  • The novel integration method with programmable high-pass filter is suggested in order to solve the problem of integration for stator flux estimation in a stator flux oriented direct vector controlled induction motor drive system. The dc offset in a pure integrator is eliminated using high-pass filter with fixed time constant, and then time constant of programmable high-pass filter is controlled with a inverter frequency for integration in a wide frequency range, considering phase lag and attenuation due to both the hardware low-pass filter and high-pass filter. The proposed method is verified with the experimental results implemented by 32-bit DSP.

동물 세포 내에서 MJ1 인티그라제에 의한 부위 특이적 재조합 (Site-Specific Recombination by the Integrase MJ1 on Mammalian Cell)

  • 이전 연구에서, bacteriophage ${\Phi}FC1$이 Enterococcus faecalis KBL703에서 UV induction을 통해 분리 동정되었으며, ${\Phi}FC1$은 phage attachment site인 attP와 bacterial attachment site인 attB 사이에서 site-specific integration을 촉매하는 integrase를 가지고 있다는 것을 밝혀냈으며 이를 MJ1이라 명명하였다. 이 연구에서는 이를 바탕으로 MJ1에 의한 site-specific integration의 효율을 Escherichia coli와 NIH3T3 cell에서 확인 하기 위해 attP, attB, MJ1을 각각의 벡터에 삽입하였다. MJ1 인테그라제에 의한 재조합을 수행하기 위해서 기질 벡터 pABLP를 $DH5{\alpha}$에 형질전환시킨 후, LB 배지에서 $37^{\circ}C$ 1시간 배양한 후 암피실린(ampicillin)과 테트라싸이클린(tetracycline) 항생제 플레이트로 pGMJ1과 pABLP 같이 가지고 있는 colony 들을 선별하여, LacZ 유전자가 불활성화 된 흰색 콜로니 개수를 세고 통계를 낸 결과 integration의 frequency가 99% 이상인 것으로 나타났다. 또한, 실제로 재조합이 일어났는 지를 확인하기 위해서 콜로니 PCR을 수행하여 재조합의 산물인 attL 150 bp을 확인하였다. PCR 산물은 염기서열분석을 통해 정확한 site-specific integration이 일어났음을 확인하였다. MJ1에 의한 integration을 보이기 위해 attP와 attB를 가지고 있는 vector를 MJ1 expression vector와 함께 NIH3T3 cell에 cotransfection 했으며 GFP를 reporter로 사용해 그 activity를 관찰하였다. NIH3T3 cell에서 GFP의 발현을 형광 현미경을 통해 알아본 결과, MJ1에 의한 sitespecific integration이 다른 accessory protein의 도움 없이 일어난다는 것을 볼 수 있었다. 마찬가지 방법으로, attR과 attL 간의 excision을 GFP로 알아본 결과, GFP는 발현하지 않았으며, 이는 MJ1에 의한 excision이 일어나지 않았음을 보여주었다. 이와 같은 결과로 볼 때, MJ1의 host만이 아니라 넓은 범위안에서도 integration을 수행할 수 있다는 것을 보여주었다. 따라서 MJ1을 이용한 site-specific integration system의 개발은 gene therapy를 위한 gene delivery system의 구축에 있어서 좋은 시작이 될 수 있다.

Retroviral integration profiles: their determinants and implications for gene therapy

  • Retroviruses have often been used for gene therapy because of their capacity for the long-term expression of transgenes via stable integration into the host genome. However, retroviral integration can also result in the transformation of normal cells into cancer cells, as demonstrated by the incidence of leukemia in a recent retroviral gene therapy trial in Europe. This unfortunate outcome has led to the rapid initiation of studies examining various biological and pathological aspects of retroviral integration. This review summarizes recent findings from these studies, including the global integration patterns of various types of retroviruses, viral and cellular determinants of integration, implications of integration for gene therapy and retrovirus-mediated infectious diseases, and strategies to shift integration to safe host genomic loci. A more comprehensive and mechanistic understanding of retroviral integration processes will eventually make it possible to generate safer retroviral vector platforms in the near future.

A Study on the Dynamic Relationship between Cultural Industry and Economic Growth

  • The cultural industry is treated as the sunrise industry in modern society. It has taken an increasing role in promoting the economic growth. Due to this, this paper attempts to explore the dynamic relationship between cultural industry and the economic growth. On the grounds of Cobb-Douglas production function, the cultural industry is regarded as a determinant such as the labor input and the capital input to impact the economic growth. Meanwhile, the quarterly datum form 2000-Q1 to 2017-Q4 are employed to perform an empirical analysis via the vector error correction model. The GDP is treated as an independent variable. The input of capital, the input of labor and the total input of cultural industry are treated as dependent variables. Furthermore, a menu of statistical approaches such as the co-integration test and the impulse response function will be used to testify the dynamic relationship between cultural industry and economic growth. Via the Johansen co-integration test, the results report that the cultural industry has a obviously positive effect on economic growth. Through the vector error correction estimation, the results also report that the cultural industry also has a significantly positive effect on economic growth, but less than that of the Johansen co-integration test. This paper provides a view that the cultural industry is a kind of a determinant to promote the economic growth. Therefore, the China's government should pay much attention to the cultural industry construction.

Ready-Made Garments (RMG) Export Earnings and Economic Development of Bangladesh: Empirical Analysis Using Vector Error Correction Model

  • Ready-made Garments (RMG) export earnings, which are almost 80% of the total exports of Bangladesh, have been recognized as one of the main catalysts for the recent development of the country. Therefore, the need to determine whether the RMG export had served as a mechanism for increasing the GDP growth as well as the economic development of the country is topical and pressing. We have applied the Johansen Co-integration test and Vector Error Correction Model (VECM) to reveal the linkage of RMG export earnings and other variables with the GDP growth rate in Bangladesh. Using data from 1990 to 2020 for Bangladesh, we have found long-run as well as short-run associations among RMG Export earnings, Foreign Direct Investment (FDI), and GDP growth. A co-integration among the variables is validated through the Johansen Co-integration test. Moreover, a causal correlation running from RMG export earnings to GDP was revealed by the Granger causality test in the long run. Finally, we estimated impulse response functions to observe the variations of model variables in response to a shock. Our result supports the proposition that RMG export earnings are one of the main growth engines in Bangladesh and this sector leads growth in other sectors also in the long term.

공적분·벡터오차수정모형을 활용한 벙커유 가격의 장기균형 수렴에 관한 실증분석 (An Empirical Analysis on the Long-term Balance of Bunker Oil Prices Using the Co-integration Model and Vector Error Correction Model)

  • This study performs a factor analysis that affects the bunker oil price using the Co-integration model and Vector Error Correction Model (VECM). For this purpose, we use data from Clarkson and the analysis results show 17.6% decrease in bunker oil price when the amount of crude oil production increases at 1.0%, 10.3% increase in bunker oil price when the seaborne trade volume increases at 1.0%, 1.0% decrease in bunker oil price when total volume of vessels increases at 1.0%, and 0.003% increase in bunker oil price when 1.0% increase in world GDP, respectively. This study is meaningful in that this study estimates the speed of convergence to long-term equilibrium and identifies the price adjust mechanism which naturally exists in bunker oil market. And it is expected that the future study can provide statistically more meaningful econometric results if it can obtain data during more long-periods and use more various kinds of explanatory variables.

멀티레이트 수치적분법을 이용한 유연다물체 동역학해석 (Flexible Multibody Dynamic Analysis Using Multirate Integration Method)

  • A Nordsick form opf the multirate integration scheme has been proposed for flexible multibody dynamic systems. It is assumed that vibrational modal coordinates in the equations of motion are treated as fast variables, whereas the relative joint coordinates are treated as slow variables. In the multirate integration, the fast variables are integrated with small step-size, and the slow variables are integrated with larger step-size. The proposed multirate integration method is based on the Adams-Bashforth-Moulton predictor-corrector method and implemented in the Nordsieck vector form. The Nordsieck form of multrate integration method provides effective step-size control and at the same time, inherits the efficiency from the Adams integration method. Simulations of a flexible gun and turret system of the military tank have been carried out to show the effectiveness and efficiency of the proposed method.

Glucoamylase 유전자 STA의 염색체내 삽입에 의한 Saccharomyces diastaticus의 glucoamylase 생성능 향상 (Improvement of Glucoamylase Productivity of Saccharomyces diastaticus by Intergration of Glucoamylase Gene, STA, into Chromosomal DHA)

  • Saccharomyces distaticus 의 glucoamylase 생성능을 증진시킬 목적으로 STA1 유전자를 YIp vector 를 이용하여 염색체에 도입해 주고자 하였다. STA1 유전자 5.8-Kb 를 YIp vector 에 재조합하여 YIp-STA 를 재작하고, S. diastaticus GMT-11 (a, ura 3, STA1) 을 숙주균주로 하여 염색체의 STA1 유전자 부위에 homologous recombination 되어 삽입하도록 형질전환을 실시하였다. 이렇게 하여 glucoamylase 생성능이 모균주에 비해 최대 6배까지 증대된 다양한 형질전환체들을 얻을 수 있었다. 그리고 glycoamylase 생성능이 증대된 형질전환체들의 염색체 DNA 를 분리하여 Southern hybridization 을 실시한 결과 YIp-STA 가 multi-copy integration 되었음을 확인하였고, 또한 도입해 준 YIp-STA 는 세포분열인 30세대기간 동안 계속되었어도 안정하게 유지되었음을 알았다.

  • This paper describes the simulation approaches employed for a vector control system of induction motor drive using SIMNON for windows program. SIMNON program tool can solve differential and difference equations for nonlinear dynamical control system. One powerful feature is its ability of allowing integration of individual program modules after each individual module is programed and tested independently. This particular feature is exploited here for an SVPWM inverter drive by real-time modeling and simulation. The suggested programs are provided a simple and complete simulation for induction motor vector drive system.

Unscented Filtering in a Unit Quaternion Space for Spacecraft Attitude Estimation

  • A new approach to the straightforward implementation of the unscented filter in a unit quaternion space is proposed for spacecraft attitude estimation. Since the unscented filter is formulated in a vector space and the unit quaternions do not belong to a vector space but lie on a nonlinear manifold, the weighted sum of quaternion samples does not produce a unit quaternion estimate. To overcome this difficulty, a method of weighted mean computation for quaternions is derived in rotational space, leading to a quaternion with unit norm. A quaternion multiplication is used for predicted covariance computation and quaternion update, which makes a quaternion in a filter lie in the unit quaternion space. Since the quaternion process noise increases the uncertainty in attitude orientation, modeling it either as the vector part of a quaternion or as a rotation vector is considered. Simulation results illustrate that the proposed approach successfully estimates spacecraft attitude for large initial errors and high tip-off rates, and modeling the quaternion process noise as a rotation vector is more optimal than handling it as the vector part of a quaternion.
