Phosphorus is one of the major plant growth limiting nutrients, despite being abundant in soils in both inorganic and organic forms. Phosphobioinoculants in the form of microorganisms can help in increasing the availability of accumulated phosphates for plant growth by solubilization. Penicillium oxalicum CBPSTsa, isolated from paddy rhizosphere, was studied for its phosphate solubilization. The influence of various carbon sources like glucose, sucrose, mannitol and sorbitol and nitrogen sources like arginine, sodium nitrate, potassium nitrate, ammonium chloride and ammonium sulphate were evaluated using liquid media with tricalcium phosphate (Ca-P), ferric phosphate (Fe-P) and aluminium phosphate (Al-P). Maximum soluble phosphate of 824 mg/L was found in the amendment of sucrose-sodium nitrate from 5 g/L of Ca-P. Mannitol, sorbitol, and ariginine were poor in phosphate solubilization. While sucrose was better carbon source in solubilization of Ca-P and Al-P, glucose fared better in solubilization of Fe-P. Though all the nitrogen sources enhanced P solubilization, nitrates were better than ammonium In the amendments of ammonium chloride and ammonium sulphate, higher uptake of available phosphates by the fungus was found, and this resulted in depletion of available P in Fe-P amendment Phosphate solubilization was accompanied by acidification of the media, and the highest pH decrease was observed in glucose amendment Among the nitrogen sources, ammonium chloride favored greater pH decrease.
The effects of inorganic salts and feather concentrations on pretense production by Bacillus megaterium F7-1 were investigated. Pretense production was dependent on the presence of phosphates in the medium. Supplementation of medium with calcium ion slightly increased protease production. The highest protease production was obtained at $1.4\%$ feather. The optimal medium contained $2.0\%$ glucose, $0.8\%$ skim milk, $0.06\%\;K_{2}HPO_{4}\%,\;0.04\%\;KH_{2}PO{4},\;0.06\%\;NaCl,\;0.03\%\;MgCl_{2}\cdot6H_{2}O,\;0.002\%\;CaCl_{2}\cdot2H_{2}O,\;and\;1.4\%$ whole feather. By using this optimized medium, increased production of the protease was achieved compared with the cases of using basal medium.
Phytases (myo-inositol hexakisphosphate phosphohydrolase; EC 3.1.3.8) are enzymes which catalyze the hydrolisys of phytate into myo-inositol and inorganic phosphates. Phytases are found in plants and a variety of microorganisms. Aspergillus species were treated with 254 nm of UV irradiation for the screening of phytase overproducing mutant strains. At 15 minute irradiation, the survivals of population were less than 5%, and UV irradiation time was decided at 20 minute for the isolation of mutant strains. Four UV mutant strains in A. oryzae (YUV-47, -169, -341, -511) and six in A. ficuum (FUV-17, -36, -69, -193, -317, -419) were isolated on PSM media containing ammonium phosphate. The specific enzyme activities of A. ficuum mutants are 110 to 140% higher than that of wild type.
Forty yeast strains isolated from soils taken from different locations in Egypt were tested for their P-solubilizing activities on the basis of analyzing the clear zone around colonies growing on a tricalcium phosphate medium after incubation for 5 days at $25^{\circ}C$, denoted as the solubilization index (SI). Nine isolates that exhibited P-solubilization potential with an SI ranging from 1.19 to 2.76 were genetically characterized as five yeasts belonging to the genus Saccharomyces cerevisiae and four non-Saccharomyces, based on a PCR analysis of the ITS1-26S region amplied by SC1/SC2 species-specific primers. The highest P-solubilization efficiency was demonstrated by isolate PSY- 4, which was identified as Saccharomyces cerevisiae by a sequence analysis of the variable D1/D2 domain of the 26S rDNA. The effects of single and mixed inoculations with yeast PSY-4 and Bacillus polymyxa on the P-uptake and growth of corn were tested in a greenhouse experiment using different levels of a phosphorus chemical fertilizer (50, 100, and 200 kg/ha super phosphate 15.5% $P_2O_5$). The results showed that inoculating the corn with yeast PSY-4 or B. polymyxa caused significant increases in the shoot and root dry weights and P-uptake in the shoots and roots. The P-fertilization level also had a significant influence on the shoot and root dry weights and P-uptake in the shoots and roots when increasing the P-level from 50 up to 200 kg/ha. Dual inoculation with yeast strain PSY-4 and B. polymyxa at a P-fertilization level of 200 kg/ha gave higher values for the shoot and root dry weights and P-uptake in the shoots and roots, yet these increases were nonsignificant when compared with dual inoculation with yeast strain PSY-4 and B. polymyxa at a P-fertilization level of 100 kg/ha. The best increases were obtained from dual inoculation with yeast strain PSY-4 and B. polymyxa at a P-fertilization level of 100 kg/ha, which induced the following percentage increases in the shoot and root dry weights, and P-uptake in the shoots and roots; 16.22%, 46.92%, 10.09%, and 31.07%, respectively, when compared with the uninoculated control (fertilized with 100 kg/ha).
Soil samples collected from abounded mines of Boryeong area in South Korea were used in isolating bacterial strains and their capacity to solubilize inorganic phosphates and heavy metal tolerance were assessed in vitro. Three different inorganic phosphate sources (Ca phosphate, Fe phosphate, and Al phosphate) and four different heavy metals (Co, Cd, Pb and Zn) each with three concentrations ($100{\mu}g\;mL^{-1}$, $200{\mu}g\;mL^{-1}$, and $400{\mu}g\;mL^{-1}$) were used. The bacterial isolates PSB-1, PSB-2, PSB-3, and PSB-4 solubilized significantly higher amount of Ca phosphate during the first five days of incubation though subsequent drop in soluble phosphorus level in the medium was observed at the later stage (after 5 days) of the incubation. Solubilization of Ca phosphate and Fe phosphate was concomitant with the acidification of the culture medium compared to the control where it remained constant. Isolated strains could solubilize Fe phosphate to certain extent ($25-45{\mu}g\;mL^{-1}$) though solubilization of Al phosphate was found negligible. All the isolates were tolerant to heavy metals (Cd, Pb, and Zn) up to the concentration of $400{\mu}g\;mL^{-1}$ except PSB-1 and PSB-8, which were shown to be vulnerable to Co even at $100{\mu}g\;mL^{-1}$. Heavy metal tolerant strains should be further evaluated for plant growth promoting activities also under field conditions in order to assess their agricultural and environmental significance.
The kinetics of growing microalga, Spirulina platensis was investigated to treat swine wastes with optimum growth conditions. Temperature was varied from 15 to 40$\circ$C at three different light intensities, 6 W/m$^{2}$, 12 W/m$^{2}$ and 24 W/m$^{2}$. The specific growth rate was increased as temperature increased up to 30$\circ$C. The activation energy was estimated as 13.5 kcal/mol by an Arrhenius relationship. 0.24 (1/day) of specific growth rate was obtained from batch cultivation with 30% swine wastes, compared to 0.31 (1/day) from clean culture. It was found that Spirulina platensis was able to reduce 70-93% of PO$_{4}$$^{3-}$ -P, 67-93% of inorganic nitrogen, 80-90% of COD and 37-56% organic nitrogen by adding various concentrations of swine wastes for 12 days of batch cultivation. Rate constants for removing nitrates and phosphates in treating swine wastes were estimated as 0.17 (1/day) and 0.14 (1/day) in the first order reaction, respectively. 1.52 (g/L) of maximum cell density was maintained at 0.20 (1/day) of dilution rate in continuous culture, adding 20% swine wastes for 30 days. The chemical composition of the biomass obtained from the process showed 58.7% of protein, 11.0% of lipid and 15.6% of ash.
This study assessed the feasibility of iron oxide nanoparticles impregnated with biochar (INPBC), derived from woody biomass, as a stabilizing agent for the stabilization of farmland soil in the vicinity of an abandoned mine through pot experiments with 28 days of lettuce growth. The lettuce grown in the INPBC amended soils increased by more than 100% and the concentrations of inorganic elements (Cu, Ni, Zn) decreased by more than 40%. As, Cd and Pb were not transferred properly from the soils to the lettuce biomass. The bioavailability of arsenic and heavy metals in the INPBC amended soils were decreased by 26%~50%. It seems that the major mechanisms of stabilization were arsenic adsorption on iron oxides, heavy metal precipitation by soil pH increasing and heavy metal adsorption on organic matter. These results revealed that the lower bioavailability of the inorganic pollutants in the soils stabilized using INPBC induced lower transfer to the lettuce. Thus, INPBC could be used as an amendment material for the stabilization of farmland soils contaminated by arsenic and heavy metals. However, a pre-review of the chemical properties of the amended soil must be performed prior to applying INPBC in farmland soil because the concentration of the nutrients in the soil such as available phosphates and exchangeable cations (Ca, Mg, K) could be decreased due to adsorption on the surface of the iron oxides and organic matter.
Acidic nucleotidase from Asfiergilius nlger has been partially purified by Sepharose CL-6B gel filtration and DEAE-Sephacel ion exchange chromatography. The optimum pH and temperature for the enzyme reaction with 5'-AMP or 3'-AMP as a substrate were 4.5 and 55%, respectively. However, the optimum temperature became 70% when p-nitrophenyl phosphate was used as a substrate. The enzyme was stable at acidic pH. The enzyme activity was not affected by addition of various nucleotides, nucleosides and inorganic phosphates. Ferric, aluminium, vanadate and molybdate ions inhibited the enzyme activity dramatically. In kinetic studies, $K_m$), values for 3'-AMP, 5'-AMP and p-nitrophenyl phosphate were 1.39 mM, 1.5 mM and 5.77 mM, respectively. The substrate efficiency ($V_{max}/K_m$) shows 3'-AMP is the prefered substrate for the enzyme among tested substrates.
The fouling of Nanofiltration membrane (NF) was examined using wastewater containing reactive black dye RB5 of 1500 Pt/Co color concentrations with 16890 mg/l TDS collected from El-alamia Company for Dying and Weaving in Egypt. The NF-unit was operated at constant pressure of 10 bars, temperature of 25℃, and flowrate of 420 L/min. SEM, EDX, and FTIR were used for fouling characterization. Using the ROIFA-4 program, the total inorganic fouling load was 1.07 mM/kg present as 49.3% Carbonates, 10.1% Sulfates, 37.2% Silicates, 37.2% Phosphates, and 0.93% Iron oxides. The permeate flux, recovery, salt rejection and mass transfer coefficients of the dye molecules were reduced significantly after fouling. The results clearly demonstrate that the fouling had detrimental effect on membrane performance in dye removal, as indicated by a sharp decrease in permeate flux and dye recovery 68%. The dye mass transfer coefficient was dropped dramatically by 34%, and the salt permeability increased by 14%. In this study, all the properties of the membrane used and the fouling that caused its poor condition are identified. Another study was conducted to regeneration fouled membrane again by chemical methods in another article (Abdel-Fatah et al. 2017).
Two phosphate solubilizing Pantoea strains (P. agglomerans and P. rodasii) were employed in elucidating their phosphate solubilizing potential under different carbon and nitrogen sources, pH, temperature and salt conditions. Plant growth promoting characteristics such as ACC deaminase activity, indole acetic acid (IAA), HCN, ammonia, and siderophore production of the two strains were assessed in vitro. Potential applicability of the strains as bio-inoculants was also evaluated in pot experiments conducted under green house conditions. Phosphate solubilization measured as the amount of phosphorous released into the medium was recorded as 810 and $788{\mu}g\;ml^{-1}$ respectively by P. agglomerans and P. rodasii. Glucose at the rate of 2% was found be the best carbon source, while $(NH_4)_2SO_4$ was the best nitrogen source for both strains. Despite a slight decrease in phosphate solubilization observed at higher temperature, pH and salt concentrations, both strains could withstand against a range of temperature ($30-35^{\circ}C$), pH (7-9) and the presence of NaCl (up to 5%) without much compromising the phosphate solubilization. Different plant growth promoting traits (ACC deaminase activity, IAA, HCN, ammonia, and siderophore production) of the strains and their ability to promote the growth of green gram seedlings indicate that both strains possess high potential to be used as bio-inoculants.
