Seed Setting and Viability and Fertility of Pollens in Families of Artificial and Natural Interspecific Hybrids in Lepidobalanus of Genus Quercus (참나무속의 인공 및 자연 종간잡종 가계의 종자결실 및 활력과 화분의 임성)

  • Interspecific hybrid seedlings by artificial crossing of Genus Quercus (Q. serrata, Q. dentata, Q. mongolica var. crispula, Q. aliena) were planted at nursery in Tottori University. Seedlings of hybrids by natural crossing(Q. fabri and $Q.{\times}mccormickii$) were selected and planted at Hiruzen district in Tottori University. Artificial interspecific hybrid $F_1$ and natural hybrid $F_1$ bloomed when they were 4 years old and 3 years old, respectively. The pollen fertility and seed viability were investigated from the bloomed individuals in 2001. The germination percentages of the pollens of artificial interspecific hybrid were more than 84% except one individual, and the extension of pollen tubes was normal. The normal seed percentages of artificial interspecific hybrid were more than 90% similar to parents. Germination percentages of normal seed of natural crossing family were more than 64%, respectively, and selfed offsprings of Q. fabri, and $Q.{\times}mccormickii$ hade high reproductive ability.

  • Reciprocal interspecific hybrids between two bitterling species Rhodeus uyekii (RU) and R. notatus (RN) were genetically identified based on the partial sequence analysis of recombination activating gene-1 (RAG-1) gene. Out of 863 bp positions analyzed, 13 nucleotide substitutions were detected between the two parental species (RU and RN genotypes). Both the induced hybrids (RU female$\times$RN male; UN genotype) and their reciprocal counterparts (RN female$\times$RU male; NU genotype) displayed the double peaks (or polymorphism) of sequence chromatograms at the 13 diagnostic positions, indicating that those hybrids were actual karyogamy derived from the two parental haploid genomes. However, it was not possible to distinguish between the reciprocal interspecific hybrids.

  • Squash cultivars of Cucurbita moschata have good fresh quality, but their viny growth habit hinders wide cultivation under structure. On the other hand, 'Zucchini' varieties of C. pepo have bush growth habit and better growth ability under the low temperature condition than C. moschata, but have inferior flesh quality. To combine desirable characteristics of the two species, many attempts have been exerted to obtain interspecific hybrids. A shootless phenomenon was found among segregating populations; the cotyledon expanded normally but further shoot growth. The inheritance of the character was investigated in F2 and BC1F2 populations. The segregation ratios indicated that the shoot-lacking characteristics is expressed by double recessive gene pairs.

  • The cytogenetic pattern of autosome and sex chromiosome after Giemas staining were examined in the hybrids between two sibling species, Drosophila melanogaster and D. simulans. The analysis of karyotype in the hybrid female between D. melanogaster females and D. simulans males could be easily distinguished the characteriation of eight chromosomes from bothe species, especially with regard to X chromosomes. The lagging duplication of Y chromosome was investigated in the interspecific hybrid males from the cross between female of Drosophila melanogaster(OR) and males of D. simulasn (K18). On the other hand, the X chromatids of D. simulans were loosely separated in the early stage of anaphase.

  • Interspecific cross between Nicotiana trigonophylla and N. tabacum cv. BY4 is highly sterile because of abnormal ovule and embryo development. In vitro culture of excised N. trigonophylla ovules after polination by N. tabacum allows significant numbers of hybrid embryos to develop into mature plants. Total yield of seedlings and number of normal seedlings were produced following in vitro culture of individual fertilized ovules of N. trigonophylla X N. tabacum at four days post-pollination on B5 medium containing 6% sucrose. Hybrids were uniform in morphology and peroxidase isozyme composition and the majority were cytologically stable: flower characteristics were generally intermediate between those of the parents. All hybrids evaluated were self-sterile.

  • In order to evaluate the applicability of enzyme electrophoresis for the identification of intra/interspecific hybride obtained by the protoplast fusion in Trichoderma, soluble proteins, intracellular soluble enzymes and extracellular $\beta$-glucosidase were analyzed by polyacrylamide gel electrophorsis. As the results, patterns of soluble protein, and isozyme patterns of peroxidase, malate dehydrogenase, and $\beta$-glucosidase in hydrids were defferent from those in parental and wild type strains. Therefore, it was established that the analysis of protein pattern by electrophoresis could be applied to isolate and identify the hybrids from the protoplast fusion.

  • This study was conducted to investigate the utility and agronomic characteristics and use of cytoplasmic. male-sterile (CMS) haploid plants derived from interspecific cross between (male-sterile NC82$\times$burley21) F$_1$ plant (Nicotiana tabacum L.) and Nicotiana africana. Abundant seeds of high germinability were obtained when Nicotiana tabacum (cytoplasmic male-sterile F$_1$ plants) is pollinated by Nicotiana africana. Most of seedlings died at the cotyledonary stage. The remaining seedlings are viable F$_1$ hybrids or maternal haploids that can be easily distinguished. Number of interspecific Fl hybrids and matermal haploids per capsule obtained from the interspecific cross between cytoplasmic male-sterile tobacco F$_1$ plants and N.africana yielded 2.2 and 0.5 plants, respectively. Out of 149 CMS haploid plants obtained from interspecific cross, 102 plants showed green type while the others were yellow type for leaf and stem. This results agreed with the genetic ratios expected among haploid plants from the F$_1$ hybrids heterozygous for two recessive genes of yellow color of burley tobacco plant. Out of 83 CMS haploid plants inoculated with TMV, 48 plants showed resistant, while the others was susceptible. It agreed with the expected genetic ratios for a single dominant gene of TMV resistance. CMS haploid plant will be useful as a source material for breeding of CMS doubled haploid lines

A Study on the Natural Interspecific Hybrid between Rhodeus notatus and R. ocellatus (떡납줄갱이 Rhodeus notatus와 흰줄납줄개 R. ocellatus의 자연 종간잡종에 관한 연구)

  • Three specimens, presumed to be natural hybrids between Rhodeus notatus and R. ocellatus, were collected from Onyangcheon Stream area, Jwabu-dong, Asan-si, Chungcheongnamdo, the Republic of Korea, and morphological and phylogenetic analyses were performed to clearly identify their parent species. The body color of the three natural hybrids was light greenish-brown on dorsal side, and the size of red area on the upper front of the dorsal fin and the outer margin of the anal fin generally showed intermediate characters between the parent species, R. notatus and R. ocellatus. Among the measurement and meristic characters, the ratio of prepectoral length and preanal length in the standard length, and the ratio of snout length, interorbital width, length of caudal peduncle and depth of caudal peduncle in the head length, and the number of longitudinal row scales were analyzed as the unique characters of natural hybrids. In the rag1 gene of nuclear DNA, the three natural hybrids were analyzed to be reflected all the single nucleotide polymorphism sites between R. notatus and R. ocellatus, and in the phylogenetic tree using the cytb gene of mitochondrial DNA, they formed the same genetic clade as R. notatus. Therefore, three specimens, presumed to be natural hybrids analyzed in this study were identified as interspecific hybrids between female R. notatus and male R. ocellatus.

Utility of Selected Non-coding Chloroplast DNA Sequences for Lineage Assessment of Musa Interspecific Hybrids

  • Single-copy chloroplast loci are used widely to infer phylogenetic relationship at different taxonomic levels among various groups of plants. To test the utility of chloroplast loci and to provide additional data applicable to hybrid evolution in Musa, we sequenced two introns, rpl16 and ndhA, and two intergenic spacers, psaA-ycf3 and petA-psbJ-psbL-psbF and combined these data. Using these four regions, Musa acuminata Cola(A)- and M. balbisiana Colla (B)-containing genomes were clearly distinguished. Some triploid interspecific hybrids contain A-type chloroplasts (the AAB/ABB) while others contain B-type chloroplasts (the BBA/BBB). The chloroplasts of all cultivars in 'Namwa' (BBA) group came from the same wild maternal origin, but the specific parents are still unrevealed. Though, average sequence divergences in each region were little (less than 2%), we propose that petA-psbJ intergenic spacer could be developed for diversity assessment within each genome. This segment contains three single nucleotide polymorphisms (SNPs) and two indels which could distinguish diversity within A genome whereas this same region also contains one SNP and an indel which could categorize B genome. However, an inverted repeat region which could form hairpin structure was detected in this spacer and thus was omitted from the analyses due to their incongruence to other regions. Until thoroughly identified in other members of Musaceae and Zingiberales clade, utility of this inverted repeat as phylogenetic marker in these taxa are cautioned.

Comparative Studies on the Developmental Capacity by Interspecific Nuclear Transplantation (이종간의 핵치환에 의한 발생수행능력의 비교 연구)

  • In order to investigate the role of nuclei and cytoplasm in early embryogenesis, interspecific nuclear transplantation was carried out Nuclei of one species of amphibian was transplanted into enucleated egg of another species. In interspecific hybrids between Rana species, development was arrested before or right after the dorsal lip formation. In intergenetic hybrids between Rana, Xenopus and axolod, developmental arrest took place at late blastula stage. Comparing the developmental capacity of each nudeocytoplasmic hybrid, the more distantly related the species, the earlier does development arrest. The general rule is that nuclear transfers between any amphibian species will form a regulary cleaved late blastula. Two plausible factors relate to limitation of tite developmental capacity of nudeocytaplasmic hybrids. One is an irreversible change in grafted nuclei and another is tunctional incompatibility between the recipient cytoplasm and transplanted nucleus. The later is postulated a more possible cause of the arrestrnent.
