  • Four different backcrossing methods were designed and 23 near isogenic lines (NILs) of 22 linkage groups were obtained using Hb as recurrent parent, the mutant gene lines which held markers as donor parents. Eleven of them had been mated with the recurrent parent for 10 times, and the others for 7∼8 times. The NILs of other 6 linkage groups are under way and had been backcrossed to the recurrent for 3∼4 times. These NILs will act important roles in the construction of molecular linkage map and gene location and positional cloning.

  • In order to develop near isogenic lines (NILs) the heterozygous rice plants for alkali digestibility value(ADV) were selected and tested in every generation from $F_5$to $F_9$ of a cross, Seratus malam/Suwon 345. Finally several sets of NILs, which were six low ADV lines and four medium-high ADV lines, were selected among $F_{10}$ lines. No differences of the plant growth characteristics, amylose content and protein content of rice grain were found between low and medium-high ADV lines. Rice flour of low ADV lines showed longer gel length in gel consistency test than medium-high ADV lines, and also showed different gelatinization characteristics measured by rapid visco analyzer compared with medium-high ADV lines. The result of genetic analysis indicated that ADV-NILs developed were differed in a pair of major gene controlling ADV, and low ADV was dominant over medium-high ADV.

  • A $F_1$ hybrid out of 143 crosses of rape (Brassica napus) using cytoplasmic-genetic male sterility yielded 5.18 tons per hectare. European and native varieties used, were found out to have 78% and 58% of genes restoring fertility, respectively. The $F_1$ hybrids of three way cross using non-isogenic maintainer showed higher heteroses than those of double cross. To produce F 1 hybrids of three way cross, three stages of crossing operations are needed and seeds produced from about 60 hectares can be covered to 60, 000 hectares.

  • A double-stranded RNA (dsRNA) mycovirus was detected in malformed fruiting bodies of Pleurotus ostreatus strain ASI2792, one of bottle cultivated commercial strains of the edible oyster mushroom. The partial RNA-dependent RNA polymerase (RdRp) gene of the P. ostreatus ASI2792 mycovirus (PoV-ASI2792) was cloned, and a cDNA sequences alignment revealed that the sequence was identical to the RdRp gene of a known PoSV found in the P. ostreatus strain. To investigate the symptoms of PoV-ASI2792 infection by comparing the isogenic virus-free P. ostreatus strains with a virus-infected strain, isogenic virus-cured P. ostreatus strains were obtained by the mycelial fragmentation method for virus curing. The absence of virus was verified with gel electrophoresis after dsRNA-specific virus purification and Northern blot analysis using a partial RdRp cDNA of PoV-ASI2792. The growth rate and mycelial dry weight of virus-infected P. ostreatus strain with PoV-ASI2792 mycovirus were compared to those of three virus-free isogenic strains on 10 different media. The virus-cured strains showed distinctly higher mycelial growth rates and dry weights on all kinds of experimental culture media, with at least a 2.2-fold higher mycelial growth rate on mushroom complete media (MCM) and Hamada media, and a 2.7-fold higher mycelial dry weight on MCM and yeast-malt-glucose agar media than those of the virus-infected strain. These results suggest that the infection of PoV mycovirus has a deleterious effect on the vegetative growth of P. ostreatus.

  • With isogenic waxy and non-waxy rice cultivars, growth responce as affected by leaf-removal treatment were examined in various fertilizer application. Vertical dry matter of culm was significantly decreased from ground upto 10cm. Culm dry matter of waxy rice was more steadily decreased than that of non-waxy rice. Starch content of culm in non-waxy rice was higher than waxy rice, and that in waxy and non-waxy rice were the lowest in three leaf-removal but no difference in single leaf-removal. Flag leaf in waxy rice and 2 nd leaf in non-waxy rice were dominantly affected yield by leaf-removal. The 1000-grain weight and rippend grain ratio of non-waxy rice were more higher than waxy rice and that in waxy and non-waxy rice were the lowest value in three leaf-removal but no difference in single leaf-removal treatments.

  • Two near-isogenic lines with dark and pale green leaves, derived from the F$\_$7/ generation of wxl26 cross were compared on photosynthetic capacity, growth parameters, grain yield and yield-related characters. Dark green-leaved lines contained much greater content of chlorophyll a and b than pale green-leaved ones, but chlorophyll a to b ratio showed no difference between them. The photosynthetic nte per unit leaf area was higher in dark green-leaved lines than in pale green-leaved ones in the flag leaves at heading stage, but that per unit chlorophyll content showed reversed result. The crop growth rate from transplanting to heading was consistantly higher in the dark green-leaved lines, resulting from their greater net assimilation rate. Dark green-leaved lines produced greater number of panicles and spikelets per hill, out yielding pale green-leaved lines, but ripened grain ratio and 1000-grain weight showed no differences between those lines.
