Transcription activator-like effector nucleases (TALENs) are powerful tools for targeted genome editing in diverse cell types and organisms. However, the highly identical TALE repeat sequences make it challenging to assemble TALEs using conventional cloning approaches, and multiple repeats in one plasmid are easily catalyzed for homologous recombination in bacteria. Although the methods for TALE assembly are constantly improving, these methods are not convenient because of laborious assembly steps or large module libraries, limiting their broad utility. To overcome the barrier of multiple assembly steps, we report a one-step system for the convenient and flexible assembly of a 180 TALE module library. This study is the first demonstration to ligate 9 mono-/dimer modules and one circular TALEN backbone vector in a one step process, generating 9.5 to 18.5 repeat sequences with an overall assembly rate higher than 50%. This system makes TALEN assembly much simpler than the conventional cloning of two DNA fragments because this strategy combines digestion and ligation into one step using circular vectors and different modules to avoid gel extraction. Therefore, this system provides a convenient tool for the application of TALEN-mediated genome editing in scientific studies and clinical trials.

As we have become more interested in 'competency' that means ability to do something around the world, the competency of the best performers has also been introduced in the university curriculum as a concept of core competency. Research continues on why this competency-based education is needed compared to existing academic-oriented education, how it can be introduced into existing curricula, and how it can be developed and evaluated in detail. This study develops and validates core competency assessment tools that can diagnose core competencies of engineering students. Therefore, this research paper conducted a literature review related to core competencies and also core competency assessment tools of university students. It seeks to explore the implications of core competency assessment tools for engineering students and then lay the foundation for competency-based teaching and learning at engineering colleges. And also it defines the concepts of core competencies and each core competency of engineering students through prior research analysis of competence, core competence, and core competence of university students. The primary core competency assessment tool consisted of sub-factors and questions of core competencies. It were modified through the expert validation of the primary one and then it was used as a core competency assessment tools for preliminary investigation. The core competency assessment tools for engineering students are consisted of 6 competencies, 22 sub-factors, and 91 questions. There are core competencies as follows: engineering basic competencies, major engineering competencies, self-management competencies, communication competencies, interpersonal competencies, global competencies. The preliminary survey was conducted on 426 engineering students attending the Engineering Education FESTA 2019. The preliminary findings were derived by conducting exploratory factor analysis, confirmatory factor analysis, question characteristics analysis, and reliability analysis for validation. The core competency assessment tools developed through this study can be used to verify the effectiveness of the curriculum and programs for students at engineering colleges. In addition, the developed core competencies, sub-factors, and questions can be utilized in a series of courses that design, conduct, and evaluate engineering curricula and programs as competency-based curriculum. The Innovation Procurement Policy was introduced as part of the strategic public procurement policy to improve firms' innovation capabilities and enhance the public sector's ability to solve social problems by designating and purchasing so-called 'innovative products.' The pilot procurement project for innovative products was first introduced in 2019, and the policy system for designating and discovering innovative products by government departments, as well as the priority purchase system, was established in 2020. Hence, this study conducted a quantitative analysis focusing on the effectiveness of the innovation procurement system in supporting firms after it was fully implemented. For this purpose, corporate financial and employment data from 2017 to 2021 were used, and propensity score matching(PSM) and difference-in-difference(DID) methods were utilized as analytical tools. The study found that the innovation procurement system contributed to corporate growth and employment and created additional public and private sales channels. Moreover, it is necessary to enhance the innovation procurement system, such as matching innovative product-producing companies with existing SME support policies, for companies to become self-sustaining after the innovative product designation ends.
Web caching server is one of the important components of any web site, as it makes the access of users to web content much faster while balancing the network and server load. There are many caching approaches proposed in order to solve this problem. Classically, we know several caching algorithms like FIFO, LRU, LRU-min and etc. Most of them have their advantages and disadvantages based on specific context. The purpose of this paper is to introduce the new concept of caching objects along with their related content and test the performance of the proposed caching method using CPN Tools simulator program. It is generally known that the use of Colored Petri Nets(CPNs) for modeling the simulation of new idea is one of the popular methods of system evaluation throughout software development projects. Moreover it could be very handy in describing the overall logic of the system. The CPN Tools is one of the most robust simulator programs which supports all necessary tools and functions to build and run the simulation model for CPN. Therefore in this paper, we will use CPN Tools software in order to build a simulator for our innovative caching approach named as web caching with related content. Then we will analyze the results derived from the simulation of our model and will make corresponding conclusions.

Many customers are no longer satisfied with mass-produced goods. They are demanding customization and rapid delivery of innovative products. Many companies are now realizing that the best way to reduce life cycle costs is to evolve a more effective product development paradigm using Internet and web based technologies. Yet there remains a gap between current market demands and product development paradigms. The existing CAD systems require that product developers possess all the design analysis tools in-house making it impractical to employ all the needed and newest tools. Hence, this paper addresses how assembly operation analysis can be embedded transparently and remotely into a service-oriented collaborative assembly design environment. A new assembly operation analysis framework is introduced and a relevant architecture and tools are developed to realize the framework. Instead of the current sequential process for verifying and validating an assembly design, a new Virtual Assembly Analysis (VAA) method is introduced in the paper to predict the various effects of joining during actual collaborative design. As a case study, arc welding and riveting processes are investigated. New service-oriented VAA architecture and its VAA components are proposed and implemented on prototype mechanical assemblies.
