Hepatic Fibrosis in Cholesterol and Sodium Cholate Diet-Fed Rats

  • Mostly, hypercholesterolemia has been focused on atherosclerosis and coronary heart disease and can be produced by intake of high cholesterol diet. However, toxic effects of cholesterol itself on liver and relationship between intake of high cholesterol diet and hepatic fibrosis have not been clearly investigated. Male Wistar rats were fed diet supplemented with 1.0 % cholesterol and 0.3 % sodium cholate for 12 weeks. Rats were sacrificed at 0, 3, 6, 9 and 12, respectively. Histopathological and blood chemical studies were performed on these animal sets. Total cholesterol, AST, ALT and LDH levels increased from week 3 and maintained around that level throughout the experiment compared to control. However, TG and albumin levels were the same or lower than those of control. Intake of high cholesterol and sodium cholate diet caused hepatic necrosis, macrophage infiltration, steatosis and fibrosis. Following feeding this diet to rats, hepatic necrosis, macrophage infiltration and steatosis markedly increased throughout the experiment, comparing to control. Collagen deposition and myofibroblasts were detected from at week 9 to 12 in the liver. Mast cell increased in proportion to the degree of hepatic damages. In conclusion, these results suggest that intake of high cholesterol diet is a risk factor on hepatic steatosis and fibrosis as well as atherosclerosis and coronary heart disease. Furthermore, this animal model for hepatic fibrosis can be use for application of anti-fibrogenic agents screening in vivo.

Suppression of DNCB-induced Dermatitis in NC/Nga Murine Model by Yanghyulyoonbu-tang (NC/Nga 아토피 피부염 생쥐 모델에서의 양혈윤부탕(養血潤膚湯) 효과)

  • Atopic dermatitis (AD) is a chronic inflammatory skin disease characterized by inflammatory cell infiltration in the skin. This study was performed to assess the therapeutic effects of YHYBT on the DNCB-induced dermatitis in NC/Nga mice, characterized by the onset of AD along with an increase the number of Th2 cells and dysregulation of inflammaroty mediators including cytokines and chemokines. YHYBT administration significantly reduced clinical dermatitis severity including pruritus, edema, eczematous and erythema. Histological findings indicated that the thickening of epidermis/dermis and dermal infiltration of inflammatory cells including mast cells were dramatically reduced. The suppression of dermatitis by YHYBT was accompanied by a decrease in the total number of immune cells in drained lymph node (DLN) and skin. Especially CD3+, CD4+ and CD3+CD69+ T cells in PBMC and DNL were greatly reduced. The level of IL-4 in CD3/CD28 activated splenocyte was downregulated, whereas that of IFN-'처리불가‘ was increased. Furthermore, the expression of eotaxin2 and CCR3 in skin were significanlty decreased. These data suggest that YHTBT may be effective therapeutic agents for the treatment of AD.

Consideration of the Groundwater Recharge Based on Environmental Isotopic Characteristics of the Small Basin in the Yeosu Area (환경동위원소를 이용한 여수지역 소유역에서의 지하수함양특성 고찰)

  • The processes and rates of groundwater recharge were studied by long-term monitoring of groundwater level and isotopic compositions of precipitation, surface water and groundwater in the Yeosu area. The isotopic compositions of surface water and groundwater were directly related to the precipitation event. It is also shown that the minimum amount of precipitation for infiltration to groundwater is about 20mm. The isotopic variations of groundwater shows that the Isotopic composition of groundwater changed by each precipitation event between June and Sep. 2000 is gradually changed without input of precipitation again. It indicates that the groundwater recharged from the upper part is mixed with the groundwater in reservoir. The infiltration rate of first precipitation event after a dry season is estimated to be 16.5% using isotopic mixing equations. It is expected that the groundwater recharge rate could be estimated more quantitavely, if the isotopic method is combined with the conservative methods.

A comparative study on characteristics of waterlevel responses to rainfall in the two aquifer systems (강우에 대한 지하수위 반응양상 비교분석 : 강원도 원주지역과 경기도 의왕지역)

  • We analyzed hydrologic time-series data obtained from a fractured aquifer in Wonju and a porous shallow aquifer in Uiwang area. Auto-correlation, spectral density, and cross-correlation functions were used for the analyses. Water level at a shallow well in Wonju was weakly auto-correlated and it was sensitive to direct infiltration from rainfall through soil zone while that at a well screened in the fractured zone showed a relative stability to an outer stress (rainfall), which was derived from a delayed transmission of recharge stress through a fracture network from a remote area. Characteristics of time-series data in Uiwang area were similar to those in the fractured zone in Wonju. This was caused by a regional recharge from a distant area rather than a direct infiltration. This study demonstrated a usefulness of time series analyses for identification of recharge mechanism.

LIDMOD3 Development for Design and Evaluation of Low Impact Development (저영향개발기법 설계 및 평가를 위한 LIDMOD3 개발)

  • In this study, the LIDMOD3 was developed to design and evaluate low impact development (LIDMOD). In the same fashion, the LIDMOD3 employs a curve number (NRCS-CN) method to estimate the surface runoff, infiltration and event mean concentration as applicable to pollutant loads which are based on a daily time step. In these terms, the LIDMOD3 can consider a hydrologic soil group for each land use type LID-BMP, and the applied removal efficiency of the surface runoff and pollutant loads by virtue of the stored capacity, which was calculated by analyzing the recorded water balance. As a result of Model development, the LIDMOD3 is based on an Excel spread sheet and consists of 8 sheets of information data, including: General information, Annual precipitation, Land use, Drainage area, LID-BMPs, Cals-cap, Parameters, and the Results. In addition, the LIDMOD3 can estimate the annual hydrology and annual pollutant loads including surface runoff and infiltration, the LID efficiency of the estimated surface runoff for a design rainfall event, and an analysis of the peak flow and time to peak using a unit hydrolograph for pre-development, post-development without LID, and as calculated with LID. As a result of the model application as applied to an apartment, the LIDMOD3 can estimate LID-BMPs considering a well spatical distributed hydroloic soil group as realized on land use and with the LID-BMPs. Essentially, the LIDMOD3 is a screen level and simple model which is easy to use because it is an Excel based model, as are most parameters in the database. This system can be expected to be widely used at the LID site to collect data within various programmable model parameters for the processing of a detail LID model simulation.

Suppression of DNCB-induced Dermatitis in NC/Nga Murine Model by Biheoonsupbang (비허온습방(脾虛蘊濕方)이 NC/Nga 아토피피부염 생쥐 모델의 피부 및 면역학적 변화에 미치는 영향)

BHOSB administration significantly reduced clinical dermatitis severity including pruritus, edema, eczematous and erythema. Histological findings indicated that the thickening of epidermis/dermis and dermal infiltration of inflammatory cells including mast cells were dramatically reduced. The suppression of dermatitis by BHOSB was accompanied by a decrease in the number of CD11b$^+$/Gr-1$^+$ immune cells in skin but not CD3$^+$/CCR3$^+$ cells. However, the number of CD3$^+$ cells was increased in BHOSB administrated NC/Nga mice. Oral administration of BHOSB significantly reduced the level of IL-6, TNF-a and eotaxin 2 mRNA in skin. These data suggest that BHOSB may be effective therapeutic agents for the treatment of AD.

Bioinformatic analyses reveal the prognostic significance and potential role of ankyrin 3 (ANK3) in kidney renal clear cell carcinoma

  • Kidney renal clear cell carcinoma (KIRC) is one of the most aggressive cancer type of the urinary system. Metastatic KIRC patients have poor prognosis and limited therapeutic options. Ankyrin 3 (ANK3) is a scaffold protein that plays important roles in maintaining physiological function of the kidney and its alteration is implicated in many cancers. In this study, we investigated differential expression of ANK3 in KIRC using GEPIA2, UALCAN, and HPA databases. Survival analysis was performed by GEPIA2, Kaplan-Meier plotter, and OS-kirc databases. Genetic alterations of ANK3 in KIRC were assessed using cBioPortal database. Interaction network and functional enrichment analyses of ANK3-correlated genes in KIRC were performed using GeneMANIA and Shiny GO, respectively. Finally, the TIMER2.0 database was used to assess correlation between ANK3 expression and immune infiltration in KIRC. We found that ANK3 expression was significantly decreased in KIRC compared to normal tissues. The KIRC patients with low ANK3 expression had poorer survival outcomes than those with high ANK3 expression. ANK3 mutations were found in 2.4% of KIRC patients and were frequently co-mutated with several genes with a prognostic significance. ANK3-correlated genes were significantly enriched in various biological processes, mainly involved in peroxisome proliferator-activated receptor (PPAR) signaling pathway, in which positive correlations of ANK3 with PPARA and PPARG expressions were confirmed. Expression of ANK3 in KIRC was significantly correlated with infiltration level of B cell, CD8+ T cell, macrophage, and neutrophil. These findings suggested that ANK3 could serve as a prognostic biomarker and promising therapeutic target for KIRC.

A Development of Groundwater Level Fluctuations Due To Precipitations and Infiltrations (강우에 의한 지하수위 변동 예측모델의 개발 및 적용)

  • In this study, a semi-analytical model to address groundwater level fluctuations in response to precipitations and its infiltration is developed through mathematical modeling based on water balance equation. The developed model is applied to a prediction of groundwater level fluctuations in Hongcheon area. The developed model is calibrated through a nonlinear parameter estimator by using daily precipitation rates and groundwater fluctuations data of a same year 2003. The calibrated input parameters are directly applied to the prediction of groundwater fluctuations of year 2004 and the simulated curve successfully mimics the observed. The developed model is also applied to practical problems such as a prediction of a effect of reduced recharge due to surface coverage change and a induced water level reduction. Through this study, we found that recharge to precipitation ratio is not a constant and may be a function of a precipitation pattern.

Noninvasive Monitoring of Bleomycin-induced Lung Injury in Rats Using Pulmonary Function Test

  • The single intratracheal instillation (ITI) of bleomycin (BLM) is a widely used method for inducing experimental pulmonary fibrosis in rat model. In the present study, pulmonary function tests (PFTs) of tidal volume ($V_T$), minute volume ($V_M$), and respiratory frequency ($F_R$) have been applied to study their possibility as a tool to monitor the progress of BLM-induced lung injury in rat model. Rats were treated with a single ITI of BLM (2.5 mg/kg) or saline (control). Animals were euthanized at 3, 7, 14, 21, and 28 days post-ITI. Lung toxicity effects were evaluated by inflammatory cell count, lactate dehydrogenase (LDH) activity in the bronchoalveolar lavage fluid (BALF), and light microscopic examination of lung injury. The PFT parameters were measured immediately before the animals were sacrificed. BLM treatment induced significant cellular changes in BALF-increase in number of total cells, neutrophils, and lymphocytes along with sustained increase in number of macrophages compared to the controls at days 3, 7, and 14. BALF LDH level was significantly increased compared to that in the controls up to day 14. On day 3, infiltration of neutrophils was observed in the alveolar spaces. These changes developed into marked peribronchiolar and interstitial infiltration by inflammatory cells, and extensive thickening of the interalveolar septa on day 7. At 14, 21, and 28 days, mild peribronchiolar fibrosis was observed along with inflammatory cell infiltration. The results of PFT show significant consistencies compared to the results of other toxicity tests. These data demonstrate that the most suitable time point for assessing lung fibrosis in this model is 14 days post-ITI of BLM based on the observation of fibrosis at 14, 21, and 28 days. Further, the progress of lung injury can be traced by monitoring the PFT parameters of $F_R$, $V_T$, and $V_M$.

A Study on Soil Slope Stability Design Considering Seepage Analysis (침투해석을 고려한 비탈면 설계에 대한 연구)

  • Most of slope failures are triggered by heavy rainfall during rainy season. If the rain keeps on for the season, the water content of the ground increases and its matric suction decrease, and then the safety factor of soil slope gets lower. The change of water table level for soil slope stability dose not describe the behavior of the soil slope in real situation, hence it may be necessary to modify the design standard for slope stability in association with rain infiltration. For correct design, economical construction, and maintenance of a soil slope, unsaturated flow analysis is needed for estimation of slope instability regarding water infiltration and soil behavior on unsaturated soil slopes. The entire soil slope cannot be saturated by prolonged rainfall and wetting band depth (saturated zone) just deepens from slope surface, hence the cause of the shallow surface slide is the wetting band depth depending on rainfall duration and intensity. Therefore, the paper presents the differences between theoretical equation and numerical analysis for wetting band depth on soil surface and its safety factor, and compares the slope stability obtained from unsaturated flow analysis with that obtained from conventional slope stability analysis.
