느타리버섯 후배양 기간과 온도처리가 발이 균일도에 미치는 영향 (The effect of post-incubation period and temperature treatment on the uniform primordia formation of Pleurotus ostreatus)

  • 느타리버섯 '수한 1호'와 '곤지 7호'품종의 균사배양중 후배양 기간과 온도처리가 발이 균일도 및 생육에 미치는 영향을 조사한 결과는 다음과 같다. 배양중 전배양 $20^{\circ}C$, 20일 동안에는 두 품종 모두 11~15일에 $24{\sim}26^{\circ}C$까지 배지 내 온도가 상승되었다가 21일부터 후배양 온도처리와 동일한 수준으로 배지 내 온도가 유지되었다. 또한, 배지내 온도가 가장 높게 상승되는 시점에서 '수한 1호'의 $CO_2$는 6,500 ppm, '곤지 7호'는 5,800 ppm까지 발생되었으며 배양중인 균사생장에는 영향이 거의 없었다. '수한 1호'의 후배양 온도 $26^{\circ}C$에서 10일, 15일간 처리시 측발이율이 대조대비 40%, 비상품화율은 10.5% 감소 되었으며, '곤지 7호'는 후배양 온도 $26^{\circ}C$, 후배양 10일 처리에서만 측발이율은 19%, 비상품화율은 9.5% 감소되었다. 따라서, 느타리버섯 두 품종의 후배양 적합온도는 $26^{\circ}C$, 적합 후배양 처리기간은 '수한 1호'는 10일, 15일, '곤지 7호'는 10일이 발이 균일도가 높았다.

Effect of Thermal Treatment on the Electrocatalytic Activities and Surface Roughness of ITO Electrodes

  • The electrocatalytic activities and surface roughness of indium-tin-oxide (ITO) electrodes have been investigated after thermal treatment at 100, 150, or $200^{\circ}C$ for 30 min, 2 h, or 8 h. To check electrocatalytic activities, the electrochemical behavior of four electroactive species (p-hydroquinone, $Ru(NH_3){_6}^{3+}$, ferrocenemethanol, and $Fe(CN){_6}^{4-}$) has been measured. The electron transfer rate for p-hydroquinone oxidation and ferrocenemethanol oxidation increases with increasing the incubation temperature and the incubation period of time, but the rate for $Ru(NH_3){_6}^{3+}$ is similar irrespective of the incubation temperature and period because $Ru(NH_3){_6}^{3+}$ undergoes a fast outer-sphere reaction. Overall, the electrocatalytic activities of ITO electrodes increase with increasing the incubation temperature and period. The surface roughness of ITO electrodes increases with increasing the incubation temperature, and the thermal treatment generates many towering pillars as high as several tens of nanometer.

Establishment of Optimal Conditions for the Hypoosmotic Swelling Test to Evaluate the Integrity of Spermatozoal Plasma Membrane in Dog

  • Hypoosmotic swelling test (HOST) is used for evaluating the plasma membrane function and fertilizing ability in mammal spermatozoa. However, HOS solutions and experimental conditions have not been determined clearly for assessing canine spermatozoa. This study was conducted to examine the HOS solutions and assay conditions, including incubation time (30 to 120 min), storage temperature (4, 17 and $20^{\circ}C$), semen status (fresh and frozen). Maximum spermatozoal plasma membrane swelling was obtained in an 150 mOsm Na-citrate/Fructose solutions with an incubation time for 45 min. The storage temperature and semen status affected the percentage of HOS positive spermatozoa. The HOS test adapted to canine spermatozoa in this study was simple and highly consistent assay with good repeatability. The optimal condition of HOST in canine spermatozoa is an 150 mOsm Na-citrate/Fructose solutions with an incubation time for 45 min regardless of semen storage temperature and semen status.

느타리버섯 병재배 배양온도 및 배양기간에 따른 생육반응 연구 (Study on the characteristics of fruit body growth according to incubation temperatures and period for oyster mushroom)

  • 느타리버섯 병재배 품종 다양화와 신품종 느타리버섯의 병재배기술 확립을 위해 춘추2호, 수한1호, 흑평의 배양온도별 적정 배양일수에 대한 연구 결과는 다음과 같다. 1. 춘추2호의 배양일수는 $23^{\circ}C$ 배양시 17일, $17^{\circ}C$에서 21일이었고 흑평은 $23^{\circ}C$$26^{\circ}C$ 배양시 17일, $17^{\circ}C$에서 22일이었으며, 수한은 22~23일로 배양온도간 비슷하였다. 2. 초발이 소요일수는 춘추2호와 흑평은 배양온도별로 4~5일 소요되었고 수한1호는 $17^{\circ}C$, $20^{\circ}C$, $23^{\circ}C$에서는 3일, $26^{\circ}C$에서는 5~6일이 소요되었다. 3. 자실체특성은 춘추2호의 경우 배양일수가 길수록 유효경수가 20~25개 정도로 많았고 수한1호는 배양일수가 짧을수록 유효경수가 많았다. 4. 춘추2호와 흑평 수량은 배양온도별로 배양기간이 길수록 높은 경향이었으나 수한1호는 배양기간이 길수록 감소하였다. 춘추2호와 흑평은 배양온도 $20{\sim}23^{\circ}C$에서 22~27일간, 수한1호는 $17{\sim}23^{\circ}C$에서 22~23일간 배양후 발이시켰을 때 수량이 가장 높았다.

프로바이오틱 유산균 발효조건 탐색을 위한 다반응 최적화의 활용 (Applying Multi-Response Optimization to Explore Fermentation Conditions of Probiotics)

  • This review serves two purposes: first, to promote the use of improved optimization techniques in response surface methodology (RSM); and second, to enhance the optimum conditions for the fermentation of probiotics. According to research in dairy science, Lactiplantibacillus plantarum K79 is a candidate probiotic that has beneficial health effects, such as lowering blood pressure. The optimum conditions for L. plantarumK79 to produce peptides with angiotensin-converting enzyme (ACE) inhibitory activity were proposed, through modeling each of ACE inhibitory activity and pH as a function of the four factors that are skim milk concentration (%), incubation temperature (℃), incubation time (hours), and starter added amount (%). To estimate optimum conditions, the researchers employed a desirability-based multi-response optimization approach, utilizing third-order models with a nonsignificant lack of fit. The estimated optimum fermentation conditions for L. plantarum K79 were as follows: a skim milk concentration of 10.76%, an incubation temperature of 36.9℃, an incubation time of 23.76 hours, and a starter added amount of 0.098%. Under these conditions, the predicted ACE inhibitory activity was 91.047%, and the predicted pH was 4.6. These predicted values achieved the objectives of the multi-response optimization in this study.

Effects of Diluent Component, Freezing Rate, Thawing Time and Thawing Temperature on Acrosome Morphology and Motility of Frozen-thawed Boar Sperm

  • This study was carried out to obtain informations regarding the effect of N-acetyl-D-glucosamine in the LEY (lactoseegg yolk) diluent according to incubation time in 5 ml maxi-straw and the effects of freezing rate, thawing temperature and thawing time in the LEN (lactose-egg yolk and N-acetyl-D-glucosamine) diluent on acrosome morphology and motility of frozen-thawed boar sperm. The study showed that the LEN diluent was higher post-thaw NAR (normal apical ridge) acrosome than the LEY diluent for 0.5 h incubation at 37$^{\circ}C$. However, there were no differences between the LEN and LEY diluents on post-thaw sperm motility according to incubation time. The straws frozen from 5.0 cm (20$^{\circ}C$/min) to 17.0 cm (1$^{\circ}C$/min) above the liquid nitrogen surface did not show any significant differences on post-thaw sperm motility. However, the straws frozen above 5.0 cm from the liquid nitrogen surface were higher NAR acrosome than those frozen above 17.0 cm. The post-thaw percentages of motile sperm and NAR acrosome were significantly higher (p<0.05) for the maxi-straws submerged for 40 or 45 sec in a 52$^{\circ}C$ water bath than for 30, 35, 50 or 55 sec. The mean sample temperatures of maxi-straws after 40 or 45 sec submersion were 20.7 or 26.4$^{\circ}C$. In conclusion, the sample temperature of the thawed semen was very important for post-thaw sperm survival in the LEN diluent of 5 ml maxi-straw. When the temperature of the thawed semen was 20.7$^{\circ}C$, the percentages of motile sperm and NAR acrosome were highest.

저온숙성 방법에 따른 생오미자 당절임 농축액인 오미자청의 파일럿 규모 생산 및 품질특성 (Pilot-scale production of Omija-cheong by low temperature incubation: An assessment of quality characteristics)

  • 전통적인 저온숙성 당절임 방법을 이용한 오미자청의 제조과정에서 최적의 배양시간을 찾기 위하여 파일럿 규모(1톤)의 연구를 시행하였다. 생오미자와 동일한 량으로 투입된 설탕은 배양시간에 따라 과당과 포도당으로 전환되었으며 오미자청에 존재하는 유리당의 조성은 68일 이상의 배양기간에서는 일정하게 유지되었다. 배양기간에 따른 오미자청의 pH는 일정한 수준을 유지한 반면, 적정 산도와 유기산의 함량은 배양시간 37일까지 지속적으로 증가한 이후 68일 이후에는 일정한 조성을 유지하였다. 생오미자에 존재하는 주요 유기산은 숙신산으로 확인되었으며 오미자청에 존재하는 유기산의 조성 역시 생오미자와 유사하였다. 오미자에 존재하는 주요 생리활성물질인 총 플라보노이드와 폴리페놀화합물은 60일 이상 배양한 오미자청에서 높은 함량을 보이고 138일까지 일정한 수준을 유지하였다. 특히, 오미자청에 함유된 총 플라보노이드와 폴리페놀화합물의 함량은 생오미자 과육에 비하여 각각 9배와 5배 정도 높은 수준으로 증가하였다. 생오미자를 저온숙성 당절임 방법을 적용하여 오미자청으로 제조 가공하는 과정에서 유리당, 유기산 및 생리활성물질 등에 대한 일정한 품질을 확보를 위해서는 최소 60일 이상의 배양기간이 필요한 것으로 확인되었다. 이러한 연구결과는 전통적인 저온숙성 당절임 방법으로 제조 가공되는 오미자청의 고품질화와 기능성 음료 개발을 위한 제조 가공공정의 표준화에 유용한 정보가 될 것으로 판단된다.

산화막을 입힌 지르코늄 합금의 수소화 반응에 관한 연구 (A Study on the Hydriding Reaction of Pre-oxidized Zr Alloys)

  • This paper presents some experimental results on incubation time for massive hydriding of Zr alloys with oxide thickness. Oxide effects experiments on massive hydriding reaction of commercial Zr alloy claddings and pre-oxidized Zr alloys with hydrogen gas were carried out in the temperature range from 300 to $400^{\circ}C$ with thermo-gravimetric apparatus. Experimental results for oxide effects on massive hydriding kinetics show that incubation time is not proportional to oxide thickness and that the massive hydriding kinetics of pre-filmed Zr alloys follows linear kinetic law and the hydriding rate are similar to that of oxide-free Zr alloys once massive hydriding is initiated. There was a difference in micro-structures between oxide during incubation time and oxide after incubation time. Physical defects such as micro-cracks and pores were observed in only oxide after incubation time. Therefore, the massive hydriding of Zr alloys seems to be ascribed to short circuit path, mechacical or physical defects, such as micro-cracks and pores in the oxide rather than hydrogen diffusion through the oxide resulting from the increase of oxygen vacancies in the hypostoichiometric oxide.

Byssochlamys fulva가 생성하는 펙틴질 분해효소에 관한 연구 I (Studies on the Pectolytic Enzymes from Byssochlamys fulva I)

  • Effects of carbon sources, incubation time, incubation temperature, initial incubation pH, and vitamin B complex on the polygalacturonase activity of Byssochlamys fulva were studied to confirm the optimum conditions for the production of that enzyme. When pectin was used as carbon source, polygalacturonase activity reached to the maximum value of 0.50 units/ml. After 5 days of incubation, polygalacuturonase activity reached to its maximum of 0.48units/ml. Polygalacturonase activities were similar between $25^{\circ}C\;and\;45^{\circ}C$, however, decreased dramatically in the outside of this range. Polygalacturonase activity was not signicicantly influenced by the variation of initial incubation pH. However, at pH5.0, polygalacturonase activity was slightly through the addition of thiamine and riboflavin, and the optimum concentrations were $10^{-2}M$ in case of thiamine and $10^{-3}M$ in riboflavin.

인삼 근부병균 Cylindrocarpon destructans (Zinssm.) Scholten의 포자 생성에 미치는 배양기간, 온도, pH의 영향 (Effect of Incubation Time, Temperature and pH on the Production of Conidia and Chlamydospore of Cylindrocarpon destrutans (Zinssm.) Scholten Causing Root Rot of Panax ginseng)

  • The effects of media, incubation time, temperature and pH on production of conidia and chlamydospore of Cylindrocarpon destructans (Zinssm.) Scholen causing root rot of Panax ginseng were studied. Microconidia of the pathogen were abundantly produced on V-8 juice agar as a solid substrate with 5.73(log conidia/mm2) and in V-8 broth as a liquid substrate with 6.65 (log conidia/ml) among media tested. No difference was observed on the length of microconidia produced from the media with a range of 9.50∼11.38 $\mu\textrm{m}$. However, tryptic soy agar produced the broadest microconidia (average 5.00 $\mu\textrm{m}$) among the media tested. All the media produced chlamydospores In a range of 1.06∼4.37 (log chlamydospores/mm2) without a significant difference in number, while V-8 juice agar produced the bigger one (18.39 $\mu\textrm{m}$ in diameter) as compared to the tested media. The fungus began to sporulate conidia after three days of incubation and reached maximum at the 8th day. It seemed to be in a stationary phase until 30 days of incubation but was decreased thereafter. Chlamydospore was produced at 4th day after incubation. Maximum production was observed at 8th day and the number seemed to be maintained during the observation period. Both conidia and chlamydospore of the pathogen were able to be spoluated at 10∼25$^{\circ}C$. However, optimum temperatures of conidia and chlamydospore formation were 15∼25$^{\circ}C$ and 10∼20$^{\circ}C$, respectively. C. destrmtans produced conida with an wide range of pH from 3.3 to 8.0 and chlamydospore from 2.8 to 8.0. Number of conidia was increased with an increase of pH up to 4.0. There was no significant difference in the number between 4.0 to 8.0. It seemed to have two optimum pH ranges, 3.3∼4.0 and 7.1∼8.0 for the chlamydospore formation.
