Bacillus licheniformis AJ 균주제제의 설사원인 미생물의 성장 억제효과 (Inhibitory Activity of Bacillus licheniformis AJ on the Growth of Diarrheal Pathogens)

  • The injibitory effect of Bacillus licheniformis AJ isolated from genitourinary normal flora as a new probiotics on the growth of diarrheal pathogens was studied. This B. licheniformis AJ inhibited the growth of E.coli O-157. Salmonella typhi and Shigella sonnei as well as the infectivity of rotavirus. However, it did not inhibit the growth of Helicobacter pyloriand human intestinal bacteria although it inhibited the harmful enzyme activity of human intestinal bacteria. B. licheniformis AJ seems to excret heat-lable growth-inhibitory protein, bacteriocin, into the media. These results suggest that B. lichenoformis AJ could be used as a new type of probiotics.

  • The fecal tryptophanase activities were $0.267{\pm}0.10$ for rats and $0.185{\pm}0.01{\;}{\mu}mole/min/g$ wet feces for humans. The activities of indole pyruvate degradation to indole, indole pyruvate lyase, of these feces were $0.051{\pm}0.02$ and $0.046{\pm}0.01{\;}{\mu}mole/min/g$ wet feces, respectively. The optimal pH values of tryptophanase and indole pyruvate lyase were 5.5-7.5 and 5.5-6.5, respectively. When the intestinal flora or E. coli HGU-3 was cultured in GAM broth having six different pH values (5 to 10), the activities of tryptophanase and indole pyruvate IYilse in the medium adjusted at pH 6 were dramatically induced by elevating the pH to 9. However, when intestinal microflora were inoculated in the medium containing lactulose, the pro¬ductions of these enzymes were dramatically inhibited and the pH of the medium was lower than that of the control.

Synbiotics (mixture of probiotics and prebiotics) ameliorates DSS-induced ulcerative colitis in vivo.

  • Ulcerative colitis (UC) is one of inflammatory bowel disease (IBD), characterized by chronic inflammatory response and dysregulation of immune function. The severity of US has been influenced by environmental factors and food habit. The immune modulatory, anti-inflammatory and steroidal medicine have been used for the treatment of UC. However, long-term administration of those medicine is accompanied with side-effect. So, it is necessary to develop the non side-effect medicine using natural product. Prebiotics influences intestinal condition and food consumption. The heredity, immunity and environmental condition are related with occurrence of UC. In recent study, UC patients had lower level of prebiotics such as Lactobacillus and Bifidobacterium compared with healthy people. Also, previous study announced that imbalance of enteric flora aggravates the severity of UC. The effectiveness of probiotics might affect colon ability and viable bacteria also could promote the proliferation of beneficial intestinal bacteria. Prebiotics, such as herbal medicine, could lead to balance of intestinal bacteria or increase beneficial bacteria. So, proper choice of herbal medicine could control the intestinal condition. This study aimed to investigate the effect of mixture of probiotics and prebiotics (synbiotics) on dextran sulfate sodium (DSS)-induced UC in vivo. The synbiotics consist of Lactobacillus buchneri, Polymnia sonchifolia and Glycine max Merr. in this study. To evaluate the effect of synbiotics, 3% DSS was administered in BALB/c mice and synbiotics was daily administered for experimental days. The administration of synbiotics regulated colon length shortening, body weight change and disease activity index effectively. Also, extract of synbiotics upregulated survival ability of Lactobacillus buchneri in gut condition. These results suggest that mixture of probiotics and prebiotics, called as synbiotics, could influence intestinal condition also regulate the colon disease. Synbiotics might be a therapeutic agent for treatment of UC.

  • This Study was conducted to investigate Muscle Test of Point Selection through CRA(Contact Reflex Analysis) Muscel Diagnosis. So this study used to compare and analyze the effects of Muscel(Deltoid Muscel of Posterior) RMS(Root Mean Squared) and MEF(Median Edge Frequency) Among Groups that do not respond to questionnaire, Tonguibogam Naegyeongpyeon Small Intestine Group, Small Intestine MeridianPathway Primary Symptom and Secondary Symptom Group and Kwanwon(CV4) meridian Principal action Group. The questionnaire is composed of 23 items. The questionnaire was intended to elicit information on assorting into 4 groups. After Survey, Subject had to Muscle test subjects. Muscle experimental methods are as follows: Holding the shoulder without contacting Kwanwon. Holding the shoulder contacting Kwanwon. The first iteration. Group 1,2,3 were increased sEMG RMS compared with First experiment and Second experiment. Group 4(Including Uterus and Intestinal Flora Problem) were decreased sEMG RMS compared with First experiment and Second experiment. This test means that it is similar to diagnosis CRA and Small intestine channel of hand taiyang muscle, not Small Intestine MeridianPathway. It is suggested that subjects with a Small Intestine problem(Uterus and Intestinal Flora Problem) shows the results of decreasing posterior Deltoid Muscular strength. It means that CRA muscle diagnosis is associated with Alarm points diagnosis. But it doesn't consider influence of fat on the surface EMG.

정상 신생아의 대변에서 생후 1주일 동안 장내세균총의 변화 (The Changes of Intestinal Normal Flora in Neonates for Seven Days Postnatally)

  • 목 적: 자궁 내에서 태아의 소화기는 무균상태에 있다가 출생 후 신생아기에 점차 장내세균의 집락화가 이루어진다. 출생 초기에 처음으로 나타나는 세균은 Staphylococcus, Enterobacteriaceae, Streptococcus가 있으며, 이후 Bifidobacterium spp.와 같은 혐기성 세균이 나타난다. 이에 저자들은 정상 신생아의 대변에서 생후 1주일 동안 장내세균총의 분포와 변화를 규명하고자 하였다. 방 법: 한림의대 강동성심병원에서 출생한 정상 신생아 15명(자연분만 3명, 제왕절개 12명)(모유수유 1명, 분유수유 8명, 혼합수유 6명)을 대상으로 하였다. 출생 첫 대변과 1일, 3일, 7일째의 대변을 채취하였다. 대변 배양 배지와 조건를 달리하여 호기성균(blood agar plate), E. coli (trypton bile X-glucuronide 배지), 그람 양성 혐기성균(phenylethyl alcohol agar), lactobacillus (MRS 배지), Bifidobacterium spp. (bifidobacterium selective agar), Clostridium difficile (cefoxitincycloserine-fructose agar)을 검출하여 배양된 균 집락의 수를 측정하여 기록하였다. 결 과: 출생 첫 대변에서는 균이 거의 검출되지 않았다. 호기성균, E. coli, 그람 양성 혐기성균은 출생 시부터 생후 7일까지 유의하게 증가하였다(p<0.05). Lactobacilli는 한명의 신생아에서 생후 7일에 검출되었다. Bifidobacterium spp.은 2명에서 검출되었다. Clostridium difficile은 생후 7일까지 검출되지 않았다. 분만 방법과 수유 방법에 따른 균 집락의 차이는 없었다. 결 론: 정상 신생아에서 생후 1주일 동안 장내세균총의 다양한 변화를 관찰할 수 있었다. 앞으로 생후 1주일 이후의 장내세균총의 변화와 더 많은 환아에서의 연구를 진행한다면 생균제 사용에 도움이 되리라 생각된다.

Effects of dietary supplementation with Pediococcus acidilactici ZPA017 on reproductive performance, fecal microbial flora and serum indices in sows during late gestation and lactation

  • Objective: This study was conducted to determine the effects of dietary supplementation with Pediococcus acidilactici (P. acidilactici) ZPA017 as a probiotic on reproductive performance, fecal microbial flora and serum indices in sows during late gestation and lactation. Methods: A total of 94 sows (Large White×Yorkshire, average 4.50 parities) were randomly allotted to two dietary treatments: control diet and the diet supplemented with P. acidilactici ZPA017 (2.40×109 colony-forming unit/kg of diets). The study started at d 90 of gestation and conducted until d 28 of lactation. Results: Compared to sows fed the control diet, supplementation of P. acidilactici ZPA017 increased the number of weaning piglets, weaning weight of litter and piglets, survival rate of piglets at weaning (p<0.05), and decreased diarrhea rate of piglets in lactation (p<0.05). Dietary P. acidilactici ZPA017 increased fecal Lactobacillus populations (p = 0.030) and reduced fecal Escherichia coli and Staphylococcus aureus populations (p<0.05) of sows at weaning. Moreover, the supplementation of P. acidilactici ZPA017 increased serum concentrations of immunoglobulin G, immunoglobulin A and total protein (p<0.05), while decreased serum haptoglobin concentration and alanine aminotransferase activity (p<0.05) of sows at weaning. Conclusion: Administration of P. acidilactici ZPA017 in diets during late gestation and lactation had positive effects on the reproductive performance, intestinal microflora balance and immunity of sows.

  • The lack of an effective treatment for Alzheimer's disease (AD) stems primarily from incomplete understanding of AD's causes. A rapidly growing number of scientific reports highlight important roles played by peripheral infections and intestinal bacterial flora in pathological and physiological functions involving the microbiome-intestine-brain axis. The microbiome controls basic aspects of the central nervous system (CNS), immunity, and behavior, in health and disease. Changes in the density and composition of the microbiome have been linked to disorders of the immune, endocrine, and nervous systems, including mood changes, depression, increased susceptibility to stressors, and autistic behaviors. There is no doubt that in patients with AD, restoration of the intestinal microbiome to a composition reminiscent of that found in healthy adult humans will significantly slow the progression of neurodegeneration, by ameliorating inflammatory reactions and/or amyloidogenesis. In the near future, better understanding of bidirectional communication between the brain and microbiota will allow the development of functional diets using specific probiotic bacteria.

  • Probiotics are live microorganisms that can be consumed by humans in amounts sufficient to offer health-promoting effects. Owing to their various biological functions, probiotics are widely used in biological engineering, industry and agriculture, food safety, and the life and health fields. Lactobacillus acidophilus (L. acidophilus), an important human intestinal probiotic, was originally isolated from the human gastrointestinal tract and its functions have been widely studied ever since it was named in 1900. L. acidophilus has been found to play important roles in many aspects of human health. Due to its good resistance against acid and bile salts, it has broad application prospects in functional, edible probiotic preparations. In this review, we explore the basic characteristics and biological functions of L. acidophilus based on the research progress made thus far worldwide. Various problems to be solved regarding the applications of probiotic products and their future development are also discussed.

Effect of microencapsulation of egg yolk immunoglobulin Y by sodium alginate/chitosan/sodium alginate on the growth performance, serum parameters, and intestinal health of broiler chickens

  • Objective: Egg yolk immunoglobulin (IgY) is an antibiotic alternative to prevent and fight intestinal pathogenic infections. This study aimed to investigate the effects of sodium alginate/chitosan/sodium alginate IgY microcapsules on the growth performance, serum parameters, and intestinal health of broiler chickens. Methods: One-day-old broilers (Ross 308) were divided into five treatments, each with 10 replicates of five chickens. The dietary treatments were maintained for 28 days and consisted of a basal diet (NC), basal diet + 500 mg chlortetracycline/kg diet (CH), basal diet + 50 mg non-microencapsulated IgY/kg diet (NM), basal diet + 600 mg low levels microencapsulated IgY/kg diet (LM), and basal diet + 700 mg high levels microencapsulated IgY/kg diet (HM). Results: Throughout the 28-day trial period, the NM, LM, HM, and CH groups increased average daily gain compared with the NC group (p<0.05), and the HM group reduced feed conversion ratio compared with the CH group (p<0.05). The LM and HM groups increased relative organ weights of thymus and spleen compared with the CH and NM groups (p<0.05). The HM group improved the duodenal, jejunal and ileum villi height (VH) and villus height to crypt depth ratio (VH:CD) compared with the CH and NM groups (p<0.05). Compared with the CH group, the HM group increased serum immunoglobulin (IgA), immunoglobulin G (IgG), superoxide dismutase, total antioxidant capacity, and glutathione peroxidase levels (p<0.05), and decreased serum malondialdehyde levels (p<0.05). Compared with the NC group, the NM, LM, HM, and CH groups reduced colonic Escherichia coli and Salmonella levels (p<0.05). and the HM group promoted the levels of lactic acid bacteria and bifidobacteria compared with the CH group (p<0.05). Conclusion: Microencapsulation could be considered as a way to improve the efficiency of IgY. The 700 mg high levels microencapsulated IgY/kg diet could potentially be used as an alternative to antibiotics to improve the immune performance and intestinal health, leading to better performance of broiler chickens.

Effect of Panax ginseng Extract on Growth Responses of Human Intestinal Bacteria and Bacterial Metabolism

  • The growth responses of a variety of human Intestinal bacteria to extracts of Pun(1.vKy'n.ieny and five other oriental medicinal Araliaceae were evaluated in vitro and in vivo. The extracts enhanced the growth of Bifidobncterilim breve and B. longum in Media with or without carbon sources, suggesting the bifid factor (5) might be involved in the phenomenon. This effect was most pronounced with water extract of p. ginseng, the growth of 27 bifidobacteria strains belonging to B. ndolexcentium, H. longlrm, and 1. breve and B. iniuntis being greatly stimurated, whereas seven B. bifidum strains and other bacteria such as clostridia and 5.fcherirhia coli had little or no ability to utilizes it (or growth. Methanol extracts of p, ginseng were found to selectively inhibit growth of various clostridia including C. perfringens and C. Paraputrificum, but this effect was not observed on other bacteria including bifidobacteria. The effect of ginseng extract intake(600 mg/day for two weeks) on the fecal microflora, pH, volatile fatty acids, ammonia, putrefactive products, and -glucuronidase, -glucosidase and nitroreductase activities, and on the blood components (triglyceride, total cholesterol and ammonia) were investigated using seven healthy human volunteers. The total concentration of fecal microflora including Bri'idobucterilim app. during the period of ginseng extract intake was significantly unaffected from the proceeding and sub sequent control periods. However, the frequency of occurrence of subjects having C. perfringens was significantly decreased. The fecal pH value was also significantly decreased, suggesting that the intake might increase the activity of Bifidobacterium spp. Other biochemical properties in faces did not changed significantly. The levels of ammonia and triglycerid in blood were decreased with ginseng extract intake. These results may be an indication of at least one of the pharmacological actions of P ginseng as an adaptogen.
