Effect of the Cultivation Technology on the Yield of Paddy Straw Mushroom (Volvariella volvacea)

  • The edible paddy straw mushroom Volvariella volvacea (Bull. ex F.) Singer has high nutritional and medicinal values. They are grown on a wide variety of agricultural by-products using different several methods. The result of this present study showed that type of bed and cultivation condition (outdoor/indoor) had effects on the yield of paddy straw mushrooms grown on the spent oyster mushroom sawdust. The treatment of circular compact bed under indoor cultivation condition (CYIC) had the best yield and biological efficiency (B.E.) with 2,119.2 g/bed and 14.5%, respectively, followed by the treatment of conventional bed under outdoor cultivation condition (COOC) with the yield and B.E. of 1,935.5 g/bed and 13.2%, respectively, but the lowest yield and B.E. were observed in the treatment of conventional bed under indoor cultivation condition (COIC) with 1,226.1 g/bed and 8.4%, respectively. Paddy straw mushrooms cultivated on spent oyster mushroom sawdust should be proper in both outdoor and indoor conditions by conventional and circular compact methods as well. However, in indoor conditions, paddy straw mushrooms should be grown in a circular compact bed to ensure the temperature of the bed is suitable for the mushroom growth.

고온피해 경감을 위한 표고 원목재배사 모델개발 (Development of Cultivation Facility Models to Reduce High Temperature Damage in Oak Mushroom (Lentinula edodes) Cultivation Using Bed-logs)

  • 표고버섯 원목재배시 고온피해를 경감시키기 위하여 임가실증지 5개소를 선정하였고, 기존 1중(비닐+차광)재배사의 내부 지붕에 2중으로 차광망을 추가 설치(개선형)하였다. 기존 재배사와 개선형 재배사 형태에 따른 내부 온습도, 균사 만연도 및 버섯 생산성을 조사하였다. 조사결과 시설 내부 최고온도는 개선형 재배사가 기존 1중 재배사 보다 평균 약 $3{\sim}5^{\circ}C$정도 낮았으나, 평균습도는 그와 달리 기존 1중 재배사와 개선형 재배사간에 큰 차이는 없었다. 특히 기존형과 개선형 재배사 모두 임내 재배장보다 평균습도가 10% 정도 낮아 건조한 불량환경이 유지되어 버섯 품질과 생산성 향상을 위해서는 지속적인 수분관리가 중요함을 알 수 있었다. 버섯목 표면, 내부의 균사 만연도를 비교한 결과 개선형 재배사가 가장 우수하였고, 임내재배사, 기존 1중 재배사가 순으로 저조하였다. 버섯 생산성을 비교한 결과 기존 1중 재배사에 비하여 117~204% 증수 효과가 있었으며, 재배사 동고 높이가 낮을수록 개선효과는 극대화 되었다. 따라서 시험결과를 토대로 고온피해 경감을 위한 원목재배사 모델 5종을 제시하였다.

원목 및 톱밥배지 버섯 재배사 내 실내 공기서 분리한 미기록 진균 보고 (Unrecorded Fungal Species Isolated from Indoor Air in the Log Bed- and Sawdust Media-based Mushroom Cultivation Houses)

  • 2016년 4월부터 6월까지 표고 원목재배 임가 4곳과 표고 톱밥재배 임가 1곳의 재배사 내 실내 공기에서 진균을 포집 및 분리 동정하였다. 총 6개의 국내 미기록 진균(Cenangium acuum, Periconia macrospinosa, Neopestalotiopsissurinamensis, Metarhiziummarquandii, Trichoderma petersenii, Trichoderma paratroviride)을 분리하였으며 이들 균류에 대하여 형태학적 특성 및 internal transcribed spacer rDNA region, large subunitregion, ${\beta}-tubulin$ 또는 translation elongation factor $1{\alpha}$유전자 염기서열에 기반하여 계통학적 분석 결과를 기술하였다.

실내재배를 위한 적상추와 청상추의 차광에 따른 생육 반응 (Growth Responses of Red and Blue Lettuce (Lactuca sativa L.) under Different Levels of Shading for Indoor Cultivation)

  • Production of lettuce under indoor cultivation is highly affected by light intensity. In this study, we used shade cloth (commercial black net) to examine the effect of these condition on growth without an associated yield penalty of container-grown lettuce. Four levels of shading treatments (0%, 35%, 55%, and 75% referred to as Cont., S35, S55, and S75 with respect to Cont.) and two lettuce varieties (red and blue) were evaluated. Variety-specific growth responses were observed with respect to different levels of shading treatments. High growth of red lettuce was occurred in Cont. treatment despite plant height and leaf length being higher than Cont. However, under 35% shading treatment blue lettuce was higher than in control plants. The highest root length was observed 0% shading (Cont.) of both varieties. These results reinforce the idea that blue lettuce is the better leafy vegetable rather than red lettuce for indoor cultivation related to lower light intensity environment conditions.

Temperature ranges for survival and growth of juvenile Saccharina sculpera (Laminariales, Phaeophyta) and applications for field cultivation

  • Saccharina sculpera is highly valued for human consumption and value-added products. However, natural resources of this kelp have decreased sharply and it is in danger of extinction. Resources recovery through cultivation is being trialed to enable the sustainable use of this species. In this study, the temperature range for survival and optimal growth of juvenile S. sculpera was identified and applied to field cultivation. This study investigated the survival and growth of juvenile S. sculpera under six temperatures (i.e., 5, 10, 15, 16, 18, and 20℃) and two light intensities (i.e., 20 and 40 µmol photons m-2 s-1) in an indoor culture experiment. In these experiments, the blade length decreased at 16℃ under the both light intensities. The thalli died at 20℃ and 20 µmol photons m-2 s-1, and at 18-20℃ and 40 µmol photons m-2 s-1. During the field cultivation, early growth of S. sculpera was highest at the 5 m depth and growth decreased as the water depth increased. When the initial rearing depth was maintained without adjustment throughout the cultivation period (from December to October), all the cultivated S. sculpera plants died during August and September. However, S. sculpera plants lowered from 5 to 15 m and grew to 90.8 ± 13.1 cm in July. The seawater temperature at 15 m depth was similar to the upper level of thermal tolerance demonstrated by juvenile S. sculpera in the indoor culture experiments (16℃ or lower). The plants were subsequently lowered to 25 m depth in August, which eventually led to their maturation in October. The present study confirmed that improved growth rates and a delay in biomass loss can be achieved by adjusting the depth at which the seaweeds are grown during the cultivation period. These results will contribute to the establishment of sustainable cultivation systems for S. sculpera.

A Study on Cultivation of Petalonia fascia (Scytosiphonales, Phaeophyta) by Vegetative Regeneration

  • To establish a cultivation method of Petalonia fascia, seeds and seedlings cultures and growth tests were performed at the Daeri aquafarm in Haeui, Shinan, Jeollanamdo, Korea. Gametes were easily released from the mature plurilocular sporangia. They developed to crustose discoidal stolons and grew to filamentous and discoidal stolons. The indoor seeding was performed by using the 100-150 ${\mu}m$long fragments of stolons on Porphyra nets and the erect thalli developed from the cuttings when the seawater temperatures were 10-15$^{\circ}C$. In the experimental cultivation in the sea, 1-2mm long plantlets were found after 15 days of cultivation; after two months thalli grew to their maximal size of 215-355 mm long blades; after three months the length of thalli began to decrease due to distal disintegration and the plant color changed to yellow and epiphytic diatoms were attached on the thalli, which deteriorated the quality of products. The cultivation of P. fascia by the regeneration of filamentous-discoidal complexes was carried out successfully for the first time in Korea.

국내 표고버섯 재배사에 바이오에어로졸로서 분포하는 진균의 농도와 종 분석 (Analysis of Fungal Concentration and Species Present as Bio-aerosols in Oak Mushroom Cultivation Houses)

  • 공기에 의해 운반되는 바이오에어러졸은 농산물 오염을 널리 일으키는 원인으로 알려져 있다. 육안으로 보이지 않는 진균 포자는 버섯 재배 중에 버섯과 인간의 건강에 해를 끼치는 바이오에어로졸이다. 본 연구는 한국의 표고버섯 재배사 실내의 공기 중에 존재하는 진균의 농도 및 종 분포에 관한 기초 자료를 얻기 위해 수행되었다. 2015년과 2016년에 국내 6 개의 다른 지역에 위치한 13 개의 표고버섯 재배 농장에서 21 차례 실내공기로부터 진균 포자를 샘플링하고 분석하였다. 공기 중 진균 농도는 $1.30{\times}10^2-1.59{\times}10^4cfu/m^3$ 범위였다. 중요하게도 20 차례의 농장방문 시료 채취 결과에서 진균 농도가 환경부의 실내공기질 권장 기준 농도인 $500cfu/m^3$를 초과하였다. 총 450 균주의 진균이 분리되었으며 46 속 33 종으로 동정되었다. 이중에 인체 위해균(4 속 4 종)과 식물병원균(10 속 13 종)이 존재하였다. 또한, 버섯 건강에 영향을 미치는 해로운 종인 Trichoderma harzianum, T. atroviride 및 T. longibrachiatum가 21 건 시료 채취 중 17 건에서 빈도 높게 검출되었다. 본 결과는 버섯생산의 재배관리를 위해서 재배사 실내 공기질을 개선해야 한다는 증거를 제시하였다.

Isolation and Characterization of Airborne Mushroom Damaging Trichoderma spp. from Indoor Air of Cultivation Houses Used for Oak Wood Mushroom Production Using Sawdust Media

  • Some species of the Trichoderma genus are reported as the major problem in oak wood mushroom production in Korea. In spite of economic loss by the fungi, scientific information on airborne Trichoderma species is not much available. To generate information for disease management development we analyzed airborne Trichoderma. A total of 1,063 fungal isolates were purely obtained from indoor air sampling of cultivation houses used for oak wood mushroom using sawdust media. Among the obtained isolates, 248 isolates were identified as Trichoderma fungi including T. harzianum, T. atroviride, T. citrinoviride, and T. pseudokoningii, by morphological and molecular analysis. T. harzianum was dominant among the four identified species. All the four Trichoderma species grew fast on solid nutrient media tested (potato dextrose agar [PDA], malt extract agar [MEA], Czapek's Dox + yeast extract agar [CYA] and cornmeal dextrose agar). Compact mycelia growth and mass spore production were better on PDA and CYA. In addition, T. harzianum and T. citrinoviride formed greenish and yellowish mycelium and spores on PDA and CYA. Greenish and yellowish pigment was saturated into PDA only by T. pseudokoningii. These four Trichoderma species could produce extracellular enzymes of sawdust substrate degradation such as β-glucosidase, avicelase, CM-cellulase, amylase, pectinase, xylanase, and protease. Their mycelia inhibited the growth of oak wood mushroom mycelia of two tested cultivars on dual culture assay. Among of eleven antifungal agents tested, benomyl was the best to inhibit the growth of the four Trichoderma species. Our results demonstrate that the airborne Trichoderma fungi need to be properly managed in the cultivation houses for safe mushroom production.

실내에서 식물과 수경시설이 온열환경에 미치는 영향 (The Effect of Plants and Waterscape Facilities on the Thermal Indoor Environment)

  • This survey is to investigate the effect of plants and waterscape facilities on the thermal indoor environment and to provide basic data for proper plant cultivation to enhance indoor landscape. The survey of the measure of comfort on the indoor environment for the residents of Taegu shows that the measure of comfort on the thermal-environment, which consist of temperature and humidity, has more negative responses than the measure on lighting . are . sound environment, which consists of air freshness, lighting condition and sound environment. The experiments on the effect of the amount of leaves and the distance of plants on the indoor thermal-environment are made. The experimental results illustrate that, as the capacity of a plant becomes greater and the distance from the plant shorter, the falling effect of temperature and the rising effect of humidity on the top of the plant are taken higher than on the side of the plant. When the same amount of leaves is set up, the distance range of the rising effect of humidity becomes wider than that of the falling effect of temperature. The investigation of the effect of waterscape facilities on the indoor thermal-environment shows that temperature and humidity of the space with installed waterscape facilities are lower and higher than without facilities, respectively.

천마 생육단계별 변온에 의한 최적온도 및 재배기간 (The ptimum temperatures during cultivation period of Gastrodia elata according to growth stages)

  • 본 연구는 실내시설재배에서 천마의 생육단계를 균사활착기, 괴경형성기, 괴경비대기, 휴면기로 구분하고, 최적 온도 및 배양기간을 설정하였으며, 괴경비대기에 변온처리를 하였다. 천마의 균사활착기는 $20^{\circ}C$에서 30일, 괴경형성기는 $25^{\circ}C$에서 120일, 괴경비대기는 $6{\sim}24^{\circ}C$에서 60일, 휴면기는 $5^{\circ}C$에서 30일로 처리하여 총 배양기간은 240일로 설정하였다. 실내시설재배는 노지재배보다 균사활착기는 30일 단축되었고, 괴경형성기는 30일 연장되었으며, 휴면기는 120일 단축됨으로써, 전체 재배기간이 약 120일 단축되었다. 따라서, 본 연구 결과, 밀폐된 실내시설재배에서 온도 조건을 제어하면 천마를 연중생산 할 수 있는 생육모델의 기초자료로 활용될 수 있을 것으로 판단되었다.
