Refolding of Proteins at High Concentration by Size Exclusion Chromatography

  • Renaturation of Lysozyme by size exclusion chromatography(SEC) to improve yield as well as the initial and final protein concentration has been studied in detail, Although urea decreases the rate of proteins refolding, it can suppress protein aggregation to sustain pathway of correct refolding at high protein concentration, and there existed an optimum urea concentration in renaturation buffer. Lysozyme was successfully refolded from initial protein concentration of up to 100mg/m1 by SEC, the yield was more than 40%. And the refolding of Interferon-${\gamma}$ was further investigated.

  • Combustion phenomenon in scale-downed combustor is investigated. As the combustor scale decreases surface to volume ratio increases and chamber size approaches quenching distance. As the combustor scales down surface to volume ratio increases resulting increased heat loss. And this heat loss can affect quenching and instability of the flame. To investigate this effect plastic mini combustor is made. Stoichiometricaly premixed Hydrogen / air gas is used as fuel. Initial chamber pressure and chamber size are varied and the effects are evaluated. Peak pressure decreased with the decrease in chamber height. As initial chamber pressure decreases peak pressure decreases and this change is more important than scale down effect till the chamber height of 1mm. With this result and further information following the experiments design parameter for micro engine can be established.

  • Modified couple stress formulation and first order shear deformation theory are used for magneto-electro-elastic bending analysis of three-layered curved size-dependent beam subjected to mechanical, magnetic and electrical loads. The governing equations are derived using a displacement field including radial and transverse displacements of middle surface and a rotation component. Size dependency is accounted based on modified couple stress theory by employing a small scale parameter. The numerical results are presented to study the influence of small scale parameter, initial electric and magnetic potentials and opening angle on the magneto-electro-elastic bending results of curved micro beam.

  • Stream Control Transmission Protocol(SCTP) is a transport layer protocol to support the data transmission. SCTP is similar to Transmission Control Protocol(TCP) in a variety of aspects. However, several features of SCTP including multi-homing and multi-streaming incur the performance difference from TCP. This paper highlights the data transfer during the initial slow start phase in SCTP congestion control composed of slow start phase and congestion avoidance phase. In order to compare the mean transfer time between SCTP and TCP, we experiment with different performance parameters including bandwidth, round trip time, and data length. By varying data length, we also measure the corresponding initial window size, which is one of factors affecting the mean transfer time. For the experiment, we have written server and client applications by C language using SCTP socket API and have measured the transfer time by ethereal program. We transferred data between client and server using round-robin method. Analysis of these experimental results from the testbed implementation shows that larger initial window size of SCTP than that of TCP brings the reduction in the mean transfer time of SCTP compared with TCP by 15 % on average during the initial slow start phase.

  • In this paper, It is performing to the elastic and elasto-plastic large deformation series analysis using a numerical method for the initial deflection effect of the aluminum alloy rectangular plate in the elasto-plastic loading area patch loading size. It is assumed a boundary condition to be a simply supported condition and consider the initial deflection amplitude, aspect ratio. It examined the critical elastic buckling load and post-buckling behaviour of aluminium alloy A6082-T6 rectangular plate. It used a commercial program for the elastic and elasto-plastic deformation series analysis. If the initial deflection amplitude is smaller, the in-plane rigidity with increasing to load is reduced from the start and occurs significantly more increasing the amplitude. More higher the aspect ratio, the initial yield strength is gradually decreased, and the plate thickness thicker and occurs larger than the thin walled plate a reduction ratio of the initial yield strength of the patch loading size as 0.5.

  • Pulverization of two different sized micro-$B_4C$ particles (${\sim}10{\mu}m$ and ${\sim}150{\mu}m$) was investigated using a STS based high energy ball milling system. Shapes, generation of the impurities, and reduction of the particle size dependent on milling time and initial particle size were investigated using various analytic tools including SEM-EDX, XRD, and ICP-MS. Most of impurity was produced during the early stage of milling, and impurity content became independent on the milling time after the saturation. The degree of particle size reduction was also dependent on the initial $B_4C$ size. It was found that the STS nanoparticles produced from milling is strongly bounded with the $B_4C$ particles forming the $B_4C$/STS composite particles that can be used as a neutron absorbing nanocomposite. Based on the morphological evolution of the milled particles, a schematic pulverization model for the $B_4C$ particles was constructed.

  • In transportation problems, various initial solution methods are known and being used. The purpose of this paper is to compare effectiveness of those methods in terms of time required in computing an initial solution. In this paper, Northwest Corner rule, Least Cost rule, Vogel's Approximation rule, Russel's Approximation rule, are compared, These rules are similar in loxio sized problems. But as the size of the problem become bigger, Russet's and Vogel's methods are found much more effective than others.

  • The preparation of SiC powder by SHS in the system of $SiO_2-Mg-C$ was investigated in this study. The effects of various processing parameters such as the initial pressure of inert gas in reactor, the content of Mg and C in mixture and the size of $SiO_2$ particles on the synthesis of SiC by SHS methode were investigated. The minimum initial pressure of inert gas in reactor for SHS reaction in this system was 5 atm, and as the pressure increased, and the concentration of unreacted Mg decreased. At 50 atm of the initial inert gas pressure in reactor, the optimum composition for the preparation of pure SiC was $SiO_2+2.5Mg+1.2C$. SiC powder synthesized in this condition had a mixture of ${\alpha}-SiC\;and\;{\beta}-SiC$ with an irregular shape and the particle size of $0.5{\sim}0.8{\mu}m$.

Characteristics of Settling and Consolidation Behavior for Non-Plastic Dredged Soils (비소성 준설토의 침강-압밀 거동 특성)

  • A series of column test with a silty marine soil mixed with Jumunjin Standard Sand were performed to investigate the characteristics of settling and consolidation of non-plastic dredged soils. Column tests were carried out by using the separable column to measure the grain size distribution of consolidated layer. Column tests were performed with changing the mixing ratio of Jumunjin Standard Sand to the silty marine soil, initial water content of slurry and initial height of slurry. Height of interface of slurry was monitored during tests and grain size distribution tests were carried out after finishing tests. Influencing factors on the particle segregation, eventually to the characteristics of settling and consolidation of non-plastic soil, were analyzed on the thesis of test results. As results of column tests, the mixing ratio of sand to the silty marine soil and the initial water content of slurry were known to affect the characteristics of settling and consolidation resulted in significant particle segregation of slurry. Initial height of slurry was found not to affect seriously to particle segregation.

A Dynamic Buffer Allocation Scheme in Video-on-Demand System (주문형 비디오 시스템에서의 동적 버퍼 할당 기법)

  • In video-on-demand(VOD) systems it is important to minimize initial latency and memory requirements. The minimization of initial latency enables the system to provide services with short response time, and the minimization of memory requirements enables the system to service more concurrent user requests with the same amount of memory. In VOD systems, since initial latency and memory requirement increase according to the increment of buffer size allocated to user requests, the buffer size allocated to user requests must be minimized. The existing static buffer allocation scheme, however, determines the buffer size based on the assumption that thy system is in fully loaded state. Thus, when the system is in partially loaded state, the scheme allocates user requests unnecessarily large buffers. This paper proposes a dynamics buffer allocation scheme that allocates user requests the minimum buffer size in fully loaded state as well as a partially loaded state. This scheme dynamically determines the buffer size based on the number of user requests in service and the number of user requests arriving while servicing current requests. In addition, through analyses and simulations, this paper validates that the dynamics buffer allocation outperforms the statics buffer allocation in initial latency and the number of concurrent user requests that can be supported. Our simulation results show that, in proportion to the static buffer allocation scheme, the dynamic buffer allocation scheme reduces the average initial latency by 29%~65%, and in a systems having several disks. increases the average number of concurrent user requests by 48%~68%. Our results show that the dynamic buffer allocation scheme significantly improves the performance and reduce the capacity requirements of VOD systems.
