General Pharmacology of $^{13}C$-Urea Powder Preparation in ${Helikat}^{TM}$

  • The pre-mixed $_{13}C$-urea powder preparation in ${Helikit}_{TM}$ for test of Helicobacter pylori was evaluated for pharmacological properties. The oral doses of the preparation used in mice were 30-fold as compared to human doses. The results obtained in the present study demonstrate that spontaneous movement, hexobarbital-induced hypnosis, rotarod performance, body temperature, acetic acid-induced writhing syndrome, chemical and electroshock convulsion, pupil size and intestinal propulsion had not been affected at the oral doses of 230, 700 and 2100 mg/kg in mice. The blood pressure was slightly elevated as given intravenously in rats at a dose of 5 mg/kg of the preparation, but respiration was not influenced at the dose. In isolated guinea pig ileum and rat fundus preparation, the preparation at a concentration of $1{\times}10^{4}$ g/ml neither caused any direct effect nor inhibited the contraction produced by acetylcholine, histamine or 5-hydroxytryptamine. These results reported here provide evidence that pre-mixed $^{13}C$ 13/C-urea powder preparation is free of general pharmacological properties performed in oral administration.

지렁이 모사 로봇의 모델링 및 실험 검증 (Modeling and Experimental Validation of Earthworm-like Robot)

  • In recent years, capsule endoscope is highlighted for the patient's convenience and the possibility of the application in the small intestine. However, the capsule endoscope has some limitations to get the image of the digestive organ because its movement only depends on the peristaltic motion. In order to solve these problems, locomotion of capsule endoscope is necessary. In this paper, we analyze the locomotive mechanism of earthworm-like robot proposed as locomotive device of capsule endoscope and derive the condition which can Judge the possibility of its mobility using theoretical analysis. Based on a biomechanical modeling and simulation, the critical stroke, that is minimum stroke of the earthworm-like robot to perform motion inside small intestine, is obtained. Also, this derived critical stroke can be validated by the moving test of fabricated earthworm-like robot. Consequently, it is expected that this study can supply useful information to design of earthworm-like robot for mobility of capsule endoscope.

흰쥐에 있어서 펙틴이 식이의 상부소화관내 이동 속도에 미치는 영향 (The Effect of Dietary Pectin on the Upper Gastrointestinal Transit Rate in Rats)

  • 식이 중의 펙틴이 식이의 상부소화관내 이동 속도에 미치는 영향을 조사하였다. 흰쥐에게 무섬유식이 또는 10% 펙틴식이를 공급하면서 식이의 이동속도 표시물질인 51-Cr를 투여하였다. 51-Cr 투여후 20분부터 6시간까지 정해진 시간 간격을 두고 동물을 희생시켜, 51-Cr이 소화관에서 이동하는 속도를 측정하였다. 51-Cr를 섭취한 뒤 3시간까지 펙틴은 51-Cr이 위에서 소장으로 이동해 나가는 속도를 유의성있게 증가시켰다. 51-Cr을 섭취한 후 3시간과 4시간 사이에 펙틴은 51-Cr이 소장을 통과하여 맹장으로 이동해 나가는 속도를 증가시켰다. 따라서 펙틴이 포도당 부하후 또는 식이섭취후 혈당치와 인슐린치의 급격한 증가를 완화시키는 효과가 펙틴이 상부소화관내 이동속도를 지연시키기 때문이라는 가설은 정확하지 않은 것으로 보인다.

Laxative effect of peanut sprout extract

  • Certain phenolic compounds are known to exhibit laxative properties. Seed sprouts, such as those of peanut, are known to promote de novo biosynthesis of phenolic compounds. This study was conducted to examine the potential laxative properties of 80% (v/v) ethanolic extract of peanut sprout (PSE), which contains a high concentration of phenolic compounds such as resveratrol. For this, SD rats were orally administered PSE while a control group was incubated with saline. Laxative effects were examined in both groups of rats. Constipation induced by loperamide in SD rats was improved by administration of PSE. Constipated rats showed increased intestinal movement of $BaSO_4$ upon administration of PSE compared to the control, and the groups administered 100 or 1,000 mg PSE/kg bw were not significantly different in transit time of the indicator. However, colon length was not statistically different among the experimental groups, although it was longer in the group incubated with 1 g PSE/kg bw compared to other groups. Further, there was no significant difference in stool number among the experimental groups. Taken together, these findings show that PSE has a laxative effect in a rat model of loperamide-induced constipation.

Longevity and Stress Resistant Property of 6-Gingerol from Zingiber officinale Roscoe in Caenorhabditis elegans

  • In order to discover lifespan-extending compounds made from natural resources, activity-guided fractionation of Zingiber officinale Roscoe (Zingiberaceae) ethanol extract was performed using the Caenorhabditis elegans (C. elegans) model system. The compound 6-gingerol was isolated from the most active ethyl acetate soluble fraction, and showed potent longevity-promoting activity. It also elevated the survival rate of worms against stressful environment including thermal, osmotic, and oxidative conditions. Additionally, 6-gingerol elevated the antioxidant enzyme activities of C. elegans, and showed a dose-depend reduction of intracellular reactive oxygen species (ROS) accumulation in worms. Further studies demonstrated that the increased stress tolerance of 6-gingerol-mediated worms could result from the promotion of stress resistance proteins such as heat shock protein (HSP-16.2) and superoxide dismutase (SOD-3). The lipofuscin levels in 6-gingerol treated intestinal worms were decreased in comparison to the control group. No significant 6-gingerol-related changes, including growth, food intake, reproduction, and movement were noted. These results suggest that 6-gingerol exerted longevity-promoting activities independently of these factors and could extend the human lifespan.

Detecting of Periodic Fasciculations of Avian Muscles Using Magnetic and Other Multimedia Devices

  • In the past, there was a theory that influenza wasn't transmitted directly from birds but was infected to humans via swains. Recently, molecular level research has progressed, and it was confirmed that the avian influenza virus can directly infected to human lung and intestinal epithelial cells. Three pandemicsin the past 100 years were also infected to humans directly from birds. In view of such scientific background, we are developing a method for screening sick birds by monitoring the physiological characteristics of birds in a contactless manner with sensors. Here, the movement of respiratory muscles and abdominal muscles under autonomic innervation was monitored using a magnet and Hall sensor sewn on the thoracic wall, and other multimedia devices. This paper presents and discusses the results of experiments involving continuous periodic noise discovered during flight experiments with a data logger mounted on a Japanese pheasant from 2012 to 2015. A brief summary is given as the below: 1. Magnet and Hall sensor sewn to the left and right chest walls, bipolar electrocardiograms between the thoracic walls, posterior thoracic air sac pressure, angular velocity sensors sewn on the back and hips, and optical reflection of LEDs (blue and green) from the skin of the hips allow observation of periodic vibrations(fasciculations) in the waves. No such analysis has been reported before. 2. These fasciculations are presumed to be derived from muscle to maintain and control air sac pressure. 3. Since each muscle fiber is spatially Gaussian distributed from the sympathetic nerve, the envelope is assumed to plot a Gaussian curve. 4. Since avian trunk muscles contract periodically at all time, we assume that the sympathetic nerve dominates in their control. 5. The technique of sewing a magnet to the thoracic wall and measuring the strength of the magnetic field with a Hall sensor can be applied to screen for early stage of avian influenza, with a sensor attached to the chicken enclosure.

루푸스 동물 모델에서 인삼부자탕(人蔘附子湯)이 미치는 영향 (Study of Insam-Buja-Tang (IBT) on MRL/MpJ-Faslpr lupus-prone mice)

  • Systemic Lupus Erythematosus(SLE) is an autoimmune disease invading the skin, joint, kidney, intestinal membrane, neurosystem and other organs. SLE is an autoimmune disease characterized by immune dysregulation resulting in the production of antinuclear antibodies(ANA), generation of circulating immune complexes, and activation of the complement system. In Korean medicine, lupus can be classified as acute arthritis, reddish butterfly erythema, asthenic disease, edema and so on. The cause and procedure of the diseases are flourishing noxious heat, excessive fire due to deficiency of yin, blood stasis due to stagnation of qi, internal movement of the liver-wind, congenital deficiency, exhausted vital-qi, which are treated by clearing away heat and cooling the blood, nourshing yin and extinguishing fire, treating flatulence and activating blood circulation, nourishing the blood to expel wind, invigorating the liver and kidney, invigorating qi and replenishing the blood. To experimentally examine the influence of Insam-Buja-Tang (Ginseng & Aconiti Extract, IBT) on the outbreak and development of lupus, lupus induce MRL/MpJ-Faslpr lupus-prone mice model was used. As IBT was orally administrated to a lupus model mouse, various tests such as the weight, urine protein, renal function, Lymph cell test of the spleen, Cytokine expression, histopathological analysis of kideny were performed to see the influence on the kidney and whether it work effectively on the immune function. The main purpose of this study is to evaluate the effect of IBT on MRL/MpJ-Faslpr lupus-prone mice model. The effect of IBT on MRL/MpJ-Faslpr lupus-prone mice that can have autoimmune disease similar to SLE in human was evaluated after IBT per oral in the present study.

비침습적 초음파 조직 탄성도 측정 시스템 개발 (Development of a Non-invasive Ultrasonic Measurement System for tissue elasticity)

  • Diseases caused by indurate tissues of the internal organs are liver cirrhosis and abdominal sclerosis. The cause of chronic gastro-intestinal disease is a digestive system disorder and a defecation disorder. They impede peristaltic movement and digestive system with the symptom that indurate tissues. The purpose of the present study was to determine the disease grade quantitatively by measuring an indurated standard of tissues and organs. For the measurement of elasticity, we designed the system that measure the displacement of the substance and approved pressure using ultrasound transducer. For verification of developed system, we compared elasticity as results of experiment between the developed system and public elasticity measurement machine at individual plastic phantoms made by plastic hardener and softener. Elasticity of the plastic phantoms is averagely 0.007MPa lower measured by developed system than Micro-indenter, and less than 10% errors. Comparing with economical value and accuracy between developed system and Micro-indenter, the system is significant of measurement for tissue elasticity. Thus, it is possible to measure a elasticity at human tissue and organ. A chronic gastro-disease as well as grade could be decided objective validity using this system.

  • Kefir exhibits antimicrobial activity in vitro against gram-positive and gram negative bacteria, as well as some fungi. The ability of LAB to inhibit the growth of closely related bacteria is well known. This inhibition of pathogenic and spoilage microbes may be due to the production of organic acids, hydrogen peroxide, acetaldehyde, diacetyl, carbon dioxide, or bacteriocins. Lactobacilli are the major contributors to acid production and, hence, a determining factor in the flavor development in kefir. Lactic acid, proteolytic activity, and acetaldehyde are the essential flavor compounds in kefir. Both acid and bacteriocins contribute to the antimicrobial activity of kefir and kefir grains. Kefir is rich in proteins, calcium, vitamin $B_{12}$, niacin, and folic acid. Many studies have investigated the benefits of consuming kefir and have shown that it is a natural probiotic, which when consumed regularly, can help relieve intestinal disorders, promote bowel movement, reduce flatulence, and improve the overall health of the digestive system. Tibetan kefir, which is different from traditional kefir, is produced in China. It has been reported to exhibit antimicrobial activity that is nearly identical to that of traditional kefir. Kefir production is considered a rapidly growing food industry in China.

Virechana karma (therapeutic purgation) in the restoration of gut microbiota concerning Amavata (RA): A scientific exposition

  • Background: Amavata is a disease that occurs as a result of the error of metabolism. Poor dietary habits and faulty Dincharya (daily regimen) and ritucharya (seasonal regimen) leading to deranged metabolism and Agni (metabolic fire) which results in the formation of Ama(undigested product of metabolism). When Amaconceals with Vata(subtle energy associated with movement) and circulates in the body under the influence of Vyana Vayu (omnipresent air)it clogs the srotasas (microchannels) and initiates the inflammatory cascade. Amavata is commonly correlated with rheumatoid arthritis (RA) while other forms of auto-immune disorders can also be included in Amavata.Dysbiosis of the gut microbiota (GM) has been connected to the onset of diverse autoimmune diseases. In this study, it was hypothesized that Panchakarma (bio-purificatory methods) based intervention such as Virechana Karma (therapeutic purgation) may influence microbiota. Materials and Methods: Various Ayurvedic literature were reviewed for the etiopathogenesis of Amavata. Different databases were searched with research papers related to Gut Dysbiosis and autoimmunity and management of RA. A connecting link between Intestinal Dysbiosis with the autoimmune mechanisms was established and it was also found that the bowel cleansing introduced a change to the GM. Conclusion: It was concluded that Virechana karma is effective in gut flora Dysbiosis. This study aims to correlate the ancient Ayurvedic principles related to Agni Bala(metabolic energy) and biopurificatory treatment modalities like Virechana karma (therapeutic purgation)with the modern concept of gut microbiota and its role in the pathogenesis of various autoimmune disorders such as rheumatoid arthritis. The article creates an understanding about principles of Ayurveda and its rationality in today's scientific world and thereby opens newer vistas of research in therapeutics from Ayurveda, which may be helpful in the management of various immune-mediated Diseases through Ayurveda.
