Degradation Ability and Population of Resistant Strains of Chlorothalonil in Upland Soil Distributed in Honam Area (호남지역 밭토양에 분포된 Chlorothalonil 내성균(耐性菌)의 밀도(密度)와 분해능(分解能))

  • This experiment was conducted to obtain the basis of degradation of remaining agricultural chemicals accumulated in upland soils of Honam district in Korea. The population. relative growth rate(RGR). chlorothalonil(TPN)-degradation ability and bacterialogical characteristics of TPN resistant strains were investigated in TPN levels of 0, 25, 50, 100 and $500{\mu}l/ml$ compared with Mancozeb. A number of TPN-resistant bacteria were differ in the area of examined and were decreased with higher levels of TPN. The resistance of bacteria was stronger in TPN than Mancozeb but the resistance of fungi was vise versa. RGR of bacteria in the culture was the highest at the level of $50{\mu}l/ml$ and the lowest in $500{\mu}l/ml$ of TPN. TPN-degradation ability of bacteda isolated in various TPN levels was varied : only 8 percentage of bacteria showed 75 percentage or more degradation ability. The higher the concentration in TPN resistance, the larger the number of strains carried great ability to decompose pesticide residues. The strains having higher decomposition ability was rod-shapes cells and senstive to heat. Analyses of the indol production, methyl red, and V-P test have given similar results, with negative reaction in all these strain, while the other biochemical characteristics were differ in the strains. Based on these, these strains might be classified into Pseudomonas sp., Corynebacterium sp., Acinetobacter sp. and Moraxcella sp.

Preparation of Functional Healthy Drinks by Ethanol Extracts from Defatted Safflower Seed Cake (탈지 홍화씨박 에탄올추출물 함유 기능성 건강음료의 제조)

  • Functional healthy drinks were processed with freeze dried powders of ethanol extract from of defatted safflower (Carthamus tinctorious L.) seed cake and some useful components of the drinks were investigated. Yield of freeze dried powder was the highest as 8.42% when it extracted with 60% ethanol (60% EFDP). Each drink contained 0.02% of freeze dried powder and ranged 10.6∼13.8% of soluble solid, 2.90∼3.68 of pH, 0.10∼0.83% of titratable acidity. ‘L’ value of drink-I (DSD-I) was the highest as 94.82$\pm$2.45, ‘b’ and ‘a’ value of drink-V (DSD-V) was highest as 27.15-2.65 and 28.67$\pm$2.69, respectively. Major free sugars of drink were 6015.3∼7918.2 mg% of glucose and 1511.4∼2091.0 mg% of sucrose. The content of citric acid was the highest as 179.2∼981.3 mg%. The content of total phenol in 60% EFDP was 99.17 mg% and that of drink-II(DSD-II) and DSD-V was 307.84 mg% and 224.06 mg%, respectively. Total flavonoid was contained as 50.29 mg% in 80% ethanol extract (80% EFDP) and 125.20 mg% in DSD-V. N-[2-(5-hydroxy-1H-indol-3-yl) ethyl] ferulamide (serotonin-I) was determined as high as 18.81 ppm in 80% EFDP and ranged 2.42∼2.89 ppm in drinks. N-[2-(5-hydroxy-lH-indol-3yl)ethyl]-p-coumaramide (serotonin-II) was determined as 30.17 ppm in 80% EFDP and ranged 3.79∼4.59 ppm in drinks. Acacetin, flavonoid compound were 9.83 ppm in amyloglucosidase hydrosis + 60% ethanol extract (A + 60% EFDP) and ranged 0.98∼1.26 ppm in drinks. Electron donating ability (EDA, %) was measured and compared with 100 ppm BHA as chemical antioxidant. EDA was 93.97$\pm$2.21% in A+60% EFDP, 94.79$\pm$2.26% in DSD-I, 94.69$\pm$1.37% in DSD-II, and 93.83$\pm$1.49% in BHA. DSD-II added with hot water extract solution from Korean ginseng and safflower yellow pigment recorded the highest sensory score.

Preparation and Antioxidant Activity of Health Drink with Extract Powders from Safflower (Carthamus tinctorius L.) Seed (홍화씨 추출분말 함유 건강음료의 제조와 항산화성)

  • Health drinks were prepared with freeze dried powder of 60% ethanol extract (60% EFDP), 60% ethanol extract after hydrolysis with amyloglucosidase (60% AEFDP) and 80% ethanol extract (80% EFDP) from roasted safflower seed. Quality characteristics and antioxidative properties were investigated. Yield of freeze dried powders were ranged in $4.67%{\sim}5.62%$. Brix, pH and titratable acidity of safflower drinks were ranged in $11.4{\sim}14.2%$, $2.83{\sim}3.34$ and $0.09{\sim}0.91%$, respectively. Content of total phenolic compounds was much more in 80% EFDP (117 mg/g) and safflower drink-I (SD-I, 440 ppm) than others. Content of total flavonoid was observed in higher level in 60% EFDP (49 mg/g) and safflower drink-V (SD-V, 138 ppm) than others. Antioxidant compounds such as N-[2-(5-hydroxy-1H-indol-3-yl)ethyl]ferulamide(serotonin-I) and N-[2-(5-hydroxy-1H-indol-3yl)ethyl]-p-coumaramide(serotonin-II) exhibited higher contents of 21.09 ppm, 33.56 ppm in 60% EFDP and of 3.83 ppm, 5.81 ppm in safflower drink-II (SD-II) than others. Content of acacetin was much more in 80% EFDP (13.53 ppm) and safflower drink-IV (SD-IV, 1.14 ppm) than others. From the DPPH test to measure antioxidant activity, it was shown that 80% EFDP and SD-I have stronger scavenging activities of 94.58% and 94.88%, respectively, while BHA standard solution does 93.88%. Among drinks, SD-II was revealed to have highest level on overall acceptance, color and flavor through sensory evaluation. These results induced that safflower seed can be used as natural antioxidant and functional food material.

Shoot-tip culture for massive production of radish foundation seeds (무 원종 대량생산을 위한 경정배양)

  • Shoot-tip culture was used to produce clonal plants of radish stock seeds. Using 6-benzyladenine (BA), the largest number of RA2 line multi-shoots were formed with an average of 14.67 shoots on 1.33 µM BA in early seedlings and 11.33 shoots on 1.78 µM BA in juvenile seedlings. The largest number of RA4 line multi-shoots were formed with an average of 11.67 shoots on 2.22 µM BA in early seedlings and 13.67 shoots on 1.33 µM BA in juvenile seedlings. There was little difference in the significance level by BA concentration in both lines. Using Thidiazuron (TDZ), the number of RA2 line multi-shoots increased with increasing TDZ concentration, forming the largest number of multi-shoots in 0.45 µM TDZ (7.0 and 3.0 multi-shoots for early and juvenile seedlings, respectively), but few multi-shoots were formed from TDZ 2.25 and 4.5 µM. RA4 line produced almost no multi-shoots in early seedlings, and 3.7 multi-shoots were produced in 0.23 and 0.45 µM TDZ in juvenile seedlings, but not at higher concentrations. Analysis of the tissue culture seedlings grown by cultivating the generated multi-shoots with Radish Foundation seeds using SSR marker revealed a weak pattern of mutation in the generated tissue culture seedlings, but there was no mutant. In addition, in terms of root roots, both RA2 and RA4 lines generally had the best rooting, number of roots, and degree of root development in 4.9 µM indol-3- butyric acid (IBA).

Examination of Root Induction Ratio for Regeneration of Alfalfa by Medium Component (알팔파 재분화에서 배지조성에 따른 뿌리 유도율 조사)

  • The alfalfa (Medicago sativa L.) callus was induced from seeds on SH medium contained $3\;mg/{\ell}$ of 2,4-D. Several regenerated alfalfa plants and many shoots were obtained by procedure of Kim et al. (1999); 1) incubation for 28~30 days on SH medium added $5\;mg/{\ell}$ of NAA and $2\;mg/{\ell}$ of Kinetin, 2) incubation for 3~5 days on SH medium added $11\;mg/{\ell}$ of 2,4-D and $1\;mg/{\ell}$ of kinetin. 3) incubation for 21~25 days on SH medium added $1.6\;g/{\ell}$ of ammonium sulfate and $5.75\;g/{\ell}$ of proline. To increase of root induction ratio on plant regeneration process of alfalfa, root induction ratio was examined on 8 kinds of medium, containing different amount of hormone and SH salt. Root induction ratio was higher on SH medium contained IBA than SH basal medium. In case 1.5 mg of IBA was added in SH medium, root induction ratio was the highest to 56.0% in this study. On the other hand, root induction ratio was higher on SH medium diminished SH salt amount to half volume and addition of IBA makes high root induction ratio, too. Thus, we conclude that the medium for root induction of alfalfa may be added $1.5\;mg/{\ell}$ of IBA and diminished SH salt amount to half volume.

Antimutagenic and Cytotoxic Effects of an Ethanol Extract of Buckwheat Sprout (메밀싹 에탄올 추출물의 항돌연변이원성 및 암세포 성장억제 효과)

  • This study was performed to determine the antimutagenic and anticancer effects of ethanol extract of buckwheat sprout using Ames test and SRB assay, respectively. An ethyl acetate fraction (200 ${\mu}/plate$) from the ethanol extract of buckwheat sprout showed inhibition rate of 80.6% against the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in Salmonella typhimurium TA100 strain. Also the ethyl acetate fraction (200 ${\mu}/plate$) showed higher antimutagenic activity than other fractions against the mutagenesis induced by 4-nitroquinoline-1-oxide (4NQO) in Salmonella typhimurium TA98 and TA100. In addition, the ethyl acetate fraction (200 ${\mu}/plate$) showed high antimutagenic effect of 80.9% and 85.9% against the mutation of TA98 and TA100 strains induced by 3-amino-1,4-dimethyl-5H-pyrido-(4,3-b)indol (Trp-P-1), respectively. The cytotoxic effects of each solvent fraction from the ethanol extract of buckwheat sprout against human cancer cell lines including lung carcinoma (A549), gastric carcinoma (AGS), breast adenocarcinoma (MCF-7), hepatocellular carcinoma (Hep3B), and colon adenocarcinoma (Colo 205) were investigated. The ethyl acetate fraction of buckwheat sprout ethanol extract at the concentration of 1.0 mg/ml showed strong cytotoxic activities of 70.3, 94.8, 79.6, 82.3, and 73.2% against A549, AGS, MCF-7, Hep3B and Colo 205 cancer cell lines, respectively.

The Effect of Enhancer on the Penetration of Indapamide through Hairless Mouse Skin (경피흡수촉진제의 영향에 따른 인다파마이드의 피부투과)

  • The chemical formula of indapamide is 3-(aminosulfonyl)-4-chloro-N-(2,3-dihydro-2-methyl-1H-indol-l-yl)-benzamide, Indapamide is an oral antipertensive diuretic agent indicated for the treatment of hypertensive and edema. Indapamide inhibits carbonic anhydrase enzyme. Transdermal drug delivery systems, as compared to their corresponding classical oral or injectable dosage form counterparts, offer many advantages. The most important advantages are improved systemic bioavailability of the pharmaceutical active ingredients (PAI), because the first-pass metabolism by the liver and digestive system are avoided; and the controlled, constant drug delivery profile (that is, controlled zero-order absorption). Also of importance is the reduced dose frequency compared to the conventional oral dosage forms (that is, once-a-day, twice-a-week or once-a-week). Other benefits include longer duration of therapeutic action from a single application, and reversible action. For example, patches can be removed to reverse any adverse effects that may be caused by overdosing. In order to evaluate the effects of vehicles and penetration enhancers on skin permeation of Indapamide, the skin permeation rates of Indapamide from vehicles of different composition were determined using Franz cells fitted with excised hairless skins. Solubility of Indapamide in various solvents was investigated to select a vehicle suitable for the percutaneous absorption of Indapamide, The solvents used were Tween80, Tween20, Labrasol, Lauroglycol90 (LG90) and Peceol. Lauroglycol90 increase the permeability of indapamide approximately 3.75-fold compared with the control. Tween80, Tween20, Labrasol, Lauroglycol90 (LG90) and Peceol showed flux of $0.06ug/cm^2/hr,\;0.4ug/cm^2/hr,\;0.21ug/cm^2/hr,\;0.72ug/cm^2/hr,\;0.29ug/cm^2/hr$, respectively.

Antioxidative and Antimutagenic Effects of the Ethanol Elrtract from Cordyceps militaris (번데기동충하초(Cordyceps militaris) 에탄을 추출물의 항산화성 및 항돌연변이원성 효과)

  • Cordyceps militaris is a parasitic fungus that has been used as a Chinese medicine for the treatment of fatigue, debility, kidney disease, tuberculosis, asthma and cardiac insufficiency etc. This study was carried out to determine the antioxidative and antimutagenic effects of Cordyceps militaris using DPPH free radical donating method and Ames test, respectively. They were extracted with ethanol and then further fractionated to n-hexane, chloroform, ethyl acetate, butanol and water, stepwise. Among five fractions, the EtOAc and BuOH fractions showed the highest electron donating activities, about 2-fold higher than other fractions. In Ames test, most of the extracts had strong antimutagenic effects against the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG), 4-nitroquinoline-1-oxide(4NQO), benzo($\alpha$)pyrene(B($\alpha$)P) and 3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indol (Trp-P-1). The EtOH extracts of C. militaris (200 $\mu\textrm{g}$/plate) showed 62.8%, 74.4% and 67.2% inhibitory effects on the mutagenesis induced by 4NQO, B($\alpha$)P and Trp-P-1, respectively, against TA98 strain, whereas 78.1%, 78.6%, 78.6% and 82.7% inhibition were observed on the mutagenesis induced by MNNG, 4NQO, B($\alpha$)P and Trp-P-1, respectively, against TA100 strain. Especially, the BuOH fraction showed the highest antimutagenic effects against mutation induced by MNNG.

Effect of Indole-3-Carbinol on Inhibition of MMP Activity via MAPK Signaling Pathway in Human Prostate Cancer Cell Line, PC3 Cells (인돌이 인체 전립선암세포 PC3 Cell 전이 관련 Matrix Metalloproteinases (MMPs) 활성과 발현에 미치는 영향)

  • We examined the effect of indole-3-carbinol (I3C, $C_9H_9NO$), an autolysis product of a glucosinolate and a glucobrassicin in vegetables, on MMP-2, -9 activities and TIMP-l and -2 inductions via microtubule-associated protein kinase (MAPK) signaling pathway in prostate cancer cell line, PC3 cells. Our results indicated that I3C inhibited cell growth of PC3 cells in dose (0,50, 100 ,${\mu}M$) and time (0,24,48 and 72 h) dependent manners. Using gelatin zymography for MMP activity, we demonstrated that I3C significantly decrease MMP-2 and -9 activities in PC3 cells. We also observed that I3C decreased the proteins and mRNA levels of MMP-2 and -9 in PC3 cells as well. Inversely, expressions of TIMP-l and -2 protein and mRNA in PC3 cells were increased by I3C in a dose dependent manner. In another experiment, we showed that I3C inhibited PC3 cells invasiveness by using marigel invasion assay and we also found that I3C suppressed MMP transcriptional activity by MAPK signaling pathways. Taken together, our results suggest that I3C may contribute to the potential beneficial food component to prevent the cancer metastasis in prostate cancer cells. (KoreanJNutr2008; 41(3): 224~23I)

Antimutagenic Effects of Browning Products Reacted with Polyphenol Oxidase Extracted from Apple (사과 Polyphenol Oxidase에 의한 효소갈변반응 생성물의 항돌연변이 효과)

  • This study was carried out to investigate the antimutagenic effects of five kinds of apple enzymatic browning reaction products(AEBRP) on mitomycin C (MMC), N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), 4-nitroquinoline-1-oxide(4NQO), benzo(${alpha}$)pyrene(B(${alpha}$)P) and 3-amino-1,4-dimethyl-5H-pyri-do [4,3-b]indol (Trp-P-1). In spore rec-assay using B. subtilis Hl7($rec^+$) and M45($rec^-$), homocatechol-AEBRP and hydroquinone-AEBRP showed strong antimutagenic effects on MMC and MNNG as the concentration of AEBRP increased. In the Ames test using S. typhimurium TA98 and TA100, hydroxyhydroquinone-AEBRP and pyrogallol-AEBRP showed strong antimutagenic effects on Trp-p-1 and B(${alpha}$)p in TA98 and TA100 in the presence of S-9 mix. Most of AEBRPs suppressed about 50% to 80% the mutagenesis in S. typhimurium TA98 induced by MNNG, however, AEBRPs except hydroxyhydroquinone-AEBRP showed antimutagenic effects of about 94% in TA100. Antimutagenic effects of the five kinds of AEBRPs on 4-NQO were more or less weak, in particular homocatechol-AEBRP exhibited the inhibitory effect of about 48% in TA98, and homocatechol-AEBRP and hydroquinone-AEBRP showed inhibitory effects of about 46% to 58% in TA 100.
