산양유를 섭취한 성인 여성의 분변에서 분리한 Bifidobacteria와 Lactobacilli의 탄수화물 발효특성 (Carbohydrate Fermentation Character of Bifidobacteria and Lactobacilli isolated from Feces of the Adult Women supplied with Goat Milk)

  • 산양유을 섭취한 성인의 분변 내 bifidobacteria의 증식 효과 및 분변에서 분리된 bifidobacteria와 lactobacilli의 당 발효 능력을 조사하였다. 1. 산양유를 섭취한 처리군이 대조군보다 bifidobacteria 세균수가 상대적으로 높았으며, 8주에서 95% 신뢰한계에서 유의성이 있었다. 2. 분변에서 분리한 균주들을 13 균주의 16S rRNA 염기서 열로 동정한 결과, 산양유를 섭취한 처리구는 B. adolescentis, B. longum, B. pseudocatenulatum, B. dentium, L. sakei 순으로 분리되었다. 대조군의 12 균주는 B. adolescentis, B. longum, L. ruminis, L. sakei, B. pseudocatenulatum 순으로 분리되었다. 3. 산양유 올리고당과 lactulose는 모든 bifidobacteria 및 lactobacilli 균주가 발효하였다. Fructooligosaccaride는 B. adolescentis 7 균주 모두 발효하였다. 조사한 B. pseudocatenulatum 4 균주, L. sakei 3 균주, B. longum 7 균주 중에서 각각 3, 2, 1 균주가 fructooligosaccharide를 발효하였다.

유산균의 장내환경개선효과 (Isolation of bifidobacteria inhibiting harmful enzymes of Korean intestinal bacteria)

  • Five hundreds of bifidobacteria were isolated from an healthy Korean and the inhibitory effects of these isolated bacteria on harmful enzymes of human intestinal microflora were examined by cocultivation of the isolated bifidobacteria with E. coli HGU-3 or total human intestinal microflora. In comparison with the results of E. coli or intestinal microflora cultivation, Bifidobacterium breave K-110, B. breve K-111 and B. infantis K-525 effectively inhibited harmful enzymes (${\beta}-glucuronidase$ and tryptophanase) of E. coli and lowered the pH of the culture media. Also they inhibited the harmful enzymes (${\beta}-glucosidase$, ${\beta}-glucuronidase$, tryptophanase and urease) and ammonia production of intestinal microflora, and lowered pH of the culture media by increasing the number of bifidobateria on intestinal microflora. The inhibitory effect of bifidobacteria on Growth of Helicobacter pylori and Rotavirus infection were exammed. Bifidobacterium K-110 and K-111 inhibited effectively them. When these isolated bifidobacteria were administered to mice, the activities of fecal harmful enzymes were inhibited and the AC and ACF formation were suppressed. Among tested bifidobacteria, B. breve K-110 had high inhibitory effect of fecal harmful enzymes and ACF formation.

Comparison of Bifidobacteria Selective Media for the Detection of Bifidobacteria in Korean Commercial Fermented Milk Products

  • This study was carried out to compare the efficacy and selectivity of TOS and BS media for enumeration of bifidobacteria in commercial fermented milk products. First, bifidobacteria was isolated from 20 fermented milk products, and all isolated bifidobacteria were identified by genomic technology as Bifidobacterium lactis. The two media significantly differed from each other with regard to the recovery of B. lactis, that is, the recovery of this organism was as much as 6 logs lower on BS medium than on TOS. When the concentration of BS solution (mixture of paromomycin sulfate, neomycin, sodium propionate, and lithium chloride) used in BS medium was reduced to 50% (BS50), a relatively high percentage recovery of bifidobacteria from pure cultures was achieved. Susceptibility tests to antibiotics and tests for selective agents for the isolated bifidobacteria and lactic acid bacteria were conducted. The BS solution inhibited some lactic acid bacteria and Bifidobacterium species, while mupirocin (MU) suppressed the growth of all tested lactic acid bacteria but not Bifidobacterium. As compared with BS50 medium, TOS with or without MU showed good bifidobacteria recovery and readily distinguishable colonies; in particular, TOS supplemented with MU had a high selectivity for bifidobacteria. In conclusion, all results suggested that TOS medium with or without MU was found to be suitable for selective enumeration of bifidobacteria from mixed cultures in fermented milk, and better in that capacity than BS medium.

Inhibitory Effects of Bifidobacterium spp. Isolated from a Healthy Korean on Harmful Enzymes of Human Intestinal Microflora

호상 요구르트 제품에 이용되는 Bifidobacteria 의 선택적 검출 방법 (Stydies on the Selective Screening Method of Bifidobacteria Used in Yoghurt)

  • In fermentation of dairy products, bifidobacteria is used in conjunction with other lactic acid bacteria, such as L. acidophilus, L. bulgaricus and S. thermophilus, rendering the enumeration of bifidobacteria difficult. In order to develop optimum conditions for selective enumeration of bifidobacteria, we examined MIC of several antibiotics against various bifidobacteria and other lactic acid bacteria. The growth of L. acidophilus, L. bulgaricus and S. thermophilus were inhibited by lithium chloride at the concentration of less than 4 mg/ml, whereas growth inhibition of bifidobacteria occurred at concentrations over 6-10 mg/ml. Tetracycline and chloramphenicol were also found to selectively inhibit growth of other lactic acid bacteria at the concentration of 1-3 $\mu$g/ml. Addition of 6 mg/ml lithium chloride, 1 $\mu$g/ml, tetracycline or 3 $\mu$g/ml chloramphenicol to medium was found to be optimal for selective enumeration of bifidobacteria. By using these three inhibitory chemicals in the TPY medium, higher number of bifidobacteria were selectively isolated than with NPNL agar and LP agar.

Hypocholesterolemic Activity of Bifidobacteria Isolated from a Healthy Korean

  • This study was undertaken to investigate the hypocholesterolemic activity of Bifidobacteria (B. breve K-110, B. breve K-111, and B. infantis K-525) isolated from a healthy Korean. The administration of B. breve K-110 and K-111 with a high cholesterol diet significantly protected the increase of serum total cholesterol and LDL cholesterol relative to that of a high cholesterol diet alone. Such a diet supplemented with 0.5% B. breve K-111 decreased serum total cholesterol and LDL cholesterol to 57 and 55%, respectively. The administration of Bifidobacteria also significantly inhibited the lipid-deposited surface in the aorta. The normalizing activity of serum cholesterol level in cholesterolemic rats was accelerated by Bifidobacteria. The normalizing activity of B. breve K-111 on serum cholesterol level was superior to that of B. breve K-110. These results suggest that Bifidobacteria in the human intestine playa role in the prophylactics of arteriosclerosis.

한국인의 분변으로부터 내산소성 균주의 분리, 동정 및 분리 균주의 특성 (Isolation and Identification of Oxygen Resistant Bifidobacterium sp. from Korean and its Characteristics)

  • Bifidobacteria have been known as beneficial inhabitant of human intestine. Therefore, bifidobacteria began to be noticed as a starter in the manufacture of fermented dairy products. Perhaps the key for effective use of bifidobacteria in commercial dairy products is the maintenance of viability of bifidobacteria during large scale preparation of starter culture and distribution of products. So we tried to obtain the bifidobacteria having suitable characteristics for using as a starter in the manufacture of fermented dairy products. Among bifidobacteria isolated from Korean, E-4 strain showed the highest resistance to oxygen. To know whether the selected strain will be fit for manufacture of fermented dairy products, we also investigated resistance of the selected strain to HCI. The selected strain, E-4, was more resistant to environmental stresses such as oxygen, H2O2 and HCI than Bifidobacterium longum known as resistant strain to environmental stresses. According to carbohydrate fermentation patterns and morphological characteristics, E-4 strain was identified as B. bifidum. In conclusion, the selected strain, E-4, was thought to be fit for manufacture of fermented dairy products.

Bifidobacterium longum BO-11을 이용한 김치의 제조 (Preparation of Kimchi Containing Bifidobacterium longum BO-11)

  • 건강한 성인의 분변에서 분리한 내산성 및 내담즙성이 우수한 B. longum BO-11을 김치 제조용 스타터로 이용하여 그 생존 능력과 김치 발효에 미치는 영향에 대해 조사하였다. 발효과정 중 bifidobacteria의 계수는 BS 배지를 사용하였으며 20일의 저장 기간 동안 균수가 약간 증가하였다. 일반 미생물은 bifidobacteria 비첨가군과 비슷하였으며 bifidobacteria 첨가군의 경우 산의 생산이 더 많았다. 김치 맛은 일반 김치보다 bifidobacteria 첨가 김치가 더 우수하였다. 이러한 결과는 bifidobacteria를 이용한 기능성 김치의 상업화 가능성을 보여준다.

Effect of Bifidobacteria on Production of Allergy-Related Cytokines from Mouse Spleen Cells

  • To study the effect of bifidobacteria on preventing allergy response, levels of IFN-$\gamma$, IgG2a, IL-4, and IgG1 were investigated in splenocytes isolated from ovalbumin (OVA)­sensitized allergic mice and BGN4-administered allergy­suppressed mice in the presence of various bifidobacterial strains. Most of the bifidobacteria, except 2A, increased production of Th I-associated immune markers, IFN -$\gamma$ and IgG2a. In addition, most of the bifidobacteria, except 2A and 19A, decreased production of IL-4, whereas the differences in the production of IgG1 were less pronounced. These results suggest that some strains of bifidobacteria may have the potential to prevent the occurrence of allergy by switching Th1/Th2-type antibodies and/or related cytokines.

한국인 분변에서 분리한 Bifidobacteria의 탈지유에서의 배양특성

  • Wild strains of bifidobacteria isolated from Korean feces were tested for their growth and acid production abilities in 10% skim milk. Growth of bifidobacteria was markedly decreased from the second transfer in the skim milk culture. When two strains, BF5 and BF33, were grown in skim milk with various supplements, the growth was enhanced by supplementation of 0.5% yeast extract, and 0.05% cysteine but not by short chain fatty acids. There was no enhancing effect of CO$_{2}$, substitution in the fermentor on growth. The viable cell counts of bifidobacteria, BF5 and BF33, were 9.76 and 9.98 logCFU/ml, respectively, after 30 hr cultivation and were diminished by 3 and 6 logs during storage at 5$\circ$C for 12 days.
