Electron microscopic study on the insulin-, glucagon-, somatostatin-, and pancreatic polypeptide secreting cells in Korean native goat (한국재래산양 췌장의 insulin, glucagon, somatostatin 및 pancreatic polypeptide 분비세포에 관한 전자현미경적 연구)

  • Ultrastructures of pancreatic endocrine cells containing glucagon, insulin, somatosratin and pancreatic polypeptide were studied in the pancreas of the Korean native goat by immunohistochemical and elecron microscopy. Glucagon immunoreatctive cells were round or fusiform in shape and contained secretory granules of 200-260 nm in diameter. The secretory granules were high in electron density and had a halo between the limiting membrane and the central granule core. Insulin immunoreactive cells were round or oval in shape, and contained various sizes of secretory granules from 135 to 300 nm in diameter. The secretory granules were low or moderate electron density and had a variform halo. Somatostatin immunoreactive cells were elliptical or fusiform shape with cytoplasmic processes. They contained the secretory granules of 140-320 nm with moderate electron densities. Pancreatic polypeptide immunoreactive cells were elliptical or fusiform and contained small secretory granules with high electron densities. The secretory granules were 120-230 nm in diameter and the least in number.

  • Pancreatic endocrine cells of the native Korean goat were investigated immunocytochemically at electron microscopic level. All glucagon-, insulin-, somatostatin- and pancreatic polypeptide(PP)-immunoreactive cells were showed chromogranin(CG) immunoreactivity in the secretory granules of each cells. In addition, bovine pancreatic polypeptide immunoreactivity was found to be colocalized in the secretory granules of the glucagon and insulin cells. These observations support that chromogranin is available as the marker of pancreatic endocrine cells on the native Korean goat and BPP colocalized in the secretory granules of glucagon and insulin cells.

Effects of Uncooked Korean Food on Blood Glucose and Antioxidant Fnzyme Activities of Streptozotocin-induced Diabetic Rats (당뇨환자용 식사대용식이 Streptozotocin으로 유발된 당뇨쥐의 혈당 및 항산화 효소 활성에 미치는 영향)

  • Objectives : This study has been carried out to understand the effect of Uncooked Korean Food(F-DM) on blood glucose and antioxidant enzyme activities in streptozotocin-induced Diabetic Rats. Methods : SD rats were separated into four groups(each with 20 rats). Except normal two group, the other two groups were injected into intra-peritoneal with streptozotocin 60 mg/kg. Experimental group was eated Feed with 25% F-DM for 4 weeks. The change of plasma glucose level, body weight were observed. After 4 weeks, liver and kidney weight, antioxidant enzyme activities, survival rate were observed with histological changes on liver, kidney and pancreas. Results : In experimental group, body weight and survival rate increased, plasma glucose level were decreased significantly. Liver and kidney weight, XOD activity were decreased in experimental group compared to control group. GSH-px and CAT activities, insulin- immunoreactive granules in p-cells were increased significantly in experimental group compared to control group. Conclusions : This study shows that the F-DM might be effective for treatment of diabetes and its complications, as well as reduction of the oxidative stress.

Effects of Saengchinyanghyoltang-gamibang on the Pancreatic Islets of the Mice Induced with Streptozotocin (생진양혈탕가미방이 Streptozotocin으로 유발된 당뇨 생쥐의 췌장에 미치는 영향)

  • This study was carried out to understand the effects of Saengchinyanghyoltang-gamibang (SGT) on pancreatic islets of the mice induced with streptozotocin (STZ). In the control group, two times injected with 50 mg/kg 572 at 24-hour intervals, a few number of insulin immunoreactive-cells are observed at the pancreatic islets of the mice. In the experimental group which administered with extract of SGT during 21-day, a number of immunoreactive-cells are observed at the pancreatic islets. According to the electron microscopic observation, $\beta-cells$ of the control group were contained a few of secretory granules, but also these granules were contained electro-lucent materials. In the experimental group, a lot o( secretory granules and well developed cell organelles are observed at the $\beta-cells$. The level of glucose was significantly decreased in the experimental group compared with control group, but the level of BUN was similar in these two groups. These results suggest that administration of SGT to the mice improved the damage of $\beta-cells$ from injected with STZ.

  • Objective : This study wascarried out to understand the effect of Sopyung-tang (SPT) on blood glucose & antioxidant enzyme activities in streptozotocin-induced diabetic rats. Methods : SD rats were separated into three groups, each with 20 rats. Except the normal group, the other two groups were intra-peritoneally injected with streptozotocin 6mg/kg. The experimental group was treated with SPT extract 500mgkg/day for 4 weeks. The normal and control groups were treated with saline 500mg/kg/day for 4 weeks. Changes of plasma glucose level and body weight were observed. After4 weeks, liver and kidney weight, antioxidant enzyme activities, and survival rate were observed with histological changes on liver, kidney and pancreas. Results : In the experimental group, body weight and survival rate increased, while plasma glucose level decreased significantly. Liver and kidney weight, XOD activity decreased in the experimental group compared to the control. GSH-px and CAT activities andinsulin-immunoreactive granules in ${\beta}-cells$ increased significantly in the experimental group compared to the control. Conclusions : This study shows that SPT might be effective for treatment of diabetes and its complications, as well as reduction of oxidative stress.
