Determination of β-Carotene in Infant Formulas by High-Performance Liquid Chromatography (HPLC를 이용한 조제유류 중 베타카로틴 함량 분석 연구)

  • A procedure based on high-performance liquid chromatography (HPLC) is described to determine ${\beta}-carotene$ in infant formulas. The method for ${\beta}-carotene$ analysis was performed on a C18 reversed-phase column using acetonitrile/methanol/dichloromethane (6:1:3, v/v/v) as a mobile phase. ${\beta}-Carotene$ was determined in HPLC with photo diode array (PDA) detector. The parameters of validation were specificity, linearity, LOD, LOQ, accuracy, precision and repeatability. The specificity was confirmed by the retention time and the linearity ($R^2$), which was over 0.999 in the range of 0.125~2 mg/L. The detection and quantification limits were 0.064 and 0.193 mg/L, respectively. The accuracy and precision of this method using an STD spiked sample were 80~119% and 1.02~2.05% respectively. The method was applied to the analysis of various infant formula and follow-up formulas products containing ${\beta}-carotene$, and all the products contained acceptable levels of ${\beta}-carotene$ for nutrition labeling.

Determination of Aflatoxin M1 and Heavy Metals in Infant Formula Milk Brands Available in Pakistani Markets

  • Aflatoxin $M_1$ ($AFM_1$) after its bioconversion from aflatoxin $B_1$ in animal liver becomes the part of milk while heavy metals get entry into milk and milk products during handling in the supply chain. Aflatoxin $M_1$ and heavy metals being toxic compounds are needed to be monitored continuously to avoid any ailments among consumers of foods contaminated with such toxicants. Thirteen commercially available infant formula milk (IFM) brands available in Pakistani markets were analyzed for the quantitative determination of $AFM_1$ and heavy metals through ELISA and atomic absorption spectrophotometer, respectively. $AFM_1$ was found positive in 53.84% samples while 30.76% samples were found exceeding the maximum EU limit i.e. $0.025 {\mu}g/kg$ for $AFM_1$ in IFM. Heavy metals lead (Pb) and cadmium (Cd) were found below the detection limits in any of the sample, whereas the concentrations of iron (Fe), zinc (Zn) and nickel (Ni) ranged between 45.40-97.10, 29.72-113.50 and <$0.001-50.90 {\mu}g/kg$, respectively. The concentration of Fe in all the tested brands was found in normal ranges while the concentrations of Zn and Ni were found exceeding the standard norms. Elevated levels of $AFM_1$, Zn and Ni in some of the tested IFM brands indicated that a diet completely based on these IFM brands might pose sever health implications in the most vulnerable community i.e., infants.

Growth and Development of Infants Fed Soy- Based Formulas over 3 Months (액상 대두 유아식을 3개월 이상 섭취한 영유아의 성장과 발달)

  • The objective of this study was conducted to investigate growth and development status of infants fed soy-based formulas over 3 months. The height and weight were measured and Z-scores were calculated by using standard of the same age groups. Their mothers were interviewed using questionnaires including general and environmental characteristics, total food intakes, soy based formula intakes and Ewha infant development screening test. Main results were as follows: 1) Nutrient intake levels of subjects were similar to or more than the level of Korean Recommended Dietary Allowances except for intakes of vitamin E (79.89% RDA), and the average status of nutrient intakes of infants were fairly good. 2) Z-scores of height for age (HAZ) and Z-scores of weight for age (WAZ), Kaup index, WLI and Ewha Infant Developmental Screening Test score of subjects were in the normal growth range. 3) There were no significant differences among soy based formula intake percentile groups in HAZ, WAZ, Kaup index, WLI and Ewha Infant Developmental Screening Test score. 4) Total energy intake was positively correlated with HAZ (p < 0.00, WAZ (p <0.00, and WLI (p < 0.05) in infants less than 12 month. Also, soy based formula energy intake was positively correlated with HAZ (p < 0.05) in infants less than 12 month. However, energy and soy based formula intake levels of infants over 12 month were not significant among variables. Considering results of this study, infants fed soy-based formulas over 3 months showed normal growth and development status. Further studies are needed to evaluate longterm growth and development in infants fed soy based formulas.

A Study of the Influence of Environmental Factors and Factors Associated with Feeding on Infants' Growth (영아의 성장발육에 영향을 미치는 환경요인 및 섭식관련 요인에 관한 연구)

  • This study was aimed to evaluate any influence of environmental factors associated with feeding on infants' growth. One hundred infants(62 males and 38 females) aged 4-9 months were measured for length and were weighed. Their mothers were interviewed using questionnaires including general and environmental characteristics and maternal feeding attitude, ideal body habitus, infant's food behavior, and infant's food behavior, and infant's temperament. A further study was conducted on 29 infants at the time their ages reached 12 months. Their dietary intakes were assessed, and blood hemoglobin and serum ferritin concentrations were measured. Z-scores of weight-for-age(WAZ) and Z-scores of height-for-age(HAZ) of 4-9 month- old infants were not correlated with age, education level, income, weight, and height of their parents. Formula-fed infants had significantly lower WAZ and body mass index (BMI) than those of breast fed infants. WAZ was significantly correlated with maternal feeding attitude, but not with ideal body habitus, infant's food behavior or infant's temperament. WAZ of 12-months after birth because of supplementary foods introduced from 6 months of age. WAZ and BMI were positively correlated with blood hemoglobin concentration. Therefore, to ensure proper growth of infants, efforts should be given to postental factors such as breast-feeding, maternal feeding attitude, adequate iron status as well as prenatal factors. Monitoring Bacillus cereus and Aerobic Bacteria in Raw Infant Formula and Microbial Quality Control during Manufacturing (영.유아용 식품원료의 Bacillus cereus와 일반세균 모니터링 및 제조공정 중 미생물 품질제어)

  • The purpose of this study was to examine the presence of Bacillus cereus, aerobic bacteria and coliforms in the raw material of infant formulas and investigate the manufacturing process in terms of microbial safety. Among ten kinds of raw infant formula material samples (n=20), Bacillus cereus appeared in two (n=4). Aerobic bacteria were not detected in raw infant formula material or maximum 4.15 log CFU/g. Eleven species of aerobic bacteria were isolated and 76% of them were Sphingomonas paucimobilis, Pseudomonas fluorescens, Rhizobium radiobactor, or Stenotrophomonas maltophilia. A Pearson's correlation analysis revealed that the most influential factors for detecting Bacillus cereus were aerobic bacteria and coliforms. In other words, when the measured values of aerobic bacteria and coliforms were higher, the possibility that Bacillus cereus would appear increased. In a regression model to predict Bacillus cereus, the rate of appearance was correlated with aerobic bacteria and coliforms, and its contribution rate for effectiveness was 86%. Improving microbial quality control by pasteurization, spray dry, popping and extrusion resulted in a decrease in the numbers of Bacillus cereus, aerobic bacteria and coliforms in the raw materials. The results suggest that a hazard analysis and critical control point system might be effective for reducing microbiological contamination.

Effects of Human Milk Fortifier on Growth and Nutritional Status of Growing Rats Fed Infant Formula (모유강화제(HMF)가 흰쥐의 성장과 영양상태에 미치는 영향)

  • This study was conducted to evaluate the efficacy of human milk fortifier (HMF) on growth and nutritional status in growing rats fed infant formula supplemented with HMF. Three week-old male Sprague-Dawley rats were divided into three groups and fed regular formula (RF), premature formula (PF) and regular formula fortified with HMF (RF+HMF) diets for 3 weeks. There was no significant difference in weight gain among groups. However, a significant increase of food intake was observed in PF and RF+HMF groups compared with RF group. With increasing food intake, the intakes of carbohydrate and protein were significantly increased in PF and RF+HMF groups. The weight of perirenal fat was significantly increased in rats fed RF+HMF; however, the weights of liver, kidney and spleen were not significantly different among groups. Although total lipids, total cholesterol, HDL-cholesterol concentrations of serum were not significantly different among groups, triglyceride was significantly increased in PF group. The triglyceride and total-cholesterol of liver were significantly increased in rats fed regular formula fortified with HMF and PF compared with RF group. Glutamic pyruvic transaminase (GPT), glutamic oxaloacetic transaminase (GOT), creatinine (Cre) and blood urea nitrogen (BUN) in serum showed no significant difference among groups. The concentration of growth hormone was significantly increased in PF group compared with other groups. The concentration of hemoglobin was significantly increased in rats fed PF and RF+HMF. These results suggest that the supplementation of human milk fortifier in growing rats may promote growth as increasing food intake and lipid contents in tissues and prevent the anemia of infants.

  • In this study, we selected supplementary vegetable resources to develpe nutritious supplementary model of korean formula for infants and children, purchased and analyzed it to use with a model. We also referred to an old book about Korean vegetable resources and searched a form of vegetable resources. There are a lot of vegetable resources of various kinds which lied scattered in mountains and fields of our country. As for these, a beyond compare menstruation activity component is becoming clear with a lot of things. It is thought that depth search and study of these vegetable resources and practical application to a infant and child supplemen is desirable. Therefore, we choosed vegetable resources ; a Capsella brusapastoris, a Codonopsis lanceolata, Dandelion, a wild plant, a Tractylis ovata(dried), a Tractylis ovata(native), a Pine plant, a burdock, a JanDae, a Plantain, a HollpDae, which gathered in our country, and we analyzied priximate aomposition of sample (moisture, ash, total carbohydrate, crude fat, crude protein, dietary fiber) and micronutrient contents of sample (calcium, Iron, flavonoids, vitamin A, vitamin C, vitamin E).

Comparison of the Heat Treatment Intensity in Infant Formulae (조제분유의 열처리 정도 비교)

  • The heat treatment indicators such as HMF contents, lactulose contents and whey protein denaturation rates were measured to refer to the heat treatment intensity of domestic market infant formulae. The HMF contents showed $21.0{\sim}43.9{\mu}mol/l:$ in the case of powder types, the HMF contents in enriched nutrient products(ii) were higher whereas in the case of liquid types they were packed in cans(i). The lactulose contents showed $2.5{\sim}11.4mg/100ml$ in the powder type and $27.0{\sim}164.8mg/100ml$ in the liquid type. There was much difference in the lactulose contents according to the product types. Compared with the ADPI standards, most of infant formulae were considered to be medium-heat class. The whey protein denaturation rates were $1.1{\sim}69.4%$ in the powder type and $37.4{\sim}71.3%$ in the liquid type.

Establishment of Choline Analysis in Infant Formulas and Follow-up Formulas by Ion Chromatograph (이온크로마토그래프를 이용한 조제유류 및 영아용·성장기용 조제식 중 콜린 함량 분석법 연구)

  • This study was conducted to establish the analysis method for the contents of choline in infant formulas and follow-up formulas by ion chromatograph (IC). To optimize the method, we compared several conditions for extraction, purification and instrumental measurement using spiked samples and certified reference material (CRM; NIST SRM 1849a) as test materials. IC method for choline was established using Ion Pac CG column and 18 mM $H_2SO_4$ mobile phase. The parameters of validation were specificity, linearity, LOD, LOQ, recovery, accuracy, precision and repeatability. The specificity was confirmed by the retention time and the linearity, $R_2$ was over 0.999 in range of 0.5~10 mg/L. The detection limit and quantification limit were 0.14, 0.43 mg/L. The accuracy and precision of this method using CRM were 95%, 2.1% respectively. Optimized methods were applied in sample analysis to verify the reliability. All the tested products were acceptable contents of choline compared with component specification for nutrition labeling. The standard operating procedures were prepared for choline to provide experimental information and to strengthen the management of nutrient in infant formula and follow-up formula.

Detection of Irradiated Milk Formulas using Electron Spin Resonance (전자스핀공명법(ESR)을 이용한 방사선조사 조제유류의 판별)

  • Electron Spin Resonance (ESR) spectroscopy has been used to detect the presence of radiation-induced free radicals in biological samples since the mid 1950s and to irradiate foods containing cellulose, crystalline sugar, and bone. Therefore, we analyzed the ESR spectrum of irradiated infant formula and its ingredients in this study. Samples were irradiated with 2 different radiation sources of $^{60}Co$ gamma rays and electron beams (EBs), and the absorbed doses were 0, 1, 3, 5, and 7 kGy. ESR measurements were performed under normal atmospheric conditions using a JEOL JES-FA100 spectrometer equipped with an X-band bridge. Irradiated infant formula showed anunsymmetrical spectrum ($g_1$=2.0050, $g_2$=2.0006); in contrast, non-irradiated samples showed asymmetrical spectrum. The ingredients of irradiated samples showed a multi-component ESR signal in glucose and lactose and a singlet-type spectrum in milk powder (g=2.0050). $R^2$ of the dose-response curve showed a fine linearity of over 0.95 across the entire sample. We also compared the spectra of identical samples irradiated with $^{60}Co$ gamma rays and EBs, because EBs can be used for food irradiation in foreign countries, although this is not permitted in Korea. However, we could not find any significant differences according to the types of radiation source. Thus, ESR spectroscopy can be used to detect irradiated infant formula and several types of primary ingredients in this formula.
