This study was conducted to investigate the effect of addition of lactic acid bacteria inoculants on quality of rye silage harvested at early heading stage. The nutritive values in lactic acid bacteria (LAB) inoculated group showed similar results to control. The pH of rye silage in LAB inoculation significantly decreased as compared to control (p<0.05). In addition, the content of lactic acid in LAB inoculation significantly increased (p<0.05), but the content of acetic acid in LAB treatments decreased. In addition, lactic acid bacterial counts in LAB inoculation significantly increased as compared to control (p<0.05). Therefore, we suggest that rye silage could be improved by LAB inoculation.
To investigate the etiology, pathogenicity and virological properties of NYJ-1-87 strain of Aujeszky's disease virus (ADV) that was isolated from the diseased piglet in Korea, the virus at $10^{6.0}TCID_{50}/0.1ml$ was inoculated intranasally and subcutaneously into 30 to 35 days-old piglets. Results obtained through the experiments were summarized as follows. 1. Ten of the infected piglets were clinically observed for 15 days. On the 2nd day post-inoculation(pi), the signs of pyrexia, anorexia and convulsion were noted. On the 4th to 7th days pi, nervous signs of incoordination and intermittent spasm were shown in the most of piglets, and one out of 5 piglets infected intranasally was died with severe nervous signs at the 7th day pi. The signs became relieved on the 8th day pi and all of remainder were completely recovered on the 13th to 14th days pi. 2. In hematological study, prominent decrease in the number of total leukocyte and lymphocyte was shown in the ADV-infected piglets on the 6th day pi. On the 8th day pi, the cell numbers were slightly increased and returned to normal level on the 10th day pi. 3. Viral excretion of the ADV-inoculated piglets was examined by swabbing of nasal and oral cavities, and rectal feces. During the periods of the 3rd to 11th days pi, the virus was excreted intermittently from nasal and oral cavities, and rectal feces. The nasal excretions were shown the highest virus concentration of $10^{5.2}TCID_{50}/0.1ml$ at the 5th day pi. 4. Recovery of the inoculated virus from various organs of the piglets that were died or experimentally slaughtered was attempted, and the virus was isolated from the tissues of brain and tonsil by the cultured cell-inoculation method. The highest recovery rate was noted in the tonsil. By indirect immunofluorescence antibody assay using ADV-monoclonal antibody, the viral antigens were detected in tissues of spleen and liver as well as brain and tonsil on the 7th to 9th days pi. The virus was not isolated from blood and the tissues of lung and kidney throughout the experiments. 5. Titers of virus neutralizing antibody in the piglets experimentally infected with ADV became increased after the 6th to 9th days pi in both of intranasal and subcutaneous inoculation showing the highest titers of 64 to 128 on the 29th day pi. When the antibody levels were measured by radial immunodiffusion enzyme assay, the reactive diameter was enlarged to be positive after the 4th to 6th days pi in both of intranasal and subcutaneous inoculation showing the largest diameter of 13 to 14mm on the 29th day pi.

BACKGROUND: Soil salinity causes reduction of crop productivity. Rhizosphere microbes have metabolic capabilities and ability to adaptation of plants to biotic and abiotic stresses. Plant growth-promoting bacteria (PGPB) could play a role as elicitors for inducing tolerance to stresses in plants by affecting resident microorganism in soil. This study was conducted to demonstrate the effect of selected strains on rhizosphere microbial community under salinity stress. METHODS AND RESULTS: The experiments were conducted in tomato plants in pots containing field soil. Bacterial suspension was inoculated into three-week-old tomato plants, one week after inoculation, and -1,000 kPa-balanced salinity stress was imposed. The physiological and biochemical attributes of plant under salt stress were monitored by evaluating pigment, malondialdehyde (MDA), proline, soil pH, electrical conductivity (EC) and ion concentrations. To demonstrate the effect of selected Bacillus strains on rhizosphere microbial community, soil microbial diversity and abundance were evaluated with Illumina MiSeq sequencing, and primer sets of 341F/805R and ITS3/ITS4 were used for bacterial and fungal communities, respectively. As a result, when the bacterial strains were inoculated and then salinity stress was imposed, the inoculation decreases the stress susceptibility including reduction in lipid peroxidation, enhanced pigmentation and proline accumulation which subsequently resulted in better plant growth. However, bacterial inoculations did not affect diversity (observed OTUs, ACE, Chao1 and Shannon) and structure (principle coordinate analysis) of microbial communities under salinity stress. Furthermore, relative abundance in microbial communities had no significant difference between bacterial treated- and untreated-soils under salinity stress. CONCLUSION: Inoculation of Bacillus strains could affect plant responses and soil pH of tomato plants under salinity stress, whereas microbial diversity and abundance had no significant difference by the bacterial treatments. These findings demonstrated that Bacillus strains could alleviate plant's salinity damages by regulating pigments, proline, and MDA contents without significant changes of microbial community in tomato plants, and can be used as effective biostimulators against salinity stress for sustainable agriculture.
Purpose ; In order to study the effect of Houttuyniae herbal acupuncture on growth of melanoma B 16 and immunity in mice, the Control group with normal saline acupunchure after subcutaneous inoculation of B16BL6 cells, the Sample I with Houttuyniae herbal acupuncture manufactured by distilled water extraction method after subcutaneous inoculation of B16BL6 cells and the Sample II with Houttuyniae herbal acupuncture manufactured by alcohol extraction method after subcutaneous inoculation of B16BL6 cells were divided. Methods ; To evauluate the effect of Houttuynias herbal acupuncture on growth of melanoma B16 in mice, the weight of mouse melanoma, body weight, spleen index( $\sqrt(Weight of spleen/Body weight){\times}100$), lymphocytes numbers in mouse peripheral blood and spleen tissue, the percentage of CD4+ T cells, CD8+ and the ratio of CD4+/CD8+ in mouse peripheral blood and spleen tissue are measured. Results ; In study of the weight of mouse melanoma, Houttuyniae herbal acupuncture groups(Sample I, Sample II) showed statistically significant inhibitory effect. And in study of effect of reduction of change in body weight of mouse, Sample I showed statistically significant inhibitory effect, too. In study of reduction of spleen index, lymphocytes numbers in mouse spleen tissue, and the percentage of CD4+ and the ratio of CD4+/CD8+ T cells in mouse peripheral blood, Houttuyniae herbal acupuncture groups(Sample I, Sample II ) showed with statistically significant inhibitory effect. Also, Sample I showed inhibitory effect of reduction of the percentage of CD8+ T cells in mouse peripheral blood and spleen tissue and Sample II showed inhibitory effect of reduction of the percentage of CD4+ and the ratio of CD4+/CD8+ T cells in mouse spleen tissue with statistical significance. Conclusions ; The inhibitory effect of reduction of lymphocytes numbers in mouse peripheral blood and IL-2 productivity, Houttuyniae herbal acupuncture groups(Sample I, Sample II) didn't show significant effect.
This study was carried out to investigate the influence of inoculation with Bradyrhizobia and lime types on the nutrient content and yield of peanut. Inoculation size of Bradyrhizobium sp. HCR-46 was $8.2{\times}10^7$ cells per seed. Slaked lime, magnesium lime, and calcium carbonate were applied in an amount required for neutralization of soil. 150kg of calcium sulfate was fertilized to 10a of the experimental field. Sowing space was $40{\times}25cm$ under vinyl mulching. Leaves and stems for assay were sampled at 100 day after sowing. The obrained results are as follows. 1. Number and dry weight of nodule as well as dry weight of aerial part of peanut plant increased by inoculation with B. sp. and were the highest in calcium carbonate application. 2. Inoculated with B. sp., the contents of T-N, $K_2O$, MgO, allantoin, ammonia, free amino acid and chlorophyll increased, but that of nitrate decreased. 3. The contents of T-N, free amino acid, and chlorophyll were higher in the treatment of calcium carbonate, those of $K_2O$, MgO, allantoin, ammonia were higher in magnesium lime application, and those of CaO and nitrate were higher in slaked lime fertilization than any other lime types. 4. Contents of total sugar and starch in stem at 100 days were higher in the treatment of uninoculation than inoculation with B. sp., and those were highest in the calcium sulfate application than the other lime types. 5. Inoculated with B. sp. length of main stem and number of pods increased significantiy. Yield of seed was higher in inoculation with B. sp. than in uninoculation by 64%, and in the order of carbonate, magnesium lime, slaked lime, in calcium sulfate and non-application was the contribution of soil treatments to yield increases.
The objective of this study was to elucidate the tolerance of woody plants to simulated acid rain in relation to mycorrhizal inoculation. Germinating seedlings of Robinia pseudoacacia were planted in 1I pots with autoclaved soil mixture of vermiculite, sand and nursery soil at 1:1:1 ratio. Each pot was inoculated with both crushed root nodules from a wild tree of the same species and commercial arbuscular mycorrhizal inoculum of Glomus intraradices at the time of planting the seedlings. Simulated acid rains at pH 2.6, 3.6, 4.6, and 5.6 were made by mixing sulfuric acid and nitric acid at 3: 1 ratio. Each pot received nutrient solution without N and P, and was also supplied with 180 ml of the one pH level of the acid rains once a week for 50 days. The plants were grown in the green house. At the end of experimental period, plants were harvested to determine contents of chlorophyll, mineral nutrients and net photosynthesis in the tissues, dry weight of the plants, and mycorrhizal infection in the roots. Mycorrhizal infection rate was significantly reduced only at pH 2.6, which meant vitality of G intraradices was inhibited at extremely low pH. Height growth, dry weight production, nodule production and chlorophyll content were increased by mycorrhizal infection in all the pH levels except pH 3.6. Particularly, mycorrhizal inoculation increased root nodule production by 85% in pH 5.6 and 45% in 4.6 treatments. But the stimulatory effect of mycorrhizal inoculation on nodule production was reduced at pH 3.6 and 2.6. Net photosynthesis was increased by mycorrhizal infection in all the pH levels. The phosphorus(P) content in the tissues was increased by 43% in average by mycorrhizal inoculation, which was statistically significant except in pH 2.6. It was concluded that mycorrhizal inoculation of Robinia pseudoacacia would enhance growth and resistance of the plants to acid rain by improving the photosynthesis, phosphorus nutrition, and more nodule production.

Moth orchid plants with yellowing blight and root rot symptoms were collected, and a total of 54 isolates of Fusarium spp. was obtained from roots and leaf bases of the diseased plants. The isolates were identified based on their morphological characteristics. Out of the 54 isolates of Fusarium spp., 42 isolates were identified as F. solani, 5 isolates as F. oxysporum, and 7 as F. proliferatum. Isolates of the three Fusarium spp. were tested for pathogenicity to moth orchid plants by artificial inoculation. All the Fusarium spp. induced root rot of the host plants. The symptoms progressed up to the basal part of the leaves, which later caused yellowing blight. The symptoms induced on the plants by artificial inoculation with the Fusarium spp. isolates were similar to those observed in greenhouses. The present study reveals that F. oxysporum, F. proliferatum, and F. solani cause root rot of moth orchid, and that F. solani is the main pathogen of the disease.

In order to assay one lot of the potency of tuberculin for intradermic use, four guinea pigs sensitized with heat killed M. tuberculosis were used by the method originally of the Agriculture Research Service Center(A. R. S.) and Bureau of Animal. Industry(B. A. I.) in the United. States of America in which guinea pigs were used on 21st day after sensitization. There is doubt whether the guinea pigs are sensitized or not. Therefore, this study has taken the method of Middle Brook-Dubos hemagglutination test, and observed the humoral hemagglutinating antibody in the blood. The results obtained are as follows. 1. The maximum titer of hemagglutinating antibody was demonstrated 30days after sensitizing the guinea pigs with killed tubercle bacilli. The maximum titer was continued for a period of 45 days and thereafter declined gradually. 2. The swelling and redness of the region of the sensitized guinea pigs was shown to be reached to the maximum size within 24 hours after intradermal inoculation with HCSM tuberculin and gradually reduced 96 hours after inoculation. The swelling and redness could not be detected by macro-observation after 14 days.
This study focused on evaluating the phytoextraction of heavy metals (Co, Pb, and Zn) induced by bioaugmentation of button mushroom compost (BMC) in Helianthus annuus (sunflower). When the potential ability of BMC to solubilize heavy metals was assessed in a batch experiment, the inoculation with BMC could increase more the concentrations of water-soluble Co, Pb, and Cd by 35, 25, and 45% respectively, compared to those of non-inoculated soils. BMC-assisted growth promotion and metal uptake in H. annuus was also evaluated in a pot experiment. In comparison with non-inoculated seedlings, the inoculation led to an increase in the growth of H. annuus by 27, 25, and 28% in Co-, Pb-, and Zn-contaminated soils, respectively. Moreover, enhanced accumulation of Co, Pb, and Zn in the shoot and root systems was observed in inoculated plants, where metal the translocation from root to the above-ground tissues was also found to be enhanced by the BMC. Evidently, these results suggest that the BMC could be effectively employed in enhancing the phytoextraction of Co, Pb, and Zn from contaminated soils.

This study was conducted to investigate the effect of antagonistic bacteria and pathogenic fungi on growth and yields of white clover(Trifo1iurn repens L.) in continuous cropping soil(CCS) and non-continuous cropping soil(NCCS). The gowth experiment of white clover was conducted at pots in a vinyl house. White clover was established by seeding into pots of 12 cm in diameter and 9 cm in depth containing 1:l mixture of soil and vermiculite with antagonistic bacteria and pathogenic fungi. In dark culture experiment, white clover lived longer in treatment of antagonistic bacteria than in treatment of control, but lived shorter in CCS than in NCCS. Dry weight of white clover was increased by the inoculation of the antagonistic bacteria(p< 0.05), but decreased by the inoculation of the pathogenic fungi(p< 0.05) both CCS and NCCS. In conclusion, bacterization of white clover with antagonistic bacteria enhances the growth and yield of white clover.
