복합 소독 공정을 이용한 E. coli 불활성화 (E. coli Inactivation using Complex Disinfection Process)

  • Conventional disinfectants and disinfection method are expensive, hazardous and often require long periods of exposure. Recently, there is growing interest in complex disinfection process as a disinfection technique in medical instruments such as endoscope, hand piece bur to improve the disinfection efficiency and conveniency. The aim of this study was to evaluate the performance of a new complex process for the purpose of disinfection of Escherichia coli in water. Three single process (electrolysis, UV and ultrasonic process) was combined dual and triple disinfection process. The order of disinfection performance for E. coli in dual process lie in: Electrolysis + UV > Electrolysis + Ultrasonic > UV + Ultrasonic process. Disinfection efficiency of E. coli and degradation of N, N-Dimethyl-4-nitrosoaniline (RNO, indicating material of OH radical formation) of dual process was higher than that of the triple process (Electrolysis + UV + Ultrasonic process). In electrolysis + UV process, disinfection tendency was well agreed with RNO degradation tendency.

Overproduction of the Escherichia coli Chaperones GroEL-GroES in Rhodococcus ruber Improves the Activity and Stability of Cell Catalysts Harboring a Nitrile Hydratase

  • Three combinations of molecular chaperones from Escherichia coli (i.e., DnaK-DnaJ-GrpE-GroEL-GroES, GroEL-GroES, and DnaK-DnaJ-GrpE) were overproduced in E. coli BL21, and their in vitro stabilizing effects on a nitrile hydratase (NHase) were assessed. The optimal gene combination, E. coli groEL-groES (ecgroEL-ES), was introduced into Rhodococcus ruber TH3. A novel engineered strain, R. ruber TH3G was constructed with the native NHase gene on its chromosome and the heterologous ecgroEL-ES genes in a shuttle plasmid. In R. ruber TH3G, NHase activity was enhanced 37.3% compared with the control, TH3. The in vivo stabilizing effect of ecGroEL-ES on the NHase was assessed using both acrylamide immersion and heat shock experiments. The inactivation behavior of the in vivo NHase after immersion in a solution of dynamically increased concentrations of acrylamide was particularly evident. When the acrylamide concentration was increased to 500 g/l (50%), the remaining NHase activity in TH3G was 38%, but in TH3, activity was reduced to 10%. Reactivation of the in vivo NHases after varying degrees of inactivation was further assessed. The activity of the reactivated NHase was more than 2-fold greater in TH3G than in TH3. The hydration synthesis of acrylamide catalyzed by the in vivo NHase was performed with continuous acrylonitrile feeding. The final concentration of acrylamide was 640 g/l when catalyzed by TH3G, compared with 490 g/l acrylamide by TH3. This study is the first to show that the chaperones ecGroEL-ES work well in Rhodococcus and simultaneously possess protein-folding assistance functions and the ability to stabilize and reactivate the native NHases.

전기+UV 공정에 의한 E. coli 불활성화 (Inactivation of E. coli by Electrolysis+UV Process)

  • This study has carried out to evaluate the performance of single (electrolysis, UV and ultrasonic process) and complex process (Electrolysis+UV, UV+Ultrasonic and Electrolysis+Ultrasonic) for the purpose of disinfection of Escherichia coli in water. The order of disinfection performance for E. coli in single process lie in: Electrolysis ${\fallingdotseq}$ UV >> ultrasonic process. OH radical was not produced in single disinfection process. Among the three kinds of complex process, disinfection performance of the Electrolysis+UV was higher than that of the other process (UV+Ultrasonic and Electrolysis+Ultrasonic). It demonstrated a synergetic effect between the UV and electrolysis. When the use of $Na_2SO_4$ as electrolyte instead of NaCl, current increase or more reaction time was needed for the complete disinfection. The disinfection performance of pre-electrolysis (20 W, 30sec) and post-UV (10 W, 30 sec) was higher than that of the simultaneous electrolysis+UV process at same electric power (30 W, 30 second).

Microwave Plasma Sterilization System 처리가 옥수수수염 차의 저장 중 Escherichia coli 및 Listeria monocytogens의 생육 저해에 미치는 영향 (Inactivation of Foodborne Pathogenic Bacteria in Corn Silk Tea Using a Microwave Plasma Sterilization System)

  • 옥수수수염 차의 MPSS 처리에 의한 위해미생물의 살균 효과에 대하여 연구하였다. 옥수수수염 차에 E. coli와 L. monocytogenes를 5~6 log CFU/mL 수준으로 접종하고, MPSS 처리한 후 25$^{\circ}C$에서 12일 동안 저장하면서 실험하였다. E. coli는 1, 2, 3 회 MPSS 처리 후 각각 1.14, 2.49, 5.72 log CFU/mL의 감균 효과를 보였고, L. monocytogenes는 MPSS 1, 2, 3회 처리 후 각각 1.93, 4.49, 6.62 log CFU/mL의 감균 효과를 보였다. 또한, E. coli와 L. monocytogenes 모두 MPSS 4회 처리 후에는 모두 사멸되었고, 저장 12일까지 검출되지 않았다. 그리고 MPSS 처리는 옥수수수염 차의 TPC에 부정적인 영향을 끼치지 않았고, 탁도는 4회 이상 MPSS 처리에 의해 향상되었다. 따라서 본 연구 결과, MPSS 처리는 옥수수수염 차의 품질 유지와 더불어 미생물 생육 억제가 가능하기에 옥수수수염 차의 살균에 MPSS를 적용할 수 있다고 판단된다.

고전압 임펄스를 적용한 막분리 공정에서의 생물막 오염 제어에 관한 연구 (A Study on the Biofouling Control in Membrane Processes Using High Voltage Impulse)

  • Although membrane technologies are widely applied to the water and wastewater treatment processes, strategy for the control of membrane biofouling is strongly required. In this study, a possibility of control of membrane biofouling using HVI(High Voltage Impulse) was verified based on the inactivation of microorganisms by the HVI. The HVI system was consisted of power supply, voltage amplifier, impulse generator and disinfection chamber and the model microorganism was E. coli. When 15[kV/cm] of electric fields was applied to the E. coli solution, inactivation of the microorganism was found. A possibility of the control of membrane biofouling using HVI was verified with experiments of membrane filtration with and without exposure of the HVI to biomass solution. Another membrane filtration experiments with the contaminated membranes by E. coli solution were carried out and indicate that the HVI could be used as an alternative method for membrane biofouling control. A series of simulation of the electric fields between electrodes and microorganisms was carried out for the visualization of the disinfection that showed where the electric fields are formed.

  • 수처리 살균에 이용되는 자외선 소독은 기존 염소소독법의 문제점인 소독부산물 생성이 없어 실질적인 대안으로 널리 수행되고 있다. 본 연구에서는 368.6[nm] 자외선을 이용하여 소독에 대해 저항성이 강한 대장균(E.coli)의 분활성화 특성을 파악하고자 하였다. 실험에 사용한 자외선 소독 장비는 358.6[nm]의 파장을 방사하는 자외선 램프를 사용하여 제작하였으며 소독 반응조에서 시간에 따른 사멸 특성을 살펴보았다 자외선으로 60분간 대장균을 조사하였을 때 초기 30분간은 자외선에 큰 영향을 밭지 않았지만 30분 이후에 대장균이 점차적으로 사멸되어 60분이 경과하였을 때 83.3[%]의 사멸률을 보여서 일반적인 자외선 램프보다 살균효율이 미약한 것을 확인할 수 있었다. 또한 동력학적 접근방법으로 구한 비활성화율 계수 k값은 0.00868[$cm^2$]/([$mW{\cdot}s$])로 나타났다.

UV-C 조사가 세척 당근의 저장 중 E. coli O157:H7과 Listeria monocytogenes의 생육저해 및 품질에 미치는 영향 (Effect of UV-C Irradiation on Inactivation of Escherichia coli O157:H7 and Listeria monocytogenes on Washed Carrot during Storage)

  • 세척 당근에 E. coli O157:H7과 L. monocytogenes를 인위적으로 접종한 후 UV-C 조사 처리에 따른 저장 중 미생물수 변화를 조사하였다. 당근에 접종된 E. coli O157:H7과 L. monocytogenes의 초기 균수가 6-7 log CFU/mL가 되게하였고, 사용된 UV-C 조사선량은 1, 3, 5, $10\;kJ/m^2$이었으며 조사된 시료는 $4{\pm}1^{\circ}C$에서 8일 동안 저장하였다. UV-C 조사는 E. coli O157:H7과 L. monocytogenes의 미생물 수에 있어서 조사선량이 증가할수록 유의적으로(p<0.05) 감소하는 것을 나타냈다. 특히, $10\;kJ/m^2$ 처리구는 대조구와 비교하여 E. coli O157:H7과 L. monocytogenes의 미생물 수를 각각 2.35, 2.38 log CFU/g 감소시켰다. 품질 변화에서는 UV-C 조사 5, $10\;kJ/m^2$ 처리구가 저장 중 당근 표면 $L^*$ value의 급격한 증가와 $a^*$, $b^*$ value의 감소 및 백화현상을 유의적으로(p<0.05) 억제시켰다. 관능적 품질 평가는 5, $10\;kJ/m^2$ 처리구가 대조구에 비해 유의적으로(p<0.05) 높게 나타났다. 따라서 본 연구결과, UV-C 조사 처리는 세척 당근의 저장 중 미생물학적 안전성 확보와 품질유지 향상에 효과적인 것으로 판단된다.

재조합 대장균에서 fadB 유사효소의 Polyhydroxyalkanoates 합성에 미치는 역할의 규명 (In Vivo Analysis of fadB Homologous Enzymes Involved in Biosynthesis of Polyhydroxyalkanoates in Recombinant Escherichia coli)

  • 재조합 E. coli를 이용한 MCL-PHA의 생산에서 fatty acid pathway로부터 PHA 생합성 전구체 물질들이 만들어진다는 사실과 함께 이에 관여하는 enzymes이 밝혀지고 있다. 본 논문에서는 protein homology search로부터 탐색된 paaG와 ydbU genes의 PHA 생합성에서의 역할을 확인하기 위하여 paaG와 ydbU gene이 각각 knock-out된 mutant E. coli strains 를 제작하였다. 제작된 mutant E. coli들은 모균주들보다 낮은 PHA 농도와 함량을 가졌으며, 이러한 결과들로부터 paaG와 ydbU는 fatty acid pathway에서 PHA synthesis의 전구체 물질들을 공급한다는 사실을 확인하였다. 또한, 새로운 FadB homologous enzyme YgfG를 탐색하였으며, ygfG gene이 overexpression된 균주와 ygfG mutant를 제작하여 PHA 합성을 실험한 결과 ygfG도 paaG와 ydbU와 유사한 역할을 한다는 사실을 밝혔다. 이러한 연구결과들은 E. coli에서의 MCL-PHA 단량체들의 합성 경로를 확인하여 효과적인 PHA 생산 균주를 제작할 수 있게 할 것이다.

Inactivation Efficiency of Escherichia coli and Listeria monocytogenes in Ground Pork by Combination of Natural Food Ingredients and High Pressure Processing

  • The objective of this study was to examine the effects of a combined treatment regarding antimicrobial food ingredients and high pressure processing (HP) on the inactivation efficiency of Escherichia coli and Listeria monocytogenes inoculated into ground pork. Ethanol extracted from garlic, leeks, onions, and ginger powder was prepared. Half of the prepared powder was irradiated at 5 kGy to see the effect of pasteurization before addition. The prepared food ingredients were added into radiation-sterilized ground pork (1%, w/w), and inoculated with E. coli and L. monocytogenes. The samples were vacuum-packed and applied with HP at 0.1 (control), 300, 450, and 600 MPa. Microbial log reduction increased with the increase of pressure up to 600 MPa. With minor exceptions, overall efficiency of HP treatment with regards to inactivation of pathogens increased. Inoculated microorganisms showed approximately 7-8 Log reductions by 600 MPa, except for L. monocytogenes treated with garlic (5.7 Log reductions). The E. coli reduction in ground pork mixed with ethanol extracted garlic showed the highest efficiency (1.86) compared to leeks (1.25-1.31), onions (1.17-1.44), and ginger (1.50-1.82) when treated at an HP of 450 MPa. There was no evidence for the advantage of pasteurization concerning the food ingredients before addition of antimicrobial food ingredients and HP. Results demonstrate that the combination of antimicrobial food ingredients and HP treatment may help improve the efficiency of sterilization in meat systems.

해수 및 담수에서 장구균과 총대장균군의 불활성화 (Inactivation Rate of Enterococci and Total Coliforms in Fresh Water and Sea Water)

  • Inactivation rates between enterococci and total coliforms were compared in order to find the suitability of enterococci as an indicator microorganism under various experiment conditions - freshwater and/or seawater, indoor and/or outdoor conditions. In case of indoor laboratory experiments, inactivation rates of enterococci ($k_D$: 0.050~0.082) were faster than those of total coliforms ($k_D$: 0.034~0.045) in freshwater matrix. In seawater matrix, however, survival rate of enterococci was longer than that of total coliforms at two out of three experiments in indoor condition. When incubated in outdoor conditions, enterococci were inactivated significantly more rapidly than total coliforms both in freshwater and seawater matrices. With these results, enterococci appear to be less suitable than total coliforms in terms of inactivation rates.
