Cocoon Yield Pattern and Analysis of Water, Soil and Leaves from Mulberry Gardens Irrigated with Polluted Water Around Bangalore, India

  • Sericulturists in the vicinity of Bangalore city irrigate their mulberry gardens with Vrishabhavathy stream water, which is polluted with domestic and industrial wastes from the city. To investigate the effect of pollutants on silkworm crops, a detailed field survey was conducted to study the cocoon yield pattern of the crops raised on mulberry irrigated with wastewater as against irrigation by bore/open wells. The villages along the stream were grouped into five test batches at about a distance of $5{\sim}8$ km from each other. The seasonal yield data with relevant information were collected through questionnaires from 117 rearers using stream water and 35 rearers using bore/open wells, the latter serving as control group. The average yield for 100 layings was 46 to 57 kg in the control group whereas in test groups, it ranged between 34 to 51 kg in the first test group and 22 to 38 kg in the rest. The difference in yield was $9{\sim}19$ kg depending on the season between control and test batches. In summer, this difference was higher, with high co-efficient of variation in test groups ($33{\sim}52$%). Further, water, soil and leaf samples were collected from selected rearers and were analyzed for zinc, copper, iron, lead and nickel. Results indicated significantly higher contents of these metals in samples from gardens using wastewater when compared control samples. Significantly (p<0.05 & p<0.01)) higher levels of zinc ($24{\sim}122$ ppm) and iron ($208{\sim}683$ ppm) were noticed in mulberry leaves during summer followed by winter and rainy season. The significance of high content of heavy metals in mulberry leaves and cocoon yield pattern of this area in relation to the quality of irrigation water is discussed.

An Analysis of Specialization Factors for Greenhouse-Grown Spinach and Lettuce (시설 시금치와 상추의 특화 요인 분석)

  • This paper analyzes the specialization factors for greenhouse-grown horticultural crops and location quotient. We derived the location quotient of two crops from the Agricultural Census in 2000, 2005, and 2010. The location quotient shows that specialization factors for spinach and lettuce became more concentrated. This means that cultivation of vegetables lean too much towards specialized regions. Implementation policies based on the level of specialization is expected to more efficiently improve the competitiveness of regional agriculture. The logit analysis showed that GRDP, age, education, average employee, and ratio of computer use had a significant impact on specialization of spinach. GRDP, age, and education negatively affected the specialization coefficient of spinach. However, average employee, and ratio of computer use positively affected the specialization coefficient of spinach. Based on these results, we set up the policy tools specializing the regional location quotient.

Effect of Preharvest Treatment of $CaCl_2$ and Alkaline-Reduced Water on the Quality of Oyster Mushroom during Storage (수확 전 염화칼슘 및 알카리환원수 처리가 느타리버섯 저장에 미치는 영향)

  • This experiment was focused to determine the effect of preharvest treatment of alkaine-reduced water (ARW) and $CaCl_2$ on the storage of oyster mushroom. ARW was prepared through electrolysis of water with $CaCl_2$ (0.1%) as salts and $CaCl_2$ (0.1%) was dissolved in deionized water. Mushrooms were harvested after two sprays of each solutions and quality factors including firmness and browning was determined during storage. The severity of browning was more greater when stored 20 days. Unlike $CaCl_2$, ARW containing 139 ug/L calcium was positive on maintaining freshness in terms of delay of firmness loss and respiration drop, keeping soluble carbohydrate content such as trehalose, and inhibiting the occurrence of browning. Browning, however, seemed to be more influenced by the developmental stage of mushroom at harvest. Even no significant difference was found, the level of ${\beta}$-glucan was low in ARW treatment compared to that of control. Further research including application and preparation methods of ARW is required to clarify the potential of ARW on keeping freshness of oyster mushroom through inhibiting tissue browning.

Identification and molecular characterization of downy mildew resistant gene candidates in maize (Zea mays subsp. Mays)

  • Downy mildew (DM), caused by several species in the Peronosclerospora and Scleropthora genera, is a major maize (Zea mays L.) disease in tropical or subtropical regions. DM is an obligate parasite species in the higher plants and spreads by oospores, wind, and mycelium in seed surface, soil, and living hosts. Owing to its geographical distribution and destructive yield reduction, DM is one of the most severe maize diseases among the maize pathogens. Positional cloning in combination with phenotyping is a general approach to identify disease resistant gene candidates in plants; however, it requires several time-consuming steps including population or fine mapping. Therefore, in the present study, we suggest a new combination strategy to improve the identification of disease resistant gene candidates. Downy mildew (DM) resistant maize was selected from five cultivars using the spreader row technique. Positional cloning and bioinformatics tools identified the DM resistant QTL marker (bnlg1702) and 47 protein coding genes annotations. Eventually, 5 DM resistant gene candidates, including bZIP34, Bak1, and Ppr, were identified by quantitative RT-PCR without fine mapping of the bnlg1702 locus. Specifically, we provided DM resistant gene candidates with our new strategy, including field selection by the spreader row technique without population preparation, the DM resistance region identification by positional cloning using bioinformatics tools, and expression level profiling by quantitative RT-PCR without fine mapping. As whole genome information is available for other crops, we propose applying our novel protocol to other crops or for other diseases with suitable adjustment.

Inactivation of Foodborne Pathogens by Lactic Acid Bacteria

  • The problems caused by foodborne pathogens are not only a concern to the food industry but also with regard to global public health. Over the years, fermentation technology has proved to be one of the cheapest and safest methods for inactivating and controlling pathogenic microorganisms in food. Scientific evidence shows that lactic acid bacteria fermentation exerts significant antimicrobial effect against pathogenic bacteria and viruses. Lactic acid bacteria metabolites such as organic acids, bacteriocins and hydrogen peroxides have adverse effects on foodborne pathogens which lead to their inhibition. These compounds do not only cause physical injuries, but also have significant effects on the pathogens' gene expression. Furthermore, the presence of lactic acid bacteria in food provides nutritional competition among foodborne pathogens, and all these factors together suppress their growth. This study reviews our current knowledge of the antimicrobial abilities of lactic acid bacteria, their molecular mechanisms, and their application for inactivating foodborne pathogens.

Assessment of Root-Associated Paenibacillus polymyxa Groups on Growth Promotion and Induced Systemic Resistance in Pepper

  • Twenty-nine P. polymyxa strains isolated from rhizospheres of various crops were clustered into five genotypic groups on the basis of BOX-PCR analysis. The characteristics of several plant growth-promoting factors among the isolates revealed the distinct attributes in each allocated group. Under gnotobiotic conditions, inoculation of pepper roots with P. polymyxa isolates significantly increased the biomass in 17 of total 29 treated plants with untreated plants. Experiments on induced systemic resistance (ISR) against bacterial spot pathogen Xanthomonas axonopodis pv. vesicatoria in pepper by P. polymyxa strains were conducted and only one isolate (KNUC265) was selected. Further studies into ISR mediation by the KNUC265 strain against the soft-rot pathogen Erwinia carotovora subsp. carotovora in tobacco demonstrated that the tobacco seedlings exposed to either bacterial volatiles or diffusible metabolites exhibited a reduction in disease severity. In conclusion, ISR and plant growth promotion triggered by P. polymyxa isolates were systemically investigated on pepper for the first time. The P. polymyxa KNUC265 strain, which elicited both ISR and plant growth promotion, could be potentially used in improving the yield of pepper and possibly of other crops.

Water-circulated Bed Cultivation of Water Wasabi I. Growth Change of Water Wasabi in Different Conditions of Water-circulated Bed (물 고추냉이 순환식 베드 재배 연구 I. 베드조건의 차이에 따른 물 고추냉이의 생장량 차이)

  • 물 고추냉이는 십자화과의 숙근성 다년생 초본식물로서 일본과 대만 등에서 재배되며 세계 여러 나라에서 식품, 가공원료, 향신료, 의약소재 등으로 이용되고 있다. 우리나라에서는 철원과 무주 등에서 재배되고 있으나 재배환경 및 기술의 한계로 규모와 생산량은 매우 저조한 실정이다. 따라서 본 연구는 물 고추냉이 순환 베드시스템 개발을 위한 기초연구로 수행되었다. 마사토의 높이와 고랑의 유무에 따라 3개 베드 (A-마사토 높이 4.0cm, 고랑 있음, B-마사토 높이 5.5cm, 고랑 있음, C-마사토 높이 7.0cm, 고랑 없음)로 시험하였다. 물 고추냉이의 생장량과 식물체 내의 양분 분석(질소, 인산, 칼륨)을 분석하였다. 그 결과 베드 높이가 가장 낮고(4cm) 고랑이 있는 베드에서 생장량이 가장 우수하였고 마사토 높이가 가장 높은(7cm) 처리구보다 생장량이 2배 이상 증가하였다. 전체적으로 엽장보다 엽폭이 길었고 심장형 잎의 특성을 그대로 나타내었다. 식물체 내의 양분 분석 결과 처리구 별로 질소, 인산, 칼륨의 함량 차이가 크게 나타났다. A 베드에서 질소 4,150, 인산 500, 칼륨 1,500ppm 에 비하여 C 베드는 질소 730, 인산 120, 칼륨 700ppm 정도로 낮았다. 대조구의 동일 지역, 동일 기간의 계류지 하우스재배(데이터 미발표)에 비해 순환식 베드 재배가 우수하였고, 특히 A 베드에서 가장 우수하여 절수, 2단 재배 등의 예상되는 효과를 감안할 때 물 고추냉이 순환식 베드 재배의 가능성이 충분하다고 판단된다.

Effects on the Storability Enhancement and Root Yield by Air Curing of the Seed Rhizome of Rehmannia glutinosa Liboschitz ex Steudel (지황 종근의 저장성 및 수량에 미치는 음건 처리의 효과)

  • Background: The use of Rehmannia glutinosa of the family Scrophulariaceae, in traditional medicine is well known. This study was conducted to elucidate the effect of air curing of the seed rhizome of R. glutinosa on its storability and yield. Methods and Results: The root of the R. glutinosa cultivar (Dagang) was harvested in at the end of November. The seed rhizomes were air cured for one to seven days. They were subsequently wrapped with newspaper and further stored in a plastic container at $1^{\circ}C$. It was observed that the weight loss and decay rates were significantly lower in the air cured seeds than in the untreated ones. Moreover, the decay rate of the control was approximately 50%, 120 days after storage. However. the decay rate of all the air cured treatment groups was less than 1%. Additionally, air curing led to an increase in the germination rate of the seeds and the root yield when compared with the untreated groups. Taken together, the ideal treatment period for air curing was found to be 3 days. Under these conditions, the germination rate and yield were 88.7% and 2,185 ㎏/10a, respectively. Conclusions: This study successfully demonstrated that the air curing of seed rhizomes can lead to a considerable increase in the storability and yield of R. glutinosa.

Deep Learning-based Rice Seed Segmentation for Phynotyping (표현체 연구를 위한 심화학습 기반 벼 종자 분할)

  • The National Institute of Agricultural Sciences of the Rural Developement Administration (NAS, RDA) is conducting various studies on various crops, such as monitoring the cultivation environment and analyzing harvested seeds for high-throughput phenotyping. In this paper, we propose a deep learning-based rice seed segmentation method to analyze the seeds of various crops owned by the NAS. Using Mask-RCNN deep learning model, we perform the rice seed segmentation from manually taken images under specific environment (constant lighting, white background) for analyzing the seed characteristics. For this purpose, we perform the parameter tuning process of the Mask-RCNN model. By the proposed method, the results of the test on seed object detection showed that the accuracy was 82% for rice stem image and 97% for rice grain image, respectively. As a future study, we are planning to researches of more reliable seeds extraction from cluttered seed images by a deep learning-based approach and selection of high-throughput phenotype through precise data analysis such as length, width, and thickness from the detected seed objects.

  • The aim of this study is to assess the potential for fusion and convergence in industrial agriculture. First, we analyzed the types of industrial agriculture and applied the agri-business and ICT agriculture. After then, we analyzed the Project Potential Index (PPI) of the agri-business and ICT agriculture using the agricultural enterprise database provided from Rural Research Institute in Korea. The results revealed that Haenam have a high potential for agri-business project because of large number of farmers and annual sales. Wanju was considered as the suitable place for ICT agriculture project because of large area of greenhouse. This study was applied only 7 study area but the methodology suggested in this study could be widely used for assessing potential project various types of industrial agriculture.
