Objective: The study was to investigate the effects of alanyl-glutamine (Ala-Gln) and glutamine (Gln) supplementation on the intestinal mucosa barrier in piglets. Methods: A total of 180 barrows with initial weight $10.01{\pm}0.03kg$ were randomly allocated to three treatments, and each treatment consisted of three pens and twenty pigs per pen. The piglets of three groups were fed with control diet [0.62% alanine (Ala)], Ala-Gln diet (0.5% Ala-Gln), Gln diet (0.34% Gln and 0.21% Ala), respectively. Results: The results showed that in comparison with control diet, dietary Ala-Gln supplementation increased the height of villi in duodenum and jejunum (p<0.05), Gln supplementation increased the villi height of jejunum (p<0.05), Ala-Gln supplementation up-regulated the mRNA expressions of epidermal growth factor receptor and insulin-like growth factor 1 receptor in jejunal mucosa (p<0.05), raised the mRNA expressions of Claudin-1, Occludin, zonula occludens protein-1 (ZO-1) and the protein levels of Occludin, ZO-1 in jejunal mucosa (p<0.05), Ala-Gln supplementation enlarged the number of goblet cells in duodenal and ileal epithelium (p<0.05), Gln increased the number of goblet cells in duodenal epithelium (p<0.05) and Ala-Gln supplementation improved the concentrations of secretory immunoglobulin A and immunoglobulin G in the jejunal mucosa (p<0.05). Conclusion: These results demonstrated that dietary Ala-Gln supplementation could maintain the integrity of small intestine and promote the functions of intestinal mucosa barriers in piglets.
The relationship between the intestinal histopathology and number and position of intraepithelial lymphocytes (IEL) was observed chronologically in the small intestine of rats experimentally infected with Echinostoma hortense. Sprague-Dawley rats were orally infected with 200 metacercariae obtained from Misgurnus anguillicaudatus. The rats each were sacrificed on the week 1, 2, 4, 6, 8 post-infection (PI) and samples of the intestine in the part of duodenum and jejunum were taken. The samples were stained with Hematoxylin-eosin and Giemsa. The intestinal histopathology was the severest after the week 1 PI and characterized by villous atrophy, crypt hyperplasia and decrease of villus/crypt(v/c) ratio, which continued until the week 8 Pl. The number of IEL dramatically decreased during the week 1 PI, but increased gradually thereafter with a slight decrease on the week 8 PI. In control rats, the great majority of the IEL were located at the basal region of the epithelium. During the early stage of infection, however, we found a considerable proportion of IEL to moved to the intermediate or apical regions of the epithelium. From the above results, it is sugested that the change of IEL number and position during the course of E. hortense infection should be closely related to the progression and recovery of the intestinal histopathology.
The regional distribution and relative frequencies of endocrine cells were studied immunogistochemically (PAP methods) in the alimentary tract of the tree frog, Hyla areorea japonica, using specific antisera against serotonin, somatostatin, bovine Sp-1/chromogranin (BCG), cholecystokinin (CCK)-8, vasoactive intestinal polypeptide (VIP), gastrin, bombesin, secretin and pancreatic polypeptide (PP). Six kinds of endocrine cells were identified in this study, These immunoreactive cells were located in the gastric glands of stomach regions and in the basal portion of the epithelium of the intestinal tract or esophagus with variable frequencies. They were spherical or spindle-shaped. Serotonin-immunoreactive cells were observed in the whole alimentary tract including the esophagus. Somatostatin-immunoreactive cells were also detected throughout the alimentary tract except the rectum. CCK-8-immunoreactive cells were observed from the pylorus to ileum. Vip-immunoreactive cells were restricted to the rectum. Bombesin-immunoreactive cells were restricted to the fundic gastric regions and gastrin-immunoreactive cells were restricted to the pylorus. However, no BCG-, secretin and PP-immunoreactive cells were demonstrated in this study. In conclusion, the regional distribution and relative frequency of the endocrine cells in the alimentary tract of the tree frog were similar to other anuran species, but some differences which may be caused by feeding habits and species specification were also observed.
The digestive system is gaining interest as a major regulator of various functions including immune defense, nutrient accumulation, and regulation of feeding behavior, aside from its conventional function as a digestive organ. The Drosophila midgut epithelium is completely renewed every 1-2 weeks due to differentiation of pluripotent intestinal stem cells in the midgut. Intestinal stem cells constantly divide and differentiate into enterocytes that secrete digestive enzymes and absorb nutrients, or enteroendocrine cells that secrete regulatory peptides. Regulatory peptides have important roles in development and metabolism, but study has mainly focused on expression and functions in the nervous system, and not much is known about the roles in endocrine functions of enteroendocrine cells. We systemically examined the expression of 45 regulatory peptide genes in the Drosophila midgut, and verified that at least 10 genes are expressed in the midgut enteroendocrine cells through RT-PCR, in situ hybridization, antisera, and 25 regulatory peptide-GAL transgenes. The Drosophila midgut is highly compartmentalized, and individual peptides in enteroendocrine cells were observed to express in specific regions of the midgut. We also confirmed that some peptides expressed in the same region of the midgut are expressed in mutually exclusive enteroendocrine cells. These results indicate that the midgut enteroendocrine cells are functionally differentiated into different subgroups. Through this study, we have established a basis to study regulatory peptide functions in enteroendocrine cells as well as the complex organization of enteroendocrine cells in the Drosophila midgut.
The reproductive cycle and the breeding season of the freshwater clam, Anodonta (Sinanodonta) woodiana (Lea) have been investigated by histological examination of the gonadal development under photomicroscopy. The materials were monthly collected from the Nakdong River for one year from September 1979 to August 1980. Sexuality of Anodonta (Sinanodonta) woodiana is dioecious, and the species are ovoviviparous. The gonads are irregularly arranged from the subregion of mid-intestinal gland in visceral cavity to reticular connective tissue of foot. The ovary is composed of a number of small ovarian sacs, The epithelium of ovarian sac has a function of the germinal epithelium. Oogonia actively proliferate along the germinal epithelium of the ovarian sac, in which young oocytes are growing. The testis is composed of a number of seminiferous tubules, and the epithelium of the tubule has function of germinal epithelium, along which spermatogonia actively proliferate. A great number of undifferentiated mesenchymal tissue and eosinophilic granular cells are abundantly distributed between the growing oocytes and spermatocytes in the early development stages. With the further development of the ovary and testis these tissues and cells gradually disappear. Then the undifferentiated mesenchymal tissue and granular cells are considered to be related to the growing of the oocytes and spermatocytes. The gonads had function year-round the individuals which have various developmental stages of gonads appearing all the time. Spawning continued year-round except for the period of high temperature of water, during August and September. The peak spawning seasons appeared twice a year between January and March, and between June and July in 1980. Individuals which have trochophore larvae in the marsupium of the adult appeared year-round except September 1579 and August 1980. The rate of individuals which have glochidia in the marsupium was 72.7 percent in May 1980 which was the highest brooding fate.
This review provides an overview of dietary nucleotides as an alternative to in-feed antibiotics for weaning pigs. Dietary nucleotides are composed of DNA or RNA molecules and are normally contained in protein-rich feed ingredient, brewer's yeast, yeast extract, and milk. Weaning pigs are suffering from several stresses, such as environmental challenges (i.e. crowding, transportation, and feeding). Such stressors can damage the intestinal epithelium and cause an invasion by Escherichia coli, secondary inflammatory responses, and post weaning diarrhea. To overcome weaning disorder, people often use antibiotics which reduce symptoms and boost growth performance. However, since antibiotics were banned due to concerns of antibiotic resistant bacteria, researchers are studying alternative materials to antibiotics. Dietary nucleotides are one of the alternative materials for replacing antibiotics and can be used in abnormal conditions, such as weaning diarrhea, low digestibility, and disease condition. Nucleotides have substances that have important roles in cell division and cell growth, affecting growth performance, intestinal condition, and immunological effect at the weaning stage. However, nucleotides' composition is very different between sources and this aspect makes it difficult to utilize nucleotides at the weaning stage. Therefore, this review paper focuses on i) the characteristics and functions of dietary nucleotides and ii) the effect of dietary nucleotides on the growth performance and immune system of pigs.
In order to know the enzyme activities of Filbricola seouzenis, an intestinal trematode of human and rodent in Korea. the enzyme histochemical method is applicated. Activities of acid phosphatase (E.C.3.1.3.2) and non-specific esterase (E.C.3.1.1) were present in microvilli and glandular cells of trlbocytic organ and the epithelium of the caecum.
Excretory-secretory products of Toxocara canis larvae have been considered as a major functional antigen in immune responses against toxocariasis. We studied ultrastructural localization of T. canis second-stage larval antigen using a seropositive human serum under immunogold electron microscopy. High-density gold particles were observed in the secretory cells, excretory duct, intestinal epithelium, and cuticle of the larval worm sections. The distribution of the positive reactions in the larval worms suggests that the nature of the antigen is excretory-secretory antigen including waste metabolites and secretory enzymes.
Glucagon-like peptide 2 receptor, a GPCR, binds with the glucagon-like peptide, GLP-2 and regulates various metabolic functions in the gastrointestinal tract. It plays an important role in the nutrient homeostasis related to nutrient assimilation by regulating mucosal epithelium. GLP-2 receptor affects the cellular response to external injury, by controlling the intestinal crypt cell proliferation. As they are therapeutically attractive towards diseases related with the gastrointestinal tract, it becomes essential to analyse their structural features to study the pathophysiology of the diseases. As the three dimensional structure of the protein is not available, in this study, we have performed the homology modelling of the receptor based on single- and multiple template modeling. The models were subjected to model validation and a reliable model based on the validation statistics was identified. The predicted model could be useful in studying the structural features of GLP-2 receptor and their role in various diseases related to them.
Glucagon like pepide-2, one of the GLPs, is involved in various metabolic functions in the gastrointestinal tract. It plays a major role in the regulation of mucosal epithelium and the intestinal crypt cell proliferation. Because of their therapeutic importance towards the diseases in the gastrointestinal tract, it becomes necessary to study their interaction with its receptor, GLP-2R. In this study, we have developed protein-protein docking complexes of GLP-2 - GLP-2 receptor. Homology models of GLP-2 are developed, and a reliable model out of the predicted models was selected after model validation. The model was bound with the receptor, to study the important interactions of the complex. This study could be useful in developing novel and potent drugs for the diseases related with GLP-2.
