Is Koi Herpesvirus (KHV) Related to the Mass Mortality Occurring among Cultured Carp, Cyprinus carpio, in Korea?

  • Since 1998, a new viral disease with high mortality has been consistently recorded in Korea in cultured carp, Cyprinus carpio. In this study, we investigated an epizootic of the disease that caused high mortality rates in carp obtained from 11 farms in Korea between 1999 and 2007. Assessment of koi herpesvirus (KHV) levels in diseased carp was carried out to determine if this virus was the etiologic agent of disease in this instance. High mortality rates in carp were recorded mainly in the spring and autumn at water temperatures between $19^{\circ}C$ and $24^{\circ}C$. Diseased fish typically showed surface discoloration, with a thick opaque mucus covering the body and gills. Protozoan parasites and bacteria were recovered from 7/29 (24%) and 2/26 (8%) of fish, respectively. Evidence of viral infection was marked; cytopathic effects (CPEs), characterized by cell rounding and an extended cytoplasm in fathead minnow (FHM) cells, were detected in 40/41 fish (98%). A high mortality rate (80%) resulted when supernatants of cell cultures showing CPEs were applied to previously healthy fish. KHV was detected by polymerase chain reaction in 6/41 fish (15%), but was not detected in supernatants obtained from cell cultures showing CPEs. These results suggest that KHV may not be the etiologic agent of the high mortality occurring among cultured carp in Korea; therefore, some other-as yet unidentified-infective agent must be responsible.

Seroprevalence of antibodies to Neospora caninum and Toxoplasma gondii in cattle in northern area of Gyeongnam (경남북부지역의 소 Neospora caninum 및 Toxoplasma gondii 항체 양성률 조사)

  • In the present study, we carried out a seropositive survey of Neospora (N) caninum and Toxoplasma (T) gondii in cattle at northern area of Gyeongnam province, Korea. Serum sample was obtained from Holstein cattle (n=248, 44 farms in 5 counties) and Korean indigenous cattle (n=120, 23 farms in 4 counties). All 368 sera were tested for antibodies to N. caninum and T. gondii by an enzyme-linked immunosorbent assay (ELISA). Seropositive of N. caninum at farm and heads were 41.8% (28/67) and 12.8% (47/368), respectively. Twenty-five cattle (6.8%) were seropositive to T. gondii, and eighteen (26.9%) of 67 studied farms had cows seropositive to T. gondii. The seroprevalence of Holstein cattle was much higher than that of Korean indigenous cattle. There was no apparent association of T. gondii seropositivity with age, while the seropositivity for N. caninum was associated with cattle age. The farm of cattle above the age of 5 years (16.8%) was more infective than less than 4 years (9.3%). This is the first report on seroprevalence of N. canium and T. gondii in Holstein and Korean indigenous cattle in Gyeongnam, Korea.

  • This survey was carried out to investigate the seroprevalence of antibodies to Neospora caninum in Korean indigenous cattle that was raised in central area province Gyeongnam, Korea. A total of 719 sera were tested for N. caninum antibodies using ELISA (Herdcheck anti-Neospora, IDEXX Laboratories Inc., Westbrook, Maine USA). Seroprevalence of individual and farm were 29.8%(214/719) and 53.2%(50/94). Regional seropositive rates of the samples were 61%(47/77), 23.3%(7/30), 13.8%(49/355), 37.6%(77/205), 65.4%(34/52) at Changwon, Jinhae, Gimhae, Miryang, Yangsan, respectively. It showed difference at the age and on the herd size of farms. A herd of cattle above the age of 5 was more infective than under 4 years. And in seroprevalence by herd size farms having under 30 heads was top(35.7%). Seropositive 214 herds of N. caninum antibidies were tested for brucellosis by test tube. Positive rate of double infection was 16.4%(35/214).

Sputum Inflammometry to Manage Chronic Obstructive Pulmonary Disease Exacerbations: Beyond Guidelines

  • Quantitative sputum cytometry facilitates in assessing the nature of bronchitis associated with exacerbations of chronic obstructive pulmonary disease (COPD). This is not assessed in most clinical trials that evaluate the effectiveness of strategies to prevent or to treat exacerbations. While up to a quarter of exacerbations may be associated with raised eosinophil numbers, the vast majority of exacerbations are associated with neutrophilic bronchitis that may indicate airway infections. While eosinophilia may be a predictor of response to corticosteroids (oral and inhaled), the limited efficacy of anti-interleukin 5 therapies would suggest that eosinophils may not directly contribute to those exacerbations. However, they may contribute to airspace enlargement in patients with COPD through various mechanisms involving the interleukin 13 and matrix metalloprotease pathways. The absence of eosinophils may facilitate in limiting the unnecessary use of corticosteroids. The presence of neutrophiia could prompt an investigation for the specific pathogens in the airway. Additionally, sputum measurements may also provide insight into the mechanisms of susceptibility to airway infections. Iron within sputum macrophages, identified by hemosiderin staining (and by more direct quantification) may impair macrophage functions while the low levels of immunoglobulins in sputum may also contribute to airway infections. The assessment of sputum at the time of exacerbations thus would facilitate in customizing treatment and treat current exacerbations and reduce future risk of exacerbations.

A Study on the User Needs for Renovation of Race Track Type of Nursing Units in K University Hospital (K대학병원 병동부 이중복도형의 개선을 위한 사용자 요구 조사연구)

  • Purpose: General hospital in korea is getting large-scaled, specialized and upgraded. So, nursing unit of race track type has been advanced along with a spatial organization, building equipment and environmental approach. This study is to search the guidelines for renovation of nursing unit with race track type in the case of K university hospital in Daejeon. Methods: 9 cases of recently opened general hospital has been analyzed for spatial and environmental design solution. Questionnaires and interviews about user needs of nursing staff, patient and visitors, have been conducted for a nursing care system and facility of nursing units in K university hospital. Results: The user needs are represented as followings. Center core public zone in each floor plan and center core nursing station and supporting areas in nursing unit are good for an adjacency and separation of spatial organization. Toilet of group patient room is necessary to equip for easy, safe and infective aspects, and so toilet install will decrease the patient number and increase the nursing care service. Hall type of station front is good for visibility and observation. It is appropriate to renovate into south facing group patient room for privacy, enough space for nursing care facility and supporting area, enough width of door to move portable medical equipment, room for medical doctor and practical student, noise absorbing of day room. Implications: Upgrading the nursing care service and facility equipment is necessary to reflect the user needs and cooperate with hospital management.

ANTIMICROBIAL SUSCEPTIBILITY TEST ON STREPTOCOCCUS VIRIDANS IN CHILDREN'S ORAL CAVITY (소아의 구강내에서 검출된 Streptococcus viridans에 대한 항균제 감수성 연구)

  • A large number of streptococci that do not fit readily into any of the established classification schemes have been relegated to a large heterogeneous group called the Streptococcus viridans, which are members of the normal flora of the mucous membranes of the body, including the oral cavity, the nasopharynx, and genitourinary tract. This group includes S. mitis, S. oralis, S. sanguis, S. salivarius, S. milleri, etc. Surveying on the literature, it has been reported that infective endocarditis, meningitis, rhabdomyolysis, cholangitis, appendicitis caused by Streptococcus viridans, which were the most important pathogen in children with malignant hematologic disease. Various antibiotics has been chosen for treatment or prophylaxis for these infections, but were generally lower antimicrobial susceptibilities because of an abuse of antibiotics and advent of resistant group. Therefore, surveillant culture must be performed to evaluate personal antimicrobial susceptibilities of intraoral microbes for proper antimicrobial choice for dental procedures. This study examined sampling from subgingival plaque of 60 chidren's microbes. The cultured bacterial isolates, Streptococcus viridans were examined 10 antimicrobial drugs with the Kirby-Bauer agar disk diffusion method. The used drugs were Penicillin, Ampicillin, Oxacillin, Cephalothin, Imipenem, Gentamicin, Erythromycin, Vancomycin, Ciprofloxacin, Clindamycin. The results were as follows : 1. Sampling Streptococcus viridans were S. mitis(65%), S. oralis(22%), S. sanguis(5%), S. intermedius(3%), S. salivarius(2%), S acidominimus(2%), Unidentified streptococcus(2%). 2. The antimicrobial susceptibility of total Streptococcus viridans : Oxacillin< Erythromycin< Pencillin=Ciprofloxacin< Cephalothin< Ampicillin< Clindamycin< Gentamicin< Imipenem=Vancomycin. 3. The antimicrobial susceptibility of S. mitis : Oxacillin=Erythromycin< Ciprofloxacin< Cephalothin< Penicillin=Ampicillin< Gentamicin< Clidamycin< Imipenem=Vancomycin. 4. The antimicrobial susceptibility of S. oralis : Oxacillin< Erythromycin< Penicillin=Ciprofloxacin=Clindamycin< Cephalothin=Gentamicin< Ampicillin< Imipenem=Vancomycin. 5. There was no significant difference in the antimicrobial susceptibility among each Streptococcus viridans group.

A Study about the Effect of Antigrowth Virus (Third Virus of Swine) on Small Animals (Antigrowth Virus (Third Vhird Virus of Swine)의 소동물에 대한 연구)

  • Durlng various experiments and investigations with antigrowth virus, I studied especially the infective and antiinfective process of it in small animals, and have got following results: 1. The rabbits are easily infected with this virus, having following five stages; incubation period of 2 to 3 days, stage of fever for 2 to 4 day, transient anemic stage, transient fcteric stage and stage of lasting iron-precipitation in the spleen. 2. According to the appearance of icteruis, I have divided the rabbits into following three groups, the first gorup, weighting less than 1000g, died after appearance of icterus, the second group, weighting 1000 to 1500 g. recoy ered after appearance of icterus, and the third group, weighting more than 1500 g. recovered without the appearance of jaundice. 3. In hematological study, marked decrease in R. B. C. and Hb were recognized but various leucocytes except lymphocytes slightly increased. 4. In pathological study, the spleen exhibited marked swelling with abundent blood and indistinct border between trabeculae and pulps at the initial stage of icterus and notable atrophy at the end stage of it. There are few iron-cells in the spleen histologically at the initial stage, but they increased in number with granuar appearance at the end stage. In the first group of rabbits above mentionel, the iron-cells increase suddenly with the death of animal and show diffuse precipitation of iron. The liver and indey have defferent aspects in proportion to icterus. If icterus appears, they show markel changes with turbidity and necrosis, but without icterus they present only slight changes. 5. The neutralining antibody is easily proved in the immunized rabbits for about one year. 6. The passive immunity has the defense immunability until 10\ulcorner 7. Gunia pigs are easily infected with this virus and get the immunity. On fowls this virus can be kept in the blood for about ten days by various inoculating methods, and the fowls obtain the defensive ability by reinoculation, but are unable to succeed this ability from generation to generation. 8. when young swine are infected with this virus, their growth are disturbed and result in atrophied swine. It is therofor presumed that the etiology of the antigrowth of these atrophied swine is similar morphological and functional changes observed in the above rabbits.

Excessive Food Restriction in Children with Atopic Dermititis (아토피피부염 영유아의 식품섭취 제한에 관한 실태조사)

  • The prevalence of Atopic Dermatitis (AD), a non-infective chronic inflammatory skin disease, is increasing worldwide. Avoiding the allergen is the basic principle in the treatment of AD. However, when the allergen is food, excessive restriction can lead to nutrition deficiency. The objective of the study was to examine the status of the dietary restriction and compare the caregiver's restriction practice with doctor's recommendation in Korean children with AD. A total of 158 children diagnosed with Atopic Dermatitis were recruited for this study. Information about foods that aggravate AD symptoms and food restriction were collected from the mothers of 158 children aged 6 month-5 year with AD using questionnaires. Food restriction recommendation by doctor was collected through medical chart. McNemar and Margianl homogeneity tests were used to detect a relationship between food restriction recommended by doctor and current practice by mother. There were significant proportion differences of food restriction for each food between by doctor and mother. We found 75.9% of children were avoiding eggs although only 61.4% were recommended for egg restriction by a doctor. Children with restriction of more than 4 kinds of food were 53.2% compared to 13.3% by doctor. Excessive restrictors tended to be younger and diagnosed at younger age. The caregivers of excessive restrictors had trends of "being older" and "having higher income". Avoidance of common foods in children without food allergy could result in malnutrition or impaired growth. Nutrition education is needed for sound practice and nutrition care in children with Atopic Dermatitis as well as interactive communication between caregivers and experts.

Pathogenicity of Entomopathogenic Nematode, Steinernema carpocapsae against Fall Webworm, Hyphanria cunea (Lepidoptera: Arctiidae) (미국흰불나방(Hyphanria cunea)에 대한 곤충병원성선충 Steinernema carpocapsae의 병원성)

  • Environmentally sound control of fall webworm, Hyphanria cunea (Drury) with entomopathogenic nematode, Sreinernema carpocapsae Pocheon strain was evaluated in the laboratory and pot. Pathogenicity of 5. carpocapsae Pocheon strain was different depending on larval stage, i.e., mortality of the 2nd instar and the $3\~4th$ instar was $100\%$ with >20 infective juveniles (Ijs)/larva in 3 days, but the 5th instar was $34\%$ with (Ijs)/larva in 3 days. Pathogenicity of 5. carpocapsae Pocheon strain was higher with increasing nematode concentration. Mortality of Hyphanria cunea larva by 5. carpocapsae Pocheon strain was not significantly different (more than $70\%$) between nematode concentration on treated trees (Malus alba and Platanus orientalis) and in pot.

Loss of a Strain-Specific Protein by Bacterial Infection in Amoeba proteus (Amoeba proteus에 있어서 박테리아 감염에 의한 변이주 특이성 단백질의 손실)

  • By two-dimensional gel electrophoresis loss of a cell-specific protein was detected in tD strain of Amoeba proteus that had been infected by symbiotic bacteria extracted from xD strain. In 50 days of experimental infection by induced phagocytosis the host amoeba lost the ability to synthesize the tD cell-specific protein even after removal of the infective bacteria and xD cell-specific protein by growing the amoebae at $27^\\circC$. By this time the host amoebae were obligately dependent on the bacteria. From these and other results (Lorch and Jeon, Science 221:549), it is clear that the incompatibility of the infected nuclei with the cytoplasm of the uninfected amoeba and the obligate dependence of the host on bacteria are due to the irreversible inactivation or the loss of the cell-specific gene by bacterial infection in this amoeba.
