A Comparative Study of Hataedock versus Probiotics on Immunomodulating Effect in Intestinal Mucosa (황련감초 하태독법과 프로바이오틱스의 대장점막 내 면역조절 효과 비교연구)

  • Objectives Hataedock is a treatment that dispels toxic heat and meconium which has been accumulated to the fetus from a pregnant mother via orally administering herbal extracts to a newborn baby. This study was conducted to compare the efficacy of Hataedock, with using the extract of Coptis japonica & Glycyrrhiza uralensis, to the early administration of probiotics for immunomodulation in the intestinal mucosa. Methods NC/Nga mice were divided into three groups; Control group (no treatment), CGT group (3-week-old mice given the extract of Coptis japonica & Glycyrrhiza uralensis), and MBT group (3-week-old mice given a Bifidobacterium). After 2 weeks, the intestinal mucosa tissues of each group of mice were observed. Immunohistochemical staining for IL-4, IL-13, CD40, $Fc{\varepsilon}RI$, $p-I{\kappa}B$, EGF, and VEGF in the intestinal mucosa was performed. Results CGT group showed 65% decrease in IL-4, 67% decrease in IL-13, 58% decrease in CD40, 72% decrease in $Fc{\varepsilon}RI$, 76% decrease in $p-I{\kappa}B$, 41% increase in EGF and 100% increase in VEGF compared to the control group. MBT group also showed 50% decrease in IL-4, 63% decrease in IL-13, 33% decrease in CD40, 53% decrease in $Fc{\varepsilon}RI$, 46% decrease in $p-I{\kappa}B$, 23% increase in EGF and 151% increase in VEGF compared to the control group. Conclusions These results suggest that both Hataedock, with using the extract of Coptis japonica & Glycyrrhiza uralensis, and early administration of probiotics were effective in regulating Th2, relieving inflammation and developing intestinal mucosal tissues. Hataedock with extract of Coptis japonica & Glycyrrhiza uralensis may be more effective for immunomodulation in intestinal mucosa than probiotics.

Gene expression profiling after ochratoxin A treatment in small intestinal epithelial cells from pigs

  • Ochratoxin A (OTA) is a well-known mycotoxin that causes disease through the ingestion of contaminated food or feed, for example, in the porcine industry. The intestinal epithelium acts as the first barrier against food contamination. We conducted a study on the exposure of the porcine intestinal epithelium to OTA. We used the intestinal porcine epithelial cell line IPEC-J2 as an in vitro model to evaluate the altered molecular mechanisms following OTA exposure. Gene expression profiling revealed that OTA upregulated 782 genes and downregulated 896, totalling 1678 differentially expressed genes. Furthermore, immunofluorescence, quantitative real-time polymerase chain reaction, and western blotting confirmed that OTA damages the tight junction protein ZO-1. Moreover, OTA activated the expression of inflammatory genes (IL-6, IL-8, IL-10, NF-kB, TLR4, and TNF-α). In summary, this study confirmed that OTA alters various molecular mechanisms and has several adverse effects on IPEC-J2 cells.

Growth of Intestinal Bacteria and Intestinal Inflammation of Sprout Extract from Common Buckwheat and Tartary Buckwheat (일반메밀과 쓴메밀의 새싹 추출물의 장내 유익균 증식 및 염증조절 효능 평가)

  • We aimed to assess the potential growth-promoting effects of buckwheat sprout on intestinal bacteria and their anti-inflammation effects in a cellular model of intestinal inflammation. The growth of Bifidobacterium longum ssp. infantis BT1 was enhanced with the addition of the sprout extract of tartary buckwheat. Further, in the inflammatory model cells cultured with Raw 264.7 cells were treated with buckwheat sprout including each 10 probiotics before the addition of lipopolysaccharide (LPS) to induce inflammation in Raw 264.7 cells. Buckwheat sprout in both Bifidobacterium longum ssp. infantis BT1 and Lacticaseibacillus paracasei LPC5 significantly reduced the production of NO and PGE2. The above results indicate that buckwheat sprout extract which contains with various physiologically active substances such as rutin, quercetin, and choline is effective in suppressing NO and PGE2 production, which are inflammation-related indicators. The present study suggests that buckwheat sprout could induce positive effects on the intestinal beneficial bacteria and in anti-inflammation.

Demethoxycurcumin from Curcuma longa Rhizome Suppresses iNOS Induction in an in vitro Inflamed Human Intestinal Mucosa Model

  • Background: It is known that inducible nitric oxide synthase (iNOS)/nitric oxide (NO) plays an integral role during intestinal inflammation, an important factor for colon cancer development. Natural compounds from Curcuma longa L. (Zingiberaceae) have long been a potential source of bioactive materials with various beneficial biological functions. Among them, a major active curcuminoid, demethoxycurcumin (DMC) has been shown to possess anti-inflammatory properties in lipopolysaccharide (LPS)-activated macrophages or microglia cells. However, the role of DMC on iNOS expression and NO production in an in vitro inflamed human intestinal mucosa model has not yet been elucidated. This study concerned inhibitory effects on iNOS expression and NO production of DMC in inflamed human intestinal Caco-2 cells. An in vitro model was generated and inhibitory effects on NO production of DMC at 65 ${\mu}M$ for 24-96 h were assessed by monitoring nitrite levels. Expression of iNOS mRNA and protein was also investigated. DMC significantly decreased NO secretion by 35-41% in our inflamed cell model. Decrease in NO production by DMC was concomitant with down-regulation of iNOS at mRNA and protein levels compared to proinflammatory cytokine cocktail and LPS-treated controls. Mechanism of action of DMC may be partly due to its potent inhibition of the iNOS pathway. Our findings suggest that DMC may have potential as a therapeutic agent against inflammation-related diseases, especially in the gut.

  • Inflammatory bowel diseases(IBD), including Crohn's disease and ulcerative colitis, are chronic inflammatory states of the intestinal tract. While the exact mechanisms inducing chronic inflammation are still unclear, it is hypothesized that the inflammation is caused in part by an inappropriate immune response to the intestinal microflora. Although inflammatory diseases are not directly linked to patient survival, symptoms of these diseases significantly decrease quality of life. The incidence rate is higher in western people than eastern people, but the incidence rate of IBD in eastern people, including Korean, is increasing. Recently, it has been reported that IL-17 is an important factor that appears to be involved in IBD induction and progression. This report reviews many recent papers reporting the relationship between IBD and IL-17, which may provide an understanding leading to new means of prevention and treatment for IBD.

  • Many modern people are exposed to chronic inflammatory diseases, such as inflammatory bowel disease (IBD), atopic dermatitis and immune disorder. Among those chronic diseases, the incidence ratio of IBD has been increased. IBD, including Crohn's disease and ulcerative colitis (UC), is known to cause abnormal inflammation in intestinal tissue. UC is accompanied by abdominal pain, bloody stool and diarrhea. Many therapeutic agents, such as sulfasalazine, corticosteroids, immunosuppressive agents, have been used for treating UC. However, those agents have side-effects and temporary effects on UC. The aim of this study was to investigate the effect of herbal medicine on UC and relationship between UC and intestinal bacteria according characteristic of herbal medicine. To determine the effect of herbal medicine on UC, various herbal medicine were chosen within oriental medicine category such as cheongyeol and onyeol medicine. In this study, we found carthami fructus, included in cheongyeol medicine, had stronger effect than onyeol medicine. Also, we determined influence of carthami fructus against lactic acid bacteria. Catthami fructus and lingon berry extracts affected the composition of mice intestinal bacteria in mice fecal. The symptoms of UC could be regulate by using herbal medicine, according to characteristic of herbal medicine. Also, herbal medicine might be change body condition to healthy by controlling intestinal bacteria composition. Herbal medicine characteristic could be a therapeutic agent by revealing relationship between intestinal bacteria and UC.

  • A partial obstruction of small intestine was diagnosed by physical and radiographic examination in a dog which had signs of anorexia, vomiting, diarrhea and dehydration The dog was treated by surgical method. String-like foreign body was located from stomach to upper jejunum. Diffuse laceration, inflammation and perforation caused by foreign body were observed in intestine. Intestinal resection was carried out to remove the foreign body from upper duodenum to upper jejunum. The dog was convalesced successfully after operation.

Carbuncle is one of Venus festering nature file that happen in skin and muscle because blood does not circulate with flag and part's flare, calorification, pain, puffiness are characteristic. Carbuncle is agreed with concept of inflammation of modern medicine. When treat carbuncle, without using surgery medical treatment, used together internal medicine surgery medical treatment. Intestines carbuncle(腸癰) is come in inflammation in abdominal cavity as kind of inflammation, partiality peritoneum festering disease round present cecum as one of inside carbuncle. I think, when treat intestines carbuncle, in case cold and moisture become stasis and heat is less, Uiibujapaedoksan(薏苡附子敗毒散) can be used. Independently of festering, Daehwangmokdantang(大黃收丹湯) can be used in case heat and extravasated blood become stasis. Saliva ulcer on the vulva is comes in impetigo In Case of young child, and is come in Venus eczema in case is general. Prognosis of saliva ulcer on the vulva widespread thing can treat by arm, leg at Lips region, but it does not treat to grow at Lips region from limb, and treatment uses Hwangryunbun(黃連粉)

Parasitic Helminth Cystatin Inhibits DSS-Induced Intestinal Inflammation Via IL-$10^+F4/80^+$ Macrophage Recruitment

  • Many immune down-regulatory molecules have been isolated from parasites, including cystatin (cystain protease inhibitor). In a previous study, we isolated and characterized Type I cystatin (CsStefin-1) of the liver fluke, Clonorchis sinensis. To investigate whether the CsStefin-1 might be a new host immune modulator, we induced intestinal inflammation in mice by dextran sodium sulfate (DSS) and treated them with recombinant CsStefin-1 (rCsStefin-1). The disease activity index (DAI) increased in DSS only-treated mice. In contrast, the DAI value was significantly reduced in rCsStefin-1-treated mice than DSS only-treated mice. In addition, the colon length of DSS only-treated mice was shorter than that of rCsStefin-1 treated mice. The secretion levels of IFN-${\gamma}$ and TNF-${\alpha}$ in the spleen and mesenteric lymph nodes (MLNs) were significantly increased by DSS treatment, but the level of TNF-${\alpha}$ in MLNs was significantly decreased by rCsStefin-1 treatment. IL-10 production in both spleen and MLNs was significantly increased, and IL-$10^+F4/80^+$ macrophage cells were significantly increased in the spleen and MLNs of rCsStefin-1 treated mice after DSS treatment. In conclusion, rCsStefin-1 could reduce the intestinal inflammation occurring after DSS treatment, these effects might be related with recruitment of IL-10 secreting macrophages.
