Holostemma annulare (Family Asclepiadaceae) is an invaluable vulnerable medicinal plant; the root tubers are used in Ayurveda medicine and by folk healers to treat various ailments. In this study, Schenk and Hildebrandt medium fortified with the cytokinins 6-benzyl adenine, kinetin, and auxins, including indole 3-butyric acid, indole 3-acetic acid, α-naphthaleneacetic acid, and 2,4-dichlorophenoxyacetic acid, were checked for their efficiency on root tuber induction from different explants. Adventitious root tubers were more successfully induced from in vitro leaf segments and shoots when cultured in Schenk and Hildebrandt medium supplemented with 0.5 mg/l of α-naphthaleneacetic acid. In addition, preliminary phytochemical analysis of in vitro root tubers and identification of different secondary metabolites were conducted. Thin layer chromatography and high performance thin layer chromatography analysis of the crude methanolic extracts of the in vitro root tuber identified the presence of lupeol, a bioactive triterpene. Adventitious root tuber induction offers a novel method for the in vitro production of bioactive metabolites that can be scaled up by bioreactors, thus ensuring the conservation and sustainable utilization of H. annulare. The study warrants further scale-up production and pharmacological investigation that can be extended for pharmaceutical needs.
본 연구는 채종원에서 개화결실이 부진한 낙엽송 (Larix leptolepis S. et Z. Gordon)의 삽목증식법을 개발하기 위하여, 삽수의 모수령, 채취부위, 클론 및 발근촉진제가 삽목 발근에 미치는 영향을 구명하고저 실시하였다. 2, 8, 16, 30년생 모수에서 7월 하순경 당년에 생장한 삽수를 채취한 후, 발근촉진제인 IBA(Indole Butyric Acid)를 1000, 2000. 5000ppm의 세가지 농도로 5초간 처리하였다. 삽수는 peatmoss, vermiculite, perlite를 1 : 1 : 1 로 배합하여 조제된 온실내 삽목상에 삽목한 후 3개월이 경과된 10월 하순에 발근여부를 조사하였다. 발근촉진제 무처리의 경우 발근율은 2년생에서 52%, 8년생에서 48%, 16년생에서 36%, 30년생에서 20%인 반면, IBA 1000ppm 처리시 가장 높은 발근율을 얻었는데 2년생 80%, 8년생 71%, 16년생 52%, 30년생 25%로 발근율이 향상되었으며, 부정근의 수와 주근의 길이가 모수령이 낮을수록 증가하였으며, IBA 처리로도 증가하였다. 시기별 생존율은 삽목후 15-45일 사이에 현저히 감소한 반면, 그 후에는 생존율이 거의 감소하지 않았으며, IBA 처리로 생존율이 향상되었다. 모수의 클론별 발근율 조사는 8년생 모수를 사용하였는데, IBA 1000ppm 처리시 강원 49호에서 40%로 저조한 반면, 충남 6, 7호와 전북 1, 9호는 67%로 가장 커서 클론간에 발근율이 차이가 있었다. 삽수의 채취부위에 따른 위치효과는 수관 상부에서 채취한 삽수가 IBA 처리시 발근율이 평균 53%로 가장 낮았으며, 수관 하부가 56%, 수관 중부는 60%의 순으로 발근율이 증가되었다.
Somatic embryos were formed from calli obtained from axillary shoots (raised from nodal segments of glasshouse-grown plants under aseptic conditions), internodal segments (from in vitro-raised plants), and root and coty-ledonary leaf segments (from in vitro-raised seedlings) after 8 weeks of initial culture. Embryo formation was the highest (97.33%) from cotyledonary leaf callus on Mura-shige and Skoog's (MS) medium containing kinetin (KN) (3 mg/L). Somatic embryo induction was lesser with different combinations of auxins while it increased to 100% in internodal segment and cotyledonary leaf calli with 6-benzyladenine (BA) (2mg/L) along with 2,3,5-triiodobenzoic acid (TIBA) (2mg/L). The shoots were induced from somatic embryos raised from root, coty-ledonary leaf and internodal segment calli grown on MS medium containing BA in combination with indole-3-acetic acid (IAA). Maximum of 66.67% cultures formed shoots on MS medium containing BA (1mg/L) in combination with IAA (2mg/L). The shoots raised from somatic embryos were rooted on MS medium supplemented with indole-3-butyric acid (IBA) (2mg/L). The plantlets transferred to the field showed 70% survival rate after one year.
This communication describes for the first time an efficient and reproducible protocol for large-scale multiplication of Bambusa nutans. Nodal segments collected from field-grown clumps and cultured on Murashige and Skoog (MS) medium supplemented with $4.4{\mu}M$ benzylaminopurine (BA) and $2.32{\mu}M$ kinetin (Kin) gelled with 0.2% gelrite yielded 80% aseptic cultures with 100% bud-break. The in vitro-formed shoots obtained after bud-break were successfully multiplied in MS liquid medium supplemented with $13.2{\mu}M$ BA, $2.32{\mu}M$ Kin, and $0.98{\mu}M$ indole-3-butyric acid (IBA). Sub-culturing of shoots every 3 weeks on fresh multiplication medium yielded a consistent proliferation rate of 3.5-fold. Shoot clusters containing three to five shoots were successfully rooted with 100% success on half-strength MS liquid medium supplemented with $9.8{\mu}M$ IBA, $2.85{\mu}M$ indole-3-acetic acid (IAA), $2.68{\mu}M$ naphthaleneacetic acid (NAA), and 3% sucrose. Plantlets grown in vitro were acclimatized and subsequently transferred to the field. Inter-simple sequence repeat analysis has confirmed the genetic uniformity of the tissue-cultured plants up to 27 passages.
The effect of sodium sulphate on shoot induction and multiple shoot formation from nodal explants of Vitex negundo L. was tested on Murashige and Skoog's (MS) medium fortified with different auxins, cytokinins and sucrose. Highest percentage $(97.78\%)$ of explants for shoot induction and multiple shoot (20.68/explant) production were observed in the combination treatment of $N^6-Benzyl$ adenine (BA) $(17.80\;{\mu}M/L)$, ${\alpha}-Naphthalene$ acetic acid (NAA) $(2.15\;{\mu}M/L)$ and $5\%$ sucrose supplemented with 100 mg/L sodium sulphate. In vitro raised shoots were rooted on the half-strength MS medium fortified with different concentrations of NAA, Indole-3-acetic acid (IAA), and Indole-3-butyric acid (IBA) alone and in combinations. Among the treatments, $4.90\;{\mu}M/L$ of IBA was found most effective $(95.56\%)$ in inducing roots. The rooted plantlets were shifted to glasshouse for acclimatization and later transferred to the field with cent percent survival. Furthermore, in vitro flowering was observed in the shoots cultured on MS medium supplemented with BA $(8.90\;{mu}M/L)$ and NAA $(1.61\;{\mu}M/L)$.
Factors affecting regeneration of different Rubus varieties (blackberry, raspberry and their hybrid) were examined and a reliable regeneration system was established. Media for stock plant maintenance were tested; different explants and media were investigated to find the best circumstances for the regeneration. The effect of the commonly used antibiotics was studied to determine the most suitable one for selection of the transformants. We found that both MS and LS media supplemented by $20\;gL^{-1}$ sucrose are suitable for the stock plant maintenance. The optimal hormone content for the stock plants is $0.125\;mgL^{-1}$ 6-benzylaminopurine (BAP) with $0.01\;mgL^{-1}$ indole-3- butyric acid (IBA). The highest regeneration rate was observed on medium containing MS salts with B5 vitamins complemented with glucose, sucrose, maltose, $10\;gL^{-1}$ each, supplemented with benzylaminopurine riboside (BAR) ($2\;mgL^{-1}$) and indole-3-acetic acid (IAA) ($0.1\;mgL^{-1}$). The regenerated shoots appeared directly from the cut edges, without callus phase. Hygromycin and geneticin proved to be good selection agents for the Rubus explants, but due to their severe effect on the tissues we propose to use marker-free constructions for the transformation.
An efficient protocol for in vitro multiple shoot bud induction and plant regeneration from mature green cotyledon derived callus tissues of Acacia sinuata has been developed. Callus formation occurs at all the concentrations of thidiazuron (TDZ) in Murashige and Skoog's (MS) medium, but 0.6 ${\mu}M$ proved to be the best with maximum callus formation frequency. Supplementation of TDZ in combination with indole-acetic acid (IAA) in MS media accelerates shoot bud organogenesis in differentiating callus tissues with 60-70% conversion of shoot buds into shoot Most efficient shoot organogenesis was recorded when TDZ induced calli were subcultured at different concentrations of 6-benzyla-denine (BA). Optimum shoot bud induction and plant regeneration from callus was achieved when 0.6 ${\mu}M$ (TDZ) induced calli were subcultured at 3.0 ${\mu}M$ (BA) where $16.6{\pm}0.74$ shoots/unit callus on obtained. Rooting in in vitro differentiated shoots was achieved when transferred to medium containing different concentration of indole-3-butyric acid (IBA) in full & half strength MS medium. The well rooted plantlets were hardened and transferred to net house with 90% survival rate.
A method for plant regeneration via organogenesis from Pelagonium inquinans leaf disc has been developed. Mature leaf explants were collected from field-grown plants and used for the induction of adventitious shoot regeneration on Murashige and Skoog (MS) medium supplemented with 3% (w/v) sucrose plus plant growth regulators. Maximum shoot organogenesis, with $11.8{\pm}1.5$ shoots (98.6%) per leaf disc, was obtained with $2\;mg/l$$N^6-benzyladenine$ (BA) and $0.5\;mg/l$${\alpha}-naphthyleneacetic$ acid (NAA) in 30 days. For rooting, the in vitro proliferated and elongated shoots were excised into 1.5-2 cm in length microcutting, which were plated individually on an half-strength MS (1/2MS) medium supplemented with 2% (w/v) sucrose plus various concentrations of indole-3-butyric acid (IBA). Shoots rooted with a frequency of 100% following culture on 1/2MS medium containing $0.5\;mg/l$ IBA.
A protocol is described for rapid multiplication of Zanthoxylum piperitum DC. (Rutaceae), an important aromatic and medicinal plant, through shoot-tip explant cultures. Murashige and Skoog (MS) medium supplemented with various concentrations of N-6-benzyladenine (BA), N-6-benzylaminopurine (BAP) and thidiazuron (TDZ), in single or in combination with ${\alpha}-naphthaleneacetic$ acid (NAA), was used to determine the rate of shoot proliferation. N-6-benzyladenine (BA) used at 0.5mg/l, was the most effective in initiating multiple shoot proliferation at the rate of 23 microshoots per shoot-tip explants after 40 days of culture. Shoot multiplication increased 1.2-fold in each successive subculture. Induction of rooting (98%) was achieved by transferring the shoots to the same basal medium containing 2 mg/l indole-3-butyric acid (IBA). Plantlets went through a hardening phase in a controlled growth chamber, prior to in vivo transfer. These results represented that possible application for the mass production of plantlets through in vitro culture system of Zanthoxylum piperitum DC.
The adsorptive removal properties of synthetic A4 zeolite were investigated against a total of 16 offensive odors consisting of reduced sulfur compounds (RSCs), nitrogenous compounds (NCs), volatile fatty acids (VFAs), and phenols/indoles (PnI). Removal of these odors was measured using a laboratory-scale impinger-based adsorption setup containing 25 g of the zeolite bed (flow rate of $100mL\;min^{-1}$). The high est and lowest breakthrough (%) values were shown for PnIs and RSCs, respectively, and the maximum and minimum adsorption capacity (${\mu}g\;g^{-1}$) of the zeolite was observed for the RSCs (range of 0.77-3.4) and PnIs (0.06-0.104), respectively. As a result of sorptive removal by zeolite, a reduction in odor strength, measured as odor intensity (OI), was recorded from the minimum of approximately 0.7 OI units (indole [from 2.4 to 1.6]), skatole [2.2 to 1.4], and p-cresol [5.1 to 4.4]) to the maximum of approximately 4 OI units (methanethiol [11.4 to 7.5], n-valeric acid [10.4 to 6.5], i-butyric acid [7.9 to 4.4], and propionic acid [7.2 to 3.7]). Likewise, when removal was examined in terms of odor activity value (OAV), the extent of reduction was significant (i.e., 1000-fold) in the increasing order of amy acetate, i-butyric acid, phenol, propionic acid, and ammonia.
