Acebutolol, a Cardioselective Beta Blocker, Promotes Glucose Uptake in Diabetic Model Cells by Inhibiting JNK-JIP1 Interaction

  • The phosphorylation of JNK is known to induce insulin resistance in insulin target tissues. The inhibition of JNK-JIP1 interaction, which interferes JNK phosphorylation, becomes a potential target for drug development of type 2 diabetes. To discover the inhibitors of JNK-JIP1 interaction, we screened out 30 candidates from 4320 compound library with In Cell Interaction Trap method. The candidates were further confirmed and narrowed down to five compounds using the FRET method in a model cell. Among those five compounds, Acebutolol showed notable inhibition of JNK phosphorylation and elevation of glucose uptake in diabetic models of adipocyte and liver cell. Structural computation showed that the binding affinity of Acebutolol on the JNK-JIP1 interaction site was comparable to the known inhibitor, BI-78D3. Our results suggest that Acebutolol, an FDA-approved beta blocker for hypertension therapy, could have a new repurposed effect on type 2 diabetes elevating glucose uptake process by inhibiting JNK-JIP1 interaction.

Dynamic Analysis of Multiple-Body Floating Platforms Coupled with Mooring Lines and Risers

  • In this study, the program to investigate the multiple body interaction effects between a floating platform and a shuttle tanker considering the coupled effect of hull (FPSO) with mooring lines and risers was developed. The coupled analysis program, which is called WINPOST-MULT using the hydrodynamic analysis results by WAMIT, was made. For the verification of WINPOST-MULT by means of numerical experiments, two multiple-body models of an FPSO-FPSO and an FPSO-shuttle tanker system are adopted. With the FPSO-FPSO model and a two-mass-spring system to idealize two identical bodies for the 100-year storm wave condition in GOM, the numerical simulations were performed to investigate the interaction effects between two identical bodies. For the more reality, the coupled analysis for the FPSO-shuttle tanker model in the tandem arrangement was carried out in the consideration of the environmental condition of the West Africa Sea as a rather mild condition. Through the case studies with interaction effect and without interaction effect by the iteration method and the combined method, it is verified that the program is a very useful tool for the analysis of the interaction problem of multiple-body system and the coupled problem of the hull/mooring/riser.

Numerical investigation of the unsteady flow of a hybrid CRP pod propulsion system at behind-hull condition

  • Flows induced by hybrid CRP pod propulsion systems (CRP-POD) are fundamentally characterized by unsteadiness. This work presents a numerical study on the unsteady flow of a CRP-POD at behind-hull condition based on CFD (Computational Fluid Dynamics). Unsteady RANS method is adopted, coupled with SST k-u turbulence model and sliding mesh method. The propeller thrusts and torques obtained by CFD is validated by model tests and acceptable agreements are obtained. The time histories of shingle-blade loads and pressures near the hull surface are recorded for the analysis of unsteady flow features. The cases of forward propeller alone and aft propeller alone are also computed to distinguish the hull-propeller interaction and propeller-propeller interaction. The results show the blade loads of both forward and aft propellers strongly fluctuate with phase angles. For the forward propeller, the blade load fluctuation is mainly governed by the hull-propeller interaction, while the aft blade load is remarkably affected by the propeller-propeller interaction in terms of the load average and fluctuation pattern. The fields of pressure, vorticity and velocity are also analyzed to reveal the unsteady flow features.

해일-조석-파랑을 결합한 폭풍해일 수치모델 개발에 관한 연구 (Study on Development of Surge-Tide-Wave Coupling Numerical Model for Storm Surge Prediction)

  • IIn this study, a wave-surge-tide coupling numerical model was developed to consider nonlinear interaction. Then, this model was applied and calculations were made for a storm surge on the southeast coast. The southeast coast was damaged by typhoon "Maemi" in 2003. In this study, we used a nearshore wind wave model called SWAN (Simulating WAves Nearshore). In addition, the Meyer model was used for the typhoon model, along with an ocean circulation model called POM (Princeton Ocean Model). The wave-surge-tide coupling numerical model could calculate exact parameters when each model was changed to consider the nonlinear interaction.

콘텐츠 제작을 위한 소프트웨어 디자인 프레임워크 (Software Design Framework for Content Creation)

  • 최근에는 터치형 단말기에서 소비되는 어플리케이션 형태의 콘텐츠 제작을 지원하는 소프트웨어가 다양하게 등장하고 있는데 이러한 콘텐츠 제작 기반의 소프트웨어는 기존의 사용자 태스크와 다른 형태를 갖는다. 사용자 인터랙션을 고려하는 시점이 콘텐츠 제작과 소비, 양 측면에서 모두 중요한 역할을 담당하기 때문이다. 이러한 변화의 흐름 가운데 본 논문은 태스크 모델링에 기반을 둔 모델 기반 유저 인터페이스 개발(Model Based User Interface Design, MB-UID)과 사용 중심 디자인(Usage Centered Design, UCD) 모델을 기반으로 콘텐츠 제작 소프트웨어 유저 인터페이스 디자인 프레임워크를 새롭게 제시한다. 본 프레임워크는 비즈니스 규칙 모델, 제작 역할 모델, 제작 흐름 모델, 제작 행동모델, 표현 모델의 다섯 단계로 구성된다. 본 연구를 통해 콘텐츠 제작시 사용자와 소프트웨어 간의 인터랙션을 효율적으로 통제함과 동시에 콘텐츠 소비시 발생 가능한 사용자 인터랙션 유형을 콘텐츠 제작 단계에서 미리 고려할 수 있다.

Evaluation of numerical procedures to determine seismic response of structures under influence of soil-structure interaction

  • In this study, the accuracy and reliability of fully nonlinear method against equivalent linear method for dynamic analysis of soil-structure interaction is investigated comparing the predicted results of both numerical procedures with the results of experimental shaking table tests. An enhanced numerical soil-structure model has been developed which treats the behaviour of the soil and the structure with equal rigour. The soil-structural model comprises a 15 storey structural model resting on a soft soil inside a laminar soil container. The structural model was analysed under three different conditions: (i) fixed base model performing conventional time history dynamic analysis, (ii) flexible base model (considering full soil-structure interaction) conducting equivalent linear dynamic analysis, and (iii) flexible base model performing fully nonlinear dynamic analysis. The results of the above mentioned three cases in terms of lateral storey deflections and inter-storey drifts are determined and compared with the experimental results of shaking table tests. Comparing the experimental results with the numerical analysis predictions, it is noted that equivalent linear method of dynamic analysis underestimates the inelastic seismic response of mid-rise moment resisting building frames resting on soft soils in comparison to the fully nonlinear dynamic analysis method. Thus, inelastic design procedure, using equivalent linear method, cannot adequately guarantee the structural safety for mid-rise building frames resting on soft soils. However, results obtained from the fully nonlinear method of analysis fit the experimental results reasonably well. Therefore, this method is recommended to be used by practicing engineers.

Seismic evaluation of soil-foundation-structure interaction: Direct and Cone model

  • The present research intends to study the effects of the seismic soil-foundation-structure interaction (SFSI) on the dynamic response of various buildings. Two methods including direct and Cone model were studied through 3D finite element method using ABAQUS software. Cone model as an approximate method to consider the SFSI phenomenon was developed and evaluated for both high and low rise buildings. Effect of soil nonlinearity, foundation rigidity and embedment as well as friction coefficient between soil-foundation interfaces during seismic excitation are investigated. Validity and performance of both approaches are evaluated as reference graphs for Cone model and infinite boundary condition, soil nonlinearity and amplification factor for direct method. A series of calculations by DeepSoil for inverse earthquake record modification was conducted. A comparison of the two methods was carried out by root-mean-square-deviation (RMSD) tool for maximum lateral displacement and story shear forces which verifies that Cone model results have good agreement with direct method. It was concluded that Cone method is a convenient, fast and rather accurate method as an approximate way to count for soil media.

분산 협업 환경에서의 콘텐츠 공유 모델 (Contents Sharing Model in Distributed Collaboration Environment)

  • 본 논문에서는 분산된 협업 환경을 통합하는 콘텐츠 공유 모델을 제안한다. 확장 가능한 고해상도 디스플레이 자원, 로컬 및 원격 콘텐츠 공유, 다중 사용자 상호작용 및 접근제어 특성들을 통합하여 결합된 하나의 모델을 제공한다. 다중 사용자의 상호 작용은 모든 사용자가 상호 작용이 가능하다는 이점이 있지만, 그로 인해 오버랩이 발생할 수 있다. 그 오버랩을 관리하기 위해 접근 제어를 모델에 적용했으나 작업 흐름의 방해가 가능하다. 따라서 이 두 가지 사항을 중점으로 모델을 평가하기 위해 상호작용성에 대한 이용자 연구를 실시했다. 그 결과를 바탕으로 제안된 콘텐츠 공유 모델은 상호작용과 그 흐름을 방해 하지 않아 작업에 집중할 수 있는 분산 협업 환경을 제공한다.

  • In this study, a fatigue damage estimation for an ice-going vessel navigating through broken ice fields was carried out. A numerical model to simulate the interaction between ice and structure developed using the finite element method was introduced. Time series of stresses calculated by the proposed model and the corresponding fatigue analysis results are presented. The numerical model enables the long time analysis through an efficient interaction model, the application of the periodic media analysis and the convolution integral, and it allows the stress time history to be extracted directly using the finite element method. To describe the probability distribution of stress amplitudes, the 2-parameter Weibull model was applied to the calculated stress time history, and the fatigue damage was calculated using the Palmgren-Miner rule. Finally, the fatigue damage considering the ice conditions of the Baltic Sea was calculated using the proposed method and LR method, and the results were compared to each other.

A Protein-Protein Interaction Extraction Approach Based on Large Pre-trained Language Model and Adversarial Training

  • Protein-protein interaction (PPI) extraction from original text is important for revealing the molecular mechanism of biological processes. With the rapid growth of biomedical literature, manually extracting PPI has become more time-consuming and laborious. Therefore, the automatic PPI extraction from the raw literature through natural language processing technology has attracted the attention of the majority of researchers. We propose a PPI extraction model based on the large pre-trained language model and adversarial training. It enhances the learning of semantic and syntactic features using BioBERT pre-trained weights, which are built on large-scale domain corpora, and adversarial perturbations are applied to the embedding layer to improve the robustness of the model. Experimental results showed that the proposed model achieved the highest F1 scores (83.93% and 90.31%) on two corpora with large sample sizes, namely, AIMed and BioInfer, respectively, compared with the previous method. It also achieved comparable performance on three corpora with small sample sizes, namely, HPRD50, IEPA, and LLL.
