Studies on Anopheles sinensis, the vector species of vivax malaria in Korea

  • Extensive previous studies on taxonomy, behavior/bionomics and control of Anopheles sinensis are reviewed and summarized. Recent molecular identification revealed that the population of An. sinensis complex includes An. sinensis, An. pullus, An. lesteri and at least two new species, and An. yatsushiroensis is synonmy of An. pullus. An. sinensis is the main vector specie of vivax malaria in Korea. Larvae of An. sinensis breed in wide range of habitats which are naturally-made clean water, stagnant or flowing; main habitats include rice fields, ditches, streams, irrigation cannals, marshes, ponds, ground pools, etc. Their host preferences are highly zoophilic. Human blood rate is very low ($0.7-1.7\%$); nevertheless An. sinensis readily feeds on man when domestic animals are not found near by. They feed on hosts throughout the night from dusk to dawn with a peak period of 02:00-04:00 hours; they are slightly more exophagic (biting outdoors); much larger numbers come into the room when light is on. Main resting places are outdoors such as grasses, vegetable fields and rice fields. A mark-release-recapture study resulted that $37.1\%$ was recaptured within 1 km, $29.4\%$ at 1-3 km, $21.1\%$ at 3-6 km, $10.3\%$ at 6-9 km and $2.1\%$ at 9-12 km distance. An. sinensis hibernate outdoors (mostly under part of dense grasses) during October-March. At the end of the hibernation period (March-April) they feed on cows at daytime. Until today any single measure to effectively control An. sinensis population has not been found. Indoor residual spray with a long-lasting insecticide can not reduce vector population densities, but shorten their life spans in some degree, so contributes to malaria control.

Environmentally-friendly control of Riptortus pedestris (Hemiptera: Alydidae) by environmental friendly agricultural materials (친환경농자재를 이용한 톱다리개미허리노린재의 친환경적 방제)

  • Recently, the rate of cultivation of such as hairy vetch was increased as a way of reducing the current soil problem. However, the occurrence of insect pest such as bean bugs, Riptortus pedestris, were increased in the field of green manure crops and their injuries to hairy vetch were observed. While minimizing insecticide use for the environmental friendly agriculture, the control of the bean bug can be utilized environmental friendly agricultural materials (EFAMs) in the green manure crop fields. The control effects of some EFAMs and most of chemical insecticides to the bean bug were significantly high. As a result of direct spray of EFAM that contained sophora extract or neem extract, the control effects of six EFAMs were higher than 70% at 120 hours after treatments. Among them, three EFAMs were showed over 90% of control effects. On the other hand, most of chemical insecticides were showed 100% of control effect against the bean bug at 48 hours after treatment. As mortality effects of EFAMs were sloely observed until 120h after treatment, we must use selected EFAMs at the beginnibgs of occurrence and entrance in the field of green manure for effective control of bean bugs.

The toxicity of an IGR class insecticide, Diflubenzuron on silkworm, Bombyx mori and abnormal symptoms (IGR계 농약 diflubenzuron의 독성과 누에이상증상)

  • Surveying, laboratory and field trial were performed to elucidate the causes of non-spinning syndrome of silkworm, Bombyx mori, abnormal symptoms of silkworm instars observed in certain sericultural fanning region. Mortality and growth of silkworm was still influenced by feeding the mulberry leaves sprayed with 10,000 fold diluted solution of standard spray of diflubenzuron WP (25%). Mortality and growth of silkworm fed with diflubenzuron-sprayed mulberry leaves with recommended spray solution were entirely affected even six weeks after spraying irrespective to the instars. Larval period of 5th instar and cocooning duration of silkworm was not affected in case 50m of buffer zone has maintained in near-by orchard field sprayed with diflubenzuron WP (25%). However, survival rate of pupae was not influenced in case 100 m of buffer zone has kept. Insect growth regulating insecticides were required to keep a certain distance of buffer zone when sprayed with water-diluted solution, 100 m from the application site at least in order to prevent contamination of the spraying drift by the chemicals.

  • Differences in susceptibility of several organophosphate, carbamate and pyrethroid insecticides to three species of aphids were determined by leaf-dipping and spray methods. The insects tested were the apterous adults of the green peach aphid (Myzus persicae), cotton aphid (Aphis gossypii) and soybean aphid (Aphis glycines). The comparison of $LC_{50}$ levels was indicated as follows; (1) The susceptibility to insecticides tested were greatly varied with the test methods. (2) The leaf dipping method is more recommendable than the spray method for insecticide screening with aphids. It was easier, more accurate and less variable than the latter. (3) The susceptibility to insecticides was greatly varied between the aphid species. Soybean aphid was more susceptible to the insecticides than green peach aphid and cotton aphid. Exceptionally, pirimicarb was not effective on the cotton aphid.

Mass Production of a Recombinant Baculovirus Expressing CpBV-ELP1 and Control of the Beet Armyworm, Spodoptera exigua (CpBV-ELP1 발현 재조합 벡큘로바이러스의 대량 증식과 파밤나방 방제 기술)

  • Cotesia plutellae bracovirus (CpBV) is a polydnavirus symbiotic to C. plutellae parasitizing young larvae of the diamondback moth, Plutella xylostella. Several CpBV genes play important roles in suppressing immune responses of the parasitized larvae. This study tested a hypothesis that the CpBV genes inducing host immunosuppression could be applied to develop a potent recombinant baculovirus. Based on a previous study, a recombinant baculovirus expressing CpBV-ELP1 (AcMNPV-ELP1) was selected and multiplied using larvae of the beet armyworm, Spodoptera exigua. The recombinant viruses were produced in a yield of $5{\times}10^{10}$ polyhedral inclusion body (PIB)/larva. The cultured AcMNPV-ELP1 exhibited a much higher pathogenicity against S. exigua larvae. However, its insecticidal activity was varied among larval instars of S. exigua, in which first and late instars were high susceptible. Spray of the recombinant baculovirus ($5{\times}10^6PIB/mL$) exhibited higher control efficacy (${\approx}$ 88%) against S. exigua larvae infesting cabbage than a chemical insecticide, tebufenozide, at 7 days after treatment. These results indicate that AcMNPV-ELP1 mass-cultured using host insect system is highly pathogenic and can be applied to develop a novel microbial control agent.

Insecticidal Activity of 7 Herbal Extracts against Black Pine Bast scale, Matsucoccus thunbergianae (솔껍질깍지벌레에 대한 7종류 한약재 추출물의 살충활성)

  • The black pine bast scale, Matsucoccus thunbergianae is one of the most serious insect pest in Japanese black pine, Pinus thunbergii forest in Korea. Insecticidal activity of 10 folds hot water extracts from 7 herbal plants (Atractylodes lancea, Eugenia caryophyllata, Lonicera japonica, Melia azedarach, Quisqualis indica, Sophora flavescens and Taraxacum mongolicum) were tested against different stage of M. thunbergianae using spray method both in laboratory and field. Efficacies of herbal extracts were different depending on stage of M. thunbergianae. Q. indica sprayed with hand sprayer produced the highest corrected mortality of 95.7% on intermidiated nymph stage, however, A. lancea produced the highest corrected mortality (51.3%) on pupae of M. thunbergianae in laboratory. Q. indica and A. lancea were highly effective against female M. thunbergianae in laboratory, however, corrected mortality was lower than other stage (40.0%). Efficacy of Q. indica was similar to effective insecticide, fenitrothion 50% EC against M. thunbergianae in field trials. These results indicated that Q. indica could be an environmental friendly control agent of M. thunbergianae.

Chemical Control of the Pine Gall Midges (Thecodiplosis japonensis Uchida et Inouye) ( I ) -Low-volume Foliar Spray of the Insecticides- (솔잎혹파리의 약제방제에 관한 연구 -고농도미량엽면살포-)

  • The experiments were carried out to evaluate the effectiveness of some insecticides in the control of the pine gall midges (Thecodiplosis japonensis Uchida et Inouye) when the low-volume of the insecticides were applied by the mist foliar sprayer. The insecticides, Salithion $(24\%\;Ec)$, Omethoate $(Folimat^{(R)}\;50\%\;Ec)$, Monocrotopos $(Azodrin^{(R)}24\%\;Ec)$, Acephate $(Ortran^{(R)}\;50\%\;Wp)$, Metalkamate $(Bux^{(R)}24\%\;Ec)$. BPMC $(Bassae^{(R)}50\%\;Ec)$ and MIPC $(Mipcin\;50\%\;Wp)$ were tested with 100, 200 and 400 times of water solution on the basis of single application at the six different intervals. Salithion was the most effective for the pressure of the gall formation by the pine gall midges and Omethoate, Monocrotophos and Metalkamate were relativey effective and others no effective. The most reliable concentration of the insecticides was 100 times of water solution, and the feasible timing of insecticide application on the basis of single application would be from May 9 to July 1 with Salithion, June 6 with Metalkamate and June 20 with Omethoate and Monocrotophos. The number of pine needles with the larval injured signs (no larvae in the needles) excluded the gall-formed needles were relatively higher in the treatments of Omethoate, Salithion and Monocrotophos than other insecticides. The treatments of Omethoate, Salithion and Monocrotophos caused the lower larval population in the gall than other insecticides.

Susceptibility of Pine Sawyer, Monochamus saltuarius Adults (Coleoptera: Cerambycidae) to Commercially Registered Insecticides (북방수염하늘소의 살충제 감수성)

  • This study was performed to investigate the toxicity of 39 registered insecticides to the susceptibility, systemic effect, and residual effect and control effect against Pine sawyer beetle, Monochamus saltuarius. Eleven kinds of chemicals such as fenitrothion, fenthion, phenthoate, phosphamidon, dinotefuran, actamiprid, clothianidin, thiacloprid, thiamethoxam, esfenvalerate+fenitrothion, and fipronil were showed 100% insecticidal activity both in body spray and twig dipping bioassay. Among these chemicals, fenitrothion and fenthion were showed 100% insecticidal activity when sprayed at 4000 times diluted solutions, and phenthoate, thiacloprid, thiamethoxam and fipronil were showed 100% insecticidal activity when sprayed at 2000 times diluted solution. Root systemic effect was showed 100% mortality in phosphamidon, clothianidin, thiamethoxam, and 77.7% in thiacloprid. In residual effect, fenitrothion and thiamethoxam were showed 80% mortality fifteen days after treatment (DAT), and fenthion, phosphamidon, clothianidin were showed 80% mortality ten DAT, fenitrothion, thiamethoxam, fipronil showed 100% mortality in seven DAT, thiacloprid was showed 100% mortality in three DAT. Fenthion and phenthoate were showed 100% mortality one DAT. In the control effect, 6 kinds of chemicals were showed 100% mortality one DAT and all chemicals showed 100% mortality three DAT.

Discussions on the Efficacy, Phytotoxicity and Residue of Organo-Arsenic Fungicides (유기비소제의 약효 및 약해와 잔류에 관한 종합적 고찰)

  • Since rice high yielding practices with blast resistant varieties induced severe and earlier infestation of sheath blight recently, it is strongly suggested to spray at least 10 days earlier than the present recommendation and more than 2 sprays from early July to early August seems to be essential for the efficient control of sheath blight in the future. Among organo-arsenic fungicides, MAFe (Neo Asozin) is known as the most effective and economic as well as with the advantages of free phytotoxicity and safe residure on rice, and applications of the mixed formulation with other fungicide and insecticide is suitable for the one-hit control of rice diseases and insect pests in connection with cooperative mass control system. Present level of arsenic $(As_2O_3)$ residue in .ice is summarized as 0.2ppm to 0.65 ppm in highest average, and there are more possibilities of extensive MAFe fungicide uses for the efficient and economic control of this disease. During the past few years, the acreage of sheath blight control has been extended several times while the increase of the fungicides used for sheath blight showing about 10 times in quantity and mort than 15 times in amount, and also the fungicide cost estimated for the control is differ from 1 to 12 billion 1on by the use of MAFe or antibiotic fungicide in a year.

Selection of Crop Protectant for Friendly Environmental Control of Spodopfera exigua (Lepidoptera: Noctuidae) (파밤나방(Spodoptera exigua)의 환경친화적 방제를 위한 작물보호제의 선발)

  • For the development of friendly environmental control of the beet armyworm, Spodoptera exigua that is too hard to control in the field, 25 insecticides were chosen from 58 registered to the beet armyworm, and bioassayed. There are 12 insecticides with neurotoxical activities, 10 with insect growth regulators and 3 Bacillus thuringiensis products. Among 12 insecticides with neurotoxical activities, mortality of S. exigua was 100% with emamectin benzoate (EC) and indoxacarb (WP) within 3 and 5 days after application, respectively. Otherwise, WG and SC of indoxacarb, Indoxacarb + etofenprox (WP) and pyridalyl (EW) were showed up to 91 %. Methoxyfenozide + spinosad (SC) was better than any other insect growth regulator as 100% mortality within 3 days after application. And methoxyfenozide (WP), tebufenozide (WP) and methoxyfenozide (SC) were 92% by 5 days. However, 3 kinds of B. thuringiensis products were showed under 35% mortality within 5 days from first spray.
