in which protein content was higher than other tissues. When the activity was expressed as percentage of control, SOD activity was increased after treating with metal ions. SOD activity in leaves and roots of metal treated plant was significantly increased under the metal ions stress conditions. In the roots of 50 ppm of Zn treated plant, SOD activity was extremly high. POD activity was inhibited with Cd and Zn treatment in all parts of the plant. However, in leaves and stems, there was marked increase in activity after treating with Cu. The patterns of SOD isozyme after metal treatment show that two bands were stained in all metal ion treated and that no new band appeared. POD isozyme band intensity resulting from the treatment of metal ions was in order of roots > stems > leaves, but there was no significant difference.

Effect of Light and Cadmium on the Activity and Isozyme Pattern of Catalase from Ric(Oryza sativa L.) (빛과 카드뮴이 벼 catalase 활성과 동위효소 발현에 미치는 영향)

  • The effects of cadmium on the catalase activity and isozyme patterns under light and dark conditions of rice(Oryza sativa L. cv. Dongjin) seedlings were examined. Cadmium treatment resulted in the notable enhancement of $H_2O_2$ contents in the seedling roots and leaves under light and dark conditions. The catalase isozyme patterns in the roots were different from those in the leaves, showing tissue-specific expression of the enzyme. Moreover, the expression patterns of catalase isozymes in the green seedling roots were different from those in the etiolated seedling roots following cadmium treatment. The increase of total catalase activity was about 16 times at 1 mM cadmium and marked inductions of the isozyme CAT1 and CAT2 contributed to this increase in the green seedling roots. On the other hand, in the etiolated seedling roots, total catalase activity was lower than that of control at 0.5 and 1 mM cadmium, even though catalase activity increased about 3 times at 0.1 mM cadmium. The 3 fold increase of total catalase activity was mainly due to the increase of CAT1, CAT3 and CAT4 at 0.1 mM cadmium. However, treatment with higher concentrations of cadmium decreased the activity of CAT2 and CAT4 in the etiolated roots. In the leaves, the catalase existed as three isozymes; one cationic isozyme CATc, one neutral isozyme CATn and one anionic isozyme CAT1 in the control. The isozyme patterns and total activities remained unaffected by cadmium under light and dark conditions in the seedling leaves. Taken together, it seems that cadmium-induced changes of catalase might be regulated by light in the roots, but not in the leaves.

  • In the rats administered mercuric chloride (1mg/kg/48 hrs i.p.), it was found that LDH$_{1}$ and LDH$_{2}$ were increased in heart, brain and kidney as well as LDH$_{5}$ increased in liver and muscle in the duration of sixteen days. After the sixteen days of administration, the LDH isozyme patterns in the above mentioned organs were found to be irregularly changed. Considerable amount of mercury accumulation in liver and kidney were found, and especially the mercury accumulation in kidney was notable.
