This study was conducted to establish an efficient screening system for resistant tomato to bacterial wilt caused by Ralstonia solanacearum. Under several conditions such as inoculation methods, growth stages of tomato seedlings, inoculum concentrations, and incubating temperatures after inoculation, development of bacterial wilt on nine resistant or susceptible cultivars of tomato was investigated. To inoculate by drenching the non-cut roots with the bacterial suspension was better to distinguish resistance and susceptibility of tomato cultivars than by drenching the cut roots using scalpel. And 'Hawaii7996' a resistant tomato to R. solanacearum showed high resistance at all the tested conditions including growth stages (3-, 6-, 8-, 10-leaf stages), inoculum concentrations ($OD_{600}=0.1-0.4$) and incubation temperatures (25, 30, $35^{\circ}C$). On the other hands, susceptible cultivars represented disease index of 3.7 and 3.9 at 6- and 8-leaf stages, respectively. At 3- and 10-leaf stages, the cultivars demonstrated lower disease severity of 2.1 and 0.5, respectively, than at 6- and 8-leaf stages. When the inoculated seedlings were incubated in growth chambers of 25, 30 and $35^{\circ}C$, disease severity of susceptible cultivars was significantly greater at 30 and $35^{\circ}C$ than at $25^{\circ}C$. In addition, the level of resistance of the tomato cultivars was not significantly affected by inoculum concentrations of $OD_{600}=0.1-0.4$. On the basis of the results, we suggest an efficient screening method to measure resistance level of tomato cultivars to bacterial wilt. The eight-leaf stage seedlings transplanted 7 days before inoculation, are inoculated with R. solanacearum by drenching the non-cut roots with a bacterial suspensions ($OD_{600}=0.4$) to give inoculum volume of 50 ml/soil l. The inoculated plants are incubated in a growth room at $30^{\circ}C$ for 12-13 days with 12-hour light a day.
A field experiment was conducted in Agronomy farm at an altitude of 1350 m. above sea level in a randomized complete block design with three replications and eight treatments on vegetable type soybean in 2005. The objective was to see the effect of vermicompost alone and in combination with Rhizobium inoculum and mineral fertilizers on the yield of vegetable soybean. The result showed higher number of nodules from the non inoculated plot, however, the nodules weight was highest from the inoculated plots. No significant difference was notice statistically on roots and shoots dry weight. Soybean grain was obtained highest (32.3%) over the non inoculated plot followed by vermicompost plus inoculated, however, there was non significant difference. There was a slight increment on grain and straw yield from the inoculated plots over the non inoculated.
Several culture conditions affecting cellulose production by a newly isolated Acetobacter sp. A9 were examined by cultivating cells under shaking cultures. The inoculum size in the range of 1-10% (v/v) did not influence cellulose production. Maximum cellulose production was obtained with 200 rpm of agitation speed. The cells grown in the 75 ml of medium in a 250-ml conical flask produced the highest level of cellulose. The strain was able to produce cellulose at 25-3$0^{\circ}C$ with a maximum at 3$0^{\circ}C$. Cellulose production occurred at pH 4.5-7.5 with a maximum at pH6.5.
This study characterizes the growth of white rot fungi Phanerochaete chrysosporium IFO 31249) and lignin peroxidase(LiP) activity in different submerged culture media. P. chrysosporium was grown in the form of pellet of various sizes from a spore inoculum under shaking liquid culture condition. While the growth of mycelia was higher under the nitrogen-sufficient culture than under the nitrogen-limited culture, ligninase activity was relatively lower. The lignin peroxidase appeared in nitrogen-limited culture and was suppressed by excess nitrogen. High level(40U/l) of lignin peroxidase activity was obtained in the growth medium containing 1.5mM veratryl alcohol, a secondary metabolite of P. chrysosporium. Lignin peroxidase production was not observed under conditions of nitrogen sufficiency or in balanced media, suggesting that control parameters could increase the activity by manipulating the secondary metabolism.
The effect of field sanitation using ecological characters of the pathogen was investigated for controlling Phomopsis seed decay in soybean. Field sanitation which was eliminated the inoculum by removing host debris, abscised petioles and cotyledones out of field, reduced remarkably infection percentage of pods and seeds by Phomopsis spp. as compared to the inoculated field. Neverthless, seed infection was 28.7% in the sanitized field. The fields sanitized by benlate application around the soybean plants also decreased seed infection with Phomopsis spp. Total seed infection including that with miscellaneous pathogens occurred as much as 75∼79% to the no application and their control values were 34∼42% over the routine application schedule. Even though it was not satisfactory, field sanitation seemed to be effective in controlling Phomopsis seed decay when infection pressure was low level. Diaporthe phseolorum va. sojae, D. phaseolorum var. caulivora and Phomopsis longicolla were mostly identified from soybean seeds and Colletotrichum truncatum, Cercospora kikuchiana were also isolated in sequence. Field sanitation did not significantly increase in soybean yield over the no application, while routine application schedule did in field.
Experiments were conducted to determine the method of nioculation and inoculum level, and the optimum age of pepper seedlings for evaluation of resistance to bacterial wilt in pepper. Injection of bacterial suspension(107~108 cells/ml) to the leaf axil of the 3rd or 4th leaf of pepper, and drenching the soil planted with pepper seedling after wounding roots with scalpel, resulted in good varietal difference in resistance to bacterial wilt. PI377688, PI358812 and PI369994 of 298 open-pollinated lines and 10 hybrids tested for resistance to bacterial wilt were highly resistant and such local cultivars as Masan, Anjinbaengi, Kimyongcho and Punggakcho moderately resistant.
Optimization of culture conditions for L-asparaginase production by submerged fermentation of Aspergillus terreus MTCC 1782 was studied using a 3-level central composite design of response surface methodology and artificial neural network linked genetic algorithm. The artificial neural network linked genetic algorithm was found to be more efficient than response surface methodology. The experimental $_L$-asparaginase activity of 43.29 IU/ml was obtained at the optimum culture conditions of temperature $35^{\circ}C$, initial pH 6.3, inoculum size 1% (v/v), agitation rate 140 rpm, and incubation time 58.5 h of the artificial neural network linked genetic algorithm, which was close to the predicted activity of 44.38 IU/ml. Characteristics of $_L$-asparaginase production by A. terreus MTCC 1782 were studied in a 3 L bench-scale bioreactor.
Lactobacillus acidophilus KFCC12731 and Saccharomyces cerevisiae KFCC32017 were incubated together in soymilk and the conditions for acid production were investigated. The acid production of Lactobacillus acidophilus was much higher when this organism was incubated with Saccharomyces cerevisiae in soymilk than when it was incubated alone. Optimum acid production by the mixed cultures of Lactobacillus acidophilus and Saccharomyces cerevisiae was achieved with the following conditions; a temperature of 34$^{\circ}C$, a 3:7-8:2 (OD 660) ratio of Lactobacillus acidophilus to Saccharomyces cerevisiae at inoculum, a 1.5% level of sucrose fortification or a 2.0-3.0 % level of skim milk powder fortification and a culture time of 12 hours or more.
Objective: The aim of the study was to isolate gossypol-degrading bacteria and to assess its potential for gossypol degradation. Methods: Rumen liquid was collected from fistulated cows grazing the experimental pasture. Approximately 1 mL of the rumen liquid was spread onto basal medium plates containing 2 g/L gossypol as the only source of carbon and was then cultured at $39^{\circ}C$ to isolate gossypol-degrading bacteria. The isolated colonies were cultured for 6 h and then their size and shape observed by microscope and scanning electron microscope. The 16S rRNA gene of isolated colonies was sequenced and aligned using National Center for Biotechnology Information-Basic Local Alignment Search Tool. The various fermentation conditions, initial pH, incubation temperature, inoculum level and fermentationperiod were analyzed in cottonseed meal (CSM). The crude protein (CP), total gossypol (TG), and free gossypol (FG) were determined in CSM after fermentation with isolated strain at $39^{\circ}C$ for 72 h. Results: Screening results showed that a single bacterial isolate, named Rumen Bacillus Subtilis (RBS), could use gossypol as a carbon source. The bacterium was identified by 16S rDNA sequencing as being 98% homologous to the sequence of Bacillus subtilis strain GH38. The optimum fermentation conditions were found to be 72 h, $39^{\circ}C$, pH 6.5, moisture 50%, inoculum level $10^7cell/g$. In the optimum fermentation conditions, the FG and TG content in fermented CSM decreased 78.86% and 49% relative to the control. The content of CP and the essential amino acids of the fermented CSM increased respectively, compared with the control. Conclusion: The isolation of a gossypol-degrading bacterium from the cow rumen is of great importance for gossypol biodegradation and may be a valuable potential source for gossypol-degradation of CSM.
Through the laboratory and vinyl house experiments, the effects of inoculum density, plant age and temperature on the incidence of Phytophthora crown rot of pepper (Capsicum annum L.) were investigated. The propagule survival was greater in the natural soil than in autoclaved soil within first 2 weeks when the sporangial suspension of the pathogenic fungus was incorporated into soil, thereafter the survivability reduced rapidly. The propagule was not detectable in 35 days by means of Papavizas selective medium neither in natural nor in autoclaved soil. At least 5 sporangia per gram soil were required to induce crown rot for 30 days old pepper seedlings. Further increase in inoculum concentration above this threshold level resulted in higher disease incidence and shorter incubation period. When the same amount of inoculum was infested, higher disease incidence was observed for younger plants until 3 weeks after inoculation. On the other hand after 4 weeks this tendency was not extended any more. Younger plants were recognized as having shorter incubation period upon infection, however, the days from first symptom appearance to complete death were not significantly different among differently aged seedlings. Exposure of inoculated pepper seedlings to $25^{\circ}C$ resulted in highest infection rates and followed by those to $30^{\circ}C\;and\;20^{\circ}C$ but no disease was found at $15^{\circ}C\;and\;35^{\circ}C$ for 10 days. When the plants previously incubated at different temperature for 10 days were moved to $25^{\circ}C$ room temperature, prior exposure to $20^{\circ}C\;and\;30^{\circ}C$ brought continuous disease development. Even those plants preincubated at $15^{\circ}C$ were diseased up to 50%. But the prior exposure to $35^{\circ}C$ induced no symptom developed, indicating no seedlings infected at all.
