Puberty Related Changes in Hormonal Levels, Productive Performance, Carcass Traits, and Their Interactions in Slovakian White Gilts

  • The aim of this experiment was to evaluate the levels of hormones (progesterone, IGF-I and IGFBP-3) in blood plasma, growth, carcass traits and their interactions of sexually immature (n = 18) and sexually mature (n = 17) gilts. To calculate average daily weight gain (ADG), gilts were individually weighed at the beginning of the trial and at slaughter (110${\pm}$10 days old). Blood concentrations of progesterone, IGF-I and IGFBP-3 were determined by RIA. The right hot carcass sides were dissected and the individual basic parts from carcasses were weighed to record the carcass traits. IGFBP-3, ADG and carcass traits were not affected by pubertal maturation. Compared to sexually immature gilts, mature gilts had higher blood concentrations of progesterone and IGF-I. High correlations were noted between levels of some hormonal substances, productive performance and carcass traits of sexually immature and mature gilts.

In situ Ruminal and Intestinal Digestibility of Crude Protein and Amino Acids in By-product Feedstuffs

  • The objectives of this study was to evaluate the degradability and digestibility of crude protein (CP), rumen undegradable protein (RUP), and individual amino acids (AA) on six by-product feedstuffs (BPF) (rice bran, RB; wheat bran, WB; corn gluten feed, CGF; tofu residue, TR; spent mushroom substrate from Pleurotus ostreatus, SMSP; brewers grain, BG) as ruminants feed. Three Hanwoo steers (40 months old, $520{\pm}20.20kg$ of body weight) fitted with a permanent rumen cannula and T-shaped duodenal cannula were used to examine of the BPF using in situ nylon bag and mobile bag technique. The bran CGF (19.2%) and food-processing residue BG (19.7%) had the highest CP contents than other feeds. The RUP value of bran RB (39.7%) and food-processing residues SMSP (81.1%) were higher than other feeds. The intestinal digestion of CP was higher in bran RB (44.2%) and food-processing residues BG (40.5%) than other feeds. In addition, intestinal digestion of Met was higher in bran RB (55.7%) and food-processing residues BG (44.0%) than other feeds. Overall, these results suggest that RB and BG might be useful as main raw ingredients in feed for ruminants. Our results can be used as baseline data for ruminant ration formulation.

Effects of Spent Mushroom Substrates Supplementation on Rumen Fermentation and Blood Metabolites in Hanwoo Steers

  • This study was designed to investigate the effects of supplementation of spent mushroom substrates (SMS) on rumen fermentation and blood metabolites in Hanwoo steers. The experiment was conducted as a duplicated Latin square design with six Hanwoo steers ($600{\pm}47\;kg$), each permanently fitted with a ruminal cannula. There were three treatments; i) control (concentrates 4.8 kg +rice straw 1.2 kg/d), ii) Pleurotus eryngiia (PE) treatment (concentrates 4.8 kg+rice straw 0.73 kg+Pleurotus eryngiia 1.20 kg/d) and iii) Pleurotus osteratus (PO) treatment (concentrates 4.8 kg+rice straw 0.73 kg+Pleurotus osteratus 1.20 kg/d). There were no major effects of different dietary treatments on rumen parameters such as pH, ammonia-N, individual and total VFA production. Parameters of N utilization, including blood urea nitrogen (BUN), total protein and albumin levels, were not significantly different among the treatments, except for creatinine. Thus, the present results indicated that protein utilization was mostly unaffected by SMS treatments such as PE and PO, even though creatinine concentration was lower in PE compared with control and PO treatments (p<0.05). The present results indicate that Pleurotus eryngii and Pleurotus osteratus could be used as a forage source to replace 40% of rice straw without any negative effects on rumen fermentation and blood metabolites in Hanwoo steers.

  • Recently, many studies on big data based smart farming have been conducted. Research to quantify morphological characteristics using image data from various crops in smart farming is underway. Rice is one of the most important food crops in the world. Much research has been done to predict and model rice crop yield production. The number of productive tillers per plant is one of the important agronomic traits associated with the grain yield of rice crop. However, modeling the basic growth characteristics of rice requires accurate data measurements. The existing method of measurement by humans is not only labor intensive but also prone to human error. Therefore, conversion to digital data is necessary to obtain accurate and phenotyping quickly. In this study, we present an image-based method to predict leaf number and evaluate tiller number of individual rice crop using YOLOv5 deep learning network. We performed using various network of the YOLOv5 model and compared them to determine higher prediction accuracy. We ako performed data augmentation, a method we use to complement small datasets. Based on the number of leaves and tiller actually measured in rice crop, the number of leaves predicted by the model from the image data and the existing regression equation were used to evaluate the number of tillers using the image data.

Oxidation of extracellular cysteines by mercury chloride reduces TRPV1 activity in rat dorsal root ganglion neurons

  • Transient receptor potential vanilloid type 1 (TRPV1) receptor plays an important role as a molecular detector of noxious signals in primary sensory neurons. Activity of TRPV1 can be modulated by the change in the environment such as redox state and extracellular cations. In the present study, we investigated the effect of the mercury chloride ($HgCl_2$) on the activity of TRPV1 in rat dorsal root ganglia (DRG) neurons using whole-cell patch clamp technique. Extracellular $HgCl_2$ reversibly reduced the magnitudes of capsaicin-activated currents ($I_{cap}$) in DRG neurons in a dose-dependent manner. The blocking effect of $HgCl_2$ was prevented by pretreatment with the reducing agent dithiothreitol (DTT). Inhibition of $I_{cap}$ by $HgCl_2$ was abolished by point mutation of individual cysteine residues located on the extracellular surface of TRPV1. These results suggest that three extracellular cysteines of TRPV1, Cys616, Cys634 and Cys621, are responsible for the oxidative modulation of $I_{cap}$ by $HgCl_2$.

동서의학의 인체에 대한 관점 비교 고찰 (The Oriental and Western Medical Study on the Viewpoint in Human System)

  • Oriental medicine which is based on internal activeness has constantly been developed for over thousands years and during that time it has encountered Western medicine that has the different system. Because of a lot of different viewpoints of how to understand human and nature, there exist lots of problems to cope with for the integration of two medicines as follows: (1) Oriental medicine regards each person as a cosmos in miniature but Western medicine is based on dualism, substantialism and mechanism. (2) Oriental medicine thinks much of totality as a cosmos in a miniature but Western medicine investigates individual structure and function of human organ and system. (3) Oriental medicine considers that organs are the core of the Oriental medical physiology and makes much account of their functions and their relationships with fundamental texture, but Western medicine gives much emphasis on the structure than functions. Despite of these differences, Oriental medicine and Western medicine should develop mutual understanding and this will facilitate the achievement of the third medicine that is suitable for the primary object of medicine that is to enhance human health and cure diseases.

  • The treatment before growing into disease is the highest level of the spirit of oriental preventive public medicine. Specially the treatmentt is very important in the life style disease of the aged society, and it is closely connected the human body-form diagnosis reflecting the character of individual physiology and pathology. The human body-form diagnosis explain systematically the color of face, the form and movement of the body, the smell and the taste, the mentality, many or few of $Yin{\cdot}Yang$, the state of pulse, the Meridian, the state of pathology and the direction of the treatment, and therefore the human body-form diagnostis method has also the practical ity on classifying the type. As the spirit of oriental preventive public medicine is based on the Meridian control method and food cooking control method, it has an important effect on the general management of the disease, the making on the beautiful face and the healthy body, the prolongation of life, the management of the individual-ordered health.

Systems-level mechanisms of action of Panax ginseng: a network pharmacological approach

  • Panax ginseng has been used since ancient times based on the traditional Asian medicine theory and clinical experiences, and currently, is one of the most popular herbs in the world. To date, most of the studies concerning P. ginseng have focused on specific mechanisms of action of individual constituents. However, in spite of many studies on the molecular mechanisms of P. ginseng, it still remains unclear how multiple active ingredients of P. ginseng interact with multiple targets simultaneously, giving the multidimensional effects on various conditions and diseases. In order to decipher the systems-level mechanism of multiple ingredients of P. ginseng, a novel approach is needed beyond conventional reductive analysis. We aim to review the systems-level mechanism of P. ginseng by adopting novel analytical framework-network pharmacology. Here, we constructed a compound-target network of P. ginseng using experimentally validated and machine learning-based prediction results. The targets of the network were analyzed in terms of related biological process, pathways, and diseases. The majority of targets were found to be related with primary metabolic process, signal transduction, nitrogen compound metabolic process, blood circulation, immune system process, cell-cell signaling, biosynthetic process, and neurological system process. In pathway enrichment analysis of targets, mainly the terms related with neural activity showed significant enrichment and formed a cluster. Finally, relative degrees analysis for the target-disease association of P. ginseng revealed several categories of related diseases, including respiratory, psychiatric, and cardiovascular diseases.

Production of Crossbred Lambs through Artificial Insemination of Non-prolific Medium Size Malpura and Avikalin Ewes Using Fresh Diluted Semen of Prolific Micro Size Garole Rams

  • Six adult Garole rams maintained under a semi-intensive system were used as semen donors for this study. Semen was collectied daily during the monsoon season with the help of an artificial vagina and examined for its quality characteristics. Ejaculates of thick consistency, rapid wave motion, ${\geq}80%$ motility and intense movement of motile spermatozoa were diluted at the rate of 1:1 with egg yolk McIllvaine glucose diluent at $30^{\circ}C$ in water bath. Estrus in ewes was detected by parading aproned rams of proven vigour at 12 h intervals. The ewes (54 Malpura and 23 Avikalin) in estrus were artificially inseminated with fresh diluted ram semen. The overall conception rate was 94.8%, (range 91.7 to 100%). The overall lambing percent was 80.5 with a range of 75.0 to 84.6%. There was no significant (p>0.05) difference in lambing and conception rate because of individual rams. Fertility was significantly lower (p<0.05) in ewes of less than two years and more than six years of age. Breed (Malpura and Avikalin) effect was not observed in conception and lambing rate (p>0.05). No significant difference (p>0.05) in birth weight and 12 month weight was observed between Garole${\times}$Avikalin and Garole${\times}$Malpura crossbred lambs but there was significant (p<0.05) difference at three month and six months body weight of both the crossbred lambs.

Production of Bovine Nuclear Transfer Embryos Using Fibroblasts Transfected with Single-Chain Human Follicle-Stimulating Hormone Gene

  • Human follicle-stimulating hormone (hFSH) is a pituitary glycoprotein that regulates follicular development and ovulation. Clinically, hFSH has been used to induce follicular growth in infertile women. The hormone is composed of heterodimers, including a common ${\alpha}$ subunit among the gonadotropin family and a hormone-specific ${\beta}$ subunit. Since assembly of the heterodimer is a rate-limiting step in the production of functional hFSH, transgenic clone cows carrying a single-chain hFSH transgene may efficiently produce functional hormone. Genes encoding the ${\alpha}$ and ${\beta}$ subunits of hFSH were linked using the C-terminal peptide sequence from the ${\beta}$ subunit of human chorionic gonadotropin. Bovine fetal fibroblasts were transfected with the gene construct, including the goat ${\beta}$-casein promoter and a single-chain hFSH coding sequence. Transfected fibroblasts were transferred into enucleated oocytes, and individual nuclear transfer (NT) embryos developed to the blastocyst stage were analyzed for the transgene by polymerase chain reaction. Seventy eight blastocysts (30.8%) were developed from 259 reconstructed embryos. Among these blastocysts, the hFSH gene was detected in 70.8% (34/48) of the embryos. Subsequent transfer of hFSH-transgenic clone embryos to 31 recipients results in 11 (35.5%) early pregnancies. However, all fetuses were lost before reaching day 180 of gestation. The results from this study demonstrated that bovine NT embryos carrying single-chain hFSH could be produced, and further extensive studies in which NT embryos are transferred to more recipients may give rise to single chain hFSH-transgenic cows for biomedical applications.
