Study on the Effect of SGHHT on Hyperlipidemia (순기활혈탕(順氣活血湯)이 고지혈증(高脂血症)에 미치는 영향(影響))

  • As a long-term goal for the development of new prescriptions for hyperlipidemia, SGHHT was examined in the present study using a rat model in which the hyperlipidemia was induced. The major parameters related to lipid metabolism were investigated and the key findings are summarized below. 1. The body weight of hyperlipidemia-induced rats began to show lower body weight beginning one week after SGHHT treatment compared to non-treated control group animals. 2. Cholesterol levels showed a significant decrease beginning three weeks after SGHHT treatment, compared to hyperlipidemia-induced control group. 3. Total cholesterol levels in SGHHT-treated animal group were significantly decreased compared to the hyperlipidemia-induced control group. 4. Glucose levels in SGHHT-treated animal group were significantly decreased compared to the hyperlipidemia-induced control group. 5. Triglyceride levels in SGHHT-treated animal group were significantly decreased compared to the hyperlipidemia-induced control group.6. SGOT levels in SGHHT-treated animal group were significantly decreased compared to the hyperlipidemia-induced control group whereas there was no significant change in SGPT levels. 7. HDL-cholesterol levels were significantly increased in SGHHT- treated animal group compared to the hyperlipidemia-induced control group. 8. LDL-cholesterol levels were significantly decreased in SGHHT-treated animal group compared to the hyperlidemia-induced control group.

The Effects of SWS(Sahyang·Woohwang·Samchilkeun) on Hyperlipidemia and Brain Damage (사향(麝香)·우황(牛黃)·삼칠근(三七根) 복합방(複合方)이 고지혈증(高脂血症) 및 뇌손상(腦損傷)에 미치는 영향(影響))

  • For the evaluation of the effect on SWS, experiments were made on hyperlipidemia induced by hypercholesterol diet, inhibitory reaction to human platelet aggregation, Pulmonary thrombosis induced by collagen and epinephrine, global cerebral ischemia induced by KCN, brain ischemia induced by MCA occlusion, cytotoxicity of PC12 cells induced by amyloid ${\beta}$ protein(25-35), and NO production in RAW cells stimulated by lipopolysaccharide. The results were obtained as follows : 1. In the experiment on hyperlipidemia, the level of serum total cholesterol, phospholipid, and LDL-cholesterol were significantly decreased while the level of triglyceride, VLDL-cholesterol, and HDL-cholesterol had no significant change. 2. In the experiment on inhibitory reaction to platelet aggregation, SWS inhibited platelet aggregation induced by ADP(36.05%), by collagen(20.4%), and by thrombin(0.6%). 3. In the experiment on pulmonary thrombosis induced by collagen and epinephrine, the protective effect was found(37%). 4. In the experiment on global cerebral ischemia, coma duration induced by KCN changed insignificantly. 5. In the experiment on MCA occlusion, the change of neurologic grades on hind limb was significant only after the operation. Besides brain ischemic area and edema ratio were significantly decreased. 6. In the experiment on cytotoxicity of PC 12 cells induced by amyloid ${\beta}$ protein, the significant protective effect was found as concentration increases. 7. In the experiment on NO production in RAW cells stimulated by lipopolysaccharide, NO was significantly decreased. According to the results, it is expected that SWS might be effective on hyperlipidemia and brain damage.

The Protective Effects of N-Acetyl-L-cysteine on Cadmium-induced Cell Apoptosis in Rat Testis

  • Cadmium (Cd) generates reactive oxygen species (ROS), which in turn cause the apoptosis of various cell types including developing germ cells in rodent testis. Ascorbic acids (AA), one of the ROS scavengers, had been reported to protect against Cd-induced apoptosis. N-Acetyl-L-cysteine (NAC), another ROS scavenger, is known to remove ROS and alleviate the Cd-induced apoptosis in various cell types. In this study we tried to elucidate how NAC affected on Cd-induced cell apoptosis in rat testis. Rats were administered with NAC before and after Cd treatment and then testicular cell apoptosis was examined. NAC treatment resulted in the reduction of Cd-induced chromosomal DNA fragmentation in agarose gel electrophoresis. Terminal deoxynucleotidyl transferase dUTP nick end-labeling (TUNEL) assay showed that treatment of NAC reduced the Cd-induced apoptosis of germ cells. The administration of NAC showed that the translocation of apoptosis inducing factor (AIF) from mitochondria to nucleus was prevented, which indicated that the mechanism of Cd-induced testicular apoptosis is mediated through the release of AIF in caspase-independent manner. Taken together, the NAC may remove Cd-induced ROS and protect ROS-induced cell apoptosis in rat testis.

  • Effects of Taeyeumjoweetang on the obestity of mouse induced by gold thioglucose. It is researched to elucidate the effects of Taeyeumjoweetang on the obesity of mouse induced by gold thioglucose and the differentiation and growth of preadipocyte, 3T3-L1 cell. The result were as follows. 1. Taeyeumjoweetang extract improved the blood level of transaminase changed by the obesity of mouse induced by gold thioglucose. 2. Taeyeumjoweetang extract inhibited the increase of liver fat and body fat induced by the obesity of mouse induced by gold thioglucose. 3. Taeyeumjoweetang extract inhibited the increase of body weight induced by the obesity of mouse induced by gold thioglucose. 4. Taeyeumjoweetang extract inhibited the growth of undifferentiate preadipocyte 3T3-L1. 5. Taeyeumjoweetang extract showed the effects of inhibition on the differentiation of preadipocyte 3T3-L1. The above results suggested that the Taeyeumjoweetang extract may be used on the obesity induced by the overgrowth and differentiation of adipocyte, and the accumulation of fat in liver and body.

Role of G-protein in the Contraction of Rabbit Trachealis Muscle (토끼 기관평활근 수축에서 G Protein의 역할)

  • Fluoride (F-), a known stimulator of G-protein, induced strong contraction in rabbit trachealis muscle. $AlCl_3\;(5{\sim}20\;{\mu}M)$, which is required for G-protein stimulation by $F^-$, potentiated the contractile response to $F^-$. $Ca^{2+}-removal$ and verapamil, a calcium channel blocker, inhibited the fluoroaluminate-induced contraction. Fluoroaluminate increased $^{45}Ca$ influx in the absence and presence of verapamil. In heparin-loaded muscle high $K^+-induced$ contraction was not affected, but acetylcholine and fluoroaluminate-induced contractions were inhibited. The fluoroaluminate-induced contraction was partially relaxed by isoproterenol, a stimulator of adenylate cyclase. Pertussis toxin partially inhibited fluoroaluminate-induced contraction and potentiated isoproterenol-induced relaxation in the presence of fluoroaluminate, but had no effect on acetylcholine-induced contraction and the isoproterenol-induced relaxation in the presence of acetylcholine. These results suggest that fluoroaluminate has the ability to stimulate at least two putative G-proteins in rabbit trachealis muscle; One causes $Ca^{2+}$ influx through the potential-operated $Ca^{2+}$ channel and the other induces intracellular $Ca^{2+}$ release by the increase of inositol-1, 4, 5-triphosphate.

  • The purpose of this research was to investigate the effect of Kamidangkwieumja(KDEJ) water extract on the allergy reaction in mice. The results were obtained as follows: 1. The passive cutaneous anaphylaxis induced by egg albumin in fat was not affected. 2. The lethal anaphylaxis induced by platelet activating factor in mice. was inhibited. 3. The degranulation of peritoneal mast cells induced by compound 48/80 was not affected. 4. The acute hind paw edema was inhibited after 2hours later when it was induced by histamine. 5. The permeability of evans blue into peritoneal cavity induced by acetic acid was not affected. 6. Arthus reaction in mice was not affected. 7. The delayed type hypersensitivity induced by SRBC was inhibited. 8. The contact dermatitis induced by DNFB was not affected. 9. The hemagglutination titer induced by SRBC was inhibited. 10. The writhing syndrome induced by acetic acid was inhibited. 11. The population of heper T cells in mice thymus was enhanced. 12. The phagocytic activity of peritoneal macrophages was enhanced. 13. The production of nitric oxide from peritoneal macrophages was not affected. These results suggest that the anti-allergy effect of KDEJ is caused by steroidlike and enhanced immune action. The steroidlike action of KDEJ correspond with steroid-applying-method that frequently used in clinic, so it is used io treatment of psoriasis. The research on anti-allergy of KDEJ might has to be continued.

Current Situation and Determinants of Induced Abortion in Korea (한국 유배우 여성의 인공임신중절의 실태 및 결정요인)

  • This research explores the current situation and determinants of induced abortion in Korea. Using 2000 National fertility Survey by Korea Institute of Health and Social Affairs, this study finds that induced abortion rate is rapidly declining so that the role of induced abortion in determining the level of fertility has disappeared. The fundamental reason for resorting to an induced abortion for Korean women is to avoid unwanted birth. Thus, we find that induced abortion is usually taken by Korean women for the purpose of limiting family size and spacing births as in any other countries. Premarital pregnancy and economic hardship are also important reason for taking an induced abortioin, especially for the first time abortion.

Clinical Efficacy of herbal medicine for Chemo-Radiotherapy-induced oral mucositis

  • Objectives: The purpose of this systematic review was to evaluate the effects of herbal medicine on Chemo-Radiotherapy-induced oral mucositis. Methods: Electronic databases were used to search for studies published through 10 years until October 2022, and a randomized controlled study was conducted to evaluate efficacy of herbal medicine on chemo-radiotherapy-induced oral mucositis. Study quality was assessed using the Cochran's risk bias tool. Results: Two-hundred and three articles were initially searched, and 11 studies (head and neck cancer, breast cancer, colorectal cancer, esophageal cancer etc. undergoing radio-chemotherapy were included in analysis. The effect of herbal medicine on chemo-radiotherapy-induced oral mucositis, 9 studies reported that herbal medicine was more effective than the placebo group or conventional treatment. One study reported that the effect of the herbal compound treatment group was similar to that of the conventional herbal medicine, and one study reported that there was no difference in effect between the two herbal medicines and the group without treatment. Conclusion: This study suggests that herbal medicine effectively relieves the symptoms of chemo-radiotherapy-induced oral mucositis. However, there is limited evidence that herbal medicine may relief chemo-radiotherapy-induced oral mucositis, so further investigation is needed.

The Study of Anti-inflammtory Mechanism with Bee Venom on Human Synoviocytes (인체(人體) 활막세포(滑膜細胞)를 대상으로 한 봉양침액(蜂藥鍼液)의 염증(炎症) 치료(治療) 기전(機轉) 연구(硏究))

  • Objective : The purpose of this study was investigation how the bee venom(BV) prevents inflammation in human cell. Methods : we induced inflammation on human synoviocyte cell by lipopolysaccharide(LPS) and sodium nitroprusside(SNP), treated the bee venom and melittin on this cell, surveyed the expression of Nisotric oxide(NO), inducible nitric oxide synthase(iNOS), Cyclooxygenease-2(COX-2), cytolic phospholipase $A_2(cPLA_2)$, Prostaglandin $E_2(PGE_2)$ and nuclear factor-${\kappa}B$(NF-${\kappa}B$), and got below conclusions. Results : Compared with control 1. Expressions of LPS-induced $PGE_2$(BV 1, $5{\mu}g/m{\ell}$) and SNP-induced PGE2(BV 0.5, 1, $5{\mu}g/m{\ell}$)were decreased significantly. 2. Expressions of LPS-induced NO(BV 0.5, 1, $5{\mu}g/m{\ell}$) and SNP-induced NO(BV 1, $5{\mu}g/m{\ell}$)were decreased significantly. 3. Expressions of LPS-induced COX-2(BV 1, $5{\mu}g/m{\ell}$) and SNP-induced COX-2(BV $5{\mu}g/m{\ell}$)were decreased significantly. 4. Expressions of LPS-induced iNOS(BV 0.5, 1, $5{\mu}g/m{\ell}$) and SNP-induced iNOS(BV $5{\mu}g/m{\ell}$) were meanless by all dose. 5. Expressions of LPS-induced $cPLA_2$(BV 1, $5{\mu}g/m{\ell}$) and SNP-induced cPLA2(BV 1, $5{\mu}g/m{\ell}$)were decreased significantly. 6. Expressions of LPS-induced NF-${\kappa}B$(BV $5{\mu}g/m{\ell}$, melittin $5{\mu}g/m{\ell}$) and SNP-induced NF-${\kappa}B$(BV 0.5, 1, $5{\mu}g/m{\ell}$, melittin 5, $10{\mu}g/m{\ell}$)were decreased significantly. 7. Expressions of LPS-induced NF-${\kappa}B$ binding activity (BV $1{\mu}g/m{\ell}$, melittin $5{\mu}g/m{\ell}$, melittin $5{\mu}g/m{\ell}$+ DTT 20mM) were decreased significantly. Conclusion : The bee venom treatments on synoviocyte showed significant changes in LPS and SNP induced NO, iNOS, COX-2, cPLA2, PGE2 and NF-${\kappa}B$, these results suggest that bee venom is effective to inflammations and establish the process of bee venom therapy, so we expect active use of bee venom to control the inflammation.

Different Mechanisms for $K^+-Induced$ Relaxation in Various Arteries

  • $[K^+]_o$ can be increased under a variety of conditions including subarachnoid hemorrhage. The increase of $[K^+]_o$ in the range of $5{\sim}15$ mM may affect tensions of blood vessels and cause relaxation of agonist-induced precontracted vascular smooth muscle $(K^+-induced$ relaxation). In this study, effect of the increase in extracellular $K^+$ concentration on the agonist-induced contractions of various arteries including resistant arteries of rabbit was examined, using home-made Mulvany-type myograph. Extracellular $K^+$ was increased in three different ways; from initial 1 to 3 mM, from initial 3 to 6 mM, or from initial 6 to 12 mM. In superior mesenteric arteries, the relaxation induced by extracellular $K^+$ elevation from initial 6 to 12 mM was the most prominent among the relaxations induced by the elevations in three different ways. In cerebral arteries, the most prominent relaxation was produced by the elevation of extracellular $K^+$ from initial 1 to 3 mM and a slight relaxation was provoked by the elevation from initial 6 to 12 mM. In superior mesenteric arteries, $K^+-induced$ relaxation by the elevation from initial 6 to 12 mM was blocked by $Ba^{2+}\;(30\;{\mu}M)$ and the relaxation by the elevation from 1 to 3 mM or from 3 to 6 mM was not blocked by $Ba^{2+}.$ In cerebral arteries, however, $K^+-induced$ relaxation by the elevation from initial 3 to 6 mM was blocked by $Ba^{2+},$ whereas the relaxation by the elevation from 1 to 3 mM was not blocked by $Ba^{2+}.$ Ouabain inhibited all of the relaxations induced by the extracellular $K^+$ elevations in three different ways. In cerebral arteries, when extracellular $K^+$ was increased to 14 mM with 2 or 3 mM increments, almost complete relaxation was induced at 1 or 3 mM of initial $K^+$ concentration and slight relaxation occurred at 6 mM. TEA did not inhibit $Ba^{2+}-sensitive$ relaxation at all and NMMA or endothelial removal did not inhibit $K^+-induced$ relaxation. Most conduit arteries such as aorta, carotid artery, and renal artery were not relaxed by the elevation of extracellular $K^+.$ Among conduit arteries, trunk of superior mesenteric artery and basilar artery were relaxed by the elevations of $[K^+]_o.$ These data suggest that $K^+-induced$ relaxation has two independent components, $Ba^{2+}-sensitive$ and $Ba^{2+}-insensitive$ one and there are different mechanisms for $K^+-induced$ relaxation in various arteries.
