토끼 적출 신동맥에 있어서 전기자극에 의한 신경성 수축작용의 neuromodulation 효과 (Neuromodulation on neurogenic contraction of electrical nerve stimulation on isolated renal artery of rabbit)

  • To elucidate the neuromodulation of neuropeptide Y and $\alpha,\;\beta$-methylene ATP on the neurogenic contraction of electrical perivascular nerve stimulation and the contractile response of noradrenaline from polygraph in the isolated renal artery of rabbit. 1. The neurogenic contraction induced by perivascular nerve stimulation was the voltage-dependent manner(10-100V) in the isolated renal artery of rabbit. 2. Neuropeptide Y(0.1uM) and $\alpha,\;\beta$-methylene ATP(1uM) increased the contractile responses of noradrenaline in the isolated renal artery of rabbit. 3. Neuropeptide Y(0.1uM) and $\alpha,\;\beta$-methylene ATP(1uM) increased the neurogenic contraction of electrical perivascular nerve stimulation in the isolated renal artery of rabbit. These results suggest that neuropeptide Y and $\alpha,\;\beta$-methylene ATP neuromodulated on the neurogenic contraction of electrical perivascular nerve stimulation on the isolated renal artery of rabbit.

Differential effects of saturated and unsaturated fatty acids on vascular reactivity in isolated mesenteric and femoral arteries of rats

  • Free fatty acid (FFA) intake regulates blood pressure and vascular reactivity but its direct effect on contractility of systemic arteries is not well understood. We investigated the effects of saturated fatty acid (SFA, palmitic acid), polyunsaturated fatty acid (PUFA, linoleic acid), and monounsaturated fatty acid (MUFA, oleic acid) on the contractility of isolated mesenteric (MA) and deep femoral arteries (DFA) of Sprague-Dawley rats. Isolated MA and DFA were mounted on a dual wire myograph and phenylephrine (PhE, $1-10{\mu}M$) concentration-dependent contraction was obtained with or without FFAs. Incubation with $100{\mu}M$ of palmitic acid significantly increased PhE-induced contraction in both arteries. In MA, treatment with $100{\mu}M$ of linoleic acid decreased $1{\mu}M$ PhE-induced contraction while increasing the response to higher PhE concentrations. In DFA, linoleic acid slightly decreased PhE-induced contraction while $200{\mu}M$ oleic acid significantly decreased it. In MA, oleic acid reduced contraction at low PhE concentration (1 and $2{\mu}M$) while increasing it at $10{\mu}M$ PhE. Perplexingly, depolarization by 40 mM KCl-induced contraction of MA was commonly enhanced by the three fatty acids. The 40 mM KCl-contraction of DFA was also augmented by linoleic and oleic acids while not affected by palmitic acid. SFA persistently increased alpha-adrenergic contraction of systemic arteries whereas PUFA and MUFA attenuated PhE-induced contraction of skeletal arteries. PUFA and MUFA concentration-dependent dual effects on MA suggest differential mechanisms depending on the types of arteries. Further studies are needed to elucidate underlying mechanisms of the various effects of FFA on systemic arteries.

  • To elucidate the effect of calcitonin gene-related peptide(CGRP), vasoactive intestinal peptide(VIP) and substance P was investigated with perivascular nerve stimulation and treatment of peptides from polygraph in the isolated renal artery of rabbit. 1. The neurogenic contraction induced by perivascular nerve stimulation was the frequency-dependent manner(264 Hz) in the isolated renal artery of rabbit. 2. CGRP and VIP caused the relaxation on the precontraction with noradrenaline($10{\mu}m$) on the presence and absence of endothelium in the isolated renal artery of rabbit. 3. Substance P caused the endothelium-dependent relaxation on the precontraction with noradrenaline($10{\mu}m$) in the isolated renal artery of rabbit. 4. CGRP and VIP inhibited the neurogenic contraction by the perivascular nerve stimulation(0.3 ms, 80 V, 50 Hz, 1 sec) on the absence and presence of endothelium in the isolated renal artery of rabbit. 5. Substance P inhibited on the neurogenic contraction by the perivascular nerve stimulation with the endothelium-dependent in the isolated renal artery of rabbit.

심룡근(心朧筋)의 반복수축현상(反復收縮現象)에 관(關)하여 (After Contraction in Isolated Cardiac Muscle)

  • Present paper is attempted to introduce the phenomenon of 'after contraction' in isolated cardiac-muscle. Papillary muscles were removed from cat right ventricle and were used as a preparation. The muscle strip was Placed in tissue bath which is kept in steady temperature of around $25^{\circ}C$ and was perfuced by Tyrode solution, saturated with 95% $O_2$ and 5% $CO_2.$ under the condition of high calcium (8.2-10.0 mM/l), low sodium (72.4-70.0 mM/l) perfusion with the administration of epinephrine (1-2 mg/l) into tile tissue bath normally triggered muscle contraction was followed by oscillatory, repetitive contractions - after contraction. The phenomenon of after contraction was augumented by decrease in tissue bath temperature and by increase in number of preceding beats and in driving rate. Authors were able to maintain the phenomenon in prominent and steady state giving proper experimental conditions such as fixed bath temperature (ranged from $22^{\circ}C\;to\;27^{\circ}C$), suitable driving rate (20 per minute in average) and perfusion of high calcium, loll sodium and 1-2 mg/l of epinephrine. In some preparations, the strength of after contraction (second contraction) reached up-to 80% of normally triggered contraction and five repetitive contractions were observed as largest number of after contractions. Intracellular action potential measured in the muscle which was beating regulary showing steady after contraction revealed no oscillating after potential in most parts of the muscle but in few cases oscillating changes of after potentials were detectable. In electrogram of the muscle preparation recorded by means of contact electrode prominent, oscillating after potentials were observable when the recorder was set at highest sensitivity. It still is not clear that whether after contraction is the phenomenon which corresponds to those changes in action potential, oscillating after potential, of the muscle preparation. Possible mechanism of the phenomenon of after contraction relating with after potential changes was proposed. Detailed results obtained from further studies on after contraction and concrete discussion on the phenomenon will be reported by authors.

Effect of Cyclic Nucleotides on Phorbol Ester-Induced Contraction in Rabbit Carotid Artery

  • This study was designed to clarify the action of cyclic nucleotides, cyclic AMP and cyclic GMP, on phorbol 12,13-dibutyrate (PDBu)-induced contraction in rings isolated from rabbit carotid artery. Arterial rings, 2 mm in width, were myographied isometrically in an isolated organ bath. PDBu produced slowly developing, sustained contraction in rabbit carotid artery, in a dose dependent manner, which was independent of extracellular $Ca^{2+}$ PDBu-induced contraction was relaxed by staurosporine, which suggests that PDBu-induced contraction is mediated by protein kinase C (PKC). $^{45}Ca^{2+}$ uptake by rabbit carotid artery was increased by PDBu during depolarization, but not in control. Isoproterenol and sodium nitroprusside (SNP) relaxed phenylephrine-induced contraction. However, SNP but not isoproterenol relaxed the contraction induced by PDBu. Acetylcholine relaxed PDBu-induced contraction in the presence of the endothelium. 8-bromo-cyclic AMP, a permeable analogue of cyclic AMP, suppressed phenylephrine-induced contraction but not PDBu-induced contraction. 8-bromo cyclic GMP relaxed both of them with dose dependency. A large dose of forskolin relaxed PDBu-induced contraction. PDBu increased cyclic AMP without considerable change in the level of cyclic GMP. Based on these findings, PDBu-induced contraction of rabbit carotid artery was relaxed by cyclic GMP more effectively than cyclic AMP, and the action of cyclic AMP could be mediated by cyclic GMP dependent protein kinase. Therefore it is suggested that the antagonistic action between protein kinase C and cyclic GMP-dependent protein kinase plays a major role in the regulation of vascular tone.

몇가지 자율신경성약물(自律神經性藥物)이 Reserpine 처리(處理) 병아리의 척출공장(剔出空腸) 내재성신경(內在性神經)에 미치는 영향(影響) (Effects of Some Autonomic Drugs on the Intramural Nervous System of Isolated Jejunum of Chiken Pretreated with Reserpine)

  • Pharmacological investigation was performed in chickens which were pretreated with reserpine. Transmural stimulations were given to the isolated jejunum of chickens and studied the responses and the effects of dibenamine, propranolol, atropine and tetrodotoxin on them. The results obtained were summerized as follows: 1. Three different patterns of response were obtained from the isolated jejunum of non-treated chickens after giving them transmural stimulation. The first pattern was contraction followed by relaxation, the second pattern was contraction only and the third pattern was relaxation only. 2. The transmural stimulation of the jejunum preparations evoked relaxation while the periarterial stimulation evoked contraction in the reserpinized chick. 3. The relaxation response to the transmural stimulation was not affected by the pretreatment with dibenamine, propranolol and atropine. 4. The relaxation response to the transmural stimulation was abolished by tetrodotoxin. The results obtained in these studies indicate that the relaxation response to the transmural stimulation is due to non-adrenergic inhibitory fibers.

중력이 혈관에 미치는 영향 (Effect of Bambusae Caulis in Liquamen on the Isolated Rat Aorta)

  • Bambusae Caulis in Liquamen has been used as a herbal medicine for the treatment of heat, paralysis of the hands or feet, or hemiplegia. In the present study, we investigated the effect of Bambusae Caulis in Liquamen to the phenylephrine (PE) induced contraction of isolated rat aorta Contractile force was measured with force displacement transducer under 1.5 g loading tension. Contractions evoked by PE 0.1 μM were significantly increased by low dosage of Bambusae Caulis in Liquamen, decreased by high dosage of Bambusae Caulis in Liquamen. L-NNA, ODQ and propranolol significantly altered the effect of Bambusae Caulis in Liquamen, but indomethacin did not change the relaxation of Bambusae Caulis in Liquamen. These results suggest that Bambusae Caulis in Liquamen can relax EP induced contraction of isolated rat aorta and that this decreasing contraction related to sympathetics.

Caerulein의 담낭수축작용에 관한 연구 (Studies on the gallbladder contraction induced by caerulein)

  • The effects of phenoxybenzamine and other related drugs were studied for their interaction with caerulein on gallbladder contraction in anesthetized animals and isolated gallbladder strips. Cholecystostomy and cystic duct ligation were made on anesthetized dog, cat and pig. Pressure changes of gallbladder were measured by a physiological pressure transducer connected to polygraph recorder. Isolated rabbit gallbladder strips were placed in a muscle chamber containing Locke-Ringer solution maintained at $38^{\circ}C$. The contractile responses were measured by a force-displacement transducer connected to polygraph recorder. Caerulein ($30{\sim}200$ ng/kg i.v.) produced marked contraction of gallbladder in situ and the cholecystokinetic potencies appear in decreasing order; dog, cat and pig. The response of caerulein was abolished by the large doses of phenoxybenzamine (15 mg/kg i.v.) but not affected with dibenamine, phentolamine or tolazoline. Cholecystokinetic effect of methacholine or barium chloride was also partially inhibited by phenoxybenzamine and the effect of caerulein was weakly inhibited intravenous injection of cyclophosphamide or papaverine. In isolated rabbit gallbladder strips, the response of contraction to caerulein were progressively inhibited by pretreatment of phenoxybenzamine along with time exposed. These results lead to the conclusion that phenoxytenzamine may inherently inhibit the contractile response of gallbladder to caerulein, and this effect was not related with ${\alpha}-adrenergic$ receptor blocking action.

복분자가 백서의 적출동맥에 미치는 영향 (Effect of Fructus Rubi on the isolated rat aorta)

  • This study is to determine the effect of Fructus Rubi to the phenylehrine (PE) induced contraction of isolated rat aorta. Contractile force was measured with force displacement transducer under 1.5g loading tension. Contractions evoked by PE 0.1 μM and KCI 65.4 mM were decreased significantly by Fructus Rubi. L-NNA, ODQ, indomethacin and atropine significantly altered the effect of Fructus Rubi, but propranolol did not change the relaxation of Fructus Rubi. These results indicate that Fructus Rubi can relax EP and KCI induced contraction of isolated rat aorta and that this decreasing contraction related to epithelium, nitric oxide, and parasympathetics.

Effect of Imipramine on Calcium Utilization of Single Cells Isolated from Canine Detrusor

  • This study is to investigate the mechanism of inhibitory effect of imipramine on the calcium utilization in single cells isolated from canine detrusor. 2 mm thick smooth muscle chops were incubated in 0.12% collagenase solution at $36^{circ}C,$ and aerated with 95% $O_2/5%\;CO_2,$ and then cell suspension was examined. Acetylcholine (ACh) evoked a concentration-dependent contraction of the isolated detrusor cells in normal physiologic salt solution (PSS), and the ACh-induced contraction was significantly inhibited by imipramine. In $Ca^{2+}-free$ PSS, ACh-induced contraction was less than those in normal PSS and it was not affected by the pretreatment with imipramine. $Ca^{2+}-induced$ contraction in $Ca^{2+}-free$ PSS was supressed by imipramine, but addition of A 23187, a calcium ionophore, overcomed the inhibitory effect of imipramine. High potassium-depolarization (40 mM KCl) evoked cell contraction, which was inhibited by imipramine. Caffeine, a releasing agent of the stored $Ca^{2+}$ from sarcoplasmic reticulum, evoked a contraction of the cells that was not blocked by the pretreatment with imipramine. These results suggest that imipramine inhibits the influx of calcium in the detrusor cells through both the receptor-operated- and voltage-gated-calcium channels, but does not affect the release of calcium from intracellular storage site.
