Antioxidative and Antiaging Effects of Dietary Yellow and Black Soybean in Rats (노란콩 및 검정콩의 섭취가 흰쥐의 항산화 및 항노화 시스템에 미치는 영향)

  • To investigate antioxidative and antiaging effects of yellow and black soybeans in vivo system, male SD rats (n=24) were fed the diets containing casein (C), yellow soybean (YS) or black soybean (BS) for 8 weeks. Experimental groups showed the preventive effect on lipid and protein oxidation, especially protein oxidation of plasma was significantly inhibited in BS group. SOD and catalase activities were not different among the groups, but hepatic glutathione peroxidase (GSH-px) activity was significantly lowered in YS and BS groups compared with control group. The contents of superoxide anion radical in cytosol were significantly lowered in experimental groups compared with control group. And hydroxyl radical was slightly lowered in soybean groups. Lipofuscin accumulation on the heart and eyes of rats was effectively inhibited by yellow soybean and black soybean, respectively. In conclusion, these results suggest that yellow soybean and black soybean have the antioxidative/antiaging effect in vivo system and they have similar activities despite different color of seed coat.

  • Due to the rapid growth of mobile communication services, many studies have been performed on the biological effects of EMF(Electromagnetic field) exposure. However, it is not easy to create the field uniformity in radio frequency. To overcome this difficulty, the electric field and SAR(Specific Absorption Rate) distribution is developed to measure the whole body exposure system. In this paper, we developed the whole body exposure system for in-vivo study utilizing reverberation chamber which has a high possibility of practical use by substituting previous the EMC chamber. The field uniformity in the test area of the designed reverberation chamber was satisfied by the simulation and measurement data. In animal study of the whole body exposure for a mouse, the results of FDTD simulation were compared with those of measurement to confirm SAR under the electromagnetic exposure. We analyze the electric field distribution in the interior of the reverberation chamber and treat these results in statistical manner. In the CDMA frequency band(PCS & Cellular band), an average value of the whole body SAR and local peak value for a experimental mouse were presented for various input power.

Effect of Polyamine Biosynthesis InhibItion on the Microbicidai and Jumoriddal Activities in Mouse Macrophage (생쥐 대식세포의 감염균 치사활성과 종양 치사활성에 미치는 Polyamine 생합성 억제의 영향)

  • The objective of this experiment was to examine the effect of polyamine depletion by polyamine biosynthesis inhibitors on microbicidal activity and tumoricidal activity in mouse mac-rophages. $\alpha$ -Difluoromethylomithine (DFMO), inhibitor of putrescine and spernidine biosynthesis, treatment in vivo for 6-8 days reduced chemiluminescence levels in thioglycollate-, lipo-polysaccharide (LPS), and BCG-treated mouse macrophages. An DFMO treatment in vitro inhibited production of tumor necrosis factor (TNF), in dose-dependent manner, and tumoricidal activity by macrophages. The effect of polyamine depletion by MO on ThF production and tumoricidal activity could be reversed by the addition of exogenous putrescine. These result indicated that the obserbed effect of DFMO on macrophage activities were mediated through inhibition of polyamines are, must be, required for optimal activities of macrophages.

The Anti-oxidative Activities of Fermented Hibiscus syriacus Extract in High Fat Diet-induced obese C57BL/6 Mice (고지방식이로 유도된 비만 C57BL/6 쥐에서 발효 무궁화 추출물의 항산화 활성)

  • The purpose of this study was to evaluate the anti-oxidative effects of fermented Hibiscus syriacus extract on obese mice. The antioxidant experiments were conducted both in vivo and in vitro. The in vitro experiment was measured through the use of nitric oxide (NO) free radical scavenging activity. The results showed that the scavenging activity of the NO free radical in fermented Hibiscus syriacus extract had higher activity than unfermented Hibiscus syriacus extract. The in vivo experiments were divided into a normal dietary group, a high fat dietary group (CON), a high fat dietary + fermented Hibiscus syriacus extract group (FH), and a high fat dietary + unfermented Hibiscus syriacus extract group. These four groups received intraperitoneal administrations for four weeks. Next, the levels of superoxide dismutase (SOD), catalase (CAT), and malondialdehyde (MDA) in a liver homogenate were measured. The results showed that the levels of SOD and CAT in FH were higher than the CON and MDA levels, which were significantly decreased in FH. Consequently, fermented Hibiscus syriacus extract had an inhibitive effect on oxidative stress and a heightening effect on antioxidant enzyme activity in obese mice, suggesting that fermented Hibiscus syriacus extract could be utilized as a useful, natural antioxidant.

  • A measurement unit and signal processing algorithm have been developed for predicting arterial oxygen saturation noninvasively. The measurement set-up was composed of a probe including light source and photodetector, optical signal processing section, LED driving circuit, PC interface software for data acquisition and data processing software. Light from the LED's was irradiated onto the finger nail bed and transmitted light was measured at different wavelengths. An effective baseline correction method was developed and measured data were analyzed by using various data processing methods and prediction algOlithms. For performance evaluation, a pulse oximeter simulator (Bio- Tek Instrument Inc.) was used as reference. The best performance in terms of the correlation coefficient and the standard deviation was obtained under the following conditions; when the arterial signals were computed in terms of area rather than peak-valley difference, and when the algorithm calculating by $In(I_p/I_v)/I_{avr}$ value for pulsation waveform was used. In in vivo test, prediction was improved when the developed baseline correction method was used. In addition, wavelengths of 660 nm and 940 nm provided better linearity and precision than wavelengths of 660 nm and 805 nm. 05 nm.

Development of Local-Exposure Systems for In Vivo Studies at Mobile-Phone Frequency Bands (이동통신 주파수 대역에서의 동물 실험용 국부 노출 장치 개발)

  • We have designed local exposure systems for long-time mice experiments in PCS and cellular frequency band(PCS: 1,762.5 MHz, cellular: 848.5 MHz). The fabricated systems are local exposure systems of carousel type, and 40 mice can be exposed at a time. In order not to give extra stress to the mice ender experiment, the systems were fabricated to meet the environmental conditions such as illumination, ventilation, noise etc. SAR measurement was performed using a temperature probe. Measurements at 3 points in the head of mouse cadaver and solid phantom were made, and it has been confirmed that the measurement results are in good agreement with the simulation results in the real exposure environment. The exposure systems are currently used for long-term mice experiments.

Effects of Plant Water Extract Codonopsis Lanceolatae on Mouse Immune Cell Activation Ex Vivo (더덕 물 추출물의 경구 투여가 마우스 면역 세포 활성에 미치는 효과)

  • Codonopsis lanceolatae has been used as one of the traditional remedies as well as food source. However, few studies on their immunomodulating effects have been reported. We previously reported that ex vivo supplementation of Codonopsis lanceolatae water extracts enhanced splenocyte proliferation compared to the control group. In order to elucidate its ex vivo effect, six to seven week old balb/c mice were fed ad libitum on a chow diet and water extracts of Codonopsis lanceolatae were orally administrated every other day for four weeks at two different concentrations (50 and 500 mg/kg B.W). After preparing the single cell suspension, the proliferation of splenocytes was determined by MTT (3- [4,5-dimethylthiazol-2-y] -2,5-diphenyl terazolium bromide) assay. The production of cytokine (IL-1${\beta}$, IL-6, TNF-${\alpha}$), secreted by macrophages stimulated with LPS or not, was detected by ELISA assay using a cytokine kit. After 48 hrs of incubation with the mitogen (ConA or LPS) stimulation, the mice splenocyte proliferation in experimental group was statistically increased at two different concentrations than that in control group. The cytokines production was more significantly enhanced at the lower supplementation (500 mg/kg B.W.) group rather than higher concentration (500 mg/kg B.W.) compared to the control group. The results of this study may suggest that the supplementation of water extract of plant mixture could regulate the immune function by increasing the splenocyte proliferation and enhance the immune function through regulating cytokine production capacity by activated macrophages in mice.

Evaluation of Usefulness on In-vivo Diode Dosimetry for Measuring the Tumor Dose of Oral Cancer Patient (구강암 환자의 종양 선량 측정을 위한 In-vivo Diode Dosimetry의 유용성 평가)

  • Purpose : This test is designed to identify the validity of treatment plan by implementing real-time dosimetry by means of dose that is absorbed into PTV and OAR when preparing doses of 3D and POP plans. Materials and Methods : In treatment. error can be calculated be comparing Exp. Dose with the actual dose, which has been converted from 'the reading value obtained by placing diode detector on the area to be measured'. Same test can be repeated using Alderson-Rando phantom. Results : Errors were found: A patient(POP plan): 197.6/199=-1.2%, B patient(3D-plan): 199.9/198.7=+0.6%, C patient: 196/200=-1.5%. In addition, considering the resulted value of measuring OAR besides target-dose for C patient showed 96/200, representing does of 47%, the purpose of protection was judged to be duly accomplished. Also it was acknowledged the resulted value of -3.7% met the targeted dose within the range of ${\pm}5%$. Conclusion : Aimed for identifying the usefulness of pre-treatment dose measurement using diode detector, this test was useful to evaluate the validity of curing because it resulted in the identification of category to be protected as well as t dose. Moreover, it is thought to have great advantage in ascertaining the dose of target, dose of which is not calculated yet. Similar to L-gram before treatment, this test is thought to be very effective so that it can bring great advantages in the aspects such as validity of curing method and post-treatment plan as well.

Tissue Inhomogeneity Correction in Clinical Application of Transmission Dosimetry to Head and Neck Cancer Radiation Treatment (두경부 방사선 치료 환자에서 투과선량 알고리즘의 임상 적용시 불균질 조직 보정에 관한 연구)

  • Purpose : To confirm the reproducibility of in vivo transmission dosimetry system and the accuracy of the a1gorithms for the estimation of transmission dose in head and neck radiation therapy patients. Materials and Methods : From September 5 to 18, 2001, transmission dose measurements were peformed when radiotherapy was given to brain or head and neck cancer patients. The data of 35 patients who were treated more than three times and whose central axis of the beam was not blocked were analyzed in this study. To confirm the reproducibility of this system, transmission dose was measured before dally treatment and then repetitively every hour during the treatment time, with a field size of 10$\times$10 cm$^{2}$ and a delivery of 100 MU. The accuracy of the transmission dose calculation algorithms was confirmed by comparing estimated dose with measured dose. To accurately estimate transmission dose, tissue inhomogeneity correction was done. Results : The measurement variations during a day were within $\pm$0.5$\%$ and the dally variations in the checked period were within $\pm$ 1.0$\%$, which were acceptable for system reproducibility. The mean errors between estimated and measured doses were within $\pm$5.0$\%$ in Patients treated to the brain, $\pm$2.5$\%$ in head, and $\pm$ 5.0%$\%$in neck. Conclusion : The results of this study confirmed the reproducibility of our system and its usefulness and accuracy for dally treatment. We also found that tissue inhomogeneity correction was necessary for the accurate estimation of transmission dose in patients treated to the head and neck.

  • For treatment of Total Skin Electron beam Therapy (TSET), measurement of dose at various conditions is need on the contrary to usual radiotherapy. When treating TSET with modified Stanford technique based on linear accelerator, the energy of treatment electron beam, the spatial dose distribution and the actual doses deposited on the surface of the patient were measured by using EBT2. The measured energy of the electron beam was agreed with the value that measured by ionization chamber, and the spatial dose distribution at the patient position and the doses at several point on the patient's skin could be easily measured by EBT2 film. The dose on the patient that was measured by EBT2 film showed good agreement with the data measured simultaneously by TLD. With the results of this study, it was proven that the EBT2 film can be one of the useful dosimeter for TSET.
