  • We examined methanol extracts prepared from the Korean plants for their inhibitory effects on rat lens aldose reductase (RLAR) activity in vitro. Among 41 plant extracts, the extracts of Lagerstroemia indica, Punica granatum, Eurya japonica, Liquidambar styraciflua, and Vitis coignetiae exhibited highest inhibitory potency, even more potent than tetramethylene glutaric acid (TMG), known as one of typical aldose reductase inhibitors $(IC_{50}\;value,\;0.51\;{\mu}g/ml)$. Especially, the extract of Lagerstroemia indica (Lythraceae), exhibited the most inhibitory potency $(IC_{50}\;value,\;0.069\;{\mu}/ml)$ on RLAR.

  • Various biological activities (enzyme inhibitory potency, lipoxygenase inhibition, tadpole narcosis, vapor toxicity and heat of vaporization) of molecules are correlated with molecular descriptors. The molecular descriptors used in this works are molecular connectivity index, Wiener distance index and ad hoc descriptor, which can encode information about branching, size, cyclization, unsaturation, hetero atom content and polarizability. It is found that calculated values from multiple regression equations are in a good agreement with experimental data on five biological activities of alcohol, ester and ketone compounds.

  • The NAD(P)-linked $3{\alpha}-Hydroxysteroid$ $dehydrogenase(3{\alpha}-HSD)$ of rat liver cytosol is powerfully inhibited by the non-steroidal anti-inflammatory drugs in rank-order of their therapeutic potency, and this observation has now been developed into a rapid screen for predicting the potency of products that show anti-inflammatory effect. Five-plants were screened by using this method. Among them, BuOH-fraction of Mucuna birdwoodiana showed strong inhibitory effect on $3{\alpha}-HSD$, and three isoflavone compounds were isolated. Inhibitory activates of isolated compounds were compared.

  • Aliphatic alkenals having 6 to 13 carbons were evaluated for antifungal activity against Saccharomyces cerevisiae. The activity was gradually increased with chain length, e.g., (E)-2-decenal and (E)-2-undecenal exhibited maximum potency, while (E)-2-dodecenal and (E)-2-tridecenal were completely inactive. Alkenals showed increasing inhibitory activity with chain length, as in the case of antifungal activity, towards glucose-induced medium acidification by the plasma membrane $H^+$-ATPase of S. cerevisiae. The group including (E)-2-nonenal, (E)-2-decenal, and (E)-2-undecenal exhibited maximum potency, but the potency of (E)-2-dodecenal and (E)-2-tridecenal demonstrated a sudden drop with respect to the former group. (E)-2-Nonenal revealed dose-responsive inhibition to the medium acidification and inhibited over 90% at a concentration of 1.25 mM ($175.3{\mu}g$/ml). In contrast to (E)-2-undecenal whose inhibitory efficiency increased with incubation time, inhibition by (E)-2-dodecenal was reversed with time. Of the tested alkenals, (E)-2-heptenal and (E)-2-octenal most highly inhibited ATP hydrolytic activity by the plasma membrane $H^+$ ATPase, while (E)-2-heptenal at 10 mM ($1121.8{\mu}g$/ml) showed an inhibitory efficacy of 93.2%.

  • To evaluate the possibility that the ginseng saponins could be developed as an anti-arteriosclerotic agent, we examined the inhibitory effects of ginseng saponins (total saponin[TS], panaxatriol[PT], panaxadiol[PD]) on the expression of c-fos mRNA and the proliferation of cultured rat aortic vascular smooth muscle cells (VSMCs) stimulated by angiotensin II (Ang II). TS and PT (1.0 mg/ml) suppressed c-fos mRNA induction in VSMCs stimulated by $10^{-5}$ M Ang II. The order of inhibitory potency was PT>TS. Ginseng saponins ($0.01{\sim}1.0$ mg/ml) inhibited the proliferation of VSMCs stimulated by Ang II in a concentration dependent manner, the inhibitory potency was TS>PT>PD at $0.1{\sim}1.0$ mg/ml. These results suggest that ginseng saponins may suppress Ang II-stimulated proliferation of aortic VSMCs which can be seen in atherosclerosis, hypertension and restenosis.

Methods Minimal inhibitory concentration(MIC) was measured by dropping to $50{\mu}l$ according to density Coptidis rhizoma extract(100%, 10%, 1%, 0.1%) compared with quantity(40g, 80g, 160g). Anti-bacterial potency was measured by the size of inhibition zone with change of volume. Results 1. MIC on Staphylococcus aureus in Coptidis rhizoma extract was showed anti-bacterial potency compared with quantity and density in 100% and 10% of all samples(40g, 80g, 160g). 2. MIC on Staphylococcus aureus in Coptidis rhizoma extract(40g, 80g, 160g) was showed anti-bacterial potency compared with quantity all samples($20{\mu}l,\;30{\mu}l,\;40{\mu}l,\;50{\mu}l$) in 100% density. 3. MIC on Staphylococcus aureus in Coptidis rhizoma extract(40g, 80g, 160g) was showed anti-bacterial potency compared with quantity all samples ($20{\mu}l,\;30{\mu}l,\;40{\mu}l,\;50{\mu}l$) in 100% density. Anti-bacterial potency of 80g Coptidis rhizoma extract decreased compared with 40g. Anti-bacterial potency of 160g Coptidis rhizoma extract decreased compared with 40g in $20{\mu}l,\;30{\mu}l$. Conclusions Coptidis rhizoma extract was showed anti-bacterial potency compare with quantity and density. In herbal drug, antibacterial potency compare with quantity and density must be studied.
