Establishment of Biotin Analysis by LC-MS/MS Method in Infant Milk Formulas (LC-MS/MS를 이용한 조제유류 중 비오틴 함량 분석법 연구)

  • This study was conducted to establish the standard method for the contents of biotin in milk formulas. To optimize the method, we compared several conditions for liquid extraction, purification and instrumental measurement using spiked samples and certified reference material (NIST SRM 1849a) as test materials. LC-MS/MS method for biotin was established using $C_{18}$ column and binary gradient 0.1% formic acid/acetonitrile, 0.1% formic acid/water mobile phase is applied for biotin. Product-ion traces at m/z 245.1 ${\rightarrow}$ 227.1, 166.1 are used for quantitative analysis of biotin. The linearity was over $R^2=0.999$ in range of $5{\sim}60{\mu}g/L$. For purification, chloroform was used as a solvent for eliminating lipids in milk formula. The linearity was over 0.999 in range of 5~60 ng/mL. The detection limit and quantification limit were 0.10, 0.31 ng/mL. The accuracy and precision of LC-MS/MS method using CRM were 103%, 2.5% respectively. Optimized methods were applied in sample analysis to verify the reliability. All the tested milk formulas were acceptable contents of biotin compared with component specification and standards for nutrition labeling. The standard operating procedures were prepared for biotin to provide experimental information and to strengthen the management of nutrient in milk formula.

Evaluation of MR Based Respiratory Motion Correction Technique in Liver PET/MRI Study (Liver PET/MRI 검사 시 MR 기반 호흡 움직임 보정 방법의 유용성 평가)

  • Purpose Respiratory motion during PET/MRI acquisition may result in image blurring and error in measurement for volume and quantification of lesion. The aim of this study was to evaluate changes of quantitative accuracy, tumor size and image quality by applying MR based respiratory motion correction technique (MBRMCT) using integrated PET/MR scanner. Materials and Methods Data of 30 patients (aged $62.5{\pm}10.2y$) underwent $^{18}F-FDG$ liver PET/MR (Biograph mMR 3.0T, Siemens) study were collected. PET listmode data for 7 minutes was simultaneously acquired with maximum average gate (MAG), minimum time gate (MTG) and non gate (NG) T1 weighted MR images. Gated PET reconstruction was performed using mu-maps generated from MAG and MTG by setting 35% of efficiency window. Maximum SUV ($SUV_{max}$), peak SUV ($SUV_{peak}$), tumor size and full width at half maximum (FWHM) in the z-axis direction of MAG, MTG and NG PET images were evaluated. Results Compared to NG, mean $SUV_{max}$ and $SUV_{peak}$ were increased in MAG 13.15%(p<0.0001), 8.66%(p<0.0001), MTG 13.27%(p<0.0001), 8.80%(p<0.0001) and mean tumor size and FWHM were decreased in MAG 14.47%(p<0.0001), 15.49%(p=0.0004), MTG 14.89%(p<0.0001), 15.79%(p=0.0003) respectively. Mean $SUV_{max}$ and $SUV_{peak}$ of MTG were increased by 0.07%(p=0.8802), 0.13%(p=0.7766). Mean tumor size and FWHM of MTG were decreased by 0.49%(p=0.2786), 0.36%(p=0.2488) compared to MAG. There was no statistically significant difference between MAG and MTG which increase total scan time for about 7 and 2 minutes. Conclusion SUV, accuracy of tumor size and spatial resolution were improved in both of MAG and MTG by applying MBRMCT without installing additional hardware in liver PET/MR study. More accurate information can be provided with the increase of 2 minutes scan time if applying MTG of MBRMCT to various abdominal PET/MR studies affected by respiratory motion.

Effect of Red-edge Band to Estimate Leaf Area Index in Close Canopy Forest (울폐산림의 엽면적지수 추정을 위한 적색경계 밴드의 효과)

  • The number of spaceborne optical sensors including red-edge band has been increasing since red-edge band is known to be effective to enhance the information content on biophysical characteristics of vegetation. Considering that the Agriculture and Forestry Satellite is planning to carry an imaging sensor having red-edge band, we tried to analyze the current status and potential of red-edge band. As a case study, we analyzed the effect of using red-edge band and tried to find the optimum band width and wavelength region of the red-edge band to estimate leaf area index (LAI) of very dense tree canopy. Field spectral measurements were conducted from April to October over two tree species (white oak and pitch pine) having high LAI. Using the spectral measurement data, total 355 red-edge bands reflectance were simulated by varying five band width (10 nm, 20 nm, 30 nm, 40 nm, 50 nm) and 71 central wavelength. Two red-edge based spectral indices(NDRE, CIRE) were derived using the simulated red-edge band and compared with the LAI of two tree species. Both NDRE and CIRE showed higher correlation coefficients with the LAI than NDVI. This would be an alternative to overcome the limitation of the NDVI saturation problem that NDVI has not been effective to estimate LAI over very dense canopy situation. There was no significant difference among five band widths of red-edge band in relation to LAI. The highest correlation coefficients were obtained at the red-edge band of center wavelength near the 720 nm for the white oak and 710 nm for the pitch pine. To select the optimum band width and wavelength region of the red-edge band, further studies are necessary to examine the relationship with other biophysical variables, such as chlorophyll, nitrogen, water content, and biomass.

Development and Feasibility Study for Phase Contrast MR Angiography at Low Tesla Open-MRI System (저자장 자기공명영상 시스템에서의 위상대조도 혈관조영기법의 개발과 그 유용성에 대한 연구)

  • Magnetic resonance angiography (MRA) techniques are widely used in diagnosis of vascular disorders such as hemadostenosis and aneurism. Especially, phase contrast (PC) MRA technique, which is a typical non contrast-enhanced MRA technique, provides not only the anatomy of blood vessels but also flow velocity. In this study, we developed the 2- and 3-dimensional PC MRA pulse sequences for a low magnetic field MRI system. Vessel images were acquired using 2D and 3D PC MRA and the velocities of the blood flow were measured in the superior sagittal sinus, straight sinus and the confluence of the two. The 2D PC MRA provided the good quality of vascular images for large vessels but the poor quality for small ones. Although 3D PC MRA gave more improved visualization of small vessels than 2D PC MRA, the image quality was not enough to be used for diagnosis of the small vessels due to the low SNR and field homogeneity of the low field MRI system. The measured blood velocities were $25.46{\pm}0.73cm/sec$, $24.02{\pm}0.34cm/sec$ and $26.15{\pm}1.50cm/sec$ in the superior sagittal sinus, straight sinus and the confluence of the two, respectively, which showed good agreement with the previous experimental values. Thus, the developed PC MRA technique for low field MRI system is expected to provide the useful velocity information to diagnose the large brain vessels.

Variation and Distribution of Anions and Cations in the Aerosols of Gyorae Forests in Jeju Island (제주도 교래휴양림 지역의 대기질의 음이온 및 양이온의 분포와 변이성)

  • This study was conducted to investigate the distribution and variation of the anion and cation number of aerosols in the A and B regions of Gyorae forests. Ions were measured using an ion number meter between 28 June and 13 July 2017. The total average number of anions and cations were $735ions/cm^3$ and $459.27ions/cm^3$, respectively, which were measured at five sites in A area at average temperature of $27.81^{\circ}C$, wind speed of 0.28 m/sec, and altitude of 455.7 m. The average number of anions and cations were $780ions/cm^3$ and $379.55ions/cm^3$, respectively, which were measured at all four sites in the B region at average temperature of $27.6^{\circ}C$, humidity of 80%, wind speed of 0.1 m/sec and altitude of 477 m. The number of anions and cations in the A and B regions was $757.5ions/cm^3$ and $419.41ions/cm^3$, respectively. The number of ions was highly variable for each measurement over time. The number of anions remained $275.73ions/cm^3$ higher than that of cations. The variance of the measured values of anions and cations between and within sites A and B was significant (p<.001) and there was a significant positive correlation between regional mean values of anions and cations. In conclusion, the temporal distribution and variation of the ion content in the Gyorae forests provide basic information regarding aerosol compositions and changes.

Lead Level in Hair of Elementary School Children in Urban and Rural Areas (국민학교 저학년 학생들의 두발중 연농도와 관련요인)

  • This study was conducted to explore the feasibility of using the lead level in the hair of children as a screening test for lead intoxication of children and as an indicator for environmental pollution. Lead concentration of hair was measured for 268 seven to nine-year-old elementary school children in Taegu city (147 children) and Gampo town(121 children). The lead level in hair was measured by atomic absorption spectrophotometer equipped with a graphite furnace atomizer. The following information was obtained for all children: sex, age, father's occupation and smoking habits, kinds of hair cleaner, fingernail biting habits, and status of the child's hands and clothing. The mean lead level in hair was $8.7{\pm}2.9{\mu}g/g$ in the urban area and $7.7{\pm}2.2{\mu}g/g$ in the rural area. There were significant differences between two groups(p<0.01). The difference of lead levels in hair measured by cleanliness of the child's hands(clean: $7.4{\pm}2.7{\mu}g/g$, dirty: $8.2{\pm}2.6{\mu}g/g$) and child's clothing(clean: $8.0{\pm}2.5{\mu}g/g$, dirty: $9.3{\pm}2.6{\mu}g/g$) were statistically significant, but other factors were not significant. In multiple regression analysis, difference of residency and age were significant variables for lead level in the hair of children. These findings suggest that measurement of lead level in the hair is a useful method for the screening of the lead intoxication of children and monitoring environmental conditions.

Study on Dosimetry Used TLD Dosimeter and Body Mass Index at Total Body Irradiation (전신조사방사선치료에서 열형광선량계를 이용한 선량 측정과 체질량지수에 관한 고찰)

  • Purpose: The aim of study is to expose a more uniform dose depending on the relationship between a body mass index in patients who underwent radiation therapy and an acquired dosimetric information by using a thermoluminescent dosimeter. Materials and Methods: Since 2006 to August 2011 we investigated 28 people who underwent radiation therapy were enrolled in AMC. Each patient was measured on the head, neck, chest, abdomen, pelvis, thigh, knee joint, and ankle joint using the thermoluminescent dosimeter. The measurement value of each points compared with the prescribed center point, abdominal point, and dose measurements of points on which to base the abdomen and the patient's body mass index (BMI) were compared with reference point, abdomen dose. Results: 28 patients on prescribed dose in the abdomen by which the center point, an average dose was $100.6{\pm}5.5%$, and the other seven measuring points with the average maximum difference among the head, neck, chest, pelvic, thigh, knee, and ankle were $92.8{\pm}4.2%$, $97.6{\pm}6.2%$, $96.4{\pm}5.5%$, $102.6{\pm}5.3%$, $103.4{\pm}7.9%$, $95.8{\pm}5.9%$, $96.1{\pm}5.5%$. The relationship of abdominal point dose and the patient's body mass index (BMI) was analyzed a scatter plot, and the result of linear relationship analysis by regression method, the regression of the dose (y) was -1.009 BMI (x) plus 123.3 and coefficient of determination ($R^2$) was represented 0.697. Conclusion: The total body irradiation treatment process was evaluated the dose deviation and then the prescribed dose by which the average abdominal dose was satisfied with $100.6{\pm}5.5%$. Results of the relationship analysis between BMI and dose, if we apply the correction value for each patients, it can be achieved more uniform dose delivery.

Seasonal Variation of the Quantity and Quality of Seston as Diet Available to Suspension-Feeders in Gosung and Kangjin Bays of Korea (고성만과 강진만에서 현탁물 섭식자에 유용한 입자물질 양과 질의 계절 변동)

  • Seasonal variation of the elemental and biochemical composition of the suspended particulate matter (SPM) was investigated in terms of quantity and quality of diets for suspension feeders from July 1999 to August 2000 in two coastal bay systems of Gosung and Kangjin Bays in Korea. No clear patterns in the seasonal variations of SPM concentration were found in these two bay systems. The results indicated that the seasonal variation of SPM could not be considered the variation of food available to suspension-feeders. The simultaneous peaks in chlorophyll a and biochemical components in summer indicated that the quantity of the particulate organic matter primarily depended on phytoplankton productivity. However, no correlation between chlorophyll a and biochemical components [particulate protein (PPr), carbohydrate (PCHO) and lipid] were found, indicating that other processes might also contribute to the particulate organic matter in the period when the phytoplankton biomass was low. High C: Chl a and C:N, and carbohydrate peaks during the autumn to spring period suggested that resuspension of surface sediments was a probable process to supply the particulate organic matter. The food material, represented by summing up the total quantity of three biochemical components, was highest in spring with minor peaks during the period from autumn to the next spring, The food index, calculated as the ratio of food material to total SPM, did not generally exceed $6\%$ with short peaks during the year. Therefore, nutritional quality of SPM in the bays are relatively poor than in other more productive coastal waters in the world. Our results confirm that the measurement of a single chemical variable cannot describe fully the nutritive value of the seston available to suspension-feeders as previously proposed, and the biochemical composition of SPM can provide effective information on its origin and nutritive Quality.

Comparison of Radiation Dose in the Measurement of MDCT Radiation Dose according to Correction of Temperatures and Pressure, and Calibration of Ionization Chamber (MDCT 선량측정에서 온도와 압력에 따른 보정과 Ionization Chamber의 Calibration 전후 선량의 비교평가)

  • This study aims to conduct the comparative analysis of the radiation dose according to before and after the calibration of the ionization chamber used for measuring radiation dose in the MDCT, as well as of $CTDI_w$ according to temperature and pressure correction factors in the CT room. A comparative analysis was conducted based on the measured MDCT (GE light speed plus 4 slice, USA) data using head and body CT dosimetric phantom, and Model 2026C electrometer (RADICAL 2026C, USA) calibrated on March 21, 2007. As a result, the $CTDI_w$ value which reflected calibration factors, as well as correction factors of temperature and pressure, was found to be the range of $0.479{\sim}3.162mGy$ in effective radiation dose than the uncorrected values. Also, under the routine abdomen routine CT image acquisition conditions used in reference hospitals, patient effective dose was measured to indicate the difference of the maximum of 0.7 mSv between before and after the application of such factors. These results imply that the calibration of the ion chamber, and the correction of temperature and pressure of the CT room are crucial in measuring and calculating patient effective dose. Thus, to measure patient radiation dose accurately, the detailed information should be made available regarding not only the temperature and pressure of the CT room, but also the humidity and recombination factor, characteristics of X-ray beam quality, exposure conditions, scan region, and so forth.

A Study on the Consumer's Service Quality Perception Based on the Types of Life-style (소비자의 라이프스타일에 따른 서비스품질 지각 차이에 관한 연구)

  • For the last decades, service quality has been studied as one of the most important tools for a service company to compete with the other companies. Based on these past researches, it has been agreed that the service quality is a basic and powerful tool to create the competitive advantage. Due to similar reason, many service marketing practitioners have been also focused on the service quality to retain the existing consumers and collect the new consumers. However, service quality is subjectively perceived by individual consumers. Consumer evaluation of service quality can be different from each other. Especially consumers with one life-style may evaluate the service quality differently from the consumers with the other life-styles. Therefore we need to know whether there are differences in service quality perception on the categories of life-style. Life-style refers to a distinctive mode of living in its aggregate and broadest sense. It embodies the patterns that were developed and emerged from the dynamics of living in a society. Since the concept of life-style and its relationship to marketing was introduced in 1963 by William Lazer, methods of measuring the life-style and their application have been developed. Life-style has been usually used to segment the marketplace because it offers marketers a unique and important view of the market. When Life-style is combined with clustering methods, life-style segmentation can generate identifiable whole persons rather than isolated fragment. Life-style segmentation begins with people instead of products and classifies them into different life-style types, each characterized by a unique style of living based on a wide range of activities, interests, and opinions(Plummer, 1974). In this study we applies the life-style segmentation based on the AIO(Activities, Interests, and Opinions) to the consumers of the large discount stores. In Korea, the large discount store market has entered into maturity stage so that the market differentiation strategy is becoming a more critical issue to the marketing practitioners. One of the most important tools to differentiate from the competitors in large discount store market is continuously to provide service of better quality than competitors. This study tries to find answers about the following questions: 1) How can we categorize the consumer life-styles in the large discount store? 2) What are the characteristics of the categorized groups? 3) Are there any differences in service quality perception among the consumers with different life-styles 4) Are there any differences in consumer behavior among them in the large discount store? For the purpose, we collected survey data from consumers and analyzed the data with the SPSS package where we had $X^2$-test, factor analysis, ANOVA, MANOVA, and cluster analysis. The survey was made during one month in the April of 2008. Among the collected 306 copies of questionnaires, 281 copies were chosen as the effective samples for empirical analysis except 25 copies with wrong responses. To identify the life-style patterns, we used the measures employed by Kim and Kwon(1999), where 44 items on a seven-point scale were used to measure factors of the life-style patterns. The Principal Component Method was used for factor extraction, and the VARIMAX orthogonal factor rotation was employed. The 7 items showing low factor loading were eliminated. The results of the factor analysis suggested that nine factors of the life-style patterns were identified as follows: 1) the equality-of-sexes and pursuit-of-independence tendency 2) self-management tendency 3) sociable tendency 4) self-display tendency 5) degree of a dilettante life 6) pursuit-of-information tendency 7) bargain hunter tendency 8) TV preference tendency 9) pursuit-of-leisure tendency. Next, after the K-means cluster analysis was performed with nine factors of the life-style patterns, the life-styles of the respondents were classified into four groups which are named as the 'progressive practicality-oriented group', 'positive success-oriented group', 'sociable ostentation-oriented group', 'stable conservation-oriented group'. The analysis results for usage behavior between the market segments showed statistically significant differences in the frequency of usage, duration time in the store, consumer satisfaction, and loyalty. Also, we tried to investigate whether the large discount store consumers differently perceive the quality of service based upon the types of life-style. To measure the service quality of large discount store, we adapted several measurement models measuring the service quality such as SERVPERF, BCP, R-SERVPERF, R-BCP. MANOVA and One-Way ANOVA were performed to confirm the difference in service quality perception based on the market segments. The results have also shown significant differences between life-style types in service quality perception. These findings show that the large discount store marketers should consider consumer life-style as one of the most important market segments for marketing and understand the difference in service quality perception between life-style types. Our findings give important implications to marketers of large discount stores as well as life-style researchers. First, this study showed there were significant differences in consumer's service quality perception and usage behavior between the types of life-style. It provides evidence that the life-style approach can be a important basis in segmenting the large discount store market and will make consumers perceive the service quality high. Second, most previous researches on service quality have been in aggregate level. However, our results imply that the future research on service quality have to focus on segment level.
