  • Antigenicity of Recombinant Granulocyte macrophage colony stimulating factor(LBD-005), a newly developed drug for hematopoietic growth, was investigated in mice and guinea pigs. 1. Mice showed production of antibodies against LBD-005 (100mg/kg) with alumin]m hydroxide gel(alum) as an adjuvant, judged by the heterologous anaphylaxis(PCA) test using rats. On the other hand, antibodies against ovalbumin(OVA) inoculated with alum were definitely detected. 2. In the studies with guinea pigs, both the inoculation of LBD-005 only and of LBD-005 with complete Freund's adjuvant(CFA) as an adjuvant did not produce positive reactions in homologous passive cutaneous anaphylaxis (PCA).

Growth Characteristics of Teratoma Shoot derived from Crown Gall Callus of Nicotiana tabacum cv. NC2326 (연초 Crown Gall Callus 유래 Teratoma Shoot의 생장특성)

  • The present study was conducted to obtain some basic information on the shoot formation from crown gall callus and the characteristics of teratoma shoot derived from crown gall induced by inoculation of Agrobacterium tumefaciens C58. Crown gall callus could be continuously cultured on the phyohormone free basic medium. The growth of crown gall callus was inhibited when BA were added to the cultural media. Shoot formation from crown gall callus fail to be initiated except teratoma shoot which induced on the phytohormone free medium after several subculture on rare occasions. Teratoma shoot could not form root and grow as normal shoot. Addition of BA to cultural media was not effective for shoot elongation, reduction in multiple shoot formation, but IBA was somewhat effective for shoot elongation of teratoma shoot, never for root formation.

Effects of Chlorhexidine Gluconate and Povidone Iodine on Wound Healing in Rabbits (Chlorhexidine Gluconate 및 Povidone Iodine이 가토의 창상치유에 미치는 효과)

  • To investigate the effect of chlorhexidine gluconate and povidone iodine on wound healing, their antimicrobial activity and their effects on wound contraction in rabbits were studied. Staphyococcus aureus (1${\times}$10$\^$7/) was inoculated on the full thickness skin defects (2${\times}$2cm) on the back. Antiseptics were applied on the defects 2 hours after Inoculation. The defects were swabbed for bacterial culture 24 and 48 hours after the application of antiseptics. Percentage of wound contraction, based on wound area measured on the day of surgery was calculated for each wound on days 1,7 and 14. The 0.5% chlorhexidine gluconate solution had more effective antimicrobial activity against Staphyococcus aureus in wounds than 0.05% chlorhexidine gluconate solution (p<0.05), and povidone iodine solution and saline (p<0.01). Percentages of wound contraction were not significantly different among different dilutions of the antiseptics and saline. Present study indicated that the antimicrobial activity of chlorhexidine gluconate in wound was superior to that of povidone iodine.

An Acute Pathophysiology of Environmental Strains of Cryptococcus neoformans Isolated from a Park in Busan

  • The present study was carried out to elucidate whether an environmental strain of Cryptococcus neoformans (environmental C. neoformans) isolated from an environmental source in a park of Busan has an acute pathophysiological effect in rats. On the second day after peritoneal inoculation of environmental C. neoformans, adverse effects occurred from the viewpoint of hematology and biochemistry. Eosinophil damages and crystal formations were found in the blood. Disturbances in cytokines production were observed in the cerebral and pulmonary tissues. Fungal budding existed in the brain, lung, liver and kidney. Tissue injury findings such as inflammation, leukocyte infiltration, bleeding, or degeneration were found in the brain, lung, liver and kidney. The present data suggest that the environmental C. neoformans can cause systematically harmful effects even for short periods of infection (two days of cryptococcal infection) and the adverse effects are summarized as immune derangements and biochemical and/or histological dysfunction and injury on major organ such as the brain, lung, liver and kidney in the immunocompetent hosts. Further studies should be focused on comparing the differences between environmental and clinical strains of C. neoformans.

Effect of Bradyrhizobium japonicum on Chlorophyll Content, Nodulation, and Plant Growth in Soybean

  • Study on effectiveness of Bradyrhizobium japonicum on soybean [Glycine max (L.) Merr.], local cultivar 'Sathiya' were carried out in the garden soil of Trib-huvan University. Different parameters like nodulation, chlorophyll content in fresh leaves and growth of plant in inoculated and uninoculated plant was studied. Pot experiment was conducted in the green house to evaluate the effectiveness of B. japonicum on soybean. It was observed that B. japonicum inoculation increased the number of nodules, shoot length of plant and total chlorophyll content in fresh leaves of soybean plant. However, root length was decreased in all inoculated plants.

Isolation of Weissella strains as potent probiotics to improve antioxidant activity of salted squid by fermentation

  • The aim of this study was to enhance the antioxidant activity of salted squid by inoculation of two Weissella spp. strains (W. cibaria FB-069 and W. viridescens FB-077) isolated from traditional Korea salted squid. The safety and probiotic potential characteristics of these two strains were evaluated. The safety of these strains was analyzed based on hemolytic activity, mucin degradation, biogenic amino production, and resistance to antibiotics. These lactic acid bacteria showed probiotic potential, including resistance to gastrointestinal tract conditions, adhesion to Caco-2 cells, and aggregation. The low-salted squid fermented with Weissella strains had consistently higher antioxidant activity through changing their amino acid profiles. Therefore, W. cibaria FB-069 and W. viridescens FB-077 might be good candidates for fermentation of salted squid to develop functional food with enhanced health benefits.

  • This study was aimed to develop a value-added fermented products from rice and apple pomace using Bifidobacterium fermentation. The Bifidobacterium fermentation system of the mixture of rice and apple pomace was developed, and the physicochemical properties of the products were investigated. After 4 different bifidobacteria were compared for their fermentation capability and sensory properties of the fermented product, Bifidobacterium FBD-13 and FBD-22 were selected as appropriate strains for the fermentation of saccharified rice solution(SRS). The optimum inoculation level was 2% and the optimum fermentation time was 42 hrs. When wet apple pomace(WAP) was added to SRS, it contributed to the improvement of sensory properties of the fermented products and the optimum mixing ratio was 40% WAP and 60% SRS in weight. For the fermentation of the mixture of WAP and SRS, Bifidobacterium FBD-27 and FBD-22 were selected as suitable strains.

  • This study was done to identify Newcastle disease virus(NDV) antigens in paraffin sections of various organs from experimentally NDV-infected chicken using peroxidase-antiperoxidase(PAP) technique. Sections were Incubated with rabbit anti-NDV polyclonal as first antibody, followed by incubation with goat anti-rabbit IgG conjugate and peroxidase anti-peroxidase ( PAP ). Positive reactions were often detected in the epithelim of trachea and in the lymphocyte of spleen at 24 hours after virus inoculation. The viral antigen was localized mainly in the cytoplasm of infected cells. The method approved to be highly specific for the indetification of NDV and allowed a precise localization of the viral antigens in infected cells.

  • This study has been conducted to assess and analyze the damage of Codonopsis lanceolata by differential inoculation levels of Meloidogyne hapla in pot and plot. As the density of M. hapla increased, early growth of C. lanceolata was inhibited 60% in vine and 54% in root. However, growth of C. lanceolata was not remarkably different by density of M. hapla in the plot test. This result may be attribute to low density of M. hapla in the plot.

Immunohistochemistry and RT-PCR for pathogenesis of Newcastle disease in chickens (닭 뉴캣슬병의 발병기전 규명을 위한 RT-PCR 및 면역조직화학적 연구)

  • The present experiment was carried out to study the pathogenesis of Newcastle disease(ND), ND virus (NDV) antigens and genes in various organs from NDV inoculated chickens were detected by immunohistochemistry and RT-PCR. Immunohistochemically, NDV antigens were detected in the spleen, thymus, cecal tonsil, proventriculus, trachea and lungs at 12 hour post-inoculation (hpi). Viral antigens were localized mainly in the cytoplasm of lymphocytes and macrophages. After 48 hpi, clinical findings of the affected chickens were open-mouth breathing, conjunctivitis, watery diarrhea and edema around the eye and neck. After 72 hpi, chickens showed muscular tremor, paralysis of the legs and wings, and coma. Histopathological results consist of multi-focal necrosis with hemorrhages in lymphoid aggregates of the intestinal tracts, necrosis of the lymphoid tissues, neuronal degeneration and necrosis, and perivascular cuffing. Using RT-PCR, virus genes were detected in the spleen and proventriculus at 48 hpi, and in the brain at 60 hpi.
