수삼물추출물의 갈변반응중 아미노산과 당류변화 (Changes in Free Amino Acids and Sugars in Water-soluble Extracts of Fresh Ginseng during Browning Reaction)

  • An aqueous extract s of fresh ginseng roots was heated at loot for 64 hrs. and the changes of color intensity, pH and the amount of free sugars and amino acids during the various intervals of the heating time were investigated. Color intensity and absorbance of the solution at 490nm were increased in proportion to the length of the heating time. Most of brown pigments produced during the treatment were water soluble, and pH 5.1 at initial stage of the solution, was slightly decreased at the final stages of the reaction. Sucrose, glucose and fructose were major free sugars in ginseng roots, and the amounts of sucrose was over 90 % of total free sugars. Sucrose. was largely decreased approximately 50%, by 64 hrs of the treatment, whereas sharp increase in the amount of glucose and fructose was observed during the reaction in the solution. The observed increase in reducing sugars, glucose and fructose was presumed due to hydrolysis of sucrose. Evidently, glucose and fructose were not important factor to control the browning reaction of the solution. Most of free amino acids and peptides except alanine and isoleucine especially arginine, serine and threonine, were sharply decreased up to 40 : 50% of the original concentration within 2 hrs. Accordingly, the content of free amino acids and peptides seems to be extremely important factor to control the browning reaction in ginseng. A free amino acid, presumed to be nor-leucine, was found in fresh ginseng root on the basis of re mention on liquid chromatography. Kinetic analysis of the browning reaction indicated a pseudo second order with respect to amino acid concentration at the initial stage.

토양미생물 복원제를 이용한 유류로 오염된 토양의 복원 (Bioremediation Efficiency of Oil-Contaminated Soil using Microbial Agents)

  • 유류로 오염된 토양을 토양미생물 복원제를 첨가한 후 다양한 조건에서20일 동안의 유류저감효과를 알아보았다. 실험조건은 유류로만 오염된 토양(DS), 토양미생물 복원제를 20%(w/w)가 되도록 첨가한 유류로 오염된 토양(DSP), 토양 미생물 복원제를 넣은 후 pH를 중성으로 보정한 유류로 오염된 토양(DSP-1), 토양미생물 복원제와 촉진제를 넣은 유류로 오염된 토양(DSP-2), 토양미생물 복원제와 촉진제를 넣은 후 pH를 중성으로 보정한 유류로 오염된 토양(DSP-3)을 설정하였다. 실험 결과 pH를 보정한 토양미생물 복원제를 첨가한 유류오염토양은 탈수소 효소 활성과 TPH 저감에서의 효능이 우수하였다. 실험이10일 경과되었을 때 탈수소 효소 활성이 가장 높은 DSP-1 토양이 TPH 역시 가장 활발히 분해했다. 결과적으로 초기 10일의 배양기간 동안 토양미생물 복원제를 첨가한 토양은 대조군에 비해 38% 가량의 TPH 저감상승효과를 볼 수 있다. 토양미생물 복원제의 첨가를 통해 초기 저감속도를 올려줄 수 있으며, 최종적으로도 비 첨가군에 비해 높은 저감효율을 기대할 수 있다. 토양미생물 복원제를 유류오염토양을 복원한다면 초기 오염물질을 빠르게 처리할 수 있지만 미생물 활성은 pH, 온도 등 환경 인자에 많은 영향을 받으므로 토양미생물 복원제의 효율을 최대화하기 위해서는 환경 인자를 분석하여 이를 바탕으로 복원을 진행한다면 오염물질 정화 효율을 향상시킬 수 있을 것이다.

탈당(脫糖)된 육계와 돼지 혈장분(血漿粉)의 상온 저장에 따른 색깔, 단백질 함량, 용해도, 기포력과 pH 변화 (Changes in Color, Protein Content, Solubility, Foaming Capacity and pH of Desugarized Brolier and Porcine Plasma Powder During Storage at Room Temperature)

  • GOD (glucose oxidase)나 제빵용 효모를 사용하여 포도당이 제거된 육계와 돼지 혈장분의 상온 저장중 색깔, biuret 단백질 함량, 용해도, 기포력과 pH의 변화를 조사하였다. 혈장분의 종류와 혈당 제거 법과는 상관없이 실험기간 동안 혈당이 제거된 혈장분은 해당 대조구보다 "L" 값이 같거나 높았으며 "a" 값과 "b"값은 낮았다(P<0.05). 축종 또는 혈당 제거 방법과는 상관없이 탈당된 모든 시료의 단백질 함량과 용해도는 상온 저장중 해당 대조구보다 항상 높았고 저장 기간이 경과할수록 감소하는 경향을 보였다(P<0.05). 대체적으로 탈당된 육계와 돼지 혈장분 기포력은 해당 대조구와 비슷하거나 높았다. 그러나 돼지 혈장분의 기포력은 육계 혈장분과는 다르게 실험기간 동안 탈당된 돼지 혈장분과 상응하는 대조구는 큰 차이를 나타내지 않았다(P<0.05). 육계 대조구와 GOD로 혈장 포도당을 제거한 육계 혈장분의 pH는 저장기간이 지남에 따라 감소하였지만 효모로 포도당을 제거한 육계 혈장분과 모든 돼지 시료의 pH는 실험 기간중 증가하였다가 감소하였다. GOD로 포도당을 제거한 육계 혈장분이 해당 육계 시료 보다 낮은 pH값을 보인 반면 돼지 혈장분에서는 대조구가 상응하는 실험구보다 항상 낮은 pH 값을 나타내었다(P<0.05). 육계와 돼지 혈장의 포도당을 제거한 후 혈장을 건조시키면 혈장분의 색깔, biuret 단백질 함량, 용해도 그리고 기포력을 증진시킬 수 있었다.

경도 두부외상 환자에서 뇌좌상주위 부종영역에서의 자기공명분광법을 이용한 대사변화 (Metabolic Changes in Pericontusional Edematous Areas in Mild Head Injury Evaluated by Proton MRS)

  • Objectives : In order to evaluate the metabolic changes associated with pericontusional edematous area in mild head injury, proton magnetic resonance spectroscopy(1H-MRS) was performed in mild head injury patients (initial GCS score 13-15) with focal brain contusion. Methods : Seven head injury patients with initial GCS 13-15(3 males and 4 females : age range 15-65 years, mean age 33 years) have underwent 1H-MRS evaluations. The patients were examined within 7 days after injury(n=7) and 2 months after injury(n=5). The region of interest(ROI) was selected in the edematous area adjacent to traumatic brain contusion upon T2-weighted MR images and a corresponding region of the contralateral hemisphere (ROC, region of contralateral corresponding hemisphere) was examined as well. The metabolic ratios of NAA/Cr and lactate/Cr were compared between ROIs, ROCs and control values. Results : In initial NAA/Cr ratios, the values of ROIs were significantly lower than those of the controls(p=0.009), but there was no difference either between ROIs and ROCs(p=0.410) or between ROCs of patients and the control (p=0.199). In lactate/Cr ratios, the ROIs in all seven patients and the ROCs in two showed increased lactate signals. The lactate/Cr ratios of the ROIs were significantly elevated as compared to those of the ROCs(p=0.02) and the control(p=0.015). In two months follow-up, lactate signals were absent or significantly reduced(p=0.015). In no patients, clinical or radiological deterioration has been observed. Conclusion : Our 1H-MRS results demonstrate that there are significant ischemic changes in pericontusional edematous areas as indicated by elevated lactate signals in the patients with mild head injury. But there were no consistent neural loss or dysfunction in these area. There findings suggest that pericontusional edematous areas can be vulnerable to secondary brain insults even in the patients with mild head injury.

폐기물 활용을 위한 종합적 처리방법의 김치쥬스 발효 향상 (Improvement of Kimchi Juice Fermentation by Combined method for Chinese Cabbage Waste Utilization)

  • The effective fermentation methods of Kimchi juice for utilization of outer layer of Chinese cabbage, an waste of Kimchi industry were studied. The Kimchi juice prepared with brining and grinding the waste of Chinese cabbage and addition of spices was fermented at $25^{\circ}C$. Addition of 5$^{\circ}C$15% fermented Kimchi juice of pH 5.4 at initial stage and pH 4.4 at middle state resulted in a significant increase in fermentation rate and solid content after 12 hours of fermentation. The combined method of enzymatic hydrolysis(0.1% viscozyme) of the brined and ground cabbage and addition of 2.0% NaCl, 1.0% sucrose and 10% fermented juice of pH 5.4 first and 4.4 during fermentation, respectively resulted in more rapid fermentation. The solid concentration was 5 times higher than control at maximum point and acidic and total flavor intensity were also significantly high.

인삼을 첨가한 동치미 쥬스 발효중 이화학적 및 미생물의 변화 (Physicochemical and Microbiological Changes in Dongchlmi Juice during Fermentaion with the Addition of Panax ginseng C.A. Meyer)

  • This study was undertaken to examine the effects of fresh ginseng on the physicochemical and microbiological changes in dongchimi juice fermented under various conditions. The pH was somewhat lower in dongchimi juice added with 2.0% and 4.0% of fresh ginseng than that without ginseng, whereas the titratable acidity was higher in dongchimi juice with 2.0% and 4.0% of ginseng addition than the control. The addition of fresh ginseng to dongchimi preparation increased the numbers of total viable bacteria, lactic acid bacteria including Leuconostac mesenterotdes in dongchimi juice during fermentation. The changes in the counts of lactic acid bacteria were similar to those of total viable cells throughout the experiment except the initial stage of fermentation. However, the number of Leucosfastoc mesenternidgs decreased after the palatable stage. Key words : Panax ginseng C.A. Meyer, dongchlmi juice, pH, titratable acidity, microbiological changes.

The Effect of Different Lupin Kernel Inclusion Levels on the Growth and Carcass Composition of Growing and Finishing Pigs

  • Two experiments were conducted to investigate the effect of different lupin kernel levels on growth performance and carcass characteristics of growing and finishing pigs. In experiment 1, a total of 54 barrows and 54 growing gilts, with an average initial body weight of $24.7{\pm}0.38kg$, was used to determine the effect of different lupin kernel levels (0, 10% and 20%; Control, LK10 and LK20, respectively) in the diets on growth performance with a $3{\times}3$ randomized complete block design for 35 days. There were no significant differences in average daily gain (ADG), average daily feed intake (ADFI) and feed:gain among treatments. In experiment 2, a total of 54 barrows and 36 finishing gilts, with an average initial body weight of $63.0{\pm}0.56kg$, was used to determine the effect of different lupin kernel levels (0, 15% and 30%; Control, LK15 and LK30, respectively) in the diets on growth performance and carcass characteristics with a $3{\times}3$ randomized complete block design for 63 days. LK30 decreased ADG and ADFI compared with the Control and LK15 (p<0.05). However, LK30 tended to improve feed:gain compared with the Control and LK15. And LK15 did not differ from the Control in ADG, ADFI and feed:gain. With inclusion of lupin kernel in the finishing diet, backfat thickness increased (p<0.01) and carcass grade tended to be improved compared with the Control.

Phosphorus Release from Sediment in Lake Sihwa and its Control

  • In this study the effects of initial pH, temperature, dissolved oxygen concentration, and sediment depth on the release of phosphorus from sediment in Lake Sihwa were investigated. No phosphorus release occurred at $10^{\circ}C$ for all pH values. DO concentrations were decreased and equilibrium was obtained 4-8 days after phosphorus release started. The DO concentrations were less than 1 mg/L. Sediment depth had little effect on phosphorus release rate. In order to control the released phosphorus, raw sludge and chalk were used. Results showed that a portion of phosphorus was removed by adsorption to chalk and raw sludge. About 90 % phosphorus removal was observed using sludge and calcinated chalk from the water sample in Lake Sihwa.

Effects of Cellulase Enzymes and Bacterial Feed Additives on the Nutritional Value of Sorghum Grain for Finishing Pigs

  • One hundred and twenty-eight finishing pigs (51.3 kg average initial body weight) were used to determine the effects of adding cellulase enzymes and lactobacillus acidophilus to sorghum-based diets on growth performance, carcass merit, and nutrient digestibility in finishing pigs. Treatments were: 1) corn-soybean meal-based positive control; 2) sorghum-soybean meal-based negative control; 3) Diet 2 with celluloytic enzymes; and 4) Diet 2 with a bacterial feed additive (lactobacillus acidophilus). There was a trend for greater average daily gain (ADG) in pigs fed com versus the sorghum treatments for day 0 to 28 (p < .09), but there was no effect of treatment (p > .15) on overall ADG (i.e., day 0 to 63). Feed consumption was not affected by treatment during the experiment (p > .19). Pigs fed the corn-soybean meal-based diet had 3.5% greater overall gain/feed than pigs fed the other diets (p < .009). Dressing percentage was not affected by treatment (p > .22), but there was a trend for backfat thickness at the last rib to be greater for pigs fed com versus the sorghum treatments (p < .09). Pigs fed the sorghum treatments had 1 % greater fat free lean index (p < .10) compared to pigs fed the corn-soybean meal-based positive control. Pigs fed com had greater apparent digestibilities of DM, N, and GE than pigs fed the sorghum treatments (p < .03), and greater DE intake (p < .07) suggesting that the increased carcass fatness for pigs fed the corn-based control diet resulted from greater energy status of those pigs. In conclusion, pigs fed the corn-soybean meal-based control diet had no improved growth performance but tended to be fatter than pigs fed sorghum. Adding cellulolytic enzymes or a bacterial feed additive to diets for finishing pigs did not affect growth performance, carcass merit, or nutrient utilization.

2,3-Dihydroxybiphenyl Dioxygenase 생산을 위한 E. coli CK1092의 배양조건 (Culture Conditions of E. coli CK1092 for the Production of 2,3-Dihydroxybiphenyl Dioxygenase)

  • PCBs를 분해하는 Pseudomonas sp. P20 균주의 pcbC gene이 재조합 된 E. coli CK1092 균주를 이용하여 2,3-dihydroxybiphenyl dioxygenase 생산에 미치는 배양조건을 검토하였다. E. coli CK1092를 2% sucrose가 포함된 LB 배지를 기본배지로 하여 질소원, 금속이온 등의 영향을 조사한 결과, $Fe^{3+}$$Fe^{2+}$$10^{-5}M$의 농도일때 효소생산이 증가되었으며 배양최적온도는 $37^{\circ}C$, 배양초기 pH 7.0일때 효소생산이 우수하였다. 배양조에서의 배양조건은 초기 pH 7.0, 통기량 1 v/v/m, 교반속도 200rpm에서 가장 효소생성이 우수하였다.
